Purification of melatonin from herbal sources and evaluation in pharmaceutical form
Saflaştırılan MEL'i farmasötik formda değerlendirmek için yumuşak tablet formu seçilmiş, karakterizasyon analizleri yapılmıştır. Özetle, bitki kaynaklı melatonin (fito-melatonin) yumuşak tablet formunda tasarlanarak başarıyla tamamlanmıştır.Melatonin (MEL) has attracted increasing scientific interest worldwide in recent years. MEL, also known as N-acetyl-5-methoxy tryptamine (C13H16N2O2), is a natural hormone that can be synthesized primarily by the pineal gland of vertebrates and secondarily by other tissues and organs. After the discovery that MEL, previously known only as an animal hormone, also exists in plants, numerous studies have been conducted on this molecule and it has been revealed that it has similar functions in plants as in animals. Research on bioactive compounds found naturally in edible plants in particular is very important in terms of nutrition and health and more studies are needed. In this context, MEL extraction was performed with solvents selected according to the literature from dried forms of Linden (Tilia tomentosa Moench), Chamomile (Matricaria recutita L.), Melissa (Melissa officinalis L.), Stevia (Stevia rebaudiana Bertoni), Sage (Salvia officinalis L.) and St. John's Wort (Hypericum perforatum L.) plants, MEL amounts in the extracts were calculated by HPLC-UV. In the Stevia rebaudiana Bertoni) plant with the highest yield, melatonin was purified and characterized by LC-MS. To evaluate the purified MEL in pharmaceutical form, a soft tablet form was selected, and characterization analyses were conducted. Forecasting the daily solar energy-based electricity generation of Düzce University campus a day in advance using deep learning methods
This study aims to forecast daily solar energy and electricity generation in the Düzce University campus area one day in advance using deep learning techniques. The rationale behind day-ahead forecasting lies in enabling proactive measures against potential issues such as overproduction or underproduction. Reliable production estimates for the following day allow for effective planning between energy generation and consumption. Particularly for large-scale consumption areas such as university campuses, this contributes significantly to optimizing energy use in balance with production. As a result, unnecessary energy consumption can be prevented, overproduction can be controlled, and economic efficiency can be improved. The study also aims to establish an infrastructure to support energy management systems. In this research, deep learning was employed to mimic the neuron-based information processing structure of the human brain for forecasting energy production from solar radiation. To develop an effective model for solar radiation prediction, the advantages of the Bidirectional Long Short-Term Memory (BiLSTM) method were utilized. The developed model aimed to forecast the hourly energy production of the next day based on past hourly energy data, thereby enabling a day-ahead prediction capability. The BiLSTM model was implemented through software tools, allowing for day-ahead electricity production forecasts specific to the region. In the model development process, a solar energy simulation software was first used to determine the energy production potential of the region. The obtained solar and temperature data were seasonally categorized and used as core inputs. These datasets were employed for training the model within a software environment. To evaluate the forecasting performance of the model, metrics such as R², MAE, MSE, and RMSE were applied. Additionally, the model's accuracy was compared with the Curve Fitting Toolbox method available in the software, and the reliability of the results was comprehensively analyzed through testing. The findings demonstrated that the BiLSTM model consistently outperformed the Curve Fitting Toolbox in all datasets, delivering higher R² values and lower error rates
Investigation of tribological properties of industrial products with different patterns produced by 3D printing using polylactic acid
PurposeWith the development of technology, the production of industrial products with 3D (3-Dimensional) printing has become widespread. Variables in production methods affect the durability of products. For this reason, studies to increase the durability of products are gaining importance. This paper aims to examine the tribological behavior of products produced in different patterns using 3D printing methods.Design/methodology/approachFriction wear tests of polylactic acid (PLA) samples produced in different patterns were carried out on a pin-on disc testing device. Friction wear tests were performed according to ASTM G-99 standard. Friction coefficient, weight loss and wear surface images were evaluated.FindingsAccording to the results obtained, it was determined that pattern differences affected the tribological properties of the samples. The lowest coefficient of friction was observed in PLA1, while the highest wear resistance was found in PLA3.Originality/valueStudies on the tribological properties of samples produced with a 3D printer are limited. In this study, the effect of samples with a 50% filling rate and different patterns on tribological properties was investigated. It was desired to determine the effect of different patterns on wear characteristics
