Digenic Inheritance of Monoallelic MUTYH and POLE Germline Variants in Adrenocortical Carcinoma: Implications for Tumorigenesis and Immunotherapy
International audienceLong-term total parenteral nutrition (TPN) results in the development of intestinal failure-associated liver disease (IFALD). Fligor et al. have depleted phytosterols from soy-based intravenous lipid emulsions in an attempt to prevent IFALD. Animal studies describe the mechanism of hepatotoxicity of phytosterols, while studies in patients associate high phytosterols with the severity of liver disease. Yet, there are unanswered questions on how much IFALD could be attributed to phytosterols. Challenges to using a phytosterol-depleted ILE include the possibility of the development of oxidation by-products in the lipid, higher cost in the face of as yet unknown clinical benefit from phytosterol depletion and challenges in performing randomised controlled trials to validate the efficacy of newer ILEs.Collision of monoallelic MUTYH and POLE germline variants in a patient with adrenocortical carcinoma who achieved a strong response to immunotherapy
Searching for dark matter with a spin-based interferometer
International audienceAxion-like particles (ALPs) arise from well-motivated extensions to the Standard Model and could account for dark matter. ALP dark matter would manifest as a field oscillating at an (as of yet) unknown frequency. The frequency depends linearly on the ALP mass and plausibly ranges from 10−22 to 10 eV/c2. This motivates broadband search approaches. We report on a direct search for ALP dark matter with an interferometer composed of two atomic K-Rb-3He comagnetometers, one situated in Mainz, Germany, and the other in Kraków, Poland. We leverage the anticipated spatio-temporal coherence properties of the ALP field and probe all ALP-gradient-spin interactions covering a mass range of nine orders of magnitude. No significant evidence of an ALP signal is found. We thus place new upper limits on the ALP-neutron, ALP-proton and ALP-electron couplings reaching below gaNN < 10−9 GeV−1, gaPP < 10−7 GeV−1 and gaee < 10−6 GeV−1, respectively. These limits improve upon previous laboratory constraints for neutron and proton couplings by up to three orders of magnitude
Alcool, méditation de pleine conscience et implémentation d'intention : évaluation (non concluante !) de l'essai contrôlé randomisé ALCOMEDIIT
International audienceL'émergence de nouvelles pratiques problématiques d’alcool chez les jeunes nécessite des stratégies de prévention innovantes. ALCOMEDIIT (pré-enregistrement OSF: https://osf.io/hc8xd?mode=&revisionId=&) est un essai contrôlé randomisé ciblant l’impulsivité, facteur déterminant du binge drinking (BD). Le groupe contrôle bénéficie d’un entretien motivationnel (EM) et d’un suivi des habitudes de consommation via une application mobile en EMA (ecological momentary assessment), permettant un recueil de données écologique. Le groupe expérimental reçoit, en complément, une introduction à la méditation de pleine conscience (MPC) et à l’implémentation d’intention (II).98 étudiant.e.s de l’Université de Caen Normandie (64 femmes et 34 hommes) agé.e.s entre 18 et 28 ans (m=21,04; s=2,21) ayant des pratiques de consommations non problématiques (score BD: m=13,38; s= 9,14), AUDIT (m=7,62; s=5,61) et craving sur 12 mois (m=4,80; s=1,59) et n’ayant pas de conséquences cognitives (BEARNI: m=22,28; s=3,07). Les analyses principales de l'étude, par GLM, n'ont pas montré d'effet significatif de l'intervention sur l'évolution du BD, de la consommation hebdomadaire, du craving, et des dimensions de l'UPPS. Aussi, pour chaque point supplémentaire sur l'AUDIT, le modèle prédit une augmentation de 0,429 du score d'évolution moyen du BD (p=.003). Cependant, pour chaque point supplémentaire au score de BD à T0, e modèle prédit une diminution de 0,745 du score d’évolution moyen du BD (p<.001).Certaines limites pourraient expliquer l'absence de résultat confirmant les hypothèses, notamment, le manque d'appropriation de la MPC par les participants ou l'impact du suivi mobile
A Disposable Dopamine Sensor Based on Oxidized Cellulose Nanofibril-Modified SPCE
International audienceDopamine is a major catecholamine neurotransmitter that plays an essential role in the functioning of the human central nervous system. Imbalances in dopamine levels are associated with neurological disorders and depression. Thus, measuring the concentration of DA in human body fluids is significantly important. In this work, TEMPO-oxidized cellulose nanofibrils (TOCNFs) extracted from marram grass (Ammophilia arenaria), harvested in the central western part of Tunisia, were utilized to modify disposable screen-printed carbon electrodes (SPCEs) for the sensitive detection of dopamine in biological fluids. Differential pulse voltammetry (DPV) measurements displayed a sensitivity of 7.92 µA/µM and a detection limit of 10 nM. The disposable TOCNF-modified SPCE presents a charge transfer coefficient, α, comparable to that of a TOCNF/graphene/AgNP composite-modified GCE. Moreover, it exhibits good repeatability (RSD = 1.9%), good reproducibility (RSD = 2.3%), and appreciable storage stability (91% of its initial response after 3 weeks). The prepared disposable sensor showed satisfactory recovery of dopamine in human urine samples

