    Sustainable mass handling in small construction companies

    Denne oppgaven tar for seg hvordan gjenbruk av stedegne løsmasser kan gi miljøgevinst og kostnadseffektivisering. Dette er gjort ved å se på to casestudier fra praksis på Kråkerøy i Fredrikstad. På disse casene er det brukt data hentet fra eget firma, hvor oppdragene ble gjennomført på vegne av private kunder. Oppgaven tar også for seg hvor mye en entreprenør kan spare i CO2 utslipp i året. Bygg- og anleggsbransjen står for utslipp av 854.000 tonn CO2eq som har gitt et stort fokus på gjenbruk og utslippsfrie byggeplasser (Direktoratet for forvaltning og økonomistyring, 2022). En del av overskuddsmassene som blir tatt ut fra ulike prosjekter blir bare dumpet til ingen nytte, på tross av at det noen steder er underskudd av løsmasser (Akershus fylkeskommune, 2021). Løsmassene vi har her i Norge er ikke-fornybare eller tar lang tid å produsere, som for eksempel jord (Landbruksdirektoratet, 2025). Det er vanskelig å måle verdien i slike masser, men oppgaven skal gi et innblikk i hvilken nytte slike løsmasser kan ha. Temaet er veldig komplekst, som er omfattet av mange lovverk (Akershus fylkeskommune, 2021). Oppgaven tar også for seg gjeldende regelverk, og nye lover som trer i kraft om kort tid.This assignment examines how the reuse of local loose materials can provide environmental benefits and cost efficiency. This has been done by looking at two case studies from practice in Kråkerøy, Fredrikstad. These cases use data obtained from a company that has carried out the projects on behalf of private clients. The assignment also assesses how much a contractor can save in CO2 emissions per year. The construction industry is responsible for emissions of 854,000 tons of CO2 equivalent, which has led to a strong focus on reuse and emission-free construction sites (Direktoratet for forvaltning og økonomistyring, 2022). Some of the surplus materials extracted from various projects are simply discarded without being put to use, even though there is a shortage of loose materials in some areas (Akershus fylkeskommune, 2021). The loose materials found in Norway are non-renewable or take a long time to produce, such as soil (Landbruksdirektoratet, 2025). It is difficult to measure the value of such materials, but the assignment aims to provide insight into the benefits of these loose materials.     Landskapsarkitekturen er en av de få disiplinene som krever et grafisk medium for å utføre sitt arbeid, ettersom arbeidet til en landskapsarkitekt innebærer blant annet å produsere plantegninger, snitt og perspektiver for å kunne kommunisere sine visjoner for et sted. I den sammenheng har håndtegningen vært et viktig medium i lang tid for å kommunisere og representere ideer. Med den digitale revolusjonen ser vi at det er bekymringer for at håndtegningen kan bli glemt hos de nyere generasjonene av landskapsarkitekter. Med tiden har også bruken av tegninger som perspektivtegninger blitt kritisert ved representasjonen av landskapet, siden de har blitt tenkt på som fotografier enn et landskap. I tillegg kan det være krevende for slike tegninger å representere landskapet siden det består av den romlige, temporale og materielle dimensjonen. Med utgangspunkt i kritikken er denne masteroppgavens problemstilling: Hvordan kan håndtegnede perspektiver og animasjon fungere som redskap for å utforske og representere den romlige erfaringen ved bevegelse gjennom anlegg? Målet med denne oppgaven er å utforske mulighetene håndtegningens teknikker og virkemidler, inkludert animasjon, tilbyr representasjon og egen forståelse av anlegg. Denne masteroppgaven har utforsket håndtegningens virkemidler og teknikker ved representasjonen og forståelsen med sekvenser i form av perspektivtegninger inspirert av Gordon Cullens Serial Vision. Representasjonen av stedene har utgangspunkt i Kevin Lynchs utvalgte lesbare elementer i byen. Den overordnede metoden for denne oppgaven er Research by design der det er brukt befaringer, tegning og animasjon. Deretter har erfaringer, refleksjoner og observasjoner fra tegne – og animasjonsarbeidet blitt diskutert opp mot problemstillingen. Oppgaven konkluderer med at håndtegningen og animasjonen kan fungere som redskap for å representere og utforske, men at de belyser ulike kvaliteter ved den romlige erfaringen ved bevegelse.Landscape architecture is one of the few disciplines which