    High Pressure/High Temperature synthesis of novel functional perovskites and their structural, magnetic, and electrical characterizations

    This doctoral thesis explores the effective solid-state synthesis of novel functional perovskites under extreme High Pressure/High Temperature (HP/HT) conditions. Specifically, it presents a comprehensive investigation of BiCu0.5Mn0.5O3 (BCMO) and KPbFeMoO6 (KPFMO), illustrating how it is possible to manipulate the chemistry of the perovskite structures to finely tune their physical properties. BCMO is characterized by a disordered mesoscale distribution of the B cations and the correlation between magnetic and electrical properties has been pointed out. In particular, BCMO displays a weak FM-like transition at 320 K and a thermally activated electrical transport, while, in the low-temperature regime, the electrons’ localization on Mn site leads to a 3D Variable Range Hopping transport and an AFM spin-glassy state at 50 K. For KPFMO a reliable cubic structure has been proposed, while further investigations are required for cationic ordering evaluation. Contrarily to BCMO, it has a more resistive character, behaving as a dielectric below 250 K and showing an AFM transition at around 50 K. Thus, this study provides an in-depth analysis of the properties of these compounds, emphasizing structural characterizations through the combination of complementary techniques such as X-rays, electrons, and neutrons diffraction and highlighting the crucial role of the crystal structure resolution process. Additionally, the magnetic and electrical properties of these perovskites are explored and compared with those of similar compounds, highlighting the pros and cons of their behaviour as multifunctional materials.Questa tesi di dottorato esplora l’effettiva sintesi allo stato solido di nuove perovskiti funzionali in condizioni estreme di alta pressione e alta temperatura (HP/HT). In particolare, essa presenta un'indagine completa su BiCu0.5Mn0.5O3 (BCMO) e KPbFeMoO6 (KPFMO), illustrando come sia possibile manipolare la chimica delle strutture a perovskite per regolare finemente le loro proprietà fisiche. BCMO è caratterizzato da una distribuzione disordinata a mesoscala dei cationi B e sono stati osservati effetti di correlazione tra le proprietà magnetiche ed elettriche. In particolare, BCMO mostra una debole transizione di tipo ferromagnetico a 320 K e un trasporto elettrico termicamente attivato, mentre, nel regime di basse temperature, la localizzazione degli elettroni sul sito del Mn porta a un trasporto di hopping 3D-variabile e uno stato spin-glass antiferromagnetico a 50 K. Per KPFMO è stata proposta una struttura cubica, mentre ulteriori indagini sono necessarie per la valutazione dell'ordinamento cationico. Contrariamente a BCMO, ha un carattere più resistivo, comportandosi come un dielettrico al di sotto dei 250 K e mostrando una transizione antiferromagnetica a circa 50 K. Questo studio fornisce quindi un'analisi approfondita delle proprietà di questi composti, enfatizzando le caratterizzazioni strutturali attraverso l'integrazione di tecniche complementari come diffrazione da raggi X, elettroni e neutroni, e sottolineando il ruolo cruciale del processo di risoluzione della struttura cristallografica. Inoltre, le proprietà magnetiche ed elettriche di queste perovskiti sono esplorate e confrontate con quelle di composti simili, evidenziando vantaggi e svantaggi del loro comportamento come materiali multifunzionali

    L'opera non realizzata di Marta Pierobon, nata a Brescia nel 1979, emerge come una performance artistica di dieci giorni, in cui l'argilla assume un ruolo centrale e preminente nell'esplorazione del concetto di spazio attraverso la “scultura performativa”. Utilizzando l'argilla come medium privilegiato, Pierobon dà forma non solo allo spazio, ma anche a una serie di personaggi e oggetti metamorfosizzati che animano un ambiente ricco di simbolismi e suggestioni. Attraverso gesti precisi e sapienti manipolazioni dell'argilla, l'artista costruisce una narrazione che sollecita lo spettatore a interagire, rielaborando parte dello spazio performativo mediante l’utilizzo dell’argilla per creare figure, immagini e sculture effimere, che si formano e si disfano, modellando e trasformando lo spazio. La performance sembra trasformarsi in un rituale attraverso una serie di abiti ideati dall’artista stessa e coinvolge attivamente i visitatori, trasformando l'ambiente in un tableau vivant in continua evoluzione, dove l'artista e il pubblico collaborano come co-autori dell'esperienza artistica. In questa cornice, lo spazio rivela la sua natura dinamica e attiva, assumendo un ruolo di protagonista nel processo trasformativo dell'opera. L’opera stimola una riflessione articolata sullo spazio, inteso come dimensione fisica, mentale ed emotiva, mettendo in luce le intricate relazioni tra individuo, arte e ambiente spaziale. Inoltre, vuole celebrare l’argilla come materia naturale strettamente legata alla pratica scultorea dell’artista e alla terra