Many Travellers mention that they were fond of watching movies, particularly before the widespread introduction of television. In their narratives they often borrow the imagery of American Westerns, which were particular favorites, to position themselves historically and imagine themselves as victors and survivors -partly because these movies feature a familiar infrastructure of horses, wagons, and tents, but also because they provide a conflict model with which Travellers can identify. John Ward recalls his grandfather's days at the time of World War I, before Travellers lived in wagons: "They could build tentsprobably seen some Indian at it in a film, a silent film, that's how they learned it." Nan Joyce remembers a man who came to take care of her and the other children every evening when her mother was in prison: "He'd get back on his horse then and ride into Portadown, it was like a cowboy movie with him riding into the sunset!" […]. When she traveled in Northern Ireland during the worst time of the "Troubles" she says she got used to the bullets and riots because it was "like seeing a cowboy picture" […]. Imagerie cérébrale et premiers succès de la psychiatrie de précision
International audienceL’imagerie par rés onance magnétique (IRM) permet d’étudier les bases neurobiologiques de maladies psychiatriques comme la schizophrénie, les troubles bipolaires, les troubles du spectre autistique ou encore les dépressions résistantes. Si des différences d’épaisseur corticale entre les patients souffrant de ces maladies et les personnes témoins ont pu être mises en évidence de façon robuste, il n’existe actuellement pas de biomarqueur des maladies psychiatriques reposant sur l’IRM. Un des enjeux de la psychiatrie de précision est de valider de tels biomarqueurs, propres à une maladie ou à une dimension clinique donnée, mais également capables de prédire l’évolution clinique ou la réponse à un traitement pour transformer la prise en charge des patients. L’IRM fonctionnelle pourrait également constituer un outil prometteur pour le ciblage individualisé de stimulations non-invasives dans les dépressions ou les hallucinations résistantes, ainsi que pour l’entraînement par neurofeedback
Endotoxin removal by the OXIRIS filter for cardiogenic shock requiring veno-arterial extra-corporeal life support: the ECMORIX randomized controlled trial
International audienceBackground Most severe cardiogenic shock requires veno-arterial extracorporeal membrane oxygenation (VA-ECMO). The OXIRIS filter, has shown potential in reducing lipopolysaccharide (LPS) levels. Our objective was to compare the efficacy of the OXIRIS filter versus the ST-150 filter in reducing LPS plasma concentration. We hypothesized that the OXIRIS filter would reduce the endotoxin burden. Methods We conducted an open-label randomized prospective study in the cardiac intensive care unit of Dijon University Hospital. Forty patients with refractory cardiogenic shock requiring VA-ECMO and renal replacement therapy (RRT) were randomized to receive either OXIRIS filter or ST-150 filter. Blood samples were collected at multiple time points. The primary outcome was LPS mass (measured 24 h after the initiation of treatment). Secondary outcomes included LPS activity, cytokine levels, and clinical outcomes. Results 20 patients were allocated to each group and analyzed. LPS plasma concentrations were not different between the OXIRIS filter and ST-150 filter groups at H24 (599 pmol/ml of esterified 3-OH fatty acids [450;734] vs 520 [456;835], p = 0.983) or when analyzing all time-points by linear mixed modelling (538 [469;723] vs 507 [434;671] at H26, 576 [513;614] vs 624 [503;724] at H48 and 632 [513;660] vs 586 [538;776] at H72, p = 0.882). No significant between groups differences were found in LPS activity, inflammation markers (IL-6, TNF-α, IL-10, MCP-1), SOFA scores, VIS scores, or 28-day mortality (13 (65%) vs 9 (45%) p = 0.21). There was high variability in LPS concentrations, suggesting heterogeneity in endotoxemia. Conclusions In patients with cardiogenic shock supported by VA-ECMO and requiring renal replacement therapy, we could not evidence any reduction in LPS blood concentration in patients treated with treatment with OXIRIS filter in comparison to ST-150 filter. Further research is required to confirm these findings and optimize endotoxin removal in this population. Trials Registration: NCT04886180. Registered 10 May 2021. Graphical Abstrac