require a graphical medium to do their work since landscape architects must amongst other things produce site plans, sections, perspectives to communicate their visions for a place. Hand drawing has for a long time in this regard been an important medium for communication and representation of ideas. In recent times, with the rise of digital technology, we see that there is a concern amongst some that hand drawing can be forgotten amongst the newer generations of landscape architects. Over time, the use of drawings such as perspective drawings has been criticized in representation of landscape, because they are first and foremost thought as images rather than landscapes. Besides this criticism of perspective drawings, drawings can have difficulties in representing landscapes since they consist of three dimensions, which are the spatial, temporal and material dimensions. Based on this criticism, the research question of this master’s thesis is: How can hand drawn perspective drawings and animation work as a tool for exploring and representing the spatial experience by movement through a site? The aim of this thesis is to explore the possibilities offered by the techniques and tools of hand drawing, which include animation, in representation and own understanding of a site. This master thesis has explored the tools and techniques of hand drawing in representation and understanding with sequences of perspective drawings inspired by Gordon Cullen’s Serial Vision. The representation of the places is based on Kevin Lynch’s elements of the city image. The method for this thesis is Research by design, where site visits, drawing and animation have been used. Then the experiences, reflections and observations from the drawing and animation process were discussed in relation to the research question. The thesis concludes that hand-drawing and animation can function as tools for representing and exploring, but that they shed light on the different qualities of the spatial experience by movement

    Discovery of Novel Antimicrobials to Combat Bovine Mastitis-Associated Pathogens

    Bovine mastitis, a common disease in dairy cattle, causes considerable economic losses in the dairy industry. This is largely due to decreased milk production, veterinary treatment and culling of infected cows. Furthermore, the disease is a leading cause of antibiotic use in the dairy industry which likely contributes to the rise in antimicrobial resistance (Touza-Otero et al., 2024; Cheng et al., 2019). Thus, to reduce the frequency of infection, as well as limit the use of antibiotics, there is a need for developing new treatment strategies. A potential alternative or addition to antibiotics is bacteriocins, which are antimicrobial peptides derived from bacteria. This diverse group of peptides exhibit potent antimicrobial activity against many mastitis-causing bacteria (Darbandi et al., 2021). However, current bacteriocins are not being used therapeutically likely because of unsuitable properties. Finding new bacteriocins with more suitable properties is therefore desired. In this thesis I describe the screening, purification and identification of the novel bacteriocin Salivaricin MA (SalMA). SalMA was discovered and identified through screening of 78 samples from the farm environment and various other samples. The samples were screened for the presence of microorganisms exhibiting antimicrobial activity against mastitis-pathogens. Promising microorganisms were isolated and the antimicrobial they produced were purified by various chromatographic methods and identified by mass spectrometry and genetic approaches. SalMA was shown to kill several strains of the mastitis pathogen Str. uberis. The bacteriocin could be purified by chromatography and was shown to be resistant to heat. More research is needed to establish if there are potential applications for SalMA or if it is suitable as a therapeutic, for example in the treatment of bovine mastitis

    CHARACTERIZATION OF THE NUTRITIONAL VALUE OF POPLAR (POPULUS SPP) LEAVES AND STEMS FOR RUMINANT FEEDING

