Chemical Composition, Antimicrobial and Antioxidant Properties of Essential Oil of Origanum vulgar ssp. Gracile
BACKGROUND AND OBJECTIVE: Nowadays, in order to improve the health and reduce the risks of chemical preservatives in the food industry, researchers have focused on the use of natural compounds, especially plants essential oils that have antioxidant and antimicrobial properties, due to the increased risk of bacteria Escherichia coli and Listeria monocytogenes in foods, in this study the antimicrobial and antioxidant properties of Origanum vulgar ssp. Gracile essential oil were investigated in vitro.
METHODS: In this experimental study, Origanum vulgar ssp. Gracile plant was prepared from Kurdistan and after drying and extraction of essential oils, components were identified using GC-MS device. Strains were prepared from the bacteriology laboratory of the Faculty of Veterinary Medicine, Tabriz University. In order to determine the minimum inhibitory and fatality concentration, various concentrations of extract were used (0.312, 0.625, 1.25, 2.5, 5 and 10 %), the minimum inhibitory and fatality concentration were determined using microdilution method. The antioxidant activity was performed by using the DPPH method and Total phenolic material was measured by Folin Ciocalteu reagent.
FINDING: Based on the results of GC-MS analysis, carvacrol (49.33%) had the most component of the extract composition. The MIC and MBC of essential oils on Escherichia coli were 1.25 and 2.5%, respectively, and 0.625 and 1.25% for Listeria monocytogenes, respectively. In addition, the antioxidant activity 12.45±0.03 μg/ml, and the content of phenol 39.13±4.13 mg of gallic acid in grams of essence was calculated.
CONCLUSION: The results of this study showed that Origanum vulgar ssp. Gracile essential oil had antioxidant properties, and the essence had an inhibitory effect on both bacteria and Listeria monocytogenesis was less resistant than Escherichia coli to the essential oil
Relationship between Obstructive Sleep Apnea (OSA) and Difficult Intubation
BACKGROUND AND OBJECTIVE: One of the challenges of patients who are candidates for anesthesia is difficult intubation, which leads to severe complications and even death after anesthesia. The aim of this study is to investigate the relationship between obstructive sleep apnea and difficult intubation through systematic review and meta-analysis.
METHODS: In this review article, observational articles about the relationship between obstructive sleep apnea and difficult intubation were extracted without time limit by searching national and international databases and the keywords were: difficult intubation, problematic intubation, Intra tracheal-endotracheal, difficult airway OSA, OSAS, obstructive sleep apnea, sleep breathing disorder, anesthesia, and their Persian equivalents. Data were analyzed using meta-analysis and fixed effects model. In order to study the heterogeneity and contradictions in the studies, Q Cochrane and I2 indices were used, respectively.
FINDINGS: Of the 72 found articles, 9 articles with a sample size of 1,126 and an average of 125 subjects were included in the study. The results of this study showed that the relationship between obstructive sleep apnea and difficult intubation is significant (OR = 3.88, CI95% = 2.69 – 5.61). In addition, the results of the analysis based on country showed that the highest and lowest odds ratios were observed in studies conducted in France and Canada, respectively.
CONCLUSION: The results of this study showed that there is a correlation between obstructive sleep apnea and difficult intubation
Second Molar Uprighting with Temporary Anchorage Devices: A Finite Element Study
BACKGROUND AND OBJECTIVE: Premature loss of mandibular first molar is a common problem in adults. Mesial tipping of second molar may occur in this situation. Various orthodontic mechanics have been proposed for molar uprighting. The aim of this study was to compare four methods of molar uprighting using Finite Element Analysis(FEM).
METHODS: In first model of this finite element study, a 0.019×0.025 inch beta-titanium segmental arch wire with a T-loop was used. In second model a miniscrew was inserted in retromolar space and force was applied using elastomeric chain. The third model was a piece of 0.016×0.022inch beta-titanium wire with a bend which was placed more occlusal than the screw. The fourth model contained a mesially inserted miniscrew with an angle of 70 degrees to bone surface and a 0.018×0.025inch beta-titanium wire with helix. Extrusion, center of rotation and stress distribution in PDL during movement was compared between methods.
FINDINGS: Buccal cusp extruded 1.36E-03, 1.13E-03, -9.74E-04 and 1.49E-03 mm in first, second, third and fourth model, respectively. Similarly, in lingual cusp, the amount of vertical displacement was at least in third model (-6.83E-04 mm). This amount in second and first method was 1.12E-03 and 4.05E-04 mm, respectively. The maximum amount of extrusion of lingual cusp occurred in fourth model (9.01E-03 mm).
Mesial and distal cusps extruded 2.12E-04 and 1.58E-03 mm in first model, -1.14E-03 and 3.80E-03 mm in second method, -2.37E-03 and 7.04E-04 mm in third design and, 1.88E-03 and 8.57E-03 mm in the fourth model.
The center of rotation was located at molar bifurcation in third model.
