Biosynthesis of silver nanoparticles from Arum dioscoridis plant leaf aqueous extract: anticancer and antimicrobial properties
This study was carried out to synthesize silver nanoparticles (AgNPs) from Arum dioscoridis (AD) leaf extract and to investigate the cytotoxic and antipathogenic effects of them. The plant material had a reducing and stabilizing effect on the synthesized nanomaterial. During the plant-mediated synthesis of nanomaterials, no substances that would cause environmental pollution were used. For the structural characterization of AD-AgNPs, Ultraviolet-visible (UV-vis) Spectroscopy, Field Emission Scanning Electron Microscopy (FE-SEM), Electron Dispersive X-ray (EDX) Spectroscopy, Fourier Transform Infrared (FT-IR) Spectroscopy, Transmission Electron Microscopy (TEM), X-ray Diffractometer (XRD), Atomic Force Microscopy (AFM) and Zetasizer analyses were performed. The produced AgNPs showed maximum surface plasmon resonance at 431.67 nm and had mostly spherical morphology. The zeta potential value of the nanomaterial was -9.76 mV and the average powder crystal size was 31.48 nm. The minimum inhibitory concentration (MIC) values (mg/L) of AD-AgNPs on Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, and Candida albicans were 0.25, 2.00, 0.125, 4.00, and 1.00, respectively. After 24 and 48 hours of application by MTT [3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazolium bromid] assay, the half-maximal inhibitory concentrations (IC50: μg/mL) of AD-AgNPs on human colon adenocarcinoma cell (CACO-2), human breast cancer cell (MCF-7), glioblastoma multiforme cell (T98-G), and healthy human umbilical vein endothelial cell (HUVEC) lines were determined as 2.977, 2.801, 5.694, 4.392; 2.115, 2.300, 2.612, 4.091, respectively.Dicle University Scientific Research Projects Coordinatorshi
Effect of cardio-gastric interaction on atrial fibrillation in GERD patients
Objective: Atrial fibrillation (AF) and gastroesophageal reflux disease (GERD) are very common in daily clinical practice. Post-prandial
AF episodes have been reported in GERD patients. Although it was reported in previous studies that it was caused by sympathovagal
imbalance, there are no studies on cardiac conduction system involvement. In this study, we aimed to evaluate whether the risk of
developing AF increases in untreated GERD patients with non-invasive electrophysiological tests.
Methods: The research was prospectively performed. Endoscopy was performed on the individuals due to reflux complaints. ECG was
recorded at 25mm/s and 10 mm/mV amplitude, and 24-hour Holter ECG (three-channel; V1, V2, and V5) was performed. ECG
parameters were measured and Holter ECG results were analyzed.
Results: A total of 120 individuals, 60 patients and 60 controls, were included. No significant statistically differences existed between
groups for hypertension, diabetes, smoking, or dyslipidemia (p>0.05). In terms of heart rate, Pmax, Pmin, QTd, and QTcd, there were
no significant differences across the two groups (p>0.05). P-wave dispersion (Pd) was substantially higher in the study group
(p=0.014). Comparing the heart rate variabilities of 24-hour Holter ECG recordings across the groups, the standard deviation of R-R
intervals (SDNN) was substantially higher in the study group (p<0.001). Low Frequency (LF) and LF/HF were significantly higher in
the control group (p<0.001 and p=0.003, respectively). AF was detected in nine individuals on Holter ECG.