    In recent years, climate change has threatened Mediterranean agriculture, especially livestock production. Agroecology, including agroforestry, has gained attention as a sustainable option. This study explored poplar leaves and stems as alternative fodder for small ruminants in the Mediterranean environment. We analyzed five poplar clones (JPT, TUCANO, IMOLA, ORION, and I214) in two farms in southern Tuscany. We measured nutritional properties (ADLom, NDFIP, DM, ADF, aNDF, ADL, CP, EE, and ASH), focusing on significant variables essential for the Cornell Net Carbohydrate and Protein System (CNCPS). Leaves showed no significant farm effects or farm × clone interactions. However, clones significantly influenced CP and NDFIP, and ASH, with JPT presenting lower CP and NDFIP but the lowest ASH. In contrast, stems had strong interactions between farm and clone for CP and NDFIP. Notably, TUCANO displayed higher CP on one farm, suggesting an adaptive advantage. In conclusion, genetic factors significantly affect nutritional value, with certain clones performing better in specific farm conditions. These findings support the integration of poplar biomass into small ruminant diets, offering a promising strategy to optimize feed resources, enhance animal productivity, and foster sustainable livestock systems in Mediterranean regions. Further research can strengthen this approach, supporting resilience under changing climatic conditions

    Forvaltningen av nasjonale turiststier i Norge: Besøksforvaltning i praksis – en analyse av seks stier med ulik forvaltningsstruktur

    Denne masteroppgaven undersøker hvordan de seks autoriserte nasjonale turiststiene i Norge, Besseggen, Trolltunga, Preikestolen, Kjerag, Fosseråsa og Rødøyløva, varierer med hensyn til besøksstrategier, bevaring av natur- og kulturverdier, samt organisatorisk og økonomisk struktur. Bakgrunnen for studien er den sterke veksten i naturbasert reiseliv de siste tiårene, og behovet for bærekraftig forvaltning av populære turmål med høyt besøkstrykk. Studien benytter en kvalitativ tilnærming med dokumentanalyse av offentlige saksdokumenter fra eInnsyn, supplert med egne feltobservasjoner. Analysen tar utgangspunkt i sentrale teorier innen bærekraft, besøksforvaltning og styringsmodeller, og søker å belyse hvilke strategier og tiltak som faktisk gjennomføres i praksis. Funnene viser at fysisk tilrettelegging i form av steinlegging, klopper, trapper og sikkerhetsinstallasjoner, er et sentralt virkemiddel for å redusere slitasje og øke sikkerhet og tilgjengelighet. Tiltakene varierer mellom stiene og tilpasses stienes topografi, besøksmengde og lokale behov. Samtidig påpekes et dilemma: Tiltak som gjør stiene mer tilgjengelige, kan også tiltrekke enda flere besøkende og svekke naturopplevelsen og villmarkspreget. Dette understreker behovet for balanserte strategier som kombinerer fysiske tiltak med besøksstyring, informasjon og overvåking. Videre viser analysen at hensyn til natur- og kulturverdier ofte er integrert i besøksstrategiene, men at tiltakene i ulik grad fanger opp mer skjulte og langsiktige økologiske effekter. Organiseringen av forvaltningen varierer betydelig, og spenner fra solide stiftelser med egne ansatte, til samarbeid mellom kommune, grunneiere og frivillige. Finansiering er ofte en utfordring, særlig i små kommuner, og inntektsmodeller som parkeringsavgifter og frivillige bidrag spiller en viktig rolle. Oppgaven konkluderer med at ordningen Nasjonal turiststi har bidratt til mer systematisk og helhetlig forvaltning, men at det fortsatt er et behov for å styrke kompetanse, finansiering og overvåking. For å sikre bærekraftig utvikling må forvaltningen være kunnskapsbasert, adaptiv og forankret i samarbeid mellom ulike aktører. Dette er avgjørende for at både naturverdiene og turopplevelsene skal kunne ivaretas i møte med økende turisme. Nøkkelord: Naturbasert reiseliv, nasjonale turiststier, stiforvaltning, offentlig–privat samarbeid.This master's thesis examines how the six designated National Tourist Trails in Norway – Besseggen, Trolltunga, Preikestolen, Kjerag, Fosseråsa, and Rødøyløva – vary in terms of visitor management strategies, conservation of natural and cultural values, and organizational and financial structures. The background for the study is the significant growth in nature-based tourism in recent decades, and the need for sustainable management of popular hiking destinations experiencing high visitor pressure. The study applies a qualitative approach through document analysis of public records from the eInnsyn portal, supplemented with field observations. The analysis draws on central theories related to sustainability, visitor management, and governance