    La tubercolosi bovina causata da Mycobacterium bovis è una malattia zoonotica che ha ripercussioni sulla sanità pubblica, sulla zootecnia e sull'economia. E’ una malattia soggetta a piano di eradicazione all’interno degli Stati membri dell’UE come recita il Regolamento (UE) 2016/429. La tubercolosi è una zoonosi distribuita a livello globale che colpisce i bovini, un’ampia gamma di mammiferi selvatici e l’uomo. Alcuni animali selvatici fungono da ospiti serbatoio nella trasmissione e nell'epidemiologia della malattia. Il cinghiale (Sus scrofa), specie in forte espansione demografica e geografica in Europa, è identificato come un importante serbatoio di M. bovis, svolgendo un ruolo cruciale nella persistenza e trasmissione della malattia nei bovini. Alcuni studi genetici hanno evidenziato la condivisione dei genotipi di M. bovis tra cinghiali e bovini, suggerendo un ciclo di trasmissione che facilita la diffusione della tubercolosi tra specie selvatiche e domestiche. La trasmissione del patogeno tra le due specie è principalmente indiretta legata alla condivisione di ambienti o risorse contaminate (come pascoli o pozze d'acqua). Lo scopo della tesi è quello di esaminare il ruolo del cinghiale nella diffusione della tubercolosi bovina in Italia e in Europa e di fornire una panoramica dei caratteri patogenetici di M. bovis, delle lesioni anatomopatologiche, delle tecniche diagnostiche e dell'epidemiologia della tubercolosi zoonotica nei cinghiali. Il lavoro di tesi evidenzia inoltre la necessità di un approccio “One Health” al controllo della tubercolosi zoonotica che comporta la collaborazione tra sanità pubblica, veterinari ed enti per la gestione della fauna selvatica per ridurre la trasmissione dell’agente patogeno tra animali e uomo.Bovine tuberculosis caused by Mycobacterium bovis is a zoonotic disease that has an impact on public health, animal husbandry and economy. It is a disease subject to an eradication plan within the EU Member States as stated in Regulation (EU) No 2016/429. Animal tuberculosis is a globally distributed zoonosis, affecting cattle, a wide range of wild mammals and humans. Some wild animals serve as reservoir hosts in the transmission and epidemiology of the disease. The wild boar (Sus scrofa), a species in demographic and geographic expansion in Europe, is identified as an important reservoir of M. bovis, playing a crucial role in the persistence and transmission of the disease in cattle. Genetic studies have highlighted the sharing of M. bovis genotypes between wild boars and cattle, suggesting a transmission cycle that facilitates the spread of tuberculosis between wild and domestic species. The transmission of the pathogen between the two species is mainly indirect. i.e. linked to the sharing of contaminated environments or resources (such as pastures or waterholes). The aim of the thesis is to examine the role of wild boars in the spread of bovine tuberculosis in Italy and Europe and to provide an overview of pathogenic characters of M. bovis, the anatomopathological lesions, the diagnostic techniques and the epidemiology of zoonotic tuberculosis in wild boars. The thesis work also highlights the need for a “One Health” approach to control zoonotic tuberculosis, which involves collaboration between public health institutions, veterinarians, and wildlife management bodies to reduce the transmission of the pathogen between animals and humans

    The neural activity of the macaque motor cortex during spontaneous walking : a neuroethological investigation

    La camminata è un comportamento che coinvolge l'intero corpo, caratterizzato dal movimento ritmico degli arti, la cui velocità e direzione vengono adattate costantemente per affrontare le sfide ambientali. Sebbene sia noto che i circuiti neurali responsabili della generazione del modello locomotorio risiedano nelle aree sottocorticali, in particolare nel midollo spinale, rimane poco chiaro il meccanismo neurale che regola e modula il ciclo del cammino in condizioni naturali. Gli studi che hanno esaminato il ruolo della Corteccia Motoria (MC) nella coordinazione della camminata hanno spesso adottato condizioni non naturalistiche, come l'utilizzo del tapis roulant, o l'impiego di modelli animali quali i gatti, lasciando non chiariti i contributi della MC alla camminata naturale nei primati. Al fine di colmare queste lacune, abbiamo registrato l'attività di singoli neuroni nella MC di macachi lasciati liberi di muoversi spontaneamente in un ampio spazio. Abbiamo osservato un'elevata attivazione neuronale (104/255 = 41%) durante la camminata