    Dynamics of SLC25A51 Reveal Preference for Oxidized NAD+ and Substrate Led Transport

    SLC25A51 is a member of the mitochondrial carrier family (MCF) but lacks key residues that have been attributed contribute to the mechanism of other nucleotide MCF transporters. Thus, how SLC25A51 transports NAD+ across the inner mitochondrial membrane remains unclear. To elucidate its mechanism, we used Molecular Dynamic simulations to reconstitute used Molecular Dynamic simulations to study reconstituted SLC25A51 homology models into lipid bilayers and generated hypotheses to test. We observed spontaneous binding of cardiolipin phospholipids to three distinct sites on the exterior of SLC25A51’s central pore and found that mutation of these sites impaired cardiolipin binding and transporter activity. We also observed that stable formation of the required matrix gate was controlled by a single salt bridge. Using simulation data and in-cell activitybiochemical assays we We identified binding sites in SLC25A51 for NAD+ and showed that its binding and specificity selectivity for NAD+ was guided by an electrostatic interaction between NAD+ the charged nicotinamide ring in the ligand and a negatively charged patch in the pore. In turn, interaction of NAD+ with interior residue E132 guided the ligand to dynamically engage and weaken the salt bridge gate, representing a ligand-induced initiation of transport

    Decreto real dirigido al Obispo de Guadalajara Antonio Alcalde y Barriga, OP, proponiendo la creación de cuatro custodias de religiosos franciscanos observantes en las provincias internas de Nueva España. Estas custodias se establecerían para mejorar la gestión espiritual y temporal de las misiones y pueblos en esa área, debido a los problemas causados por la división actual de responsabilidades entre obispos y religiosos. Cada custodia tendría su propio líder y vicarios para supervisar de cerca a los misioneros. Se planea establecer casas de hospicio en las sedes de cada provincia para albergar y capacitar a los religiosos. Esto se hace con la esperanza de promover la difusión del cristianismo entre los indígenas y mejorar la vida de los fieles en esas regiones. Las custodias no serían fundaciones formales y su existencia dependería de la voluntad real. También se destaca la importancia de evitar que los religiosos en las misiones perciban derechos parroquiales, lo que ha causado conflictos. La propuesta fue inicialmente presentada por Fray Antonio de los Reyes de la Orden de San Francisco Observantes y presidente de las misiones de la Sonora en la Pimería Alta. —— Royal decree addressed to the Bishop of Guadalajara Antonio Alcalde y Barriga, OP, proposing the creation of four custodies of observant Franciscans in the internal provinces of New Spain. These custodies would be established to improve the spiritual and temporal management of missions and towns in that area to address the problems the current division of responsibilities between bishops and friars has caused between the two groups. Each custody would have its own leader and vicars to closely supervise the missionaries. He proposes to establish convents in the headquarters of each province to house and train the friars. This is done with the hope of promoting the spread of Christianity among the Indigenous people and improving the lives of the faithful in those regions. The custodies would not be formal foundations, and their existence would depend on the will of the sovereign. He also emphasizes the importance of preventing the friars in the missions from receiving parochial rights, which has caused conflicts. Friar Antonio de los Reyes of the Order of Observant Franciscans and president of the missions of Sonora in the Pimería Alta initially presented the proposal. 9 f. (18 p.