models, and aims to shed light on which strategies and measures are actually implemented in practice. The findings show that physical facilitation—such as stone paving, boardwalks, staircases, and safety installations—is a key tool for reducing wear and tear, and for improving safety and accessibility. The measures vary between trails and are adapted to each trail’s topography, visitor volume, and local conditions. At the same time, a dilemma emerges: improvements that increase accessibility can also attract more visitors, potentially weakening the sense of wilderness and natural experience. This highlights the need for balanced strategies combining infrastructure with visitor regulation, information dissemination, and monitoring. The analysis also shows that natural and cultural values are often integrated into visitor strategies, but that the implemented measures vary in their ability to address more subtle and long-term ecological impacts. The organizational models differ significantly, ranging from dedicated foundations with permanent staff to collaborations between municipalities, landowners, and volunteers. Financial sustainability remains a challenge, particularly for small municipalities, and funding models such as parking fees and voluntary contributions play an important role. The thesis concludes that the National Tourist Trail scheme has contributed to more systematic and holistic management, but that there is still a need to strengthen competence, funding, and monitoring. To ensure sustainable development, management must be knowledge-based, adaptive, and rooted in collaboration between multiple stakeholders. This is essential to safeguard both the natural values and the quality of visitor experiences in the face of growing tourism.     A review of the effects of clinical communication training on consultation duration, and veterinarians’ and pet owners’ satisfaction with the consultation

    Aim: To examine whether communication skills training (CST) in clinical communication affects the consultation regarding time consumption, as well as veterinarian and owner satisfaction. Method: The databases Web of Science, Scopus and Ovid were searched, the articles were screened in Rayyan. Results: The search yielded 1781 hits which, after deduplication and screening, resulted in eight included articles. These eight articles resulted in a total of ten individual measurements, as one study measured all three outcome variables. Six studies investigated the effect of CST on the duration of the consultation, of which only two found a statistically significant change. Three studies investigated the effect of CST on owner satisfaction, whereas only one found a statistically significant increase. Only one study investigated the effect of CST on veterinarian satisfaction and found a significant increase. Discussion: Increases in time consumption were only found in veterinarian studies, this may be due to veterinarians having more considerations in owner communication but may also be due to the level of previous CST. All included studies are based on relatively small populations, which results in low internal validity. Conclusion: There is insufficient evidence to conclude whether CST affects duration, veterinarians’ satisfaction, or owners’ satisfaction with the consultation.Formål: Undersøke hvorvidt opplæring i klinisk kommunikasjon har en effekt på konsultasjonen med tanke på tidsbruk, samt veterinærens og eiers tilfredshet. Metode: Litteratursøket ble utført i Web of Science, Scopus og Ovid. Artiklene ble screenet i Rayyan.     Modelling the Impact of Biochar and Fertilizer on Maize Production in Kenya Using the FAO AquaCrop Model

    Afrika sør for Sahara står overfor en rekke utfordringer knyttet til klimaendringer, som alle påvirker matsikkerheten. Det er et akutt behov for bærekraftige løsninger som kan bidra til å øke matproduksjonen og styrke robustheten i landbrukssystemene. Denne oppgaven undersøker effekten av å bruke biokull i kombinasjon med mineralgjødsel på maisproduksjon i Embu, Kenya, ved bruk av vekstmodellen AquaCrop, utviklet av FNs organisasjon for ernæring og landbruk (FAO). Som en del av BICEPS-prosjektet undersøker studien om denne integrerte strategien for jordforbedring kan øke avlingsnivået og motstandskraften mot klimarelaterte stressfaktorer, innenfor rammen av bevaringsjordbruk (CA). Studien fant at det