The capacity to investigate the genetic basis of phenotypes in non-model species such as Atlantic salmon (Salmo salar) has improved thanks to recent advancements in genomic technologies, especially CRISPR-based genome editing. Large-scale CRISPR-based screening is proven to be a powerful tool to investigate genome-phenotype associations and could prove invaluable in studying the Atlantic salmon gene-biology association. However, genomic approaches are complicated by the complex nature of the Atlantic salmon genome, in addition to the potential genomic incompatibilities of applying mammalian-based transduction systems to the cold-adapted species. Given these challenges, optimising strategies for viral transduction of Atlantic salmon cells requires careful consideration of different effects. One such effect is temperature, which plays a crucial role in various biological processes, including viral transduction efficiency. Therefore, this study investigates how temperature affects Atlantic salmon's lentiviral transduction efficiency. By examining temperature variations, we aim to determine their impact on viral uptake, genome integration, and overall effectiveness on transduction efficiency. To document the effect of elevated temperatures on the fitness of Atlantic salmon cells, the thermal tolerance of Atlantic salmon fibroblast cell line is systematically characterised by monitoring growth rates across a gradient of temperatures. Thus, potential temperature candidates are established by determining the optimal range for transduction that still supports cellular viability without inducing detrimental stress responses. Following, the selected temperatures are used to assess the effect of temperature on lentiviral efficiency in Atlantic salmon cells. Building on these findings, the result reveals a distinct temperature-dependent effect on the early stages of lentiviral infection in Atlantic salmon cells, particularly affecting viral entry and reverse transcription. However, while initial infection appears successful, long-term analysis suggests deficient incorporation of lentiviral payloads into the salmon genome. These findings highlight potential incompatibilities between the mammalian-based lentiviral system and the Atlantic salmon genome, which may stem from nuclear transport or genome integration incompatibilities. To overcome these challenges, further research is needed to clarify the molecular mechanisms that govern lentiviral genome incorporation in Atlantic salmon cells

Winter maintenance in urban areas : consequences for the environment and spring cleaning

This thesis explores how winter road maintenance affects the environment in Norwegian urban areas. Every winter, large amounts of salt and sand are spread across the roads, and snowplows are out nearly around the clock. This is necessary to keep roads safe and accessible — but it also leaves a mark. Salt ends up in soil, ditches, and water bodies, and over time it can affect plants, soil life, and water quality. When the snow melts in spring, large amounts of leftover grit remain along the roads. Traffic crushes this material into fine dust, which gets stirred up on dry days and settles as a visible layer on city surfaces and building facades. Throughout this project, I’ve looked at how winter maintenance impacts air, water, and the urban environment. I’ve used reports and professional literature from sources such as the Norwegian Public Roads Administration, NILU, and the Environment Agency to build a realistic understanding. The goal has been to identify the real challenges — and what steps can actually make a difference. Several solutions already exist. Better planning, smarter salt use, quicker spring cleaning, and promoting studless tires are proven measures. In addition, we need to rethink how we design and maintain urban spaces: use materials and layouts that make it easier to manage snow, prevent flooding, and ensure that cities function well even on the coldest days. In summary, this thesis shows that good winter maintenance is about more than just clearing snow. It’s also about the environment, the economy, and how we want our cities to work — all year round.B-L

Co-Creating Inclusive Public Spaces for Children and Youth

This master’s thesis explores universal design and inclusive public space, with a particular focus on children and youth. The project centers on the co-creation of a temporary playground at Økern Torgvei 2 in Oslo, currently an unused gravel site adjacent to a temporary swimming facility. Økern is part of Hovinbyen, one of Oslo’s largest transformation areas, where major development projects often overshadow the needs of the present-day community. In contrast, this thesis proposes a small-scale, temporary intervention aimed at improving local quality of life while larger visions remain unrealized. Through co-creation - a participatory design method emphasizing collaboration and shared authorship - I engaged local users in the design process. Existing participation material from nearby projects formed the foundation of the initial concept. These were followed by three workshops during the detailed design phase: two with children from nearby primary schools and one with the parent of a child with disabilities, to address needs related to universal design. A physical 3D model of the site was used as a tool to facilitate discussion and support concrete, grounded input. The thesis demonstrates that co-creation can offer real influence when implemented at the right moment in the process - when there is still flexibility for change, but also a tangible framework to respond to. I also found that reusing existing participation material is a valuable, underused resource that not only improves design quality but also helps combat participation fatigue - particularly relevant in transformation areas where repeated consultation can lead to disengagement. The playground design emphasizes both inclusive and active play, accommodating a wide range of user needs and preferences. While not every element is universally accessible, the approach reflects a commitment to social sustainability by offering something for everyone. The project ultimately argues for a more strategic, efficient, and meaningful use of co-creation in the planning and design of public spaces