indipendentemente dalla superficie di appoggio—che fosse terreno piano o condizioni più impegnative come una struttura sopraelevata. Questi neuroni hanno mostrato diversi modelli di attività correlati alle fasi del passo, la maggior parte raggiungevano il picco durante la fase di appoggio della mano controlaterale all'emisfero registrato, e diversi hanno mostrato una modulazione durante la progressione dei passi. Stimolando elettricamente i siti associati alle risposte legate al cammino, abbiamo osservato non solo movimenti del braccio, ma anche movimenti prossimali, suggerendo che il ruolo della MC nella coordinazione della camminata potrebbe coinvolgere non solo aspetti distali legati al posizionamento degli arti, ma anche adattamenti posturali. Inoltre, è emerso che la dinamica neurale tipica del cammino è in parte condivisa con quella osservata durante l’atto di raggiungimento di un oggetto, soprattutto nella fase precedente il contatto della mano con il suolo o l'oggetto. In sintesi, questi risultati indicano che la MC svolge un ruolo fondamentale, e precedentemente sottovalutato, anche nella coordinazione della camminata spontanea in condizioni naturali.Walking is a whole-body behavior characterized by a rhythmic movement of the limbs, whose velocity and direction are constantly adjusted to address environmental challenges. While the neural circuits that produce walking pattern are known to lie in subcortical areas, particularly in the spinal cord, little is known about the neural mechanism that shape and adjust the walking cycle in natural conditions. Studies that have investigated the role of Motor Cortex (MC) in walking coordination either adopted the unnatural condition of treadmill or used cats as animal models, leaving unclear the MC’s contribution to natural walking in primates. To fill these gaps, we recorded single-neuron activity from the MC of macaques left free to move spontaneously in a large enclosure. We found a high number of neurons (104/255 = 41%) activated during walking, irrespective of walking surface—be it flat ground or more challenging conditions like elevated structure. These neurons exhibited diverse firing patterns relative to the step cycle, with many peaking in activity during the landing phase of the hand contralateral to the recorded hemisphere, and several neurons were modulated by the progression of steps. When electrically stimulated, the sites exhibiting walking-related responses elicited arm movements, but also proximal movements, suggesting that the role of MC in coordinating walking might involve not only distal aspects related to limb placement but also postural adjustments. Furthermore, we found that the neural dynamics typical of walking are partly shared with those recruited during goal-directed reaching, particularly in the phase preceding the contact of the hand with the ground or the object. Taken together, these findings indicate that the MC exerts a fundamental, and previously underestimated, role also in coordinating spontaneous walking in natural conditions

    Research of salivary biomarkers for risk stratification in patients with head and neck squamous cell carcinoma

    L’elaborato presentato ha l’obiettivo di dimostrare la necessità di trovare nuove metodiche, come la ricerca di biomarcatori biologici salivari per valutare la prognosi, la sopravvivenza e il rischio di recidiva di pazienti affetti da carcinoma squamocellulare del cavo orale, da utilizzare sia nel periodo di follow-up che in fase preoperatoria. Questa necessità deriva dall'elevata incidenza e mortalità di questi tumori: sono, infatti, la sesta causa di morte per cancro a livello mondiale, e in Europa la loro incidenza ammonta a 150.000 nuovi casi all'anno, con una mortalità di 70.000 casi all'anno. Inoltre, una migliore stratificazione aiuterebbe a prendere la giusta decisione terapeutica. Uno degli istotipi tumorali della regione testa–collo, più grossolanamente stratificati è certamente il carcinoma squamocellulare del cavo orale (OSCC), in particolare quello non correlato all'infezione da HPV (HPV-negativo). Infatti, la sopravvivenza complessiva dei pazienti con OSCC è del 60%, ma varia enormemente (dal 10% all'80%) in relazione alle dimensioni del tumore e alle caratteristiche biologiche. Da ciò si evince che è essenziale al momento della diagnosi differenziare i tumori con un comportamento biologico meno aggressivo da quelli con un comportamento biologico più aggressivo. I dati raccolti per l’elaborazione della seguente tesi sono stati estratti dalle cartelle di pazienti trattati presso l’Unità Operativa di Chirurgia Maxillo-Facciale di Parma; le informazioni raccolte comprendono il referto istopatologico, la cartella clinica chirurgica e il referto del percorso diagnostico terapeutico assistenziale (PDTA) del paziente. Dalle