var næringstilførselen (fra gjødsel) som var hoveddriveren bak økt avling, snarere enn biokullets effekt på jordas vannlagringsevne. To behandlinger ble analysert: én med 3 tonn biokull per hektar og gjødsel (BC3F), og en kontroll uten noen tilførsel. Feltdata ble samlet inn og deretter brukt til å kalibrere simuleringer i AquaCrop for å vurdere avlingsutvikling, vannbalanse i jorda og effekten av endret såtid. Modellen, etter kalibrering, klarte å gjenskape observert biomasse for begge behandlinger. BC3F- simuleringen viste betydelig høyere biomasse (7,48 t/ha) og kornavling (3,26 t/ha) sammenlignet med kontrollen (2,60 t/ha og 1,15 t/ha), noe som understreker viktigheten av næringstilgang. Disse resultatene samsvarte tett med observert biomasse på henholdsvis 7,27 t/ha for BC3F og 2,37 t/ha for kontrollen. Modellen hadde imidlertid utfordringer med å reprodusere observerte verdier for bladdekke, særlig under næringsstress. Dette skyldes trolig AquaCrops forenklede tilnærming til fertilitetsstress og mulig avvik mellom feltmålt bladdekke og modellens interne definisjon. Statistiske analyser av jordas vannretensjon viste ingen signifikante forskjeller mellom behandlingene, noe som indikerer at biokull ikke hadde noen målbar effekt på jordas vannlagringsevne i dette forsøket. Mangelen på en ren biokull-behandling gjorde det vanskelig å isolere biokullets uavhengige effekt på avling. En hypotetisk AquaCrop-simulering uten gjødsel ble forsøkt for å undersøke dette, men modellen klarte ikke å generere realistiske resultater på grunn av kalibreringsutfordringer og fravær av feltdata til validering. Simuleringer med alternative sådatoer viste at tidligere såing kunne forbedre avling og redusere vannstress under vekstsesongen 2024. Oppsummert viste studien at biokull alene ikke forbedret jordas vannretensjon, men at kombinasjonen med gjødsel hadde en positiv effekt på avlingene. Funnene understøtter at forbedret næringstilgang er avgjørende for å øke maisproduksjonen, og støtter bruken av integrert jordfruktbarhetsforvaltning som en strategi for klimarobust landbruk. Fremtidig forskning bør inkludere mer detaljert modellering av næringsstoffer, flerårig feltvalidering og scenario-baserte AquaCrop-simuleringer for å undersøke biokullets effekt under ulike innsatsnivå, for å kunne vurdere dets rolle som bærekraftig landbruksstrategi i Afrika sør for Sahara.Sub-Saharan Africa is facing a number of challenges due to climate change, that all affect food security. There is an urgent need for sustainable solutions that can help increase crop production and support resilient agricultural systems. This thesis explores the impact of applying biochar in combination with mineral fertilizer on maize production in Embu, Kenya, using the crop growth model AquaCrop, developed by the United Nation’s FAO. As part of the BICEPS project, the study investigates whether this integrated soil amendment strategy can enhance crop productivity and increase resilience to climate-related stress as a conservation agriculture (CA) practice. The study found that added nutrients (from fertilizer) were the primary driver of yield improvements, rather than biochar’s effect on soil water retention. Two treatments were analyzed: one with 3 tons of biochar per hectare and fertilizer (BC3F), and a control with no inputs. Field data were collected and used to calibrate simulations in AquaCrop to evaluate yield development, soil water balance, and the effects of sowing date variation. The model, after calibration, reproduced observed biomass values for both treatments. The BC3F simulated significantly higher biomass (7.48 t/ha) and grain yield (3.26 t/ha) compared to the Control (2.60 t/ha and 1.15 t/ha), underlining the importance of nutrient supply. This matched very closely with the observed biomass of 7.27 ton/ha for BC3F and 2.37 for the Control. However, the model had difficulty reproducing observed canopy cover, especially under stressed conditions. This was likely due to AquaCrop’s simplified fertility stress approach and potentially a mis-match between field-measured canopy cover and the internal definition in AquaCrop. Statistical analysis of the soil water retention data showed no significant difference between treatments, indicating that biochar had no measurable effect on water-holding capacity in this experiment. The lack of a biochar-only treatment limited the ability