    Assessment of Nasal Structure Using CT Imaging of Brachycephalic Dog Breeds

    The response to treatment of brachycephalic airway syndrome (BAS) varies among brachycephalic dog breeds. We hypothesized that variations in nasal structure are one of the factors responsible for this difference. To confirm this variation, we measured the ratio of the airway cross-sectional area to the total nasal cavity area (AA/NC) in three brachycephalic dog breeds. Head CT images of French bulldogs, shih tzus, and pugs were retrospectively collected. Four specific transverse planes were used to calculate AA/NC ratios. Fifty brachycephalic dogs were included in the study: French bulldogs (n = 20), shih tzus (n = 20), and pugs (n = 10). The AA/NC ratio of Shih Tzus was larger in the rostral nasal cavity and smaller toward the caudal area, whereas the other two breeds showed an inverse tendency. The results obtained from the current research indicate that the AA/NC ratio can be used to evaluate the structure of the nasal cavity. Moreover, analyzing the point with the smallest AA/NC ratio can be useful in quantifying nasal airway obstruction and the severity of BAS. These results will be useful in understanding the complexity of BAS pathophysiology and in the implementation of treatment.journal articl

    Development of Online Science Teaching Material for Decision Making and Consensus Building on “Pet Regenerative Medicine”

    オンライン形式での意思決定や合意形成を促すための試みとして,ペットの再生医療についてのオンライン理科教材を開発した.参加者(生徒・学生)の意思決定や合意形成を促すため,コンセンサス会議の手法を大幅に簡略化しつつ取り入れた.開発教材は,27枚の指導用スライド(パワーポイントファイル)と,生徒用ワークシート(ワードファイル)から構成される.再生医療について4つの技術(ES細胞・iPS細胞・遺伝子治療・ゲノム編集)の観点から順に基礎知識を学んだうえで,ヒトとペットの両者に対する日本での4つの技術の推進/規制の是非を考え,グループに分かれて話し合い活動を行うものである.中高理科の免許取得をめざす教職課程学生を対象としたオンライン試行の結果,概ね良好な反応が得られた.一方で,具体的な改善点も複数把握され,実際の中学生を想定すると,さらなる改善の必要がある.特に,オンライン授業で生徒主体の話し合い活動をどのように促進するのかについては,さらなる工夫や検討が必要である.journal articl

    Large granular lymphocyte lymphoma in the skin and urinary bladder of a dog

    A 10-year-old female Cavalier King Charles Spaniel presented with hematuria, pollakiuria and skin rash. Based on the histopathological and cytological examination of the skin and bladder mucosa, the dog was diagnosed with large granular lymphocytic (LGL) lymphoma of the bladder and skin. The dog responded well to the initial chemotherapy with nimustine for 3 months. Since recurrence of skin erosion and bladder wall thickening were observed, the dog was subsequently administered chemotherapy with other anticancer drugs, including chlorambucil, vincristine, doxorubicin, L-asparaginase, cytosine arabinoside, and cyclophosphamide. The dog survived for 11 months and died due to tumor-related disseminated intravascular coagulation. This is the first report of a canine case of LGL lymphoma in the skin and bladder.journal articl

    The matrilineal transmission of maternal behavior has been reported in several species. Studies, primarily on rats, have suggested the importance of postnatal experience and the involvement of epigenetic mechanisms in mediating these transmissions. This study aims to determine whether the matrilineal transmission of maternal behavior occurs in mice and whether the microbiota is involved. We first observed that early weaned (EW) female mice showed lower levels of maternal behavior, particularly licking/grooming (LG) of their own pups, than normally weaned (NW) female mice. This difference in maternal behavioral traits was also observed in the second generation, even though all mice were weaned normally. In the subsequent cross-fostering experiment, the levels of LG were influenced by the nurturing mother but not the biological mother. Finally, we transplanted the fecal microbiota from EW or NW mice into germ-free (GF) mice raising pups. The maternal behaviors that the pups exhibited toward their own offspring after growth were analyzed, and the levels of LG in GF mice colonized with microbiota from EW mice were lower than those in GF mice colonized with microbiota from NW mice. These results clearly indicate that, among maternal behavioral traits, LG is intergenerationally transmitted in mice and suggest that the vertical transmission of microbiota is involved in this process. This study demonstrates the universality of the intergenerational transmission of maternal behavioral traits and provides new insights into the role of microbiota.journal articl

    Preparation of Monoclonal Antibodies Specifically Reacting with the Trichothecene Mycotoxins Nivalenol and 15-Acetylnivalenol via the Introduction of a Linker Molecule into Its C-15 Position

    Nivalenol (NIV) is a trichothecene mycotoxin that is more toxic than deoxynivalenol. It accumulates in grains due to infection with Fusarium species, which are the causative agents of scab or Fusarium head blight. An immunoassay, which is a rapid and easy analytical method, is necessary for monitoring NIV in grains. However, a specific antibody against NIV has not been prepared previously. To establish an immunoassay, we prepared NIV, introduced a linker, and generated antibodies against it. NIV was prepared from a culture of Fusarium kyushuense obtained from pressed barley through chromatographic procedures with synthetic adsorbents and silica gel. NIV was reacted with glutaric anhydride, and the reaction was stopped before mono-hemiglutaryl-NIV was changed to di-hemiglutaryl-NIV. 15-O-Hemiglutaryl-NIV was isolated via preparative HPLC and bound to keyhole limpet hemocyanin (KLH) using the active ester method. Two different monoclonal antibodies were prepared by immunizing mice with the NIV-KLH conjugate. The 50% inhibitory concentration values were 36 and 37 ng/mL. These antibodies also showed high reactivity in a direct competitive enzyme-linked immunosorbent assay and specifically reacted with NIV and 15-acetyl-NIV but not with deoxynivalenol and 4-acetyl-NIV.journal articl

