Main uterine pathologies in adult female pet rabbit
Le patologie dell’apparato riproduttore, specialmente quelle a carico dell’utero, sono i disturbi più comuni nella coniglia domestica (Oryctolagus cuniculus) adulta non sterilizzata. I problemi del tratto genitale spesso non manifestano una sintomatologia evidente o caratteristica e per questo motivo possono risultare come reperti occasionali in corso di sterilizzazione oppure rendersi evidenti solo quando la patologia è in uno stadio avanzato. Per formulare una diagnosi è essenziale praticare un’attenta visita clinica e avvalersi di indagini strumentali quali radiografie ed esame ecografico, seguiti da esame istologico del tessuto uterino. Risulta di fondamentale importanza la prevenzione mediante ovarioisterectomia precoce. Le due patologie uterine più frequenti sono senza dubbio l’iperplasia endometriale e l’adenocarcinoma uterino. La possibile correlazione tra le due condizioni è ancora dibattuta, ma ad oggi si tende a considerare l’iperplasia come uno stadio precanceroso. Nella genesi di queste condizioni patologiche, oltre all’età potrebbero essere coinvolti altri fattori quali la predisposizione genetica e l’assunzione con la dieta di interferenti endocrini ad azione estrogenica.The pathologies of the reproductive system, especially those affecting the uterus, are the most common disorders in adult intact domestic rabbits (Oryctolagus cuniculus). The genital tract disorders may not manifest clear or characteristic symptoms, and, for this reason, they may be incidental findings during neutering or become apparent only when the pathology is in an advanced stage. Formulating a diagnosis requires a careful clinical examination and the use of diagnostic imaging as radiography and ultrasound, followed by histological examination of the uterine tissue. Prevention through early ovariohysterectomy is of paramount importance. The two most frequent uterine pathologies are undoubtedly endometrial hyperplasia and uterine adenocarcinoma. The possible correlation between these two conditions is still debated, but currently endometrial hyperplasia tends to be considered a precancerous stage. In the genesis of these pathological conditions, besides age, other factors such as genetic predisposition and dietary intake of endocrine disruptors with estrogenic action may be involved
Pompei, capsula del tempo dell'Impero Romano: analisi paleogenomica dei resti umani rinvenuti nell'antica città
La peculiare storia che ha vissuto l’antica città di Pompei ha fatto sì che essa abbia oggi un’importanza senza eguali in tutto il mondo. Per questo motivo il Parco Archeologico di Pompei è stato incluso all’interno dei patrimoni UNESCO. Gli scavi, già a partire dal XVIII secolo, hanno portato alla luce non solo ben preservati edifici pubblici e privati, ma anche numerosi resti umani riferibili alle vittime dell’eruzione del complesso Somma-Vesuvio avvenuta nel 79 e.c. Lo studio antropologico dei resti delle vittime è stato accompagnato, fin dalla metà degli anni ’90, dall’analisi del loro DNA limitato però alle tecniche in uso in quel periodo. Grazie alla convenzione firmata con il Laboratorio di Ricerche Applicate di Pompei, nel laboratorio di Antropologia Molecolare e Paleogenomica dell’Università degli Studi di Firenze sono stati campionati per questo progetto di ricerca più di 170 reperti, in maggioranza ossa e denti, allo scopo di analizzare il genoma degli individui che vivevano o si trovavano in quel preciso periodo a Pompei. Grazie all’applicazione di protocolli specifici per il recupero di molecole di DNA degradate e all’impiego delle più sofisticate tecnologie di sequenziamento NGS (Next Generation Sequencing), è stata rilevata un’ottima preservazione del DNA endogeno che, in alcuni casi, supera anche il 90%. Con un approccio multidisciplinare è stato studiato un ristretto gruppo di individui, alcuni dei quali legati da una relazione di parentela, che hanno perso la vita all’interno della stanza di una domus Pompeiana a causa del crollo del tetto e dell’inalazione dei fumi vulcanici. Per