Conclusion: Pd duration and risk of developing AF were higher in GERD patients
Synthesis of Drug Loaded Magnetic Nanocomposite and Investigation of Cytotoxic Properties
This aims of study to develop drug delivery systems providing an alternative solution to cancer chemotherapy. Firstly, carboxymethyl dextran which has magnetic properties (MD), was synthesized as stated in the literature.1-3 Its structure was elucidated with various analytical devices. Then, loading studies were carried out on the nanocomposite with topotecan (TP) (in the range of 0.2-40 µM in phosphate buffer at pH 5) as an anticancer drug. The cytotoxic properties of MD, TP, drug loaded MD (MD-TP), and free TP were studied on human prostate cancer cell (Du145) and human healthy prostate cell (PNT1A) line. The synthesized MD and MD-TP nanocomposites were characterized by different analytical devices such as FTIR, XPS, UV-VIS, VSM, TEM, SEM and DLS. According to the FTIR results, the presence of peaks belonging to functional groups in the expected places indicates that MD was successfully synthesized and TP was successfully loaded on nanocomposite. According to TEM image, the MD particles showed a spherical structure, and also, the particle size was measured as 9 ± 1.8 nm. The SEM image of MD indicated that the nanoparticles were aggregated. Furthermore, after the loading TP, the brightly colored particles of TP were observed on some parts of the surface of MD. Moreover, DLS measurements of the nanoparticles were taken before and after loading the drug. Due to high agglomeration of the magnetic nanoparticle, it showed polydispersity. In addition, cytotoxicity effects of drug loaded nanocomposites on Du145 and PNT1A cell lines were investigated and the results were found to be statistically significant. The topotecan loaded MD-TP nanocomposite have shown cell viability to be reduced by up to 20 %. It has been shown that synthesized polymer coated superparamagnetic nanocomposites with these properties can be used in drug targeting studies
Evaluation of Right Ventricular Global Longitudinal Strain in COVID-19 Patients After Intensive Care Unit Discharge
Background and Aim: Using two-dimensional speckle tracking echocardiography (2D-STE), the ventricular functions of hospitalized coronavirus
disease-2019 (COVID-19) patients were assessed. However, there is limited information about cardiac functions in the first year after recovery
from the intensive care unit (ICU). This research aims to assess the right ventricular functions of COVID-19 patients and their changes within the
first year after ICU discharge using 2D-STE.
Materials and Methods: The study was conducted prospectively. The study included 68 consecutive patients and 70 control patients.
Echocardiography was performed in the ICU and the first year after discharge from the hospital. Right ventricular global longitudinal strain
(RVGLS) was measured using the 2D-STE method.
Results: The mean age of the study group was 48.67±8.10 and 37 (54.4%) patients were males. There were no substantial differences across the
groups, including age, gender, body mass index, heart rate, diabetes, dyslipidemia, and smoking (P > 0.05). A substantially significant positive
correlation was detected between right ventricular dimension (RAD) (r = 0.644, P < 0.001), right ventricular diastolic dimension (RVDD) (r =
0.573, P < 0.001), ferritin (r = 0.454, P < 0.001), D-dimer (r = 0.305, P = 0.011) values and RVGLS in the in-hospital and after-discharge first-year
groups. The RVGLS values of the control, in-hospital, and after-discharge first-year groups were -20.36±3.06, -16.98±3.78, and -17.58±6.45,
indicating a statistically significant difference across the groups (P < 0.001). Tricuspid annular plane systolic excursion was higher in the control
group (P < 0.05).
Conclusion: RVGLS was found to be depressed during the in-hospital period and showed no improvement in the 1 year post discharge
Is there an intergenerational difference in the perception of tourist guide service quality?