analisi dei dati emerge come alcune caratteristiche istopatologiche dei campioni operatori siano correlate significativamente alla sopravvivenza del paziente, come il grading e il parametro pT, della stadiazione patologica TNM. Tuttavia, il sistema di stadiazione utilizzato finora nella pratica clinica (sistema di stadiazione TNM) risulta non sufficiente alla valutazione della sopravvivenza e della stratificazione del rischio di recidiva dei pazienti affetti. Questa limitazione è dovuta principalmente al fatto che questo sistema di stadiazione non tiene conto né delle caratteristiche individuali del paziente né delle caratteristiche biologiche del tumore. Alcuni dei vantaggi associati a un modello di previsione basato su marcatori biologici possono essere offerti dall’identificazione precoce della neoplasia, consentendo una diagnosi più tempestiva e la scelta di un regime terapeutico più appropriato per un determinato paziente personalizzando la terapia. Tra i biomarker salivari quelli potenzialmente più importanti per la ricerca del carcinoma orale sono mRNA, microRNA e proteine. Tuttavia, è importante precisare che i modelli di previsione basati su marcatori biologici devono essere validati accuratamente e integrati con altre informazioni cliniche necessarie per stabilire l’appropriato iter terapeutico. Ad oggi, non solo è oggetto di studio la ricerca di biomarcatori predittivi specifici e sensibili del carcinoma del cavo orale, ma anche la tecnica con cui ricercarli, indipendentemente dalla metodica di brushing o raccolta salivare. L’Unità Operativa (UO) di Chirurgia Maxillo-Facciale dell’Azienda Ospedaliero-Universitaria di Parma, in collaborazione con il dipartimento di Medicina Sperimentale dell’Ateneo dell’Università degli Studi di Parma, ha avviato uno studio prospettico di raccolta e analisi di campioni di saliva prelevati in pazienti trattati chirurgicamente per carcinoma squamoso del cavo orale e seguiti in programmi di follow-up dalla stessa UO. Lo scopo è quello di identificare dei biomarcatori genetici, in particolar modo miRNA, attraverso il campionamento non invasivo del liquido biologico salivare e la tecnica brushing. L’identificazione di un marker salivare predittivo specifico e sensibile validerebbe un nuovo modello prognostico, utile per migliorare la stratificazione dei OSCC e quindi anche le decisioni terapeutiche che ne derivano.The paper aims to demonstrate the necessity to find new methods, such as the search for salivary biological biomarkers to assess the prognosis, survival, and risk of recurrence of patients with squamous cell carcinoma of the oral cavity, to be used both in the follow-up period and preoperatively. This need stems from the high incidence and mortality of these cancers: they are, in fact, the sixth leading cause of cancer death worldwide, and in Europe their incidence amounts to 150,000 new cases per year, with a mortality of 70,000 cases per year. In addition, better stratification would help make the right treatment decision. One of the most grossly stratified cancer histotypes of the head and neck region is certainly oral squamous cell carcinoma (OSCC), particularly that unrelated to HPV infection (HPV-negative). In fact, the overall survival of patients with OSCC is 60%, but it varies greatly (10% to 80%) depending on tumor size and biological characteristics. Hence, it is essential at the time of diagnosis to differentiate tumors with less aggressive biological behavior from those with more aggressive biological behavior. The data collected for the elaboration of the following thesis were extracted from the records of patients treated at the Operative Unit of Maxillofacial Surgery in Parma, Italy; the information collected includes the histopathological report, the surgical medical record, and the report of the patient's diagnostic therapeutic care pathway (PDTA). Data analysis shows that some histopathological features of the operative specimens are significantly correlated with patient survival, such as grading and the pT parameter, of the TNM pathological staging. However, the staging system used so far in clinical practice (TNM staging system) appears to be insufficient for the assessment of survival and risk stratification of recurrence of affected patients. This limitation is mainly due to the fact that this staging system does not take into account either individual patient characteristics or the biological characteristics of the tumor. Some of the advantages associated with a prediction model based on biological markers can be offered by early identification of the malignancy, allowing earlier diagnosis and the selection of a more appropriate treatment regimen for a given patient by personalizing therapy. Among salivary biomarkers, the potentially most important ones for oral carcinoma detection are