CONCLUSION: The maximum amount of extrusion in both mesiodistal and buccolingual path was seen in fourth model. The best type of movement was found in third model, in which minimum extrusion occurred and center of rotation located at molar bifurcation
The Effect of Hydroalcoholic Extract of Cuscuta Europaea on Breast Cancer Cells (Mcf7) in Comparison with Kidney Epithelial Cells (Vero cell line)
BACKGROUND AND OBJECTIVE: Studies have shown that plants of Family Cuscutaceae can affect the growth and development of cancer cells. It is important that the herbs used for non-cancerous cells do not have cytotoxic effects. The present study was conducted to evaluate the effects of hydroalcoholic extracts of Cuscuta europaea on the viability of breast cancer cells (MCF7) compared to non-cancerous kidney epithelial cells (Vero cell line).
METHODS: In this experimental study, Vero cells and MCF7 cells were prepared from Pasteur Institute Cell Bank and divided into the control group and the groups treated with doses of 0.01, 0.1, 1 and 10 mg / ml. 48 hours after exposure, the cytotoxic effect of the extract was measured by MTT.
FINDINGS: Compared with the control group, exposure to dose of 0.01, 0.1, 1 and 10 mg/ml of hydroalcoholic extract of Cuscuta europaea reduced the viability of MCF7 cells (54.47±0.35, 55±0.42, 44.62±0.38, and 4.64±0.73%, respectively) and Vero cells (58.42±0.45, 48.61±0.55, 43.14±0.23, and 5.35±0.27%, respectively) (p<0.001).
CONCLUSION: The results of this study showed that the hydroalcoholic extract of Cuscuta europaea can simultaneously cause toxicity in non-cancerous cells, although it has toxic effects on breast cancer cells, and in this regard, the use of this extract in the treatment of breast cancer does not seem favorable due to its toxic effects on non-cancerous cells
An Evaluation of the Association between Periodontal Disease and Preeclampsia in Pregnant Women
BACKGROUND AND OBJECTIVE: In the etiology of preeclampsia, one of the most common causes of maternal mortality is the release of inflammatory and immunological factors. On the other hand, periodontal disease also leads to persistent gingivitis. Therefore, the present study was conducted to evaluate the association between periodontal disease and preeclampsia.
METHODS: This case-control study was conducted among 80 pregnant women with preeclampsia (case group) and 80 healthy pregnant women as control group. The mean of periodontal disease indices, including Plaque Index (PI), Clinical Attachment Loss (CAL), and Gingival Bleeding Index (GBI) was measured and compared between the two groups.
FINDINGS: There was no significant difference between the two groups in terms of mean age, BMI, gestational age and number of pregnancies. The mean CAL in the case group was 14.14±1.0 mm and in the control group was 13.14±1.0 mm (p=0.68), the mean PI score in the case group was 1.18±1.13 and in the control group was 1.18±0.17 (p=0.87), and the mean GBI in the case group was 14.2±7.8% and in the control group was 14.3 ± 3.1% (p=0.35).
CONCLUSION: The results of this study showed that the difference between the mean of periodontal disease indices in preeclampsia group and control group was not statistically significant
Immunization Evaluation of Type III Secretion System Recombinant Antigens and Shiga Like Toxin Binding Subunit of E. coli O157:H7
BACKGROUND AND OBJECTIVE: Escherichia coli enterohemorrhagic (EHEC) causes diarrhea, hemolysis colitis, and uremic hemolytic syndrome. Due to the dangers of antibiotic treatment in the EHEC O157: H7 infection, vaccines offer a promising way to prevent infection. The purpose of this study was to investigate the immunity of recombinant proteins Intimin, EspA and Stx2b against infection of E. coli O157 H7.
METHODS: In this research study, expression of recombinant Intimin, EspA and Stx2b proteins E.coli BL21 DE3 was induced with IPTG. Recombinant proteins were examined by Western Blot and purified using the affinity Ni-NTA chromatography. Subcutaneous and intraperitoneally immunization of mice was performed and the antibody titer was determined by ELISA. The mice were orally infected with bacteria and the bacterial loss and mortality of the mice were examined.
FINDINGS: SDS-PAGE analysis showed expression of proteins with 11,17 and 34 kDa molecular weight. The recombinant proteins were confirmed by Western Blot. ELISA end point titer for humoral response to EspA, Intimin and Stx2b was 25600. The loss of bacteria in mice was 102 cfu / ml and mortality rate were reduced to 60 Percent.
CONCLUSION: The results showed that immune response against EspA, Intimin and Stx2b proteins protects animals against challenges using E. coli O157: H7
The Effect of Tricine Compound Extracted from Allium atroviolaceum Boiss. On growth and apoptosis in PC3 cell line
BACKGROUND AND OBJECTIVE: Tricine, as a flavonoid compound in many food sources, has anti-inflammatory and anti-proliferative effects in some cancer cell lines. Considering the importance of using natural anti-cancer drugs in therapy-resistant cancers such as prostate cancer, the aim of this study is to investigate the effect of tricine on cell growth and proliferation and induction of apoptosis in PC3 human prostate cancer cell line.