    Force plate analysis of ground reaction forces in relation to gait velocity of healthy Beagles

    OBJECTIVE To evaluate changes in ground reaction forces (GRFs) in relation to gait velocity using 2 force plates (FPs) for healthy Beagles. ANIMALS 18 healthy Beagles were included (body weight, 10.45 ± 1.28 kg; age, 26 ± 11 months). PROCEDURES Ten GRF parameters were measured at three gait velocities (walk, 0.9 to 1.2 m/s; trot 1, 1.6 to 2.0 m/s; and trot 2, 2.1 to 2.5 m/s): peak lateral force (PLF), peak medial force (PMF), lateral impulse (LI), medial impulse (MI), peak propulsive force (PPF), peak braking force (PBF), propulsive impulse (PI), braking impulse (BI), peak vertical force (PVF), and vertical impulse (VI). RESULTS As velocity increased, the PVF of all limbs increased, the VI of all limbs decreased, and the PPF of the forelimbs increased. At all velocities, PBF and BI were significantly higher than the PPF and PI in forelimbs; however, PBF and BI were significantly lower than the PPF and PI in hindlimbs. There were no significant differences in the PLF, PMF, LI, and MI of the forelimbs and hindlimbs among all velocities. The PLF was significantly higher than the PMF of forelimbs during trot 1 and trot 2. CLINICAL RELEVANCE These results may be useful when comparing healthy Beagles with diseased ones when premorbid data are not available. Because the forelimbs are mainly responsible for the braking force, it is suggested that weight bearing is more stable in the forelimbs than in the hindlimbs, which are mainly responsible for the propulsive force, and that a greater force is generated laterally than medially during trot.journal articl

    Biological features of canine cancer-associated fibroblasts and their influence on cancer cell invasion

    Cancer-associated fibroblasts (CAFs) play an essential role in tumor invasion and metastasis. In dogs, the biological features of CAFs have not been well characterized. The purpose of this study was to investigate differences in the biological activities of canine CAFs and normal fibroblasts (NFs), and their influence on the migration and invasion of cancer cells. Canine CAFs and NFs were harvested from surgically-resected malignant epithelial tumor tissues and skin tissues of dogs. A wound-healing assay was conducted to compare the migratory and invasive abilities of canine CAFs and NFs. The results of this study showed that canine CAFs have a greater migratory and invasive ability than NFs. To observe the indirect and direct interactions between fibroblasts and cancer cells, Boyden chamber assay and 3D co-culture with collagen gel were conducted. The number of migrated and infiltrated cancer cells co-cultured with canine CAFs was greater than that with NFs. In the 3D co-culture, cancer cells showed noteworthy proliferation on the surface of gels containing canine CAFs and invasion into the gel. On the other hand, no infiltration of cancer cells into the gel containing NFs was observed. It was suggested that canine CAFs activate migration and invasion of cancer cells and promote the infiltration of cancer cells into collagen gels.journal articl

    History and development of staining methods for skeletal muscle fiber types

    The contractile and metabolic properties of skeletal muscles depend on the composition of muscle fibers. There are two major fiber types: type 1 and type 2. Type 2 fibers are further subdivided into type 2A, 2X, and 2B fibers. Muscle fiber type composition is an important property that affects sports performance and metabolic ability in humans, and meat quality in domestic animals. In this review, we summarize the history of muscle fiber type classification based on various staining methods for skeletal muscle sections. The history illustrates the development of an experimental method to detect myosin heavy chain (MyHC) proteins, which are the most common marker molecules for muscle fiber type. Metabolic enzymes, such as nicotinamide adenine dinucleotide-tetrazolium reductase and succinate dehydrogenase are also described for histochemical staining combined with myosin ATPase staining. We found an improvement in the quality of antibodies used for immunostaining of MyHC, from polyclonal antibodies to monoclonal antibodies (mAbs) and then to mAbs produced by synthetic peptides as antigens. We believe that the information presented herein will assist researchers in selecting optimal staining methods, dependent on the experimental conditions and purposes.journal articl

    Sphingomyelin maintains the cutaneous barrier via regulation of the STAT3 pathway

    Epidermal tissues play vital roles in maintaining homeostasis and preventing the dysregulation of the cutaneous barrier. Sphingomyelin (SM), a sphingolipid synthesized by sphingomyelin synthase (SMS) 1 and 2, is involved in signal transduction via modulation of lipid-raft functions. Though the implications of SMS on inflammatory diseases have been reported, its role in dermatitis has not been clarified. In this study, we investigated the role of SM in the cutaneous barrier using a dermatitis model established by employing Sgms1 and 2 deficient mice. SM deficiency impaired the cutaneous inflammation and upregulated signal transducer and activator of transcription 3 (STAT3) phosphorylation in epithelial tissues. Furthermore, using mouse embryonic fibroblast cells, the sensitivity of STAT3 to Interleukin-6 stimulation was increased in Sgms-deficient cells. Using tofacitinib, a clinical JAK inhibitor, the study showed that SM deficiency might participate in STAT3 phosphorylation via JAK activation. Overall, these results demonstrate that SM is essential for maintaining the cutaneous barrier via the STAT3 pathway, suggesting SM could be a potential therapeutic target for dermatitis treatment.journal articl
