The type I and III interferon responses restrict infection with tick-borne orthoflaviviruses through IFI6
Tick-borne orthoflaviviruses (TBOVs) are a growing global health concern. Several representatives of this viral family cause fatal disease in humans with increasing case numbers throughout the last decades. The innate immune response, especially interferon (IFN)-dependent signaling, is an essential part of the human defense system that counteracts infection with TBOVs and other viruses. Even though they activate the same signaling cascade, IFNs belonging to the type I and III families trigger differing gene expression patterns. Which genes the two IFN families induce to restrict infection with TBOVs remains poorly characterized. Here we show that type I and III IFNs are both capable of restricting TBOV infection of human cell lines in a cell type-specific manner. Infection of C57BL/6J mice with knockouts for either IFN type I or III receptors further underscored the critical role of IFN signaling in controlling TBOV replication in vivo . To assess the contribution of single genes to controlling TBOV infection in human cells, we used a CRISPR/Cas9-KO-based screening approach. This strategy identified IFI6 as a central player for IFN type I- and III-driven responses against TBOVs. We further defined IFI6 as an ER-resident protein that restricts TBOV replication at a post-entry step. Our work thus opens new perspectives for targeting weak points in the life cycle of TBOVs and other orthoflaviviruses, potentially paving the way for the development of new antiviral therapeutics. One Sentence Summary Type I and III interferons are crucial for protection against tick-borne orthoflavivirus infection in vitro and in vivo , both relying on IFI6 as a main antiviral effector
Fiches d'aide a la coopérationhese fact sheets are designed to aid reflection and action in structuring cooperation in the context of dealing with situations of abuse towards vulnerable adults. Although it involves a multitude of local actors with varied roles and missions (town halls, departmental social services, regional health agencies, law enforcement, public prosecutors, associations, etc.), the fight against abuse was not truly coordinated by a national public policy until 2024. The TACT (Traitement des Alertes de maltraitance en Coopération sur les Territoires) action research project consisted of collecting and comparing the practices tested and promoted by actors in the field encountered in several territories. It appears that the handling of reports of abuse against vulnerable adults poses two main challenges: the coordination of the various actors who receive reports of abuse; and the cooperation of a group of actors contributing to local policy and the monitoring of each situation. These two issues are often unclear in the field, since the same public authorities and actors are involved in both missions, sometimes in redundant and poorly identified areas. This is all the more true given that, prior to the so-called ‘Ageing Well’ law of April 2024, the need for coordination was strongly emphasised in political discourse in response to strong social demand, without any single institution or body being identified as having authority in this area. The recent inclusion in the law of units for collecting and processing reports aims to address the issue of coordination, which has already been the subject of measures that have not been implemented in a satisfactory and sustainable manner. Based on the findings of the TACT action research on the current functioning of the processing of reports of abuse of vulnerable adults, here are nine reference sheets to facilitate the implementation of the measures of the so-called ‘Ageing Well’ law of 8 April 2024. We refer to the collegial bodies whose mission is to organise the operational work between all the actors involved in monitoring situations of abuse as ‘TACT commissions’. TACT commissions are not required by law and do not have to be created from scratch: they are the result of a review of the collegial bodies that local actors have already set up and tested. The composition, scope of mission, operational functioning and name of these commissions are not predefined. These reference sheets have been designed to support this capitalisation and promotion of cooperation practices by guiding stakeholders' questions.
History-dependent and frequency-dependent dielectric nonlinearities induced by polar nanoregions in a thin film of 0.5 ( Ba 0.7 Ca 0.3 TiO 3 ) − 0.5 ( BaZr 0.2 Ti 0.8 O 3 )
International audienceIn this article, dielectric nonlinearities in 0.5(Ba0.7 Ca0.3 TiO3 ) − 0.5(BaZr 0.2 Ti0.8 O3 ) thin film are studied using impedance spectroscopy and harmonic measurements, as a function of the AC measuring field, at different frequencies and upon cycling. The measurements reveal that the pinching of the hysteresis loop, characterized by a phase angle of the third harmonic close to −270 deg, is stronger for low frequencies. This confirms that the pinching is induced by the presence of polar nanoregions (PNRs), whose responses are also strongly dependent on frequency. When repeating the measurement, the PNR contribution changes since the phase angle of the third-harmonic response evolves from pinched to conventional relaxor. This shows that strong changes in the PNR configuration can be induced, even for low AC fields. First-order reversal curves (FORCs) confirm the presence of a pinched hysteresis loop. When repeating the FORC measurement a second time, the distribution drastically changes and corresponds to a soft ferroelectric. The asymmetry of the Preisach plane measured using FORCs is confirmed by a proposed measurement strategy: unipolar impedance measurements

International audienceLanguage acquisition involves the ability to switch perspectives according to the partners’ roles in the speech context. This ability, involving morphosyntactic and pragmatic aspects, is fundamental in the acquisition process of personal pronouns (PersP). Our aim is to analyze, in spoken French, the acquisition of comprehension and production of PersP according to different perspectives (1p, 2p, 3p) and functions [subject (S); direct object (DO) and indirect object (IO) clitics]. 