METHODS: In this experimental study, the PC3 cell line was prepared and cultured from the Pasteur Institute of Iran. Extraction and purification of tricine were done by column chromatography and recrystallization of Allium atroviolaceum extract. The apoptotic effect of tricine at concentrations of 60, 80, 100, 120 and 140 μM was evaluated by MTT method. The apoptotic effect was evaluated in the cell group treated with IC50 concentration of tricine and untreated cells (control group) using Annexin-V kit and flow cytometry.
FINDINGS: The viability of cells at different tricine concentrations were 85.66±1.52, 76±3.60, 66.33±4.16, 44±3.60, and 36.66±3.21, respectively (p<0.01). The IC50 concentration of tricine was 117.5±4.4 μM for PC3 cell line. The apoptosis rate in PC3 cells after 48 hours of treatment with IC50 concentration of tricine was 24.3±0.58%, which was not significant in comparison with control cells (23.3±0.58%).
CONCLUSION: The results of this study showed that tricine resulted in cell death in the PC3 cell line, but the cell death mechanism was not apoptotic
The Relationship between Gestational Diabetes and Polycystic Ovary Syndrome
BACKGROUND AND OBJECTIVE: Women with polycystic ovary syndrome (PCOS) are at risk of insulin resistance and pregnancy complications. The aim of this study is to determine the relationship between gestational diabetes and polycystic ovary syndrome.
METHODS: This cross-sectional study was performed on 126 pregnant women with PCOS and infertility history who became pregnant after stimulation of ovulation and referred to Imam Khomeini Hospital in Ahvaz. These patients underwent screening with 75 g OGTT (Oral Glucose Tolerance Test) during the first trimester and during 24-28 weeks of gestation. They were divided into two groups of PCOS with gestational diabetes and without gestational diabetes. Variables such as age, gestational diabetes, parity and BMI were measured and the relationship between BMI and gestational diabetes was examined.
FINDINGS: The mean age of patients in the two PCOS groups with and without gestational diabetes was 26.09±2.03 and 27.07±3.03 years, respectively. Of the 126 pregnant women with PCOS, 30 (23.8%) patients were diagnosed with gestational diabetes in the first trimester of pregnancy and 11 (8.7%) patients during 24-28 weeks of gestation. Overall, 41(32.5%) patients had gestational diabetes and 85 (67.5%) patients did not have gestational diabetes. There was no significant difference in the mean BMI between the two PCOS groups with and without gestational diabetes.
CONCLUSION: The results of the study showed that more than one third of women with PCOS experience gestational diabetes during pregnancy
Investigating the Expression of EGFR And FGFR4 Genes in Patients with Lung Cancer
BACKGROUND AND OBJECTIVE: Lung cancer is a disorder that is caused by genetic and epigenetic changes and activates oncogenes and inactivates tumor suppressor genes. The aim of this study is to quantitative evaluation of EGFR and FGFR4 genes expression level in blood samples of lung cancer in compare with normal people to investigate the role of these two genes as biomarkers during lung cancer diagnosis and screening.
METHODS: This case-control study was performed on 50 blood samples of lung cancer patients compared with 50 normal controls.. Total RNA from Blood samples were extracted and cDNA is synthesized. The specific primers for detection of markers are designed and expression level of BRIP1, PALB2 in presence of gene GAPDH by using Real Time PCR method was quantitatively studied.
FINDINGS: Significant increase was observed in the expression of target biomarkers in cancer patients compared to control population. Results showed quantitative increase of FGFR4 and EGFR genes with 4.46 and 3.03 fold respectively for lung cancer in compare with normal samples (p=0.003). Also, there was a significant relationship between grade of the disease and biomarkers expression level, so that with increasing the stage and degree of severity of cancer, the expression of biomarkers increased (p=0.003).
CONSLUSION: Based on this study results we could predict the expression level of (EGFR, FGFR4) gens in suffered patients quantitatively which could use as biomarker indicator during screening of lung cancer samples
The Effectiveness of Acceptance and Commitment Group Therapy on Psychological Capital in Cardiovascular Patients in Babol
BACKGROUND AND OBJECTIVE: Psychological capital is one of the positive psychological indices that is defined by the characteristics of optimism, perseverance, positive self-efficacy, and tolerance. This study was conducted to evaluate the effectiveness of acceptance and commitment group therapy on psychological capital in cardiovascular patients in Babol.
METHODS: This cross-sectional study was performed among 24 cardiovascular patients who referred to governmental and non-governmental medical centers of Babol in two groups of 12 based on pretest-posttest design. The therapy based on acceptance and commitment was performed for the experimental group in 8 sessions. Participants were re-evaluated one month after the sessions, and psychological capital was evaluated according to self-efficacy, hope, tolerance and optimism subscales.
FINDINGS: The results of the study showed that there was a significant difference between the subscales of the psychological capital in the experimental and control groups in tolerance (3.53±0.48 vs. 2.88±0.67) (p6.03) and optimism (40.6±0.56 vs. 2.94±0.66) (p>26.459).
CONCLUSION: The present study showed that acceptance and commitment therapy could be considered as an effective intervention in increasing the psychological capital of cardiovascular patients