to isolate biochar’s independent effect on yield. A hypothetical AquaCrop simulation without fertilizer inputs was attempted to explore this scenario, but the model could not produce realistic results due to calibration challenges and the absence of observed data for validation. Simulation of alternative sowing dates indicated that earlier planting may improve yield and reduce water stress during the 2024 season’s weather. These results suggest that agronomic management strategies such as integrated nutrient use and sowing date adjustments could enhance maize performance in smallholder systems. Nonetheless, the simulations also revealed AquaCrop’s limitations in fully capturing biochar-soil-plant interactions, particularly under nutrient stress. In conclusion, while biochar alone did not improve soil water retention, its combination with fertilizer proved effective for yield enhancement. These findings support that boosting nutrient availability is key to increase maize production, supporting the use of integrated soil fertility management in climate-resilient maize production. Future work should include more detailed nutrient modeling, multi-year field validation, and scenario-based AquaCrop simulations to explore biochar’s effects under varying input conditions, in order to better assess its role in sustainable intensification strategies in sub-Saharan Africa

    An evaluation of the quantification of mineral content using SlateNorm software

    This study explores applying mineral norm calculations to evaluate the reliability of determining shale mineral composition from whole-rock geochemical analysis. Under-standing the mineralogical composition of shales like alum shale, known for high heavy metal/organic content and acid-producing capabilities (acid rock drainage), is critical for assessing shale related environmental risks and geotechnical challenges. While traditional methods like X-ray diffraction (XRD) can be both time-consuming and expensive, mineral norm calculations offer a potentially cheaper alternative. In this study, the practicality and accuracy of an existing norm calculation method originally developed for roofing-slate assessments, SlateNorm (Wagner et al., 2020), which estimates mineralogy from whole-rock geochemical composition was tested and compared with associated quantitative XRD mineral compositions. Data were compiled from four published shale datasets, consisting of 150 samples in total. The geochemical data required modifications (documented in this study) to meet SlateNorm input requirements. SlateNorm outputs were compared against XRD results using absolute relative difference, Lin’s Concordance Correlation Coefficient (rc), and a whole-dataset scatterplot showing absolute relative difference between SlateNorm and XRD plotted against XRD wt.% for each mineral within each sample (mineral-sample pair), including a modified accuracy threshold. Results indicated SlateNorm calculations deviated significantly from XRD, with a total of nearly one third (31%) of mineral-sample pairs exceeding the established accuracy threshold. Systematic over- and under-estimations (chlorite, clays) and large, erratic errors for quartz, feldspar, and pyrite were common. Lin's rc values were generally poor (mostly <0.70), indicating weak concordance with XRD. SlateNorm's relatively poor performance is likely due to algorithm limitations (e.g., ignoring structural water (H₂O⁺), applying assumptions more suited for roofing slates) and potentially also due to the necessary input data modifications. As a result, SlateNorm, in its current form is not considered a practical tool for reliable shale mineral identification, quantification, or assessing ARD potential based on its mineralogical output. Future work should address algorithm limitations (e.g., incorporating H₂O⁺ for input, and pyrrhotite output) and validate against high-resolution methods. The overall most reliable method probably still involves Automated Mineralogy Systems described in Chapter 4.2, and despite time and cost requirements it might be useful to apply these systems, even at a smaller scale, to evaluate their usefulness for shale (e.g., alum shale) mineral quantification