la prima volta è stata studiata la variabilità genomica di un esteso campione della popolazione a Pompei ricostruendo il genoma di 12 individui con una profondità media di sequenziamento 1x. La ricostruzione di questi genomi a basso coverage ha spinto verso l’applicazione di metodi di analisi che considerano l’incertezza associata ai genotipi piuttosto che la loro chiamata diretta. Dall’analisi delle componenti principali (PCA) e dell’Admixture emerge in città la presenza di una grande variabilità genomica se si confronta con quella delle altre popolazioni già studiate sia di epoche precedenti nello stesso territorio, sia coeve in più punti del continente Europeo. Infatti, a Pompei si ritrova un insieme di varianti genetiche identificate in precedenza non solo nella penisola Italiana, ma anche al di fuori di essa. I risultati delle analisi genomiche trovano ampio supporto dalle testimonianze storiche che vedono la crescita di Pompei successivamente al trasferimento della nobiltà dalla capitale dell’impero, anche per fini militari, e dai ritrovamenti archeologici all’interno della città indicandola come una delle più attive città commerciali e portuali del bacino del Mediterraneo.The ancient city of Pompeii has a worldwide interest for its unique history, so that it is included in the UNESCO World Heritage. It represents a time capsule of Roman antiquity because buildings, monuments, sculptures, paintings and mosaics were preserved and remained intact, as well as food and personal objects related with daily life. Many human remains, belonging to people who died during the eruption of Vesuvius in 79 CE, have been unearthed thanks to the intense excavations of the archaeological site. Bone microstructures seem to be well preserved under 5-6 meters of pyroclastic materials, which also slowed down the DNA degradation process due to oxidative decay. Indeed, a first shallow shotgun sequencing of more than 170 human remains (mostly bones and in some cases even teeth) demonstrated a good preservation of ancient DNA (aDNA), exceeding even 90% in a few cases. This is the first time that a great number of individuals who lived or were in Pompeii for other reasons at the time of the eruption are studied from a population genomic perspective.
Using a multidisciplinary approach, we studied human remains found in the same archaeological context, the so-called “Stanza degli Scheletri”. We were able to identify the minimum number of individuals who found shelter inside the room and a first-degree relationship between two of them. In addition, we observed high genomic variability even when studying a small sample of the population.
Moreover, twelve genomic libraries with a good percentage of endogenous DNA preservation (>20%) were selected to perform a deep shotgun sequencing with the aim to obtain an average coverage of 1x. We used a whole-genome approach to analyse the genomic variability of individuals found in Pompei and to make comparisons between them and other ancient Eurasian populations published so far. The Principal Component Analysis (PCA) and the Admixture demonstrate the presence of an heterogeneous populations. In Pompeii we found genetic variants already observed in pre-Roman Italic populations and in coeval individuals from different parts of Europe. These results are supported by historical documents and archaeological findings that show Pompeii as one of the most active commercial and port cities of the Mediterranean
Phylogeny, ontogeny and anatomo-physiology of the human social brain
La vita quotidiana dell’uomo del XXI secolo ruota attorno al lavoro, alle scuole, agli ospedali, al supermercato, al cinema, allo sport. Il presente elaborato si sofferma ad analizzare la peculiarità umana che ha permesso tali invenzioni: la socialità.
Si esordirà con un’indagine di carattere filogenetico, compresa una disamina delle dinamiche sociali e comunicative dei nostri parenti più prossimi, al fine di ricostruire le tappe evolutive che hanno condotto l’homo sapiens verso ciò che Edward O. Wilson ha definito “eusocialità”.