This master’s thesis explores how small and medium-sized real estate companies in the Oslo region adapt their business models to the green transition and the requirements of the EU Taxonomy. The backdrop for this study is the real estate sector’s significant contribution to greenhouse gas emissions and resource consumption, and the increasing need to balance economic profitability with social responsibility. The EU Taxonomy and stricter demands for sustainable development have introduced new rules of the game—posing challenges, but also creating new opportunities for innovation and green competitiveness. The aim of this thesis is to investigate how innovation and creativity can help address the trade-offs between financial returns and sustainability, and how these dynamics are reflected in the business models of small and medium-sized actors in commercial real estate. The central research question is: How and to what extent do business models address the green transition and the EU Taxonomy? How are these frameworks implemented in practice, and what challenges and opportunities arise as companies seek to balance profitability and responsibility through sustainable solutions? The analysis is structured around four research questions: * How are business models adapted to the requirements of the EU Taxonomy and the green transition? * What exogenous factors influence the implementation of green solutions, and what barriers arise? * How do companies balance economic profitability and social responsibility? * How does the EU Taxonomy influence decision-making processes and strategic choices? The study is based on qualitative case interviews and document analysis from five companies, and the findings are examined through three theoretical frameworks: Ostrom’s Institutional Analysis and Development (IAD) framework, the property theory’s sequence model, and the Business Model Canvas. The results show that all companies are affected by the EU Taxonomy, but in different ways. Some have integrated sustainability as a strategic core, while others are adapting more reactively. The taxonomy is seen as a driver of change, yet it is also perceived as complex and resource- intensive. Key barriers include uncertain profitability, difficulty understanding regulations, and lack of expertise. Meanwhile, opportunities lie in green positioning, access to capital, and market adaptation. A central finding is that sustainability cannot only be reduced to technical measures like energy efficiency or environmental certification. It also involves culture, leadership, and the capacity for innovation. Despite limited resources, small and medium-sized actors hold a unique strategic position. Their proximity to projects and decision-makers enables them to quickly test and implement new solutions. The conclusion is that small and medium-sized real estate companies have real potential to act as drivers in the green transition. However, to succeed, they require better tools, enhanced competence, and supportive frameworks that make it easier to make sustainable choices. Sustainability must be understood not merely as environmental responsibility, but as a strategic and value-driven practice—one where innovation and social responsibility go hand in hand with long- term economic development. Within this context, the EU Taxonomy serves both as a regulatory framework and as a strategic tool for navigating the green transition

More children and adolescents are failing to meet the recommended daily activity levels set by the Norwegian Directorate of Health. Schools are identified as a favorable arena to counteract this trend, and Physically Active Learning (PAL) is one approach to increasing physical activity during the school day. In this master’s thesis, a teaching unit for Chemistry 1 has been designed with a focus on PAL, incorporating outdoor spaces as a learning environment in parts of the program. The purpose of the study is to investigate how the teaching unit “Chemistry’s Footprint”, a unit that includes PAL, is experienced by students and the teacher. This purpose is specified through the research question: How do frameworks and scaffolding contribute to students’ experiences in the teaching unit “Chemistry’s Footprint”? The study is a single-case study in which “Chemistry’s Footprint” was designed and implemented in one Chemistry 1 class. The unit combined review tasks and physical activity using the mobile application Fotspor (“Footprint”). Data was collected through a survey completed by all students in the class, two focus group interviews with selected students, and an in-depts interview with the teacher. The data material was analyzed using conventional content analysis. The results show that both students and the teacher highlight several strengths and weaknesses of the teaching unit. Students point to variation from traditional chemistry instruction, well-designed discussion tasks, and increased focus due to physical activity as strengths. The teacher also noted positive discussions and increased student engagement as benefits. As for weaknesses, students reported challenges related to academic scaffolding during the activities, weather conditions, and the overall efficiency of the program, while the teacher experienced having less oversight of the class’s progress. Based on the results, the study discusses the frameworks and scaffolding provided in “Chemistry’s Footprint”, and how they contributed to the students’ experiences with the unit. The study concludes that appropriate frameworks and sufficient support structures are necessary for the successful inclusion of PAL in chemistry education. The connection between the learning environment and the learning outcomes must be clarified for the students, and support structures that guide students’ academic discussions must be designed and further developed