Krav om bærekraftsrapportering kan gradvis påvirke aktører i en hel verdikjede, også innen den jordbruksbaserte matsektoren. Problemstillingen som undersøkes, er hvordan lovpålagt bærekraftsrapportering og EUs taksonomi kan påvirke aktører i den norske matsektoren, både direkte og indirekte gjennom indirekte effekter. Oppgaven undersøker utvalgte markedsregulatorer i den norske jordbruksbaserte matsektoren og primærnæringen. Formålet er å analysere om lovpålagt bærekraftsrapportering (CSRD) hos aktører høyere opp i verdikjeden medfører ringvirkninger nedover, og påvirker primærnæringen. Denne problemstillingen er viktig fordi rapporteringskravene kan påvirke kostnader, produksjonspraksis og bærekraftige forretningsmodeller i matsektoren. Økt innsikt vil være verdifull for aktører i verdikjeden, inkludert bønder og ledere, ved at de kan tilpasse seg og møte de nye kravene på en hensiktsmessig måte. Studien vil benytte en kvalitativ metode med semi-strukturerte intervjuer av fagfolk og ledere i næringsmiddelindustrien, og bønder, for å samle inn perspektiver og erfaringer rundt temaet. Resultatene viser at CSRD har medført en del merkbare konsekvenser for de største aktørene i den jordbruksbaserte matsektoren, i form av økte kostnader og behov for ny kompetanse. Samtidig ser vi muligheter for økt transparens, styrket tillit til norsk jordbruk og bedre klimadokumentasjon. Bøndene opplever i dag få indirekte effekter, og det er viktig å holde rapporteringsbyrden høyt oppe i verdikjeden. På denne måten kan bøndene fokusere på produksjon av norsk mat. Oppgaven konkluderer med at den lovpålagte bærekraftsrapporteringen treffer de tiltenkte gruppene slik situasjonen er i dag, uten nevneverdige indirekte effekter hos for eksempel primærnæringen. Samtidig må de rapporteringspliktige i dag bruke mye ressurser på omstilling og rapportering. Det vil kreve mer forskning for å undersøke om kommende krav vil gi indirekte effekter og om andre parter enn topp og bunn i verdikjeden blir berørt.The implementation of the Corporate Sustainability Reporting Directive (CSRD) into Norwegian legislation marks a new chapter for the business sector. The requirements for sustainability reporting may gradually influence actors throughout entire value chains, including the agriculture-based food sector. This thesis investigates how mandatory sustainability reporting, and the EU Taxonomy may affect actors in the Norwegian food sector, both directly and indirectly. The study focuses on selected market regulators within the Norwegian agriculture-based food sector and primary production, aiming to analyze whether CSRD at higher levels of the value chain creates downstream ripple effects that impact the primary sector. Reporting requirements may influence costs, production practices, and sustainable business models, and increased insight can help actors such as farmers and industry leaders adapt effectively. The study uses a qualitative method based on semi-structured interviews with professionals in the food industry and farmers. Findings show that CSRD has led to noticeable consequences for major actors, including higher costs and a need for new competencies, but also offers potential for greater transparency, trust in Norwegian agriculture, and improved climate reporting. Farmers currently experience few indirect effects, and it is important to keep the reporting burden concentrated at higher levels of the value chain. The thesis concludes that CSRD currently reaches its intended target groups without significant spillover effects on the primary sector, though further research is needed to assess potential future impacts across the entire value chain

    Studier av nye genetiske faktorer som påvirker β-laktamsensitivitet i meticillin-resistente Staphylococcus aureus (MRSA)

    Staphylococcus aureus er en opportunistisk patogen som det er knyttet stor bekymring til på bakgrunn av bakteriens enestående evne til å utvikle resistens mot antibiotika. Et viktig grep i kampen mot antibiotikaresistens er å identifisere nye genetiske mål som kan svekke bakterien,enten alene eller i kombinasjon med andre allerede eksisterende antibiotika. beta-laktamantibiotika er et av de mest brukte mot S. aureus. Målet i denne oppgaven er å identifisere hittil ukjente gener som påvirker sensitivitet for beta-laktamer i MRSA stammen JE2, i tillegg til videre arbeid med et allerede oppdaget gen ved tidligere arbeid i forskningsgruppen, SAOUHSC_01975. Videre studeres disse genene for å forstå hvordan antibiotikasensitiviteten påvirkes. Utgangspunktet for arbeidet var en CRISPRi-sekvenseringsknockdownscreen utført i melk, hvor relativ sgRNA-frekvens ble brukt for å identifisere gener som potensielt påvirker sensitiviteten for penicillin G. Det ble gjort et utvalg av 23 kandidatgener, som videre ble validert ved MIC-assay for beta-laktamer. 