The perceived service quality offered by tourist guides is regarded as a significant determinant in travelers' overall satisfaction with their tour experience. The objective of this study is to ascertain thevariations in the opinion of service quality among tourists from different generations with regards to tourist guides. The qualitative research method of interview technique was employed within the parameters of this study. The data for this study were obtained from a sample of 30 participants who engaged in guided tours. These individuals belonged to the X, Y, and Z generations, and their responses were gathered through semi-structured interviews. The study's demographic comprises travelers belonging to Generations X, Y, and Z who have actively engaged in guided tour experiences. The study employed stratified purposive sampling as its sampling technique. The data were subjected to analysis using the MAXQDA 2022 software. Based on the findings of the investigation, it was determined that there exist variations in the perception of tourist guide services across different generations
Investigation of Yield and Quality Performance of Durum Wheat Genotypes Grown in Arid Conditions
İnsan beslenmesinde önemli bir yere sahip olan makarnalık buğday dünya çapında gıda
güvenliği bakımından stratejik bir bitki konumundadır. Makarnalık buğdayda kurak koşullarda
yüksek verim ve kalite potansiyeline sahip genotiplerin belirlenmesi amacıyla CIMMYT
tarafından temin edilen 110 genotip ile kurulan denemede tane verimi, protein içeriği, nişasta
içeriği, gluten içeriği, yağ içeriği, parlaklık L ve renk b* parametreleri incelenmiştir. Denemede
elde edilen sonuçlara göre; tane verim değeri 49,6-246,4 kg/da, protein içeriği %11,8-15,4,
nişasta içeriği %82,3-86, yağ içeriği 2,0-2,7, gluten içeriği %24-32,5, L değeri 54,75-61,22, b*
değeri 24,37-27,87 aralığında değişim göstermiştir. Frekans dağılım grafiğine göre tane
veriminde 51, protein içeriğinde 47, nişasta içeriğinde 59, gluten içeriğinde 50, L değerinde 59
ve b* değerinde 55 genotip ortalama değerin üzerinde yer almıştır. Seleksiyon başarısını
artırmak için yapılan Cluster analizine göre genotipler 12 grup altında ve özellikler ise 2 büyük
grup altında toplanmıştır. Bu gruplamada 19 ile 104 nolu genotipler birbirine en uzak, 13 ile 43
no’lu genotipler en yakın benzerliğe sahip olmuştur. İncelenen özellikler arasındaki korelasyon
analizi sonuçlarına göre tane verimi ve kalite değerleri arasında ilişki bulunmazken, protein ve
gluten içeriği arasında önemli doğrusal ilişki bulunmuştur
The Effect of One-Lung Ventilation on Cognitive Functions: Cross-Sectional Research
Amaç: Postoperatif kognitif disfonksiyon (POKD), majör cerrahi sonrası özellikle ileri yaş olmak üzere tüm yaş grubu hastalarda sıkça görülür ve aylarca sebat edebilir. Etiyolojisi multifaktöriyel olmakla birlikte, serebral oksijen sunum ve tüketim dengesini etkileyen faktörler suçlanmaktadır. Bu nedenle hipoksi gibi ciddi komplikasyonlarla seyredebilen tek akciğer ventilasyonu (TAV) uygulanan torasik cerrahi hastalarında POKD'yi mini-mental durum testi ile değerlendirerek hastalarda POKD'nin değerlendirilmesi amaçlandı. Gereç ve Yöntemler: Bu çalışmaya 20 yaş ve 72 yaş arası, torasik cerrahide TAV adayı hastalar dâhil edildi. Anestezi protokolü tüm hastalarda aynı şekilde uygulandı. Kognitif değerlendirme operasyondan 1 gün önce, operasyondan 1 ve 3 gün sonra yapıldı. Bu amaçla MiniMental Durum Değerlendirme Testi kullanıldı. Bulgular: Çalışmaya 30 hasta dâhil edildi. TAV uygulanan hastaların %26,6'sında POKD tespit edildi. Hastaların yapılan mini-mental durum değerlendirme testi, ameliyat öncesi ile ameliyat sonrası 1. gün ile 3. gün arasında kognitif disfonksiyon açısından fark tespit edildi. Ameliyat sonrası 1. gün ile ameliyat sonrası 3. gün arasında fark tespit edilmemiştir. Sonuç: Toraks cerrahisinde TAV uygulanan hastalarda erken dönem kognitif disfonksiyon görülmektedir. Bunda cerrahi süre, anestetik ilaçlar, hastayla ilgili parametreler etkili olmakta olup, hastalar intaroperatif yakın takip edilmeli ve ameliyat sonrasında da hastalar kognitif fonksiyonları açısından yakından izlenmelidir
Clinical and Genetic Spectrum of Myotonia Congenita in Turkish Children
Background: Myotonia congenita is the most common form of nondystrophic myotonia and is caused by Mendelian inherited mutations in the CLCN1 gene encoding the voltage-gated chloride channel of skeletal muscle.