The theme of this study revolves around the Engineer’s Guidance in Legal Clarification and Boundry Determination Cases in the Land Consolidation Court. In order to answer the research question, the study was conducted by using qualitative methods. The study is confined to addressing legal clarification and boundry determination, which constitutes chapter 4 of The Land Consolidation Act. The purpose of the guidance is to assist the parties in being able to safeguard their interests, and to contribute to a correct ruling in the case based on the facts and the applicable rules, cf. The Land Consolidation Act § 6-16. The study’s research question is: How do engineers provide guidance in legal clarification and boundry determination cases? The main research question is specified by three sub-questions: * What qualifications does the engineer possess to provide guidance? * What experiences does the engineer have in providing guidance pursuant to § 6-16 in the Land Consolidation Act and § 11-5 in the Dispute Act? * What are the differences between the various court districts regarding the engineer providing guidance, and what factors may influence these differences? The engineers demonstrate a good general understanding and knowledge of guidance, but primarily on a general level. Based on the engineers’ expertise and experience from the Land Consolidation Court, they appear to be qualified to provide guidance in legal clarification and boundry determination cases. The engineers seem to have experience providing guidance under § 6-16 of the Land Consolidation Act and § 11-5 of the Dispute Act, though this guidance does not appear to be directly based on the formulation of the law and could benefit from a more practical approach. However, it is difficult to determine precisely which circumstances influence the engineer provides guidance. It appears to a large extent that this is dependent on the individual engineer and the routines and organization of each respective Land Consolidation Court. We conclude that the guidance varies between the different Land Consolidation Courts, and that the engineer provides more guidance than they themselves may be aware of. This guidance is considered informal. Guidance is provided by asking the parties questions about the evidence and its significance. It appears that the engineers provide guidance with caution

What Triggers a Special Agricultural Safeguard (SSG)?: Analyzing the Use of SSGs by the EU and the US

    Get PDF
The special safeguard (SSG) is an exception to the market access (MA) commitments under the World Trade Organization (WTO) Agreement on Agriculture (AoA) that has caused disagreement in WTO trade negotiations. It allows the member countries that have reserved the right to their use the imposition of heightened tariff rates on selected agricultural commodities in specified cases of a surge in import volume or a fall in import price. This thesis quantitatively analyzes the invocation of price-based SSGs by the European Union (EU) and the United States (US) during 1995-2023, and whether developing countries’ exports have been adversely affected by the price-based SSGs invoked by the EU and the US. A panel data framework is applied to a probit model for binary response to estimate the effects on the response probability, that is the probability of a SSG action being taken, from selected economic factors. The regression results suggest that real GDP, the relative import price, import growth and the COVID-19 pandemic have affected the EU’s decision to invoke a price-based SSG. The relative import price, import growth and domestic production volume are estimated to have affected the decision of the US to invoke a price-based SSG. Developing countries’ exports are implied to have been adversely affected by the price-based SSGs invoked by the EU. No such effect is found from the price-based SSGs invoked by the US