mRNA, microRNA, and protein. However, it is important to point out that predictive models based on biological markers must be validated thoroughly and integrated with other clinical information needed to determine the appropriate treatment course. Nowadays, not only is the search for specific and sensitive predictive biomarkers of oral cancer being investigated, but also the technique by which to search for them, regardless of the method of salivary brushing or collection. The Operative Unit (OU) of Maxillofacial Surgery of the Azienda Ospedaliero-Universitaria of Parma, in collaboration with the Department of Experimental Medicine of the University of Parma, has started a prospective study of collection and analysis of saliva samples taken in patients surgically treated for squamous cell carcinoma of the oral cavity and followed in follow-up programs by the same OU. The aim is to identify genetic biomarkers, especially miRNAs, by noninvasive sampling of salivary biological fluid and brushing technique. The identification of a specific and sensitive predictive salivary marker would validate a new prognostic model, which would be useful for improving the stratification of OSCC and thus also the resulting therapeutic decisions

    Dinamiche del microbioma intestinale nei primati tra ecosistemi tropicali

    L'impatto antropico sugli ecosistemi tropicali ha portato a una significativa perdita di biodiversità e i primati non umani risultano essere ad alto rischio di estinzione. Secondo l'Unione Mondiale per la Conservazione della Natura (IUCN), quasi un terzo delle specie di primati non umani è classificato come a rischio di estinzione, e la causa principale è la perdita dell'habitat preferito e delle risorse alimentari. La perdita di habitat ha inevitabilmente influenzato la dieta e il microbiota intestinale di queste specie, portando a una perdita di biodiversità a livello microscopico. Il microbiota intestinale e la sua collezione di genomi (microbioma intestinale) svolgono un ruolo essenziale nella salute dell'ospite, svolgendo funzioni quali la digestione, la fermentazione di composti complessi, la modulazione del sistema immunitario e la difesa da agenti patogeni esterni. La disbiosi intestinale, dovuta alla perdita o all'acquisizione di nuovi microrganismi, è spesso associata a infiammazioni e malattie. Le ricerche sul microbiota intestinale di specie animali selvatiche che vivono in ambienti colpiti da attività umane (ad esempio, la deforestazione) hanno mostrato perdite di ricchezza o cambiamenti nella composizione del microbiota intestinale. Tuttavia, mancano indagini più approfondite sul significato funzionale di queste perdite e la comprensione delle dinamiche tra i microrganismi intestinali dell'ospite e quelli dell'ambiente circostante consentirebbe di chiarire l'impatto delle attività umane sulla salute dell'ospite. Utilizzando primati non umani come specie modello, questa tesi si propone di (i) descrivere l'attuale comprensione delle dinamiche dei simbionti intestinali e le loro conseguenze sulla salute dell'ospite; (ii) indagare il contributo dei fattori estrinseci e intrinseci alla variazione del microbiota intestinale; e (iii) esplorare l'impatto dei fattori estrinseci sul microbioma e le conseguenti variazioni nelle funzioni metaboliche dell'ospite. • Capitolo 1: Questa rassegna si concentra sugli studi dei microrganismi intestinali più abbondanti (batteri) insieme ad altri simbionti intestinali (funghi, archei, elminti e virus) per comprenderne le associazioni, le interazioni e gli effetti sulla salute dell'ospite. Questo capitolo passa in rassegna le attuali conoscenze sulle interazioni e le associazioni tra batteri e vari simbionti. Evidenzia come i cambiamenti di alcuni simbionti intestinali rispetto ad altri, così come le fluttuazioni tra più simbionti dovute a fattori intrinseci o estrinseci, possano avere un impatto sull'ospite. Queste interazioni e i loro effetti sono spesso complessi e non facilmente comprensibili. • Capitolo 2: analizza la variazione del microbiota intestinale (batteri e funghi) di una popolazione di babbuini gialli (Papio cynocephalus) in relazione al tipo di habitat (fattore estrinseco) e al sesso (fattore intrinseco). Lo studio ha osservato che le variazioni del microbiota intestinale sono influenzate dal tipo di habitat e che la diversità delle comunità batteriche e fungine è potenzialmente mediata dal sesso, sottolineando l'importanza di combinare fattori biologici ed ecologici nelle analisi del microbiota. • Capitolo 3: fornisce la prima identificazione completa delle comunità batteriche e fungine nel Sanje mangabey (Cercocebus sanjei), una specie a rischio di estinzione endemica dei Monti Udzungwa. Il capitolo esplora la potenziale contaminazione dei microrganismi intestinali da parte dei microrganismi