F. nucleatum が誘導する上皮間葉転換に対するレスベラトロールの影響
大阪歯科大学Osaka Dental University博士(歯学)Fusobacterium nucleatum, an anaerobic opportunistic pathogen associated with periodontitis, is believed to be associated with the development of oral squamous cell carcinoma (OSCC). Previous studies reported that the initial host molecular alterations induced by F. nucleatum infection may contribute to a predisposition for malignant transformation through epithelial-mesenchymal transition (EMT). Resveratrol (trans-3,4,5-trihydroxystilbene) is a polyphenol that regulates human lipid metabolism, inhibits platelet aggregation, protects cardiovascular tissue, and has anti-inflammatory and antioxidant effects. Accumulating evidence suggests that resveratrol can reverse EMT and inhibit the invasion and migration of human tumors through diverse mechanisms and signaling pathways. In this study, wound healing assays and quantitative real-time reverse transcription polymerase chain reaction (real-time RT-PCR) were performed to assess the migratory ability and EMT-relative mRNA expression of human oral squamous carcinoma-3 cells, an OSCC cell line. Our study demonstrated that F. nucleatum could trigger EMT, and resveratrol may prevent the effect of F. nucleatum in the OSCC cell line HSC-3.doctoral thesi
日本人学生と中国人留学生との甘味感受性について
大阪歯科大学Osaka Dental University博士(歯学)Studies have shown that the sugar content of sugar-sweetened beverages (SSBs) is associated with an increased risk of dental caries and erosion. Interestingly, the market shares of SSBs differ significantly between Japan and China. This study aimed to investigate the disparities in eating habits, especially sensitivity to sweetness, between Japanese and Chinese students. A total of 100 healthy Japanese and Chinese participants aged 19 – 34 years without any taste disorders were surveyed. The participants were asked to complete a questionnaire regarding their eating habits and undergo a sweetness sensitivity test. We found that Japanese students tended to consume a higher proportion of sweet snacks and sugar-free beverages in their daily choices, whereas a lower proportion of Chinese students consumed the same (p < 0.01). Additionally, compared with Chinese students, Japanese students were generally less concerned about the use of artificial sweeteners (p < 0.05). However, the longer Chinese students lived in Japan, the less likely they were to worry about artificial sweeteners. This study revealed differences in the sensitivity to sweetness between Japanese and Chinese students. These findings may help promote the development of public health initiatives, aimed at reducing the prevalence of tooth decay and related oral or general diseases.doctoral thesi
レセプトデータを用いた口腔・咽頭がん患者に対するセツキシマブの治療実態に関する疫学研究
大阪歯科大学Osaka Dental University博士(歯学)This study aimed to examine the medical costs associated with oral and pharyngeal cancers using a large administrative claims database in Japan. Economic evaluation is crucial for maintaining universal health coverage in Japan. This study used the administrative claims database of 21,736 patients diagnosed with oral or pharyngeal cancer between April 2005 and December 2020. We calculated propensity scores based on age, sex, cancer site, lymph node metastasis, and first treatment. Using propensity score matching, we selected 195 participants from each group and compared treatment cost with and without cetuximab. The average monthly medical cost was 525,551 and 285,239 yen for the cetuximab administration and non-administration groups, respectively. When compared by sex, age, and cancer site, costs remained significantly higher for oral and pharyngeal than for salivary gland cancers. Among the conditions during treatment, 11.8% of patients experienced hypomagnesemia due to cetuximab administration, while 7.7% exhibited common acne. Patients in the cetuximab administration group were more commonly diagnosed with cancer-related pain, oral mucositis, and difficulties with oral intake. The cost for patients with oral cancer is related to patient characteristics and treatment patterns. Acknowledging the high medical expenses and need for careful attention to complications that may occur is essential.doctoral thesi
下顎枝矢状分割術後の NSD を惹起する因子について
大阪歯科大学Osaka Dental University博士(歯学)In this study, we investigated the factors affecting postoperative neurosensory disturbance (NSD) in patients who underwent sagittal split ramus osteotomy (SSRO).The subjects were 92 patients with 184 sides who underwent SSRO in our department from February 2021 to January 2022. NSD was evaluated preoperatively and 1, 3, 6, 12 months postoperatively using subjective and objective assessments. We examined whether eachdata obtained from medical records and preoperative CT images would affect NSD. Gender, age at surgery, Total surgical time for SSRO, and amount of blood loss were not factors associated with NSD. Factors that influence postoperative NSD include the direction of mandibular bone movement, the presence of inferior alveolar nerve exposure, the thickness of the mandibular bone, the distance from the maxillary tuberosity to the mandible, and the distance from the mandibular canal to the outer cortical bone.doctoral thesi
思春期前期の子どもの健康に関する要因分析
大阪歯科大学Osaka Dental University博士(口腔科学)We examined factors related to early adolescent health using the method of structural equation modeling. The study utilized self-administered questionnaire responses from 4,063 pairs of caregivers and their eighth-grade children taken from the “Child Living Conditions Survey” conducted by the local government in the Kanto region of Japan. The motivation of early adolescents to learn was strongly influenced by the educational investments made by their households. The responsive caregiver attitude contributed to the establishment of a rhythm of life and the enhancement of physical and mental well-being. The characteristic feelings of anxiety and loneliness during adolescence encouraged media use, which had a significant impact on worsening the child’s physical condition, and increased consumption of sweetened beverages. Households had an important influence on restricting media use. We found that the child’s household situation had a significant influence on whether the child had dental caries. Regardless of their household economic situation or dental health, they experienced an increase in motivation to learn and improved physical health through interactions at school. We think that it is necessary to further investigate adolescent-specific characteristics and the household economic situation which both impact the child’s total health.doctoral thesi
MSC培地を用いたiPS細胞からのMSC分化誘導
大阪歯科大学Osaka Dental University博士(歯学)Mesenchymal stem cells (MSCs) and induced pluripotent stem cells (iPS) cells have great potential as cell sources for tissue engineering and regenerative medicine This study aimed to investigate whether iPS cells can be differentiated into MSCs using MSCGM, a commercially available MSC culture system. The cells were characterized by flow cytometry, immunostaining, and gene expression analyses. We also examined their potential to differentiate into osteoblasts and chondrocytes. Our results showed that iPS cells cultured in MSCGM (iPS-MSCGM) exhibited a fibroblast-like morphology and expressed CD73 and CD90 genes, as well as positive markers for CD73, CD90, and CD105. Moreover, iPS-MSCGM cells demonstrated the ability to differentiate into osteoblasts and chondrocytes in vitro. This study demonstrates a new and simple method for inducing the differentiation of iPS cells to MSCs using MSCGM.doctoral thesi