Molekylær foredling for sykdomsresistens hos jordbær

The garden strawberry, Fragaria × ananassa, is a highly popular and economically important berry crop cultivated in various climates worldwide. It first occurred during the 1700s in western Europe as the result of accidental hybridization between the South American F. chiloensis and the North American F. virginiana. These two species are adapted to vastly different growing conditions and made F. × ananassa a genetically diverse crop. Desirable complementary traits derived from the parent species have enabled the selection and domestication of F. × ananassa over 300 years breeding history. Improved strawberry cultivars have historically been obtained through traditional crossbreeding, where the most desirable offspring from crosses of superior individuals are selected based on phenotypic evaluations. This process has been relatively slow, but intensive selection significantly increased fruit size and yield. Recently established F. × ananassa genomes and genotyping arrays have enabled molecular breeding tools that can make strawberry breeding more efficient. Maintaining genetic diversity is essential for ensuring successful breeding and long-term adaptability to emerging challenges. However, neglect of disease resistance breeding, due to reliance on chemical treatments, has led to reduced resistance among current strawberry cultivars. Given the decrease in genetic diversity within breeding populations, there is an urgent need to find new sources of resistance and other important traits. To address this issue, a population genetics analysis of domesticated and wild Fragaria species was performed. There was a distinct genetic difference between F. × ananassa and its wild relatives. Cultivars clustered into five groups that reflected breeding history, with old European cultivars and new Californian cultivars being the most genetically separated. Despite this, a noticeable preference for Californian cultivars in recent European breeding was observed. Genetic pedigree analyses revealed that many cultivars were true-to-type and have been preserved for decades. Several other cultivars were identified as not true-to-type, including some cultivars that may be lost forever. Taxonomic classifications at the species and subspecies level were confirmed, but discrepancies between the assigned taxa and the genetic patterns were identified for some accessions. The population genetic analysis revealed several distinct groups that may harbor valuable genetic variation. Powdery mildew, caused by the fungus Podosphaera aphanis, is a common disease that many strawberry cultivars are highly susceptible to. Genetic analysis of powdery mildew resistance in a Nordic strawberry breeding population revealed substantial variation between accessions. Genomic selection was evaluated for its potential to improve selection processes. A prediction accuracy of 0.40 was achieved when genomic selection was used to predict the powdery mildew resistance of a testing population that differed from the training population. The ability of strawberry cultivars to resist powdery mildew infections can be influenced by genetic variation within P. aphanis populations. Powdery mildew samples from various regions of Norway were thus analyzed to identify potential genetic variation. The genetic analysis showed that there are at least two genetically distinct P. aphanis groups in Norway. Genetic differences between the groups could not be explained by geographic separation, although samples from one group originated from a specific location. This research project shows that there are several genetically unique strawberry groups with potential beneficial traits, that powdery mildew resistance can be improved through genomic selection, and that at least two genetically distinct strawberry powdery mildew groups exist.

DBRow: A Density-Based algorithm for autonomous navigation within crop rows.

    Get PDF
This thesis presents DBRow, a density-based algorithm designed to enhance autonomous navigation within crop rows. As agricultural robotics increasingly plays a crucial role in boosting productivity and addressing labour shortages, efficient navigation systems are essential for optimising operations such as planting, harvesting, and maintenance. DBRow combines Simultaneous Localisation and Mapping (SLAM) with Density-Based Spatial Clustering of Applications with Noise (DBSCAN) to overcome the limitations of previous navigation approaches that relied solely on Light Detection and Ranging (LIDAR) data for NMBU's FRE participation. Through a series of experiments conducted in simulated environments and controlled indoor settings, the performance of DBRow was evaluated using Dijkstra, Theta*, and A* path planning algorithms across various terrains. The results demonstrate that A* generally provides faster navigation and better safety outcomes, while Theta* offers shorter paths at the expense of plant safety. Dijkstra, although slower, shows robustness in rough terrains. These findings highlight the importance of terrain characteristics in influencing algorithm performance. The thesis underscores the need for further testing to achieve statistically significant results and suggests future work to implement DBRow on PEIK for the upcoming competition. Enhancements in LIDAR preprocessing and the integration of machine learning models for stem detection are recommended to improve navigation accuracy. Additionally, adapting the system to automate manual tasks, such as pesticide application, could significantly enhance its utility in agricultural operations

Diptera larvae and Nematoda were extracted from soil samples collected in the foreland of the receding Hardangerjøkulen glacier near Finse, central south Norway. Samples were standardized by being taken in snowbed habitats with Salix herbacea L. vegetation. Diptera larvae were sampled in twenty plots from 32 to 227 years age, complemented by five plots with about 10,000-year-old soil. Nematodes were studied in soils of 4, 37, 39, 62, 78, 119, and 204 years age. There was a rapid colonization in young soils of both Diptera larvae and nematodes. Brachycera larvae were sparsely represented, and Sciaridae and Chironomidae larvae were most numerous in soils younger than 50 years. Genera of Chironomidae larvae were Bryophaenocladius, Pseudosmittia, Parasmittia, and Smittia. The number of nematode taxa increased from six in the youngest soil to fourteen in the oldest. Bacterial feeders were dominated by the genus Rhabditis sensu lato and fungal feeders by Tylenchus sensu lato. The plant-parasitic Paratylenchus sp. was present at 4 years, with highest abundance at 39 years. The abundance of omnivores (subfamily Dorylaiminae) did not vary between soil ages, but predators (fam. Mononchidae and genus Tripyla) were more abundant at 78 and 119 years.Succession of soil-living Nematoda and Diptera larvae (Chironomidae, Sciaridae, and Brachycera) near a melting glacier in Southern NorwaypublishedVersio