Radiofrequency Induced Time-Dependent Alterations in Gene Expression and Apoptosis in Glioblastoma Cell Line
The widespread use of wireless communication technologies has increased human exposure to radiofrequency electromagnetic fields (RF-EMFs). Considering the brain's close proximity to mobile phones and its entirely electrical transmission network, it emerges as the organ most profoundly impacted by the RF field. This study aims to investigate the potential effects of RF radiation on cell viability, apoptosis, and gene expressions in glioblastoma cells (U118-MG) at different exposure times (1, 24, and 48 h). To achieve this, we designed and implemented an in vitro RF exposure system operating at a frequency of 2.1 GHz, specifically for cell culture studies, with an average specific absorption rate (SAR) of 1.12 +/- 0.18 W/kg determined through numerical dosimetry calculations. Results reveal a significant influence of a 48 h exposure to a 2.1 GHz RF field on U118-MG cell viability, gene expression, and the induction of caspase (CASP) dependent apoptosis. Notably, increased CASP3, CASP8, and CASP9 mRNA levels were observed after 24 and 48 h of RF treatment. However, only the 48 h RF exposure resulted in apoptotic cell death and a significant elevation in the BAX/BCL-2 ratio. This observed effect may be influenced by extended exposure durations surpassing the cell's doubling time. The increased BAX/BCL-2 ratio, which acts as a key switch for apoptosis, and the heterogeneous morphology of the astrocyte-derived U118-MG cell line may also play a role in this effect. The authors received no specific funding for this work. The authors declared that no funding is associated with the study described in this article. The authors would like to thank Melek Yaman and Zubeyir Elmazoglu for their assistance during the experiment
ON SOME INEQUALITIES RELATED TO OPEN NEWTON-COTES FORMULAS FOR DIFFERENTIABLE CONVEX FUNCTIONS WITH APPLICATIONS
In this paper, first, we prove a novel integral identity involving a single time-differentiable function. Then, we prove some new inequalities associated with one of the open Newton-Cotes formulas for differentiable convex functions. The newly established inequalities can be helpful in finding the error bounds of one of the open Newton-Cotes formulas. Finally, some applications of the inequalities are also presented in the context of open Newton-Cotes formulas.National Natural Science Foundation of China [11971241]This work is partially supported by the National Natural Science Foundation of China Grant No. 11971241
American Medical Missionary Activities in Basra in the Late Ottoman Period: Lansing Memorial Hospital
One of the most important cities of Iraq on the eastern border of the Ottoman Empire was the province of Basra, where international trade was active and which could also be considered as a gateway to India. Basra was also of high importance for the United States of America because it was the trade centre of the East and the gateway to the South Asian market, and because of the presence of oil in the neighbouring province of Khuzestan. In the vacuum created by the Ottoman Empire's loss of power in the region in the 19(th) century, America, which wanted to create an area of dominance for itself, carried out activities here. Accordingly, the Arabian Mission, founded by American Protestant missionaries at the end of the 19(th) century, established a station in Basra, which was identified as its first target for education and health activities. In 1911, it opened a hospital in Basra under the name of Lansing Memorial, and Protestant doctors and nurses affiliated with the Arabian Mission started to serve there. This study focuses on the activities of the Arabian Mission in Basra starting from the end of the 19(th) century. The primary sources of this research are the documents of the Ottoman Archives and the publications of the Arabian Mission, while literature on the Arabian Mission has also been used
Analysis of Latife Tekin's novels in the context of Kristeva's Semiotic Chora and Derrida's Différance concepts
Bu çalışmada, Latife Tekin'in romanlarındaki dil; Julia Kristeva'nın semiyotik khora, Jacques Derrida'nın différance kavramları bağlamında analiz edilmiştir. Latife Tekin'in Sevgili Arsız Ölüm'den 2022' de yazdığı son romanı Zamansız'a kadar, dil daima merkezde olmuştur. Edebiyatı, "dili kullanarak dilin dışına çıkmak" olarak tarif eden Tekin'in romanlarında dil; dil-öncesi, dişil, arkaik, sessel, ritmik, şiirsel, metaforik, psişik, özellikleriyle anlamın sınırlarını zorlar. Latife Tekin's novels present a multi-layered literary structure where semiotic and symbolic orders intertwine, pushing the boundaries of language. Latife Tekin's novels present a multi-layered literary structure in which semiotic and symbolic orders are intertwined and the limits of language are pushed. In this study, the language in Latife Tekin's novels is analyzed through close reading in the context of Julia Kristeva's semiotic chora and Jacques Derrida's différance. Language has always been at the center of Latife Tekin's novels, from her first novel, Dear Shameless Death to Timeless, written in 2022. In Tekin's novels, which she defines literature as "going beyond language by using language" , language pushes the boundaries of meaning with its pre-linguistic, feminine, archaic, rhythmic, poetic, metaphorical, psychic features. In Tekin's works, semiotic elements express the unconscious, intuitive and bodily-rhythmic experiences of the individual, while the symbolic order points to the normative dimension of language. In this context, both Kristeva's semiotic chora approach and Derrida's concept of différance point to the constantly transforming, non-fixed nature of language, the ambiguity of meaning and the postponed and variable nature of language in the production of meaning. In this thesis, a multi-layered literary analysis of Latife Tekin's novels in the light of Kristevian semiotic khora and Derrida's différance is presented. These eleven novels were analyzed in the context of semiotic, symbolic, thetic, genotex and phenotex, which are elements of semiotic chora, and deferral and différence, which are elements of différance. It has been observed that the common denominator of these novels, which correspond to different phases of Latife Tekin's novelism, analyzed within the scope of this thesis is the semiotic chora and différance features of the novels
The ecological effects of recreational activities on soil and vegetation cover in Yedigöller National Park
With the increase in population and urbanization, as well as the development of technology, people's demand for outdoor recreation activities is increasing. For this reason, national parks and other protected areas, which have rich natural and cultural resources and offer recreational opportunities to users, are visited by many people. However, these visits have a negative environmental impact and may cause deterioration in the resource values of natural areas. Scientists call studies that examine, evaluate and observe visitor impacts in natural areas recreation ecology studies. Such information is critical to identifying and describing impacts, easily understanding the consequences of impacts, and mitigating, restoring, and managing the impact. The factors that determine recreational impacts are the type of use, amount, time, and characteristics of the area. The aim of this study is to determine the effects on soil structure depending on recreational activity types in Yedigöller National Park, which has sensitive ecosystems, and to ensure sustainable use of the area
The Effects of Individual Components of E-Cigarettes on Ion Transport and Airway Surface Liquid Height in Human Bronchial Epithelial Cells
Background and Objectives: The rising popularity of new-generation electronic cigarettes (e-cig) like JUUL necessitates a better understanding of their impact on respiratory and other body systems, as the effects of JUUL's components remain unclear. This study aimed to investigate the effects of JUUL components on ion channels and airway surface liquid (ASL) height in human bronchial epithelial cells (HBECs). Furthermore, the cytotoxic effects of these components were investigated in human embryonic kidney 293T (HEK293T) cells. Materials and Methods: The components tested included nicotine salt (NicSalt), benzoic acid (BA), sodium hydrogen tartrate (NaTar), propylene glycol/vegetable glycerin (PG/VG), freebase nicotine (FBNic) and nicotine salt+benzoic acid (NicSalt+BA). Each component was prepared at 100 mu M, and HBECs were exposed for 24 h to measure ASL height, short-circuit current (Isc), and transepithelial electrical resistance (TEER). Results: Initial exposure (0 h) to these substances did not significantly alter ASL height. However, after 2 h, FBNic-treated HBECs exhibited a significant reduction in ASL height compared to NicSalt and other tested substances, with the most pronounced decrease observed at the 6th hour. This effect persisted over prolonged exposure, suggesting a cumulative impact on airway hydration and epithelial function. Additionally, adenosine administration did not induce a significant increase in ASL height. NicSalt, BA, and FBNic were found to disrupt ion balance in HBECs, affecting ion channels and ASL homeostasis while significantly decreasing TEER. In terms of cytotoxicity, NicSalt, and benzoic acid demonstrated minimal cytotoxicity at low concentrations, whereas FBNic showed significantly higher cytotoxicity at moderate levels. Conclusions: In conclusion, this study highlights that e-cigarette components can disrupt airway surface liquid homeostasis by affecting ion channel activity, compromise epithelial barrier integrity by reducing transepithelial electrical resistance, and emphasize the importance of their cytotoxic effects.NIH/FDA [HL120100]NIH/NHLBI [HL135642]NIH [5-U54-CA156733-10]Scientific and Technological Research Council of Turkey (TUBITAK)The 2214/A Doctoral Research Fellowship ProgrammeScientific and Technological Research Council of Turkey (BIdot;DEBThis work was funded by NIH/FDA (HL120100), NIH/NHLBI (HL135642), and NIH (5-U54-CA156733-10).
