Multiphysics Simulation and Experimental Investigation of the Densification of Metals by Spark Plasma Sintering
Multivariant experimental investigations and multiphysics microstructural modeling of the spark plasma sintering process of metallic powders have been performed up to a relative density of approximately 80%. In comparison, the effect of sintering temperature, pressure, and particle size on the interparticle contact area growth and axial shrinkage of cylindrical specimens of copper and nickel particles is measured in laboratory scaled tests. Herein, for the first time all relevant for sintering phenomena are considered simultaneously: the fully coupled thermo-electro-mechanical modeling of the spark plasma sintering processes, additionally taking into account for lattice, grain boundary, surface diffusion, electromigration, and thermomigration, has been carried out. The computational analysis of various physical phenomena allows to identify dominant and insignificant mechanisms. The two-level numerical simulation includes the modeling of the sintering setup at the macroscopic level and the neck formation process in particle chain systems at the microscopic level. The results of the numerical simulations show a very good agreement with the experimental data. Therefore, the impact of electrical and mechanical loads as well as of particle size on microscopic distribution of temperature, inelastic strain, and on densification has been studied by the finite element simulations
M‐CSF directs myeloid and NK cell differentiation to protect from CMV after hematopoietic cell transplantation
Therapies reconstituting autologous antiviral immunocompetence may represent an important prophylaxis and treatment for immunosuppressed individuals. Following hematopoietic cell transplantation (HCT), patients are susceptible to Herpesviridae including cytomegalovirus (CMV). We show in a murine model of HCT that macrophage colony‐stimulating factor (M‐CSF) promoted rapid antiviral activity and protection from viremia caused by murine CMV. M‐CSF given at transplantation stimulated sequential myeloid and natural killer (NK) cell differentiation culminating in increased NK cell numbers, production of granzyme B and interferon‐γ. This depended upon M‐CSF‐induced myelopoiesis leading to IL15Rα‐mediated presentation of IL‐15 on monocytes, augmented by type I interferons from plasmacytoid dendritic cells. Demonstrating relevance to human HCT, M‐CSF induced myelomonocytic IL15Rα expression and numbers of functional NK cells in G‐CSF‐mobilized hematopoietic stem and progenitor cells. Together, M‐CSF‐induced myelopoiesis triggered an integrated differentiation of myeloid and NK cells to protect HCT recipients from CMV. Thus, our results identify a rationale for the therapeutic use of M‐CSF to rapidly reconstitute antiviral activity in immunocompromised individuals, which may provide a general paradigm to boost innate antiviral immunocompetence using host‐directed therapies
Clinical improvement of Long-COVID is associated with reduction in autoantibodies, lipids, and inflammation following therapeutic apheresis
In the aftermath of the COVID-19 pandemic, we are witnessing an unprecedented wave of post-infectious complications. Most prominently, millions of patients with Long-Covid complain about chronic fatigue and severe post-exertional malaise. Therapeutic apheresis has been suggested as an efficient treatment option for alleviating and mitigating symptoms in this desperate group of patients. However, little is known about the mechanisms and biomarkers correlating with treatment outcomes. Here, we have analyzed in different cohorts of Long-Covid patients specific biomarkers before and after therapeutic apheresis. In patients that reported a significant improvement following two cycles of therapeutic apheresis, there was a significant reduction in neurotransmitter autoantibodies, lipids, and inflammatory markers. Furthermore, we observed a 70% reduction in fibrinogen, and following apheresis, erythrocyte rouleaux formation and fibrin fibers largely disappeared as demonstrated by dark field microscopy. This is the first study demonstrating a pattern of specific biomarkers with clinical symptoms in this patient group. It may therefore form the basis for a more objective monitoring and a clinical score for the treatment of Long-Covid and other postinfectious syndromes
Deep learning trained on lymph node status predicts outcome from gastric cancer histopathology: a retrospective multicentric study
Aim
Gastric cancer (GC) is a tumour entity with highly variant outcomes. Lymph node metastasis is a prognostically adverse biomarker. We hypothesised that GC primary tissue contains information that is predictive of lymph node status and patient prognosis and that this information can be extracted using deep learning (DL).
Methods
Using three patient cohorts comprising 1146 patients, we trained and validated a DL system to predict lymph node status directly from haematoxylin and eosin–stained GC tissue sections. We investigated the concordance between the DL-based prediction from the primary tumour slides (aiN score) and the histopathological lymph node status (pN). Furthermore, we assessed the prognostic value of the aiN score alone and when combined with the pN status.
Results
The aiN score predicted the pN status reaching area under the receiver operating characteristic curves of 0.71 in the training cohort and 0.69 and 0.65 in the two test cohorts. In a multivariate Cox analysis, the aiN score was an independent predictor of patient survival with hazard ratios of 1.5 in the training cohort and of 1.3 and 2.2 in the two test cohorts. A combination of the aiN score and the pN status prognostically stratified patients by survival with p-values <0.05 in logrank tests.
Conclusion
GC primary tumour tissue contains additional prognostic information that is accessible using the aiN score. In combination with the pN status, this can be used for personalised management of GC patients after prospective validation
Magnetically Enhanced Thin-Film Coarsening by a Magnetic XPFC Model Allowing to Decouple Magnetic Anisotropy and Magnetostriction
External magnetic fields provide a macroscopic control mechanism to influence the microstructure of polycrystalline materials. Herein, the influence of strong magnetic fields on grain growth in thin films is modeled with a magnetic extended phase field crystal model. The magneto–structural effects are incorporated into the correlation function in reciprocal space. With this approach, magnetic anisotropy, magnetostriction, and mobility of grain boundary can be controlled and a variety of geometrical and topological properties consistent with experimental results can be determined
Cyclist’s travel distances and risk of falls in snowy and icy conditions in German cities
Introduction: So far, no studies are known that estimate distance-based risks for cyclist falls in snowy/icy conditions compared to other conditions to account for differences in cycling levels in the different weather situations. Method: The number of cyclist falls was gathered from retrospective surveys in Germany. Cycling distances were obtained from the German Household Travel Survey “Mobility in cities – SrV,” assigned to meteorological data, and validated against counts and own surveys. The number of falls per distance cycled and Risk Ratios for snowy/icy versus other weather conditions were estimated. Results: An average decrease of 53% in the distance travelled per person and day is estimated for snowy/icy days versus other days. This decrease is lower in regions with higher general cycling mode shares. We find average risks of falls from 9.5 to 16 (field surveys) up to 76.5 falls per 10,000 km (online survey) and average Risk Ratios for cycling in snowy/icy conditions of 20 (field survey conducted in times of other weather) to 36 (field survey conducted in times of snow/ice) and 38 (online survey conducted in times of snow/ice). The risk of suffering an injury in the event of a fall is lower in snowy/icy compared to other weather conditions. Conclusions: Seeing the current trend of growing general cycling levels in Germany, we expect more cycling in winter and, in case of unchanged winter weather and maintenance, a substantial increase of cyclist falls. The reduced risk of being injured in the event of a fall in snowy/icy conditions does not outweigh the higher risk of falling in the first place. Practical Applications: Improved winter maintenance on cycling facilities can help increase winter cycling and reduce the risk of falls at the same time
