    Metabolomic Signature in Men with Central Serous Chorioretinopathy Using UHPLC-MS

    International audienceCentral serous chorioretinopathy (CSCR), the fourth leading cause of retinal disease, tends to affect men and is favored by corticosteroids. Untargeted systemic metabolomics was analyzed in 60 men with CSCR and 60 age-matched controls using UHPLC-MS. The analysis of sera revealed a total of 722 metabolites that differed significantly between the CSCR and control groups, with 592 being downregulated and 130 being upregulated. The metabolic profile exhibited incomplete breakdown products of protein digestion or catabolism, incomplete fatty acid beta-oxidation, and alterations in antioxidant metabolism. A substantial upregulation of three primary fatty acid amides was observed, along with a downregulation of long-chain acylcarnitines and kynurenines. The reduction in long- and medium-chain acylcarnitine, sphingosine-1-phosphate (S1P), tryptophan, and kynurenine, all of which are upregulated by glucocorticoids, does not support the hypothesis of excess glucocorticoid in patients with CSCR. In conclusion, we report here that patients with CRSC present a metabolic signature that could be useful as a complement to specific endocrine studies to better understand the relationship between corticoids and the pathology.     Measurement of the top-quark production cross-section and charge asymmetry at LHCb

    International audienceThe first measurements of the top- and antitop-quark differential production cross-sections and the top-quark charge asymmetry in the forward region are presented, using proton-proton collision data collected by the LHCb experiment at a centre-of-mass energy of 13 TeV corresponding to an integrated luminosity of 5.4 fb1fb^{-1}. The total production cross-sections of top and antitop quarks are also determined. Measurements are performed using the μ+b-jetμ+b\text{-jet} final state within a fiducial region defined by a b-jetb\text{-jet}p_{\text{T, jet}}>50 GeV and pseudorapidity 2.2<η_{\text{jet}}<4.0,, with the muon from the WW-boson decay required to have p_{\text{T},μ}>25 GeV and pseudorapidity 2.0<η_μ<4.5. The muon and bb-jet system must satisfy p_{T}(μ+\text{jet}) > 20 GeV. The measured integrated production cross-sections for the top and antitop quarks are σt=0.95±0.04±0.08±0.02σ_{t} = 0.95 \pm 0.04 \pm 0.08 \pm 0.02 pb, σtˉ=0.81±0.03±0.07±0.02σ_{\bar{t}} = 0.81 \pm 0.03 \pm 0.07 \pm 0.02 pb, where the first uncertainty is statistical, the second systematic, and the third accounts for the luminosity uncertainty. The top-quark charge asymmetry is measured to be ACt=0.08±0.03±0.01A_C^{t} = 0.08 \pm 0.03 \pm 0.01, where the first uncertainty is statistical and the second is systematic. These results are consistent with next-to-leading-order Standard Model predictions