大阪歯科大学Osaka Dental University博士(歯学)Glucose is important role for cellular functions. However, the behavior of osteoblasts in low glucose condition remains unclear. Therefore, this study aimed to elucidate the effects of low glucose conditions on the biological functions of osteoblasts. MC3T3-E1cells as one of osteoblast-like cells were cultured in MEM with five different glucose concentrations (0, 25, 50, 75, and 100 mg/dl). Then, low glucose conditions decreased the live cell number, proliferation, and migration of MC3T3-E1 cells. Alkaline phosphatase activity, mineralization, runx2, and osteocalcin production were also suppressed, whereas autophagy and extracellular signal-regulated kinase expression was increased under low glucose conditions. These results suggest that glucose may regulate osteoblast proliferation and differentiation.doctoral thesi

大阪歯科大学Osaka Dental University博士(歯学)Previous reports indicate that correcting the rounding of digitally abutment teeth through a ridgeline reconstruction procedure enhances the fit of CAD/CAM crowns. However, the impact of this procedure on the crown fit improvement for digital abutment teeth acquired with scanners of different specifications remains unclear. In this study, we investigated the effect of the ridgeline reconstruction procedure on the fit improvement of CAD/CAM crowns made from digital abutment teeth acquired with two types of 3D optical scanners. Preformed artificial abutment teeth for jacket crowns were scanned with two types of 3D optical scanners, and a ridgeline reconstruction procedure was applied to the marginal region of the digital abutment tooth. CAD/CAM crowns were fabricated from the acquired digital abutment teeth and placed on the abutment teeth using a silicone fit test material. The internal gap between the abutment teeth and crown at the occlusal surface center, buccal and lingual shoulders, and the area corresponding to the cement space at the inner slope of the occlusal surface were then calculated from the thickness of the silicone specimens. Statistical analysis revealed that the ridgeline reconstruction procedure improved the fit of CAD/CAM crowns fabricated with optical scanners, irrespective of the 3D optical scanner type used.doctoral thesi

大阪歯科大学Osaka Dental University博士(歯学)Peri-implantitis infections, which comprise inflammatory diseases occurring in tissues around dental implants, can lead to significant complications, such as implant instability and loss.This study aimed to elucidate the diversity of peri-implantitis microbiota and its association with patient metadata, focusing on clinical characteristics and health status. A total of 103 samples from 15 individuals, including patients with systemic diseases, such as diabetes and hypertension, were analyzed using high-throughput sequencing of the V3-V4 regions of the 16S rRNA-encoding gene.This study found significant diversity in the microbiota between peri-implantitis and natural teeth. Furthermore, the diversity varied with clinical indicators, such as bleeding on probing and plaque control, indicating the potential role of oral hygiene in managing peri-implantitis.The impact of systemic diseases, such as hypertension and diabetes, on microbiota diversity was also evaluated, revealing that these conditions might influence the oral microbial environment.These findings underscore the importance of considering individual patient profiles and systemic health conditions in peri-implantitis management and highlight the need for comprehensive treatment approaches.doctoral thesi
チタンインプラント上にEr:YAGレーザーデポジション法によって成膜されたハイドロキシアパタイト膜がM2型マクロファージの分極化と骨形成に与える影響
大阪歯科大学Osaka Dental University博士(歯学)In a previous study, we successfully coated hydroxyapatite (HAp) onto titanium (Ti) plates using the erbium-doped yttrium aluminum garnet pulsed-laser deposition (Er:YAG-PLD) method. In this study, we performed further experiments to validate the in vitro osteogenic properties, macrophage polarization, and in vivo osseointegration activity of HAp-coated Ti (HAp-Ti) plates and screws. Briefly, we coated a HAp film onto the surfaces of Ti plates and screws via Er:YAG-PLD. The surface morphological, elemental, and crystallographic analyses confirmed the successful surface coating. The macrophage polarization and osteogenic induction were evaluated in macrophages and rat bone marrow mesenchymal stem cells, and the in vivo osteogenic properties were studied. The results showed that needle-shaped nano-HAp promoted the early expression of osteogenic and immunogenic genes in the macrophages and induced excellent M2 polarization properties. The calcium deposition and osteocalcin production were significantly higher in the HAp-Ti than in the uncoated Ti. The implantation into rat femurs revealed that the HAp-coated materials had superior
osteoinductive and osseointegration activities compared with the Ti, as assessed by microcomputed tomography and histology. Thus, HAp film on sandblasted Ti plates and screws via Er:YAG-PLD enhances hard-tissue differentiation, macrophage polarization, and new bone formation in tissues surrounding implants both in vitro and in vivo.doctoral thesi