Objective: The study aimed to describe the clinical and genetic spectrum of Myotonia congenita in a large pediatric cohort.
Methods: Demographic, genetic, and clinical data of the patients aged under 18 years at time of first clinical attendance from 11 centers in different geographical regions of Türkiye were retrospectively investigated.
Results: Fifty-four patients (mean age:15.2 years (±5.5), 76% males, with 85% Becker, 15% Thomsen form) from 40 families were included. Consanguineous marriage rate was 67%. 70.5% of patients had a family member with Myotonia congenita. The mean age of disease onset was 5.7 (±4.9) years. Overall 23 different mutations (2/23 were novel) were detected in 52 patients, and large exon deletions were identified in two siblings. Thomsen and Becker forms were observed concomitantly in one family. Carbamazepine (46.3%), mexiletine (27.8%), phenytoin (9.3%) were preferred for treatment.
Conclusions: The clinical and genetic heterogeneity, as well as the limited response to current treatment options, constitutes an ongoing challenge. In our cohort, recessive Myotonia congenita was more frequent and novel mutations will contribute to the literature
The future of tourism and hospitality labour: challenges, requirements, trends, skills and the impact of technology
This study was designed to fill two main research gaps: the need for a multidimensional perspective on the future of tourism and hospitality (T&H) labour and the lack of studies from the perspectives of key stakeholders who are representatives of tourism organisations and involved in policymaking. Therefore, it unveils the perceptions of policymakers responsible for tourism organisations in Portugal towards the future of T&H labour. Following an exploratory qualitative case study approach based on semi-structured interviews with eleven policymakers, the results illustrate that the most prominent challenges that the COVID-19 pandemic created are the damage to practical ability, finding a skilled and experienced workforce, and attracting tourism labour back to the sector. However, there are some requirements to overcome such challenges in the future. Moreover, future trends and skills for T&H employment were revealed as new working models, digitisation and robotisation, and the expected skills such as management, analytical, digital marketing, and customer behaviour analysis. Finally, technology was found to have positive and negative impacts on T&H employment. The results and implications will benefit Portugal and different destinations in understanding the dimensions of T&H labour for the future and developing future actions, strategies and policies
Evaluation of Pediatric Patients Receiving Home Health Services
Background: Home health service is the provision of health and care and follow-up services to patients in line with the recommendations of physicians, in the environment they live with their families, by the healthcare team in a way that meets their medical needs, including rehabilitation, physiotherapy and psychological treatment. This study was conducted to evaluate the medical conditions of pediatric patients receiving home health services. Methods: This descriptive study includes pediatric patients under the age of 18 who received home health services from Diyarbakır Pediatric Hospital in Turkey. The files of patients who applied to the home health unit between 2017 and 2020 and were actively followed up were reviewed retrospectively. The diagnoses, reasons for demand and medical needs of the patients were examined. Results: The mean age of 372 patients who received home health services was 10.9±4.3 years. 46.5% of the patients were diagnosed with epilepsy, 46.2% with mental retardation, 22.3% with cerebral palsy. Home health services are mostly requested for examination (49.2%), diaper (37.4%), medicine (14.8%) and formula (14.8%) reports. While 74.8% of girls are fully bedridden, 62.7% of boys are fully dependent (p<0.05). Most of the patients who are bedridden are provided with personal care by their families. 35.6% of bedridden patients and 29.6% of semi-dependent patients need medical care (p<0.05). While 63-67% of bedridden patients need rehabilitation, 38% of not bedridden patients need rehabilitation (p<0.05). Conclusions: Most of the patients receiving home health services are bedridden and have neurological diseases. As bed dependency increases, the need for medical care increases