    This thesis in ergonomics draws on an intervention conducted within a city administration to experiment with, and then institutionalize, a professional dialogue aimed at improving the quality of work. By articulating the framework of organizational development in the clinic of activity (Clot et al., 2024) with the framework of organizational geneses (Folcher et al., 2022) stemming from the instrumental approach (Rabardel, 1995), it analyzes how this genre of professional dialogue is appropriated by actors and collectives to transform work, up to the co-design of a new organization. The results show that, thanks to the analysis of the instrumental genesis of dialogue, occupational and cross-functional collectives cross boundaries, thereby setting the organization in motion (creating new roles, tools, tasks, and spaces). The thesis specifies the dialectical relationship between instrumental and organizational geneses and discusses methods to “provoke” these dynamics in the service of health, efficiency, and the ecological sustainability of work.Esta tesis en ergonomía se apoya en una intervención realizada en un ayuntamiento para experimentar y luego institucionalizar un diálogo profesional orientado a la calidad del trabajo. Al articular el marco del desarrollo organizacional en la clínica de la actividad (Clot et al., 2024) con el de las génesis organizacionales (Folcher et al., 2022) derivado del enfoque instrumental (Rabardel, 1995), analiza cómo los actores y los colectivos se apropian de este género de diálogo profesional para transformar el trabajo hasta el codiseño de una nueva organización. Los resultados muestran que, gracias al análisis de las génesis instrumentales del diálogo, los colectivos profesionales y transversales cruzan fronteras, poniendo así en movimiento a la organización (creación de nuevas funciones, herramientas, tareas y espacios). La tesis precisa las relaciones dialécticas entre las génesis instrumentales y las génesis organizacionales y discute métodos para “provocar” estas dinámicas al servicio de la salud, la eficacia y la sostenibilidad ecológica del trabajo.Cette thèse en ergonomie s’appuie sur une intervention menée dans une collectivité territoriale pour expérimenter puis instituer un dialogue professionnel afin d’agir sur la qualité du travail. Articulant le cadre du développement organisationnel en clinique de l’activité (Clot et al., 2024) et le cadre des genèses organisationnelles (Folcher et al., 2022) issu de l’approche instrumentale (Rabardel, 1995), la thèse analyse l’appropriation de ce genre de dialogue professionnel par les protagonistes et les collectifs pour transformer le travail jusqu’à la co-conception d’une nouvelle organisation. Les résultats montrent que, grâce à l’analyse des genèses instrumentales du dialogue, le collectif métier et les collectifs transverses franchissent des frontières, mettant ainsi l’organisation en mouvement (création de nouvelles fonctions, de nouveaux outils, de nouvelles tâches et de nouveaux espaces).     International audienceRegulation of hepatic amino acid metabolismAnne-Catherine Maurin and colleaguesProteostasis team, Human Nutrition Unit, INRAE/Clermont-Auvergne University, France___________________________________________________________________Marked changes in amino acid (AA) homeostasis have been associated with cachexia, due to high AA requirements related to tumor growth, immune response and associated metabolic changes. As the body does not have a dedicated system for storing AA, the adaptive breakdown of fibrillar proteins in skeletal muscles mobilizes AA to meet these requirements, contributing significantly to cachexia. The liver plays a central role in regulating protein/AA homeostasis, releasing many AA used by other organs and tissues and producing the majority of proteins secreted into the blood. In the liver during cachexia, highly AA-consuming processes are upregulated or induced, such as the massive production of positive acute phase proteins (APP)1 in response to inflammatory cytokines. Two major signaling pathways are involved in detecting intracellular AA levels and regulating AA utilization and supply accordingly2. The mTORC1 complex stimulates protein synthesis (notably via rpS6 phosphorylation) and inhibits autophagy when AA is abundant. Conversely, AA scarcity activates GCN2-dependent eIF2α phosphorylation, selectively reducing protein synthesis while upregulating the transcription factor ATF4, which stimulates the expression of genes involved in AA supply and utilization. Interestingly, these two pathways interact with each other. In addition, the eIF2α-ATF4 signaling pathway can be triggered by mitochondrial or endoplasmic reticulum stress, both of which have been reported in the liver during cachexia.In our recent study using the C26 mouse model, we found that pre-cachectic stage of cancer progression was already associated with a strongly induced production of positive APP and reduced levels of most AA in systemic circulation. At the same time, the liver exhibited increased expression of positive APP, decreased albumin expression and upregulated autophagy3. At the onset of cachexia, we observed simultaneous activation of rpS6 and eIF2a signalings in the liver, as well as increased expression of ATF4 target genes involved in AA synthesis and transport, and autophagy.     ZTF-SEDm Type Ia supernova sample for Twins Embedding spectrophotometric standardisation

    International audienceThis paper has two aims: the first one is to build a large homogeneous spectrophotometric Type Ia supernova (SN Ia) sample, using 3069 spectra from the second Zwicky Transient Facility data release (ZTF DR2). Using this sample we reproduce, as the second objective of the paper, the Twins Embedding (TE) spectrophotometric standardisation method, which led to an exceptionally low value of 0.073 mag for the intrinsic scatter. We improve the flux-calibration accuracy of the SEDm SN Ia spectral sample using the ZTF photometric data, which are calibrated at the percent level. We then apply the three steps of the TE parameterisation to a subset of 783 ZTF SN spectra near maximum light, and analyse the resulting standardisation methods. The precision of the phase correction model, which is the first step of the TE, is estimated at 0.01 mag in g band, using ZTF data. Despite the challenge posed by the ZTF spectrum extraction pipeline, we apply a first standardisation in color based on the second step of the TE, the Read Between The Lines (RBTL). When considering the scatter due to the redshift error and the flux calibration error, we estimate a 0.129 mag Hubble residual scatter for this ZTF sample as an upper limit. As expected from the low spectral quality, the final TE standardisation based on three non-linear parameters did not improve the overall dispersion. We release 1897 flux calibrated spectra of 1607 SNe Ia with an estimated photometric accuracy of 0.07 mag. We further demonstrate the ability to apply a spectrophotometric standardisation with limited quality spectra. The RBTL standardisation is more efficient than that of SALT with one less parameter, and the resulting host steps are consistent with zero, making it less prone to astrophysical bias. For future spectroscopic surveys, a better spectral quality would enable the full TE standardisation to be computed. (Abridged