Tale ricostruzione culminerà con l’analisi dei modi e dei motivi per cui il cervello umano si è evoluto nel corso della sua storia, fino ad arrivare all’organo oggi accuratamente studiato dalle neuroscienze. Si procederà poi ad un vaglio della letteratura scientifica relativa ai substrati neurofisiologici soggiacenti all’intersoggettività, incentrando l’attenzione su neuroni e sistemi di tipo mirror, i quali consentono di comprendere le azioni altrui attraverso un meccanismo di risonanza motoria. Infine, verranno esaminati alcuni aspetti ontogenetici che sottolineano, ancora una volta, come la socialità rappresenti un elemento essenziale della natura umana.The daily life of 21st-century humans revolves around work, schools, hospitals, supermarkets, cinemas, and sports. This paper focuses on analysing the human peculiarity that has enabled such inventions: sociality.
We will begin with a phylogenetic investigation, including an examination of the social and communicative dynamics of our closest relatives, in order to reconstruct the evolutionary steps that led Homo sapiens towards what Edward O. Wilson has defined as "eusociality."
This reconstruction will culminate in an analysis of the ways and reasons why the human brain has evolved throughout its history, leading to the organ now meticulously studied by neuroscience. We will then proceed with a review of the scientific literature on the neurophysiological substrates underlying intersubjectivity, focusing on neurons and mirror systems, which allow us to understand the actions of others through a mechanism of motor resonance.
Finally, we will examine some ontogenetic aspects that emphasize, once again, how sociality represents an essential element of human nature
Peptide Nucleic Acid (PNA)-mediated self-assembly for nano-biotechnology
In the research work of this PhD thesis PNA probe were employed as building blocks to create supramolecular self-assembled architectures to produce biotechnological applications. A significant portion of the work was dedicated to the assembly of new nanostructures with controlled positioning of functional moieties. A second part was devoted to the use of PNA-based assemblies for the realization of lab-in-fiber optical biosensors, and for the creation of a novel approaches in RNA-targeting therapies
Neuropsychological predictors of cerebral amyloidosis in patients with cognitive disorders
La possibilità di definire in <vivo= la presenza di depositi di beta-amiloide nei pazienti con Malattia di Alzheimer (AD) mediante PET amiloide e biomarcatori su liquor ha permesso la transizione da una diagnosi clinica ad una clinico-biologica, migliorando significativamente accuratezza diagnostica e selezione dei pazienti con disturbi cognitivi. Tuttavia, tali metodiche diagnostiche avanzate sono costose e/o scarsamente disponibili nella pratica clinica quotidiana, limitando la possibilità di accesso ad una diagnosi di precisione per la maggior parte di pazienti. E' pertanto necessario trovare strumenti diagnostici accurati, ma di pronta e facile applicabilità anche nei setting ambulatoriali che possano effettuare un efficace screening e vicariare in molti casi le informazioni biologiche ottenute da PET e puntura lombare. Per semplicità di uso, fruibilità, accuratezza, e basso costo la testistica neuropsicologica si presta come strumento diagnostico in grado di raccogliere questa sfida. Scopo di questo studio di tesi è quindi quello di verificare la capacità predittiva di una batteria neuropsicologica nei confronti del risultato di una PET amiloide e quindi indirettamente della presenza o assenza di patologia amiloide, in pazienti con disturbi cognitivi in fase precoce.
Materiali e Metodi:
Lo studio ha analizzato retrospettivamente 118 pazienti consecutivi con diagnosi di Mild Cognitive Impairment - MCI o demenza lieve che erano stati sottoposti nel corso del work up diagnostico per disturbi cognitivi anche ad un esame neuropsicologico completo e ad una PET amiloide nel sospetto di AD.