del suolo, evidenziando un significativo trasferimento delle comunità fungine saprotrofe del suolo nell'intestino. Ciò suggerisce un'associazione tra la geofagia, un comportamento di consumo del suolo osservato nel Sanje mangabey, e la sua dieta altamente frugivora. La geofagia è un comportamento adattivo che può avere varie funzioni, come quella di alleviare lo stress gastrointestinale. I nostri risultati suggeriscono che questi benefici possono essere in parte determinati dalla presenza di microrganismi nel suolo e dalla loro interazione con il microbioma intestinale. • Capitolo 4: indaga la diversità delle funzioni metaboliche dei microrganismi intestinali in due ospiti di primati non umani, il colobo rosso Udzungwa (Piliocolobus gordonorum) e i babbuini gialli, utilizzando metodi di sequenziamento estensivi (metagenomica). Lo studio ha rilevato una forte associazione tra le funzioni metaboliche osservate e le diverse diete degli ospiti. Sono state osservate differenze nelle funzioni metaboliche anche all'interno della stessa specie ospite che vive in tipi di habitat contrastanti, suggerendo che le diete specializzate possono essere maggiormente influenzate dalle differenze di habitat rispetto alle diete generaliste. Nel complesso, questa tesi fornisce una visione completa della complessità delle variazioni microbiche intestinali in termini di struttura, composizione e funzioni metaboliche. Sottolinea l'importanza di studiare queste variazioni nelle specie ospiti selvatiche, spesso minacciate dalle attività umane. Questi risultati possono migliorare la consapevolezza nel campo della conservazione e aiutare a sviluppare strategie per mitigare gli effetti del disturbo antropico.The anthropogenic impact on tropical ecosystems has led to a significant loss of biological diversity, with non-human primates facing a high risk of extinction. According to the World Conservation Union (IUCN), almost one third of non-human primate species are classified as endangered, with the main driver being loss of preferred habitat and food resources. Habitat loss has inevitably affected the diet and gut microbiota of these species, leading to a loss of biological diversity at the microscopic level. The gut microbiota and its genome collection (gut microbiome) play an essential role in host health, performing functions such as digestion, fermentation of complex compounds, modulation of the immune system, and defense against external pathogens. Gut dysbiosis, due to the loss or acquisition of new microorganisms, is often associated with inflammation and disease. Research on the gut microbiota of wild animal species living in human-affected environments (e.g., deforestation) has shown losses in richness or changes in gut microbiota composition. However, deeper investigations into the functional significance of these losses are missing, and understanding the dynamics between host gut microorganisms and those of the surrounding environment would also elucidate the impact of human activities on host health. Using non-human primates as model species, this thesis aims to (i) describe the current understanding of gut symbiont dynamics and their consequences on host health; (ii) investigate the contribution of extrinsic and intrinsic factors to gut microbiota variation; and (iii) explore the impact of extrinsic factors on the microbiome and the resulting variations in host metabolic functions. • Chapter 1: This review focuses on studies of the most abundant intestinal microorganisms (bacteria) in conjunction with other intestinal symbionts (fungi, archaea, helminths, and viruses) to understand their associations, interactions, and effects on host health. This chapter reviews the current understanding of interactions and associations between bacteria and various symbionts. It highlights how changes in some intestinal symbionts relative to others, as well as fluctuations among multiple symbionts due to intrinsic or extrinsic factors, can impact the host. These interactions and their effects are often complex and not easily understood. • Chapter 2: Investigates the variation in gut microbiota (bacteria and fungi) of a population of yellow baboons (Papio cynocephalus) in relation to habitat type (extrinsic factor) and sex (intrinsic factor). The study observed that gut microbiota variations are affected by habitat type, with bacterial and fungal community diversity potentially mediated by sex, emphasizing the importance of combining biological and ecological factors in microbiota analyses. • Chapter 3: Provides the first comprehensive identification of bacterial and fungal communities in the Sanje mangabey (Cercocebus sanjei), an endangered species endemic to the Udzungwa Mountains. The chapter explores the potential contamination of gut microorganisms by soil microorganisms, highlighting