12 av 23 viste avvikende sensitivitet for penicillin G sammenlignet med MIC-verdi til villtype. Fra denne gruppen ble valgt tre gener valgt ut som videre ble testet mot et utvalgt antibiotikapanel. To av disse tre ble ekskludert på grunn av antatt påvirkning fra kjente essensielle gener nedstrøms. Det ble derfor arbeidet videre med SAOUHSC_02650 og SAOUHSC_01975 med mål om å tilegne mer kunnskap om molekylære og funksjonelle egenskaper. Videre arbeid besto av plasmidkonstruksjon, oppreguleringsanalyser og fluorescensmikroskopi. SAOUHSC_02650 ble valgt på bakgrunn av redusert vekst ved knockdown, og en predikert memebranlokalisasjon. Nedregulering førte til en moderat, men reproduserbar økning i følsomhet for både penicillin G og oxacillin. Prediksjoner ved UniProt ble benyttet for å undersøke egenskaper, og sammen med eksperimentelle funn kan det antyde at genet koder for et transmembrant protein, og kan være involvert i celleveggstabilitet, transport eller signaloverføring. SAOUHSC_01975 ble undersøkt videre som en mulig regulator med en mulig effekt på celledeling. Nedregulering viste 2-folds endring for beta-laktamer. Bioinformatikk, og resultater fra dette arbeidet antyder at det kan bidra til å regulere uttrykket av celleveggrelaterte enzymer som Sle1 og antibiotikasensitivitet. Avslutningsvis antyder resultatene at begge genene kan være involvert i prosesser som påvirker cellevegghomeostase og antibiotikasensitivitet, og kan representere mulige mål for fremtidig antibiotikautvikling.Staphylococcus aureus is an opportunistic pathogen associated with great concern due to its unique ability to develop resistance to antibiotics. An important step in the battle against antibiotic resistance is to identify new genetic targets that can weaken the bacteria, either alone or in combination with other existing antibiotics. b-lactamantibiotics are among the most widely used treatments against S. aureus. The goal of this thesis is to identify previously unknown genes that affect sensitivity to b-lactams in the MRSA strain JE2, in addition to further work with an already discovered gene by the research group, SAOUHSC_01975. Furthermore, the study seeks to investigate these genes to understand how antibiotic sensitivity is affected. This work was based on a previous CRISPRi sequencing knockdown screen performed in milk, where relative sgRNA frequency was used to identify genes that potentially affect sensitivity to penicillin G. A selection of 23 candidate genes was made, which were further validated by MIC-assay for b-lactams. 12 of 23 showed deviating sensitivity to penicillin G compared to the MIC-value of the wild type. From this group, three genes were selected which were further tested against a selected antibiotic panel. Two of three were excluded due to presumed influence from known essential genes downstream. Due to exclusion of the two previously mentioned, further work proceeded with SAOUHSC_02650 and SAOUHSC_01975 with the aim of acquiring more knowledge about molecular and functional properties. Further work consisted of plasmid construction, upregulation analyses and fluorescence microscopy. SAOUHSC_02650 was selected based on reduced growth upon knockdown and a predicted membrane localization. Downregulation led to a moderate, yet reproducible increase in susceptibility to both penicillin G and oxacillin. Predictions by UniProt were used to investigate properties, and together with experimental findings, it may suggest that the gene encodes a transmembrane protein, and may be involved in cell wall stability, transport or signa transmission. SAOUHSC_01975 was further investigated as a regulator with a possible effect on cell division. Downregulation showed a 2-fold change for b-lactams. Bioinformatics and results from this work suggests that it may contribute to regulating the expression of cell wall-related enzymes such as Sle1 and antibiotic sensitivity. Additionaly both SAOUHSC_02650 and SAOUHSC_01975 may be involved in processes that affect cell wall homeostasis and antibiotic sensitivity and may represent possible targets for future antibiotic development
