Fabrication and evaluation of aptamer-conjugated paclitaxel-loaded magnetic nanoparticles for targeted therapy on breast cancer cells
Targeted drug delivery vehicles make it possible to deliver anti-cancer drugs to the cells or tissues of interest. Aptamers are peptide or oligonucleotide molecules that can serve as targeting elements of drug carriers. In the current study, we evaluated the capacity of an aptamer-based drug carrier to deliver Paclitaxel (PTX) to cancer cells. After being synthesized, SPIONs@PTX-SYL3C aptamer was characterized using different methods, including differential light scattering (DLS), infrared spectroscopy (FTIR), Transmission electron microscopy (TEM), X-ray diffraction (XRD), Thermal gravimetric analysis (TGA), and vibrating sample magnetometer (VSM). Encapsulation efficiency (EE) and loading efficiency (LE) were also evaluated. The carrier was applied on 4T1, MCF 7, and MCF-10A breast cell lines to evaluate its drug delivery potency and specificity. EE and LE were calculated to be 77.6 and 7.76, respectively. MTT results revealed that aptameric SPIONs@PTX was more toxic than non-aptameric SPIONs@PTX. Flowcytometry analysis and DAPI staining confirmed that SPIONs@PTX-Aptamer had higher cell internalization rate when compared to non-targeted SPIONs@PTX. Our results indicate that aptamer-conjugated SPIONs@PTX has a good capacity in recognizing its target cells and inhibiting their growth and division. © 2021, The Author(s), under exclusive licence to Springer Nature B.V. part of Springer Nature
Establishing a possible association between somatic measurements and penile dimensions
Background: Concerns and doubts about body image seem to be rather common among adolescents and young men, particularly. Many of them express great preoccupation about the normalcy of their attributes of masculinity. However, their worries are less concentrated in the aspect of their secondary sexual characters and in most cases are focused on the dimensions of their genitalia, particularly their penis. The feeling of sexual inadequacy may result in low elf-esteem among those men who consider their penis, and therefore their masculinity, sexual maturity, and prowess, inadequate. Methodology: A descriptive, survey-based, and cross-sectional study was conducted among 1110 males aged 18�50 years from five countries. The measurements were made using a standardized approach that consisted in using a flexible tape extended from the pubic bone to the tip of the glans for the length and around the middle of the shaft (circumference). The study was conducted online during three consecutive months. Results: Among the individuals of the sample, the mean reported body height is 176 cm ranging from 163 cm to 195 cm with a standard deviation of 10.02 cm. The reported body weight for them ranges from 55 kg to 105 kg with a mean of 77.5 kg and a standard deviation of 13.4 kg. The reported penis's length among the men in the sample ranges from 10.5 cm to 14.82 cm with a mean of 12.66 cm and a standard deviation of 3.4 cm. Conclusion: Length and circumference of the penis can be measured by equations to assess the abnormality depending on height and weight in relation to the constant. These three equations can be used in clinical and diagnostic setting to help clinicians and doctors to take an idea about the patients� cases and how to deal with them. © 2021 Sexologie
Physico-chemical changes and maturity evaluation of composts from wood residue mixed with sewage sludge and chicken manure
Purpose Using the effective approaches for modifying the recycled wood as a novel bulky agent improves the quality of soil amendment. This study aimed to compare the stability and maturity of the soil amendments produced by the compostation of forest industrial waste and sewage sludge on seed germination. Method Three materials, namely sawdust, sewage sludge, and chicken manure were mixed at different ratios (dry weight basis) to reach the initial bulk density of 0.40 Kg.L-1, the temperature of 30 °C, the C/N ratio of 25, and the moisture content of 60 . A pilot-scale composting process was applied to monitor the aeration rate, temperature, and moisture contents during the process over time. The comparison of physico-chemical, phytotoxicity, and germination indices among the samples was performed in three repetitions. Results The results of this study indicated that the sustainable conditions (i.e., the temperature of 70 °C, aeration rate of 0.30 L.Kg-1DM.min-1, and moisture content of 50-60 ) can have a significant effect on the thermophilic stage for compost curing without any inhibitory repercussion. Increasing the germination index of cress (Lepidium sativum) up to 79 proved that the toxicity of industrial sewage sludge was declined through the elimination of heavy metals. Conclusion This study revealed that the availability of bulky agents such as wood residue can reinforce the microbial activity by continuously decreasing the C/N ratio to the minimum value of 13.2. © 2021, Islamic Azad University. Telomere shortening occurs in patients with ectodermal dysplasia
Ectodermal Dysplasia (ED) is a rare congenital disorder that is characterized by sparse hair, lack or absence of sweat glands, and rod-shaped teeth, which are diagnosed by two or three of these features. There are approximately 200 subtypes of ED have been identified. One of the rare and severe subtypes of ED is Dyskeratosis Congenita (DC). A mutation in the telomere-maintain genes causes this disorder. Since this rare disorder is a subset of ED disease, this study aimed to examine telomeres in these patients and expect to see telomere shortening in ED patients. Study on SARS-CoV-2 strains in Iran reveals potential contribution of co-infection with and recombination between different strains to the emergence of new strains
We aimed to describe SARS-CoV-2 strains in Iranians from nine distributed cities infected during two months expanding late 2020 and early 2021 by genotyping known informative single nucleotide in five PCR amplicons. Two variants associated with haplotype H1 (clade G) and nine additional variants associated with other haplotypes were genotyped, respectively, in RNA isolates of 244 and 85 individuals. The variants associated with the H1a (GR) and H1b (GH) haplotypes were most prevalent, indicating a significant change in infection pattern with passage of time. The most important findings were that recombinant genomes and co-infection, respectively, were surmised in 44.7 and 12.9 of the samples extensively genotyped. Partners of many of the recombinations were relatively common strains. Co-existing viruses were among those currently circulating in Iran. In addition to random mutations, co-infection with different existing strains and recombination between their genomes may significantly contribute to the emergence of new SARS-CoV-2 strains. © 2021 Elsevier Inc
Naringin Chelates Excessive Iron and Prevents the Formation of Amyloid-Beta Plaques in the Hippocampus of Iron-Overloaded Mice
Metal chelating agents are antioxidant agents, which decrease the reductive potential and stabilize the oxidized metal ion form. In this study, we evaluated the naringin capacity in chelating iron and preventing amyloid-beta plaque formation in the hippocampus of iron-overloaded mice. Thirty-five NMRI male mice (8�10 weeks old) were provided. The mice were classified into five groups. Iron dextran was administered as i.p. injection (100 mg/kg/day) four times a week for four subsequent weeks. The treated groups received 30 and 60 mg/kg/day naringin for a month. After histological processing, the brain sections were stained with Perls� stain kit for iron spots, and Congo red was used to stain the brain and hippocampus for amyloid-beta plaques. 30 mg/kg/day of naringin was shown to decrease nonheme iron in an efficient manner; iron content in this group decreased to 16.83 ± 0.57 μg/g wet weight, a quantity as low as that observed in the normal saline-receiving group. The nonheme iron content in the mice receiving 60 mg/kg/day of naringin was 20.73 ± 0.65 μg/g wet weight. In addition, Aβ plaque numbers in CA1, CA3, and DG areas of the hippocampus decreased significantly following treatment with 30 or 60 mg/kg/day naringin. Naringin has a strong iron chelation capacity and is able to reduce the formation of amyloid plaques. So it can be useful for neuroprotection and prevention of Alzheimer�s disease. © Copyright © 2021 Jahanshahi, Khalili and Margedari
Identification of differentially expressed micrornas in primary esophageal achalasia next-generation sequencing
Molecular knowledge regarding the primary esophageal achalasia is essential for the early diagnosis and treatment of this neurodegenerative motility disorder. Therefore, there is a need to find the main microRNAs (miRNAs) contributing to the mechanisms of achalasia. This study was conducted to determine some patterns of deregulated miRNAs in achalasia. This case-control study was performed on 52 patients with achalasia and 50 nonachalasia controls. The miRNA expression profiling was conducted on the esophageal tissue samples using the next-generation sequencing (NGS). Differential expression of miRNAs was analyzed by the edgeR software. The selected dysregulated miRNAs were additionally confirmed using the quantitative reverse transcription polymerase chain reaction (qRT-PCR). Fifteen miRNAs were identified that were significantly altered in the tissues of the patients with achalasia. Among them, three miRNAs including miR-133a-5p, miR-143-3p, and miR-6507-5p were upregulated. Also, six miRNAs including miR-215-5p, miR-216a-5p, miR-216b-5p, miR-217, miR-7641 and miR-194-5p were downregulated significantly. The predicted targets for the dysregulated miRNAs showed significant disease-associated pathways like neuronal cell apoptosis, neuromuscular balance, nerve growth factor signaling, and immune response regulation. Further analysis using qRT-PCR showed significant down-regulation of hsa-miR-217 (p-value = 0.004) in achalasia tissue. Our results may serve as a basis for more future functional studies to investigate the role of candidate miRNAs in the etiology of achalasia and their application in the diagnosis and probably treatment of the disease. © T�BİTAK
Color change of primary teeth following exposure to an experimentally synthesized liposomal nano-encapsulated ferrous sulfate drop versus the commercially available iron drops
Objectives: This study aimed to assess the color change of primary teeth following exposure to an experimentally synthesized liposomal nano-encapsulated ferrous sulfate drop compared with the commercially iron drops. Materials and methods: In this in-vitro study, liposomal nano-encapsulated ferrous sulfate (lipo-nano-ferr) drop was first synthesized. Next, 110 extracted primary anterior teeth were randomized into two equal groups (sound and demineralized). Each group was randomly divided into 5 subgroups (n = 11) and exposed to Irofant, Feroglobin, Sideral, and lipo-nano-ferr drops for 540 cycles. One subgroup was remained in artificial saliva as the negative control. Tooth color was analyzed before and after the interventions visually, and the rate of iron adsorption was analyzed by atomic absorption spectrometry (AAS), and energy-dispersive X-ray spectroscopy/scanning electron microcopy (EDS/SEM). Data were analyzed using t-test, one-way ANOVA, and LSD test. Results: Iron adsorption was significantly higher by the demineralized specimens than the sound specimens in all subgroups (P < 0.001). A significant difference was noted in iron adsorption among the four subgroups in each of the sound and demineralized groups (P < 0.001). Iron adsorption in the lipo-nano-ferr subgroup was significantly lower than that in Irofant and Feroglobin groups (P < 0.001). The visual inspection results were similar to AAS results. EDS-SEM showed that the atomic percentage of iron in Sideral and lipo-nano-ferr groups was significantly lower than that in Irofant and Feroglobin groups (P < 0.001). Conclusion: Use of nano-encapsulation technology in production of iron drops can significantly decrease the resultant tooth discoloration, causing a clinically negligible color change. © 2021 Japanese Society of Pediatric Dentistr
Building a cancer biobank in a low-resource setting in Northern Iran: The Golestan Cancer Biobank
Background: We aim to present the development and the initial results of the Golestan Cancer Biobank (GoCB), in a low resource setting in northern Iran. Methods: The GoCB protocol and its standard operation procedures (SOP) were developed according to internationally accepted standards and protocols with some modifications considering the limited resources in our setting. The main biological samples collected by the GoCB include blood sample, urine sample, fresh endoscopy tissue sample, fresh surgical tissue sample and formalin fixed paraffin embedded (FFPE) tissue sample. The GoCB collects patients' demographic data, tumor characteristics as well as data on risk factors. We developed a specific GoCB software for management of patient data and biological sample information. The GoCB dataset is annually linked with the Golestan cancer registry dataset to add complementary data (e.g., survival data). Results: The GoCB started collection of data and biological samples in December 2016. By November 2020, a total number of 1217 cancer patients participated in the GoCB. The majority of the GoCB participants (n = 942, 77) were those with gastrointestinal and breast cancers. Data on risk factors were successfully collected in 684 (56.2) of the participants. Overall, 3563 samples were collected from the GoCB participants and 730 samples were used in 7 national and international research projects. Conclusion: We considered specific strategies to overcome major limitations, especially budget shortage, in the development and maintenance of a cancer-specific biological repositories in our setting. The GoCB may be considered as a model for the development of biobank in low- and middle-income countries (LMICs). © 2021 Academy of Medical Sciences of I.R. Iran. All rights reserved