Risultati:
Il campione di studio aveva un’età media di 70,2 anni (DS: ± 7,68), un'istruzione media di 10,85 anni (DS: ± 4,70) ed un punteggio medio al MMSE di 25,27 (DS: ± 2,83). 76 partecipanti (64,4%) erano positivi all'amiloide-PET e 42 (35,6%) erano
negativi. 89 pazienti (75,4%) erano MCI, 29 (24.6%) demenze lievi. I punteggi ai subitems del Free and Cued Selective Reminding Test (FCSRT), alla copia differita della Figura di Rey e nella fluenza verbale differivano significativamente tra pazienti amiloide + e amiloide -. Nell'intero campione, il subitems IFR del FCSRT mostrava la migliore capacità predittiva (AUC 0.70), seguito da fluenza verbale (AUC 0.66) e copia differita della Figura di Rey (AUC 0.63). Nei pazienti più giovani (≤ 75 anni) e più istruiti (> 8 anni), le prestazioni predittive dei test miglioravano significativamente, raggiungendo per il subitem IFR del FCSRT una AUC, rispettivamente di 0.75 e 0.88. Nell'intero campione il modello predittivo che includeva età, IFR, copia differita della Figura di Rey e fluenza verbale raggiungeva una AUC di 0.76.
Discussione:
Anche nell'era della diagnosi biologica, i test neuropsicologici si confermano preziosi strumenti diagnostici, in grado di fornire indirettamente informazioni anche sui substrati patologici dei disturbi cognitivi. Tra i test esaminati, Il FCSRT si dimostra come il più accurato nel predire il risultato della PET amiloide, con valori di accuratezza molto buoni soprattutto nei pazienti più giovani ed istruiti.Even in the age of biological diagnosis, neuropsychological tests are confirmed as valuable diagnostic tools, able to indirectly provide information also on the pathological substrates of cognitive disorders. Among the tests examined, FCSRT is shown to be the most accurate in predicting the result of amyloid PET, with very good accuracy values especially in younger and educated patients
On Singularities in Linear Elasticity
This thesis investigates stress singularities within the framework of linear elasticity, focusing on their occurrence, interpretation, and implications in two fundamental cases: an infinite elastic wedge under a moment at its vertex and an elastic disk subjected to sectorial thermal expansion. Stress singularities, where stresses theoretically approach infinity, represent critical regions that influence material failure. Although singularities are mathematical idealizations, they highlight the limitations of classical elasticity and provide insights into real-world structural behavior.
The first study addresses the wedge problem, where an infinite elastic wedge is subjected to a concentrated couple at its vertex. Classical solutions, such as Carothers' formulation, predict a quadratic singularity in the stress field. However, at a critical wedge angle, this solution exhibits spurious behavior known as the wedge paradox. This work proposes a novel interpretation of the stress states by reframing the problem in terms of auxiliary wedges and dipole forces, offering a generalized solution that remains valid for all wedge angles. This approach resolves the paradox by demonstrating that the singular stress state corresponds to a system of dipoles with no resultant moment at the critical angle.
The second study explores thermal stress singularities in an infinite elastic disk with a sector experiencing uniform temperature rise. The thermal mismatch between the heated sector and the surrounding material generates stress discontinuities, leading to a logarithmic singularity at the sector boundary. While classical analysis of this infinite geometry yields divergent solutions, introducing a finite length scale, such as the disk radius, constrains the stress field. The study employs dislocation arrays to model thermal mismatches and validates the theoretical predictions using finite element simulations. This approach bridges the gap between idealized infinite models and finite geometries encountered in engineering applications, such as glass panels subjected to differential heating.
This research emphasizes that stress singularities, while mathematically infinite, can be interpreted to provide physically meaningful insights into structural reliability and failure mechanisms. By combining analytical, numerical, and dimensional analysis techniques, the study offers robust frameworks for addressing singularities in practical engineering contexts, including fracture mechanics, thermal stress analysis, and material design
Non-covalent polymeric networks: via hydrogen bonds and electrostatics interactions.
“Supramolecular chemistry has been defined as chemistry beyond the molecule, as it aims at designing and implementing functional chemical systems based on molecular components held together by noncovalent intermolecular forces”. This definition provided by the Nobel Price, Jean-Marie Lehn, lays the foundation of a new branch of chemistry based on the non-covalent interactions and the effect of such bonds on a system. Even though these noncovalent interactions are generally weak, they have a significant impact on the behavior of self-assemble complex structures. Supramolecular interactions include electrostatic interactions, hydrogen bonds, ion-dipole and dipole-dipole interactions, metal coordination, hydrophobic forces, and van der Waals forces. This thesis describes the use of electrostatic interactions and hydrogen bonds as tools for the formation of polymeric networks in polyolefins, featuring new thermal, mechanical, and electric properties. Polyethylene (PE) is the most widely used thermoplastic polymer and the leader in most applications in the global plastic industry. It exhibits strong resistance to solvents, exceptional flexibility, low cost, lightness, and simplicity of processing. However, its use is limited by its low strength and stiffness, low upper service temperature, stress cracking, and poor UV resistance. Additionally, its apolar nature makes it unsuitable for any coating and adhesive application unless a surface treatment is applied (like flame and corona treatment). To overcome those issues, and drastically improve its properties, crosslinking of polyethylene is performed.
Chapters 1 and 2, report the synthesis of a new type of polyethylene-based ionomer, where a few mole percent of positively and negatively charged groups, are covalently bound to the polymer backbone. Amino-terminated methacrylates and methacrylic acid, present in the form of ion pairs, are used as comonomers (ion pair comonomer, IPC) together with ethylene, in a high-temperature/high-pressure process. Specifically, Chapter 1 will present a study of the polymerization reaction of a polyethylene-based ionomer using different reaction conditions, such as temperature, IPC content, in the presence/absence of a chain transfer agent (CTA). The introduction of ionic groups into the PE matrix is an effective way to increase its hydrophilicity and thus its adherence properties to aluminum. In Chapter 2 polyethylene-based ionomers are demonstrated to feature a thermo-mechanical and dielectric property portfolio that is comparable to crosslinked polyethylene (XLPE), which may enable the design of more sustainable high-voltage direct-current (HVDC) power cables, a crucial component of future electricity grids that seamlessly integrate renewable sources of energy.
Chapters 3 and 4, present the use of multiple hydrogen bonding as a tool to compatibilize immiscible polymers. Chapter 3 introduces two-photon microscopy (2PM) as an original technique to investigate the compatibilization between PE-HEMA and EVOH at the sub-micrometer level, both on the surface and in the bulk. 2PM allows to visualize polymer blending through 3D images of the obtained films. Compatibilization was performed in solution, upon functionalization of PE-HEMA with 1.4% molar of ODIN, a fluorescent molecule able to form multiple hydrogen bonding with EVOH and to act as fluorescent probe. Different blends were synthesized, and the obtained films analyzed by 2PM. For all compositions, it was demonstrated that ODIN is evenly distributed both on the surface and in the bulk. 2PM analysis of the thermally reprocessed specimen revealed that repeated reprocessing allows the reformation of ODIN dimers as the most stable H-bonding array in the solid state, partially reversing the compatibilization.
Chapter 4 discusses the possibility to create polymer blends acting as oxygen barrier, to evaluate the applicability of a monolayer instead of a multilayer as a solution to produce monolayer packaging films to substitute triple-layer films. Ethylene-co-vinyl alcohol (EVOH) which is already used in the food and pharmaceutical industry for its oxygen barrier property, and polyethylene-co-hydroxyethyl methacrylate (PE-HEMA), which acts as a water barrier, have been utilized. The study involved the permeability characterization of the blends produced in Chapter 3 as well as the production of new binary mixtures produced by functionalizing PE-HEMA with phenyl-urea and combining it with EVOH. The following assessment of the blends, paying special attention to the oxygen permeability tests, shows an unequal distribution of the two polymers within the blends. The PE-HEMA fraction is mainly present at the surface, while EVOH is concentrated in the inner bulk. This self-segregation in the films leads to improved oxygen barrier properties
Algoritmi di controllo, percezione e coordinamento per advanced driver-assistance system: implementazioni e simulazioni ottimizzate
The future of urban mobility is undergoing changes with the development of intelligent cities and the increased use of autonomous vehicles. The transition to this new paradigm is a gradual process over several years or decades, but progress has been made through the implementation of smart sensors and communication infrastructure. The development of Advanced Driver-Assistance Systems (ADAS) with growing autonomy is also in progress.
In this thesis, two main aspects are addressed: coordination algorithms to manage the smart city traffic flow and the perception-control pipeline of autonomous vehicles with specific emphasis on the localization and planning phases.
With regards to the first aspect, several novel algorithms are proposed that exploit the new smart city capabilities to address typical problems such as Traffic Lights and Intersection Management, Parking Management, and Emergency Vehicles Management. The work proposed in this thesis is a study of the current situation in which autonomous or able-to-communicate vehicles and traditional vehicles that are not able to communicate with city infrastructure co-exist. This is a crucial aspect since mixing ADAS and traditional vehicles impacts the algorithm design.
The proposed algorithms are tested in a simulated scenario in order to study unexpected behaviors since traffic flow is a complex system and some events can trigger unpredictable consequences. The results show that the proposed algorithms improve the city's livability by decreasing the waiting time at traffic lights, reducing the parking search time, and the emergency vehicle response time.
In regards to the localization and planning stages, the emphasis is placed on the execution time of the algorithms, as it is a critical aspect. If the perception and control pipeline takes too long, the intended maneuver may become outdated due to changes in the environment, potentially causing safety risks. In light of this, novel implementations for localization and planning algorithms are proposed, which make extensive use of the GPU as an accelerator in order to reduce computational time. The GPU is leveraged to parallelize the algorithm and minimize memory access. Additionally, the use of different floating-point precision types is investigated to assess the impact on the results.
The proposed implementations of the ORB-SLAM algorithm for the localization phase and the Frenet Path Planner algorithm for the planning phase show a consistent speedup, compared to the previously published CPU-based implementations of the algorithms.Il futuro della mobilità urbana sta cambiando con lo sviluppo di città intelligenti e l'aumento dell'uso di veicoli autonomi. Il passaggio a questo nuovo paradigma è un processo graduale che dura diversi anni o decenni, ma sono stati fatti progressi grazie all'implementazione di sensori intelligenti e infrastrutture di comunicazione. È in corso anche lo sviluppo di sistemi avanzati di assistenza alla guida (ADAS) con crescente autonomia.
In questa tesi si affrontano due aspetti principali: gli algoritmi di coordinamento per gestire il flusso di traffico delle smart city e la pipeline di percezione-controllo dei veicoli autonomi, con particolare attenzione alle fasi di localizzazione e pianificazione.
Per quanto riguarda il primo aspetto, vengono proposti diversi algoritmi innovativi che sfruttano le nuove funzionalità delle smart city per affrontare problemi tipici come la gestione dei semafori e delle intersezioni, la gestione dei parcheggi e la gestione dei veicoli di emergenza. Il lavoro proposto in questa tesi è uno studio della situazione attuale in cui coesistono veicoli autonomi o in grado di comunicare e veicoli tradizionali che non sono in grado di comunicare con le infrastrutture cittadine. Si tratta di un aspetto cruciale, poiché la commistione tra veicoli ADAS e tradizionali influisce sulla progettazione degli algoritmi.
Gli algoritmi proposti sono stati testati in uno scenario simulato per studiare comportamenti inaspettati, poiché il flusso del traffico è un sistema complesso e alcuni eventi possono innescare conseguenze imprevedibili. I risultati mostrano che gli algoritmi proposti migliorano la vivibilità della città diminuendo il tempo di attesa ai semafori, riducendo il tempo di ricerca del parcheggio e il tempo di risposta dei veicoli di emergenza.
Per quanto riguarda le fasi di localizzazione e pianificazione, l'enfasi è posta sul tempo di esecuzione degli algoritmi, in quanto si tratta di un aspetto critico. Se la pipeline di percezione e controllo richiede troppo tempo, la manovra prevista può diventare obsoleta a causa dei cambiamenti dell'ambiente, causando potenzialmente rischi per la sicurezza. Alla luce di ciò, vengono proposte nuove implementazioni per gli algoritmi di localizzazione e pianificazione, che fanno ampio uso della GPU come acceleratore per ridurre i tempi di calcolo. La GPU viene sfruttata per parallelizzare l'algoritmo e ridurre al minimo l'accesso alla memoria. Inoltre, viene analizzato l'uso di diversi tipi di precisione in virgola mobile per valutare l'impatto sui risultati.
Le implementazioni proposte dell'algoritmo ORB-SLAM per la fase di localizzazione e dell'algoritmo Frenet Path Planner per la fase di pianificazione mostrano una velocità consistente, rispetto alle implementazioni degli algoritmi basati su CPU pubblicate in precedenza
Sustainability in the livestock sector. Possible mitigation systems of environmental impact of breeding
La sfida più grande per il settore agro-zootecnico è ridurre i compromessi tra incremento della produttività e sostenibilità a lungo termine. In ragione della crescente domanda alimentare stimolata dall’aumento della popolazione mondiale e dalla globalizzazione, il settore zootecnico dovrà far fronte ad una richiesta alimentare senza precedenti che richiederà, oltre all’aumento di prodotti alimentari di origine animale, di intensificare lo sfruttamento delle risorse naturali, generando un aumento dell’impatto ambientale del settore.
L’attenzione mondiale è rivolta ormai da molti anni agli effetti negativi generati nei confronti dell’ambiente dalle attività antropiche e sono stati di conseguenza sviluppati piani mondiali per riuscire a risolvere, o per lo meno migliorare, i danni ambientali che stanno scatenando una catena di eventi degenerativi terrestri. Tutti i settori partecipano alla produzione di emissioni e generano impatto ambientale, compreso il settore zootecnico che, grazie alla sua espansione mondiale e alla differenziazione del settore, ha un impatto ambientale significativo.
I settori zootecnico svolgono anche molteplici ruoli positivi che rientrano in tutti gli Obiettivi di Sviluppo Sostenibile (OSS) dell’Agenda globale 2030, con interazioni e anche compromessi molteplici e diversificati. Lo sviluppo di nuove tecniche e tecnologie per migliorare il settore zootecnico, sia dal punto di vista ambientale ma anche del benessere animale e sostegno alle comunità, rientra negli obiettivi di sviluppo sostenibile e può aiutare a migliorare molti aspetti della sostenibilità in quanto i suoi obiettivi sono intimamente interconnessi e migliorandone alcuni questi possono influenzare anche tutti gli altri.The biggest challenge for the agro-zootechnical sector is to reduce the
trade-offs between increased productivity and long-term sustainability.
Due to the growing food demand stimulated by the global population and
globalization, the livestock sector will have to cope with an unprecedented food
demand that will require, in addition to the increase in food products of animal
origin, to intensify the exploitation of natural resources, generating an increase in
the environmental impact of the sector.
The worldwide attention is now turned for many years to the negative
effects generated towards the environment by anthropic activites and have been
developed as a result world plans to be able to solve, and, or at least improve, the
environmental damage that is triggering a chain of degenerative terrestrial events.
All sectors partecipate in the production of emissions and generate environmental
impact, including the livestock sector which, thanks to its worldwide expansion and
differentation of the sector, has a significant environmental impact.
Livestock sectors also play multiple positive roles that fall under all
Sustainable Development Goals (SDGs) of the Global Agenda 2030, with multiple
and diverse interacRons and also trade-offs. The development of new techniques
and technologies to improve the livestock sector, both from an environmental point
of view but also from an animal welfare and community support point of view, it is
part of the sustainable development goals and can help to improve many aspect
Sviluppo di modelli ex vivo per lo screening di nuove formulazioni colloidali oftalmiche
Il trattamento delle patologie oculari che colpiscono il segmento posteriore dell’occhio richiede l’impiego di approcci altamente invasivi come le iniezioni intravitreali: infatti, l’attraversamento delle barriere oculari rappresenta ancora oggi una sfida per la somministrazione dei farmaci.
Negli ultimi anni l’associazione tra i nanosistemi e la somministrazione perioculare, meno invasiva della intravitreale, ha suscitato attenzione per la sua elevata potenzialità. Diversi sono i nanosistemi colloidali proposti e studiati; tuttavia, la ricerca preclinica è carente di strumenti robusti, quali i modelli oculari ex vivo, che permettano lo screening di nanoparticelle in condizioni biorilevanti.
Nella presente tesi sono stati perseguiti tre obiettivi.
Il primo è stato lo sviluppo di nanosistemi lipidici caricati con corticosteroidi (in particolare desametasone acetato e desametasone) per il trattamento del segmento posteriore dell’occhio. Precisamente, partendo da uno studio pre-formulativo, sono stati selezionati eccipienti compatibili con la somministrazione oculare e sono state preparate e caratterizzate delle Nanostructured Lipid Carrier (NLC) e delle microemulsioni. Inoltre, è stato applicato un approccio basato sul disegno sperimentale (DoE) per creare modelli matematici che consentano di predire le caratteristiche chimico-fisiche dei nanosistemi in base alle condizioni sperimentali.
Il secondo obiettivo è stato lo sviluppo e la validazione di un setup ex vivo, rappresentato dall’occhio suino intero, per il miglioramento degli studi formulativi in adempimento del principio delle 3R (Replacement, Reduction and Refinement) nella sperimentazione animale. Dopo una preliminare caratterizzazione dei bulbi oculari suini, sono stati realizzati due dispositivi con la tecnica della stampa 3D. Grazie a questi due dispositivi è possibile applicare la formulazione (liquida, semisolida o solida) sulla sclera, mantenendo il bulbo oculare in condizioni controllate di temperatura e umidità per 18 ore. In aggiunta, un metodo per estrarre i farmaci nel corpo vitreo è stato messo a punto.
Terzo ed ultimo obiettivo è stata la valutazione ex vivo delle formulazioni preparate, utilizzando il classico approccio basato sull’utilizzo della sclera e della coroide isolati montati su cella di Franz ed il nuovo modello di occhio intero.Overcoming the ocular barriers is still a challenging task since currently available treatments for diseases affecting the back of the eye require highly invasive approaches (i.e., intravitreal injections).
In the recent years, association of nanosystems with less-invasive periocular administration routes has been recognized as a possible strategy to treat these conditions. Despite the increasing number of colloidal nanosystems currently under development, preclinical research still lacks robust screening and reproducible setups in biorelevant conditions, such as ex vivo ocular models.
The present thesis has pursued three different aims.
The first was the development of lipid-based nanocarriers intended for drug delivery to the posterior segment of the eye and loaded with corticosteroids (namely, dexamethasone acetate and dexamethasone). Starting from pre-formulative studies for adequate ocular-compatible excipients selection, nanostructured lipid carriers (NLC) and microemulsions were prepared and physiochemically characterized. In addition, a novel approach based on design of experiments (DoE) was applied to create mathematical models that predict physicochemical characteristics of the nanocarriers, allowing to fine-tune the nanocarrier to the intended administration route.
The second aim was the development and validation of an ex vivo setup for the improvement of preclinical studies in the ophthalmic field in accomplishment of the 3Rs (Replacement, Reduction and Refinement) in animal experimentation. After a preliminary characterization of porcine eyeballs, a 3D printing approach allowed to manufacture two devices for trans-scleral drug delivery evaluation on whole ocular bulbs. As a result, a setup allowing for the application of liquid, semisolid, or solid formulations to porcine sclera, at a controlled temperature and humidity for up to 18 hours, was achieved. In addition, a methodology for tissue manipulation and drug quantification from the vitreous body was validated.
The third and final aim was the ex vivo evaluation of the developed formulations. The classical approach based on isolated porcine sclera and choroid mounted in Franz-type diffusion cells was then followed by the novel ex vivo whole-eye setup