110 children aged from 6 to 13 participated. Comprehension and production of PersP were evaluated through an experimental protocol consisting of various ecological situations of the children’s daily life, in a comic strip format. Sentences had two levels of difficulty: D1, involving S and DO; and D2 involving S, DO and IO PersP. We found an age effect on the scores for the two tasks. Scores for expected answers were higher in the comprehension (1) than in the production task (2). We identified a significant increase in the mean scores, after an estimated breakpoint of 8 years for (1), and after the two estimated breakpoints of 8 and 10 years for (2). In (2) we found significantly better performances for D1 than for D2 sentences. In D1, unexpected answers occur more significantly on the DO than on the S, and in D2 they occur first on the DO, then on the IO and finally on the S. In D1 sentences, the DO is pronominalized but challenging for gender; in D2 sentences it is mainly omitted, like for IO. After the age of 6, children’s performances in the acquisition of PersP’s comprehension and production increase with age. At the age of 8 for the comprehension task, and at the ages of 8 and 10 for the production task we identified a significant increase in the children’s performances. In this last task, the difficulties could be explained by its computational complexity in terms of morphosyntactic and pragmatic constraints The difficulties were focused on the use of object pronouns and mainly DO. We suggest that, in D2 sentences, children focus primarily on the partners of the interaction involved in the speech context (designated by S and IO), rather than on the object of the interaction (DO)
From settling to rheology and hydration: How fine fractions govern the behavior of cement-based suspensions
International audienceThis study investigates the role of fine particles in governing macroscopic behaviors of suspensions, in particular, cement-based pastes. Granulometric analyses were performed on both raw and binary systems of Portland cement with limestone filler re-grinded or not (LF2, LF1), and fly ash (FA) at various substitution rates (0–100 % vol) to modify fine particle content and identify a particle size threshold. Based on granulometric analyses, a threshold of 5 μm was justified and chosen for subsequent investigation. Suspension stability was assessed using Turbiscan Lab, measuring both settling velocity and supernatant clarity. The results indicate that systems with a higher fine fraction, such as those with LF2 (97.6 %), exhibit enhanced stability, as evidenced by significantly reduced settling velocity compared to LF1 (54.4 %) that demonstrates moderate stability, and FA (34.4 %) that displays pronounced instability. Rheological measurements were performed on raw and 50 % binary systems, and the results revealed that systems with a higher fine fraction exhibit a higher storage modulus G', notably LF2 compared to LF1 and FA, and OPC-50LF2 compared to OPC-50LF1 and OPC-50FA, indicating a denser structural network. Additionally, systems with a high fine fraction displayed a lower percolation threshold confirming the enhanced ability of fine particles to form interconnected networks. Isothermal calorimetry further showed that replacing 50 % of OPC with LF2 and LF1 leads to an increase in the intensity of the main hydration peak by 58 % and 47 %, respectively, compared to pure OPC, indicating that the nucleation-precipitation effect is more pronounced when more fine particles in the system
International audienceThe methodological challenge of engaging project stakeholders intersects with the application of the project-grounded research approach, which leverages design tools to create projects that integrate research objectives. The Urocyclus research project, focused on agricultural recycling of human urine, employs this approach to investigate its stakeholders. Drawing on this project-grounded research, the paper demonstrates how different categories of stakeholders are motivated to speak through the project and examines the conditions under which this occurs. The design project creates a novel in situ context that encourages participants to express themselves during the inquiry process. The elements designed and produced within the project are analysed in the research’s methodological framework to uncover the connections and interactions that enable the collection of diverse forms of discourse. The conceptual framework of Urocyclus is also presented, clarifying the socio-ecological issues underpinning the project and its stance regarding social acceptability. Finally, the strengths, weaknesses, and limitations encountered during the project-grounded research experimentation are critically examined. While the situated nature of this method constitutes a strength, its grounding in the field and the active participation it requires can also pose challenges to its implementation. Moreover, the dissemination of this approach presents a pedagogical challenge. As project-grounded research remains in its early stages, the analysis developed in this article contributes to strengthening empirical knowledge on the subject and outlines potential directions for advancing design research.
Exposure to a glyphosate-based herbicide does not alter maternal care and offspring quality in the European earwig
International audienceSublethal exposure to pesticides can affect the reproduction and population dynamics of many non-target organisms, such as insects. Among these pesticides, glyphosate-based herbicides (GBHs) were long thought to have no effect on insects because their active compounds can only affect plants and microorganisms. However, a growing body of evidence suggests that GBH can directly or indirectly alter a wide range of fitness-related traits in insects. In this study, we tested whether exposure to the GBH Roundup© affects maternal care behaviour and juvenile development, locomotion and immunity in the European earwig, an insect commonly found in vineyards and orchards. First, we exposed female earwigs to Roundup at concentrations ten times below, equal to and ten times above the normal application rate (NAR) and then measured the expression of maternal care in terms of egg collection, egg and juvenile care and defense. We also measured maternal self-grooming and locomotor activity, and the development time, weight and size of newly produced juveniles. In a second experiment, we exposed earwig juveniles to a control solution or to Roundup and then measured their locomotion, the expression of genes involved in their development (the juveniles hormone pathway), and their survival after exposure to a fungal pathogen. Overall, our results showed no significant effect of Roundup on any of the parameters measured. This finding suggests that direct exposure to a GBH formulation may not necessarily induce behavioural, physiological and developmental alteration in this species. It also calls for future studies to explore the underlying mechanisms behind this apparent lack of sensitivity. More generally, these results highlight the importance of assessing the impact of pesticide use and the factors driving potential resilience across a wide range of non-target organisms to ensure sustainable agricultural practices