a significant transfer of soil saprotrophic fungal communities to the gut. This suggests an association between geophagy— a behavior of consuming soil observed in the Sanje mangabey—and its highly frugivorous diet. Geophagy is an adaptive behavior that may have various functions, such as relieving gastrointestinal stress. Our results suggest that these benefits may be partly determined by the presence of microorganisms in the soil and their interaction with the gut microbiome. • Chapter 4: Investigates the diversity of metabolic functions of gut microorganisms in two non-human primate hosts, the Udzungwa red colobus (Piliocolobus gordonorum) and yellow baboons, using in-depth sequencing methods (metagenomics). The study found a strong association between observed metabolic functions and the distinct diets of the hosts. Differences in metabolic functions were also observed within the same host species living in contrasting habitat types, suggesting that specialized diets may be more affected by habitat differences than generalist diets. Overall, this thesis provides a comprehensive view of the complexity of gut microbial variations in terms of structure, composition, and metabolic functions. It emphasizes the importance of studying these variations in wild host species, which are often threatened by human activities. These findings can improve awareness in the field of conservation and aid in developing strategies to mitigate the effects of anthropogenic disturbance

    The widespread manufacturing of nanomaterials and nano-enabled products (NEPs) has led to concerning human and environmental exposure levels. The lack of knowledge regarding the occupational, consumer, and environmental potential adverse effects of this burgeoning technology has ignited serious safety issues and highlighted the importance of a comprehensive risk evaluation. Safety and Sustainability by Design (SSbD) is a concept that thanks to a prevention approach aims to reduce the hazards at the early stages of product development. The combined implementation of safety measures and risk assessment will have to meet regulatory infrastructures to deliver SSbD guidelines and tools. To reach this goal it will be crucial to monitor the effects of these products and their manufacturing processes on human and environmental health. Structure-properties correlations must be studied to determine general guidelines and deliver safer and more sustainable design alternatives to the actual nanomaterials. A proper design of the synthesis step directly affects the structure of the nanomaterials allowing to building of the desired physicochemical, functional, and safety profiles. In this context, the design or re-design of a nanomaterial is an early-stage measure that can effectively improve the safety and sustainability of the product creating a bridge between design space and functional properties. A full life cycle assessment is required to link the nanomaterials design step to the nano-enabled products manufacturing, use phase, and end of life. To contribute to this research field, during this work, three main topics were assessed: the synthesis of antimicrobial silver nanoparticles and their implementation into several case studies, the synthesis of gold-platinum nanoparticles for application as a catalyst in biomass valorization, and the production of biopolymeric scaffolds embedding natural-derived active phases for the wastewater remediation. Three AgNPs synthesis methods using different capping agents were studied: - Quaternized hydroxyethyl cellulose is a natural-derived biopolymer with positive charged quaternary ammonium groups that confers antibacterial activity to this compound. - Curcumin is a natural-derived organic molecule with intrinsic antimicrobial and antioxidant properties. - Cyclic lipopeptide biosurfactant derived from bacterial metabolism which possesses antimicrobial properties thanks to its cell wall lysis capability. A wide-ranging physicochemical characterization (TEM, XRD, ICP-OES, UV-Vis, DLS, ELS) was performed to correlate synthesis parameters to physicochemical and functional properties. The so-obtained AgNPs were tested against Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus and Listeria innocua) bacteria, demonstrating excellent antibacterial activity against all the tested bacterial strains. Antiviral tests were performed against the SARS-CoV-2 enveloped virus and BK polyomavirus non-enveloped virus, proposing an antiviral mechanism of action of AgNPs involving the interaction of AgNPs with the viral envelope inhibiting the attachment to cell’s receptors. The most active AgNPs variants were selected for the implementation into nano-enabled products within different case studies: antimicrobial textiles, paper, and biopolymeric film and scaffolds. The antimicrobