Development of a Survey Instrument for Measuring L2 Motivational Change Over Time: A Preliminary Rasch Analysis
Although learner motivation has been a dominant theme in English education, few studies have investigated how learner motivation fluctuates over time regarding their L2 motivational self-system. Moreover, the factors that distinguish the motivation to learn English amid demotivated, amotivated, and motivated learners warrant further investigation. In the current study, a questionnaire was developed, and Rasch analysis was conducted to determine the reliability and validity of the questionnaire and to collect interval data to be utilized in subsequent analyses. Thirty-six Japanese university learners were requested to complete an online questionnaire. The findings demonstrated that a vast number of items needed to be removed because of repetition and low validity. Several items were added to the questionnaire to examine positive emotion, relationship, ought-to L2 self, deficient ability belief, and why learners regained their motiva tion to study English. In addition to the process by which the initial questionnaire was developed, analyzed and validated, how the revised questionnaire can be used in future research will be discussed.departmental bulletin pape
Exploring ways to promote comprehensive learning ―Considering the current situation and challenges in setting research topics―
The purpose of this study is to clarify the actual situation in schools regarding how inquiry tasks are set in comprehensive learning and how teachers address problems related to setting inquiry tasks. In the first stage of the research, through organizing and analyzing previous research and practices related to comprehensive learning, we will examine methods for setting inquiry tasks for comprehensive learning in schools and ways to continue and develop inquiry learning. In the next stage of the research, we investigate the setting of inquiry tasks in comprehensive learning for public elementary school teachers. Through the analysis of the survey results, we aim to clarify the problems that hinder child-centered inquiry learning in comprehensive learning at school, and specific measures to address them. An Empirical Study on Verbal Communication Techniques Used by Early Childhood Educators with Young Children
This study aims to formalize the “verbal communication techniques” used by experienced early childhood educators. The results of a questionnaire survey revealed three key findings: First, educators combine various verbal techniques based on children’s “desired behaviors” and “behaviors requiring attention,” particularly in the case of three-year-olds. Second, educators tend to repeatedly use the same verbal techniques. Third, educators frequently use sentence-ending particles when addressing children. These findings suggest that the differences in verbal techniques between experienced educators and students of early childhood education largely stem from differences in the “interpretation and meaning-making of the caregiving context.”departmental bulletin pape

近年、検察の杜撰な捜査が社会的問題となっている。問題の背景として、法務・検察の組織体制、とりわけ人事の在り方が指摘されている。そこで、本稿は「法務・検察幹部の出世コースはどのようになっているのか」という「現状確認型」リサーチクエスチョンを立てる。まずは実態の把握が必要であると考えたからである。本稿の目的は法務・検察幹部の出世コースに関する基礎データを提供することである。本稿は、法務・検察幹部の出世コースを分析するための基礎作業として、次のようなデータセットを作成する。①同一職位の歴代人物(縦)の長期時系列データと同一人物のキャリア・パス(横)の長期時系列データを組み合わせたデータセットを作成する。②分析期間は1970年~ 2024年とする。1970年以前は法務・検察の幹部人事に公職追放が強く影響しており、その影響を排除するため、1970年以降と設定する。③分析対象は、検事総長・次長検事・高検検事長(高い序列の認証官ポスト)、法務事務次官・刑事局長・官房長(法務省内の幹部ポスト)、「現場派」の出世コースとして重要な東京地検検事正、「関西検察」における出世コースとして重要な大阪地検検事正である。④キャリア・パスの職位レベルは、課長・地検レベルから事務次官・検事総長というように、下位レベルから上位レベルまでを射程に入れる。departmental bulletin pape

Development of data repositories for glycan-related resources
創価大学博士(工学)The importance of large-scale omics analyses has been growing more and more to understand the precise interactions among biological molecules. To perform omics-wide analyses, sharing experimental data from researchers is extremely valuable. Therefore, many public data repositories have been developed in the life sciences, including PRIDE and PeptideAtlas in proteomics and GlyTouCan, GlycoPOST and UniCarb-DR in glycomics, to promote sharing and accumulating data from researchers under the FAIR (Findability, Accessibility, Interoperability, and Re-usability) data principles. However, one of the missing pieces is the accumulation of glycoconjugate data, including glycopeptides, glycoproteins, glycolipids, and glycosides. GlyTouCan, for example, assigns unique identifiers for glycan structures, including monosaccharide compositions. To assign such unique identifiers to glycoconjugates, we have developed a novel data repository called GlyComb. Researchers can currently register glycopeptides and glycoproteins by specifying an amino acid sequence or UniProt ID, glycosylation sites, and glycan information to GlyComb in TSV format. Because GlyComb is built on top of Semantic Web technologies, users will be able to collect the specified glycoconjugate-associated information easily using the SPARQL query language. In addition, existing glycomics data repositories required further collaboration enhancements in order to optimize user convenience. In particular, UniCarb-DR and GlycoPOST are both similar repositories for glycomics experimental data, but they were developed and maintained independently. Therefore, in this study, in addition to developing
a novel data repository for glycoconjugate data, and I have enhanced the integration of UniCarb-DR and GlycoPOST for integrating the results of glycomics mass spectrometry experiments. As a result, these developments enable glycoscientists to more easily submit glycoconjugate data and mass spectrometry data in the glycosciences into the worldwide informatics infrastructure.doctoral thesi
Analyses of signaling molecules in the axotomized rat facial nucleus
創価大学博士(工学)Axotomy of the rat facial nerve causes downregulation of motoneuron-specific molecules, including choline acetyltransferase, the vesicular acetylcholine transporter and GABAAR1, in surviving motoneurons. Subsequently, resident microglia are activated and proliferate. However, it is still unclear which signaling molecules are involved in these responses. In this study, I investigated the changes and localizations of immediate early genes (IEGs), transcription factors (CREB/ATF family) and mitogen-activated protein kinases (MAPKs). Immunoblotting and immunohistochemical analyses revealed the following. Among the IEGs, c-Jun was increased in injured motoneurons, but c-Fos did not. Among the CREB/ATF family members, p-CREB was significantly decreased in injured motoneurons. The levels of p-CREB/CREB and p-ATF2/ATF2 were immunohistochemically increased in microglia. Among MAPKs, p-ERK1/2 and p-JNK1 were decreased in injured motoneurons. p-p38 and p38 were markedly increased in microglia. In vitro experiments revealed that p38, CREB, and ATF2 were activated in proliferating microglia. These results strongly suggested that c-Jun is involved in the survival and repair of motoneurons, but p-CREB/CREB, p-ERK/ERK and p-JNK/JNK are associated with the downregulation of motoneuron-specific molecules. On the other hand, p-p38/p38, p-CREB/CREB, and p-ATF2/ATF2 were suggested to be closely involved in the activation/proliferation of microglia. Furthermore, I investigated whether the level of microglial proliferation is dependent on the degree of motoneuronal insult. By administrating glial cell line-derived neurotrophic factor, N-acetyl cysteine or salubrinal at the transection site, I found that these reagents ameliorated the increase in c-Jun and the reductions in levels of p-CREB and functional molecules in the injured motoneurons. In tandem with these changes, the levels of a microglial marker. i.e., Iba1, and the levels of cFms, proliferating cell nuclear antigen (PCNA), and p-p38/p38 were significantly downregulated in microglia. These results demonstrated that the recovery of motoneuron resulted in the reduction of microglial proliferation. I thus conclude that the degree of neuronal injury regulates the levels of microglial proliferation in adult rat facial nucleus.doctoral thesi
