Exploring the GSTP1 inhibition potential of photosensitizer compounds for enhanced cancer treatment in photodynamic therapy
Photodynamic therapy (PDT) has gained considerable attention in cancer treatment due to its non-invasive nature and the ability of photosensitizers to generate reactive oxygen species upon light activation, leading to tumor destruction. Glutathione S-transferase P1 (GSTP1) is a key enzyme in chemotherapy resistance, often overexpressed in various cancers, and its inhibition of GSTP1 presents a promising strategy to enhance cancer treatment. This study is aimed at assessing the potential of prominent photosensitizers as GSTP1 inhibitors through molecular docking analysis to strengthen the efficacy of PDT. The photosensitizers were docked into the active site of GSTP1, and their binding affinities, inhibition constants (Ki), and molecular interactions were assessed. Among the tested photosensitizers, zinc phthalocyanine, hypericin, and temoporfin emerged as the top candidates, exhibiting binding energies of - 10.8, - 10.2, and - 9.8 kcal/mol, along with Ki values of 0.012, 0.033, and 0.064 µM, respectively. These compounds outperformed the reference inhibitor ethacrynic acid, which had a binding energy of - 6.6 kcal/mol and a Ki of 14.35 µM. These findings suggest that the dual action of these photosensitizers provides a promising strategy for combating cancer and overcoming treatment resistance
CAP-RNAseq: an integrated pipeline for functional annotation and prioritization of co-expression clusters
Abstract Cluster analysis is one of the most widely used exploratory methods for visualization and grouping of gene expression patterns across multiple samples or treatment groups. Although several existing online tools can annotate clusters with functional terms, there is no all-in-one webserver to effectively prioritize genes/clusters using gene essentiality as well as congruency of mRNA-protein expression. Hence, we developed CAP-RNAseq that makes possible (1) upload and clustering of bulk RNA-seq data followed by identification, annotation and network visualization of all or selected clusters; and (2) prioritization using DepMap gene essentiality and/or dependency scores as well as the degree of correlation between mRNA and protein levels of genes within an expression cluster. In addition, CAP-RNAseq has an integrated primer design tool for the prioritized genes. Herein, we showed using comparisons with the existing tools and multiple case studies that CAP-RNAseq can uniquely aid in the discovery of co-expression clusters enriched with essential genes and prioritization of novel biomarker genes that exhibit high correlations between their mRNA and protein expression levels. Anevrizmatik Subaraknoid Kanamada Ultra-Erken ve Erken Tedavinin Etkileri: Tek Merkezli Retroprospektif Çalışma
Amaç: Anevrizmal subaraknoid kanamaların (aSAK) cerrahi tedavisinin zamanlaması konusundaki fikir ayrılıkları günümüzde hâlen devam etmektedir. Bu tartışmaya hastalığa bağlı sınırlamaların sebep olduğu aşikâr olup; konuya yönelik çok sayıda araştırma mevcuttur. Bu çalışmalar özellikle erken ve ultra erken cerrahiyi desteklemekle beraber; sonuçlar bu iki zamanlama içindeki tedavi seçeneklerinden hangisinin (endovasküler tedavi (EVT) veya cerrahi tedavi (CT)) üstün olduğu konusunda da henüz net değildir. Gereç ve Yöntemler: Bu çalışma, Trabzon Kanuni Eğitim Araştırma Hastanesi Beyin ve Sinir Cerrahisi Kliniğinde Haziran 2021 ile Haziran 2022 tarihleri arasında, aSAK tanısı ile tedavi edilen 50 hasta üzerinde retroprospektif olarak yapılmıştır. Hastalar çalışmamızda tedavi yöntemine bakılmaksızın (EVT veya CT) kanamanın ilk altı saat içinde tedavi edilenler ultra erken tedavi (Grup 1) ve altı saat-yirmi dört saat arasında tedavi edilenler erken tedavi (Grup 2) olmak üzere iki ayrı gruba ayrıldı. Bulgular: aSAK tanısıyla EVT veya CT ile tedavi edilen 50 hastanın 31’i erkek (%62) ve 19’u kadın (%38) idi. Grup 1, 14’ü EVT ve 12’si CT ile olmak üzere toplam 26 hastadan; Grup 2 ise 13’ü EVT ve 11’i CT ile olmak üzere toplam 24 hastadan oluşmaktadır. Grup 1 ve Grup 2’deki hastalara gerek EVT gerekse CT ile tedavi uygulanması durumunda istatistiksel olarak tedavinin tipi ve zamanlaması açısından birbirine üstünlüğü olmadığı görülmüştür (Sırasıyla p=0,678, p=0,680). Sonuç: Bu çalışma, aSAK tanılı hastaların klinik derecesine bakılmaksızın hem erken hem de ultra erken dönemde yapılan her iki müdahalenin (EVT veya CT) sonuçları açısından farklılık olmadığını göstermemiştir.</jats:p
Acute rheumatic fever: a single center experience
Aims: Acute rheumatic fever continues to be a serious public health issue globally and in our country. The aim was to evaluate the socioeconomic, clinical characteristics, and echocardiographic findings of patients diagnosed with acute rheumatic fever in the Sancaktepe district of Istanbul province, and to compare them with Turkiye and global data. Methods: The study was conducted retrospectively by reviewing the hospital records of 22 patients who presented to the pediatric cardiology outpatient clinic of Sancaktepe Training and Research Hospital and were diagnosed with acute rheumatic fever between March 2018 and October 2021. Demographic and socio-economic data, complaints, physical examination findings, clinical and laboratory features, initial echocardiographic findings at presentation, and follow-up data of the patients were identified. Results: Out of the 22 patients diagnosed with acute rheumatic fever, 13 (59.1%) were male, with a mean age of 131.9 months ±32.9. The patients were followed up for an average of 23.2±11.5 months. Most patients (36.3%) presented during the winter months. The average number of individuals living in the patients’ households was found to be 5.8±1. It was observed that 95% of the mothers and 45.5% of the fathers had completed primary education, none of the mothers were employed, and all employed fathers received minimum wage. A family history of acute rheumatic fever was present in 18.8% of the patients. When echocardiographic findings were evaluated, valve involvement was detected in 90.9% of the patients. While single valve involvement was observed in the majority (72.7%) of patients, mitral valve involvement was most common (81.8%). During the follow-up period, complete resolution was observed in 45.5% of patients with mild mitral regurgitation and 66.6% of patients with mild aortic valve regurgitation. Conclusion: While the majority of the data in the study are consistent with the literature, attention has been drawn to the challenges associated with the low socioeconomic status of our patient population. With the increase in socioeconomic status in our country, as in developed countries around the world, the incidence of disease may decrease.</jats:p
A Novel Model Based on Ensemble Learning for Phishing Attack
With the increase in the speed of the internet environment and the development of the infrastructures used, people have started to perform most of their work online. As much as this makes life easier, it also increases the possibility of being attacked by malicious people. Attackers can activate a phishing attack that aims to steal information from victims by creating copied, fake websites. While this attack is very old and somewhat simple, it can still be effective due to low IT literacy. People can enter their information on these fake websites out of spontaneity or ignorance or good intentions and be exposed to Phishing attacks. The compromise of a user's account information also puts at risk the security of the organization or institution to which it is connected. In this study, we propose a new machine learning-based ensemble model with feature selection methods to detect phishing attacks. Also, an ablation study is presented to measure the effect of different feature selection methods. The proposed model which we named as NaiveStackingSymmetric (NSS) is analyzed using the widely used accuracy (ACC), the area under curve (AUC), and F-score metrics as well as the polygon area metric (PAM), and it is shown that it outperforms other studies in the literature using the same dataset.</jats:p
Effects of Sports Massage on Post-Workout Fatigue
This study was conducted to evaluate the effect of sports massage on post-exercise fatigue. The meta-analysis method was used in the study. Meta-analysis is a statistical method used to combine the results of different research studies to answer a similar research question. It includes randomized controlled trials (RCTs) and observational studies examining the effect of sports massage on post-exercise fatigue. The study was designed in accordance with the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) guidelines. The findings indicate that sports massage is effective in reducing post-exercise fatigue symptoms and accelerating athletes' recovery processes. Most of the studies included in the analysis demonstrate that sports massage reduces muscle pain, improves muscle function, and minimizes inflammatory responses. However, there are some differences and debates in the research regarding the effectiveness of sports massage. Nevertheless, there is strong evidence overall for the potential of sports massage to reduce post-exercise fatigue and accelerate athletes' recovery processes. However, further randomized controlled trials and research in various populations are needed to optimize the effectiveness of sports massage and standardize application protocols. Future studies should focus on different sports disciplines and athlete profiles to evaluate the effectiveness of sports massage in a broader context.</jats:p
Histological Scoring Systems for the Assessment of the Degree of Lung Injury in Rats
Acute respiratory distress syndrome (ARDS) is a serious pulmonary reaction with well-defined clinical parameters in humans triggered by many causes besides bacterial and viral pneumonia. However, there is no definitive definition of ARDS parameters in the experimental animal model. With its 2010 workshop report, the American Thoracic Society defined the main histopathological features that determine the presence of ARDS in laboratory animals, such as changes in parenchymal tissue, altered integrity of the alveolar capillary barrier, inflammation, and abnormal lung function. Understanding these parameters, scoring tissue lesions is used to convert observational pathological data into semi-quantitative or quantitative data for statistical analysis and improved precision.</jats:p
Systematic Assessment of Student Autonomy in Science Process Skills: 9th Grade Physics Curriculum and Textbook Analysis
This study aimed to determine how well a physics textbook aligns with the curriculum -the first skill-oriented one in Türkiye- regarding science process skills and student autonomy. A content analysis of both the curriculum and the textbook was conducted using the Science Process Skills Inventory. Findings revealed that the 9th grade physics curriculum emphasized skills such as “collecting and interpreting data”, “communicating”, “measuring”, and “experimenting” while it neglected “observing”, “predicting”, and “inferring”. The curriculum aimed to develop these skills by encouraging students to plan and take responsibility for their learning activities. The content analysis showed that the physics textbook primarily focused on “collecting and interpreting data”, and “measuring” with frequent emphasis on “observing”, “classifying”, “inferring”, and “modeling”. However, it largely ignored “hypothesizing”, and “defining and controlling variables”. The skills were presented in highly structured activities, limiting student autonomy. The results indicated an alignment between the curriculum and the textbook in the inclusion of “collecting and interpreting data”, “measuring”, and “communicating”. However, there was a notable inconsistency in the level of openness. While the curriculum expected students to engage in designing scientific activities, the textbook provided step-by-step procedures that restricted student autonomy to create their scientific processes.</jats:p
On Generalized Fibospinomials: Generalized Fibonacci Polynomial Spinors
Spinors are important objects in physics, which have found their place more and more after the discovery that particles have an intrinsic angular momentum shape and Cartan’s mathematical expression of this situation. Recent studies using special number sequences have also revealed a new approach to the use of spinors in mathematics and have provided a different perspective for spinor research that can be used as a source for future physics studies. The purpose of this work is to expand the generalized Fibonacci quaternion polynomials to the generalized Fibonacci polynomial spinors by associating spinors with quaternions, and to introduce and investigate a new polynomial sequence that can be used to benefit from the potential advantages of spinors in physical applications, and thus, to provide mathematical arguments, such as new polynomials, for studies using spinors and quaternions in quantum mechanics. Starting from this point of view, in this paper we introduce and investigate a new family of sequences called generalized Fibospinomials (or generalized Fibonacci polynomial spinors or Horadam polynomial spinors). Being particular cases, we use (r,s)-Fibonacci and (r,s)-Lucas polynomial spinors. We present Binet’s formulas, generating functions and the summation formulas for these polynomials. In addition, we obtain some special identities of these new sequences and matrices related to these polynomials. The importance of this study is that generalized Fibospinomials are currently the most generalized sequence in the literature when moving from Fibonacci quaternions to spinor structure, and that a wide variety of new spinor sequences can be obtained from this particular polynomial sequence.</jats:p
PSİKOLOJİK YIPRANMA VE MESLEK HASTALIKLARI: BİBLİYOMETRİK BİR ARAŞTIRMA
Günümüzde örgütler, psiko-sosyal risk faktörlerinin oluşturduğu yeni çalışma ortamında, teknolojide hızlı değişimlerin yaşandığı, ruh sağlığı konusunda farkındalığın arttığı, çeşitlilik ve katılıma daha fazla vurgu yapılan bir dünyanın karmaşıklıklarıyla başa çıkmak zorundadır. Neo-liberal çalışma krizinin yol açtığı çeşitli sorunların yanı sıra artık pandemi sonrası geriye dönülmez bir biçimde norm haline gelmiş olan yeni çalışma koşulları eski şartlarda düşünmeyi imkânsız kılmaktadır. Son yıllarda artan şekilde karşılaştığımız depresyon, anksiyete, tükenmişlik, intihara eğilimlilik vb. şikayetler nedeniyle hastaneye başvuran kişilerin örneğin bankacılar gibi spesifik meslek gruplarında artış göstermesi bu mesleğin yapısı gereği bu tür psikolojik şikayetlere neden olup olmadığı, bu tür bilgi yoğun ve beyaz yaka mesleklerde performans yönetimi ve örgüt yapısı nedeniyle, daha çok mavi yaka, üretim sektörü ve kirli işlerle ilişkilendirilen meslek hastalıkları ve iş güvenliği konusunun genişleyerek, yeni tanımlanabilecek meslek hastalıklarının olabileceğine ilişkin kanıtlar sunmaktadır.Buna göre bu çalışmanın amacı psikolojik yıpranma ve meslek hastalıkları ilişkisini incelemektedir. Buna göre psikolojik yıpranma ve meslek hastalıkları kavramları ve bu kavramlar arasındaki ilişki bibliyometrik yöntemle kavramsal haritalar aracılığıyla incelenmeye çalışılmıştır. Bibliyometrik verilerin analiz edilmesinde WOS veri tabanı ve R programından yararlanılılmıştır. Çalışmanın sonucunda teori ve uygulayıcılar açısından öneriler geliştirilmesi hedeflenmektedir.</jats:p
