ABSTRACT.- Uruguay's dairy can potentially enhance milk productivity competitively, but intensification risks elevating nitrogen (N) surplus, heightening environmental concerns. This study quantified farm-gate N inputs and outputs, calculating N surplus (input-output) and N use efficiency (NUE=output/input) for 17 commercial modal dairy systems identified in the 2014 and 2019 national surveys and six prospective intensified systems based on experimental pastoral farmlets achieving near-maximal rainfed productivity. .-.-.-.-.-.-.-.-.-. .-.-.-.-.-.-.-.-.-. @ 2024 Agrociencia Urugua

Carbon footprint of mixed farming crop-livestock rotational-based grazing beef systems using long term experimental data.

ABSTRACT.- In the context of ever-growing demand for food and associated concerns regarding the environmental impacts of high-input agricultural systems, there is growing interest in mixed farm enterprises to deliver greater sustainability compared with mono-enterprise production systems. However, assessments of such systems are complex and require high-resolution data to determine the true value and interconnectivity across enterprises. Given the scarcity of information on mixed crop-livestock systems and the difficulties of its analysis, we perform life cycle assessment using temporally high-resolution data (2019-2022) from a long-term experiment in South America to evaluate the 'cradle-to-farmgate exit' greenhouse gas emissions intensities of four rotational crop-livestock systems. Systems evaluated were continuous cropping: 2 years of continuous cropping; short rotation: 2-year continuous cropping plus 2-year pasture; long rotation: 2-year continuous cropping followed by 4-year pasture; and forage rotation: continuous pasture. © The Author(s) 2024

The essential kinase TgGSK regulates centrosome segregation and endodyogeny in Toxoplasma gondii

Intracellular replication is crucial for the success of apicomplexan parasites, including Toxoplasma gondii. Therefore, essential players in parasite replication represent potential targets for drug development. We have characterized TgGSK, a glycogen synthase kinase homolog that plays an important role in Toxoplasma endodyogeny. We have shown that TgGSK has a dynamic localization that is concurrent with the cell cycle. In non-dividing parasites, this kinase is highly concentrated in the nucleus. However, during division, TgGSK displays a cytosolic localization, with concentration foci at the centrosomes, a key organelle involved in parasite division, and the basal end. Conditional knockdown of TgGSK determined that it is essential for the completion of the lytic cycle and proper parasite division. Parasites lacking endogenous protein levels of TgGSK exhibited defects in division synchronicity and the segregation of the nucleus and apicoplast into forming daughter cells. These phenotypes are associated with defects in centrosome duplication and fission. Global phosphoproteomic analysis determined TgGSK-dependent phosphorylation of RNA-processing, basal end, and centrosome proteins. Consistent with the putative regulation of RNA-processing proteins, global transcriptomic analysis suggests that TgGSK is needed for proper splicing. Finally, we show that TgGSK interacts with GCN5b, a well-characterized acetyltransferase with roles in transcriptional control. Conversely, GCN5b chemical inhibition results in specific degradation of TgGSK. Thus, these studies reveal the involvement of TgGSK in various crucial processes, including endodyogeny and splicing, and identify acetylation as a possible mechanism by which this essential kinase is regulated. Importance: While infection with the parasite Toxplasma gondii is largely asymptomatic in healthy adults, severe disease and death can result in immunocompromised individuals and in those infected congenitally. With minimal treatments for toxoplasmosis available, it is crucial to study parasite-specific processes to identify new drug targets. This study investigated the protein TgGSK, uncovering its essentiality for parasite proper division and survival. We performed an in-depth study of the functional role of this kinase. Importantly, TgGSK was shown to bear higher homology to plant proteins than its mammalian counterparts, which may allow for specific targeting of this protein.National Institutes of Health grants R01AI149766, R01DK124067, and R21AI164619 to G.A.Pasteur Network, G4 grantFOCEM (MERCOSUR 645 Structural Convergence Fund), COF 03/11. Recent co-evolution of two pandemic plant diseases in a multi-hybrid swarm.

    No full text
Most plant pathogens exhibit host specificity but when former barriers to infection break down, new diseases can rapidly emerge. For a number of fungal diseases, there is increasing evidence that hybridization plays a major role in driving host jumps. However, the relative contributions of existing variation versus new mutations in adapting to new host(s) is unclear. Here we reconstruct the evolutionary history of two recently emerged populations of the fungus Pyricularia oryzae that are responsible for two new plant diseases: wheat blast and grey leaf spot of ryegrasses. We provide evidence that wheat blast/grey leaf spot evolved through two distinct mating episodes: the first occurred ~60 years ago, when a fungal individual adapted to Eleusine mated with another individual from Urochloa. Then, about 10 years later, a single progeny from this cross underwent a series of matings with a small number of individuals from three additional host-specialized populations. These matings introduced non-functional alleles of two key host-specificity factors, whose recombination in a multi-hybrid swarm probably facilitated the host jump. We show that very few mutations have arisen since the founding event and a majority are private to individual isolates. Thus, adaptation to the wheat or Lolium hosts appears to have been instantaneous, and driven entirely by selection on repartitioned standing variation, with no obvious role for newly formed mutations. © 2023, The Author(s)
