Visualizing Fluorescent Fusion Protein Fate by Cell-Cycle Induced Recombination
Protein dynamics can be observed through the construction of fluorescent fusion proteins and subsequent fluorescence microscopy. Typically, individual proteins are indistinguishable and observations are limited to the entire population of that protein. Recent advancements have been made which can distinguish proteins by inducing a fluorescent color change to already synthesized proteins. These proteins can be differentiated between those produced before and after the color change event. However, induction of this color change must be controlled extracellularly. This thesis explores a system that uses intracellularly controlled, inducible genetic recombination to change the color in a fluorescent fusion protein expressed in S. cerevisiae for fluorescence microscopy. This work aims to construct a system for observing protein partitioning in dividing cells, where reversible recombination is induced via cellular control with every cell cycle and proteins are distinguished between “old” or “new” during development and division. Additionally, this work aims to further characterize a previously constructed system with extracellular galactose-induced irreversible recombination to elucidate the molecular dynamics of recombination. These findings will be used in the cell cycle dependent system to ensure recombination is precisely timed. This system will provide a tool to gain insight into the partitioning of cellular contents during division via fluorescence microscopy, which has broad implications in cellular biology and biochemistry

Combinatorial Synthesis of Protein-Polymer Conjugates and Protein-Drug Conjugates for Targeted Drug Delivery
Targeted drug delivery systems like protein-polymer conjugates (PPCs) and protein-drug conjugates (PDCs) offer a promising alternative to traditional cancer treatments by decreasing off-target effects and lowering the dosage required for effective treatment. PPCs and PDCs are both robust systems that offer a variety of biological applications. PPCs are able to load multiple drugs per polymer chain, thus lowering the dosage required when compared to PDCs, which are limited in their loading. Small molecules have minimal effects on the conjugated protein if the conjugation site is away from the binding interface, whereas polymers can either stabilize or destabilize the protein, vary pharmacological and chemical properties of the PPC, or it could block binding of the protein to cell receptors. In this thesis, we proposed to develop a combinatorial approach to synthesize a library of amine-reactive amphiphilic copolymers and probe the influence of polymer chemistry and reactivity on the preparation and function of PPCs in a biological context. Further, we proposed to design and synthesize a cleavable disulfide linker for protein-drug conjugates that aims to expand the design space for protein-small molecule conjugates. We also investigated the release rates and kinetics of the cleavable linker. We will be targeting the integrin αvβ3 receptor present on solid tumor cells for targeted drug delivery via engineered Fn3 proteins with specific binding to integrin αvβ3. The Fn3 proteins will either be conjugated to polymers that can have stabilizing effects on the protein and/or carry cytotoxic payloads, or conjugated directly to cytotoxic payloads via a cleavable disulfide linker

The activists in the first wave represent a wide variety of women’s rights activism, from participation in the 1970s women’s movements to work with Roma women. The activists in the second wave were affiliated with the Cassero, a local LGBTQ+ resource center, and primarily identify themselves as queer activists
Examining Information Systems Use to Facilitate the Workplace Accommodation Process
BACKGROUND: The workplace accommodation process is often affected by ineffective and inefficient communications and information exchanges among disabled employees and other stakeholders. Information systems (IS) can play a key role in facilitating a more effective and efficient accommodation process since IS has been shown to facilitate business processes and effect positive organizational changes.
OBJECTIVE: Since there is little to no research that exists on IS use to facilitate the workplace accommodation process, this paper, as a critical first step, examines how IS have been used in the accommodation process.
METHODS: Thirty-six interviews were conducted with disabled employees from various organizations. Open, axial, and selective coding were part of the analysis. Fuzzy set qualitative comparative analysis was used to identify different levels of IS use based on participants’ descriptions. RESULTS: An IS used in the workplace accommodation process consists of electronic request form, accommodation checklist, special budget, specific role, ancillary service, formal policy and procedure. There are different levels of IS use in the current accommodation process. The high-level IS use often results in a better accommodation performance than the low-level IS use, including high efficiency, high effectiveness, and low emotional tolls. Nevertheless, the high-level IS use often uses a specific, inflexible template as well as disregards human elements in the accommodation process.
CONCLUSION: This work provides implications that future IS design should raise awareness of disability and accommodation, account for individual differences, involve multiple stakeholder inputs, as well as address the fundamental social issues in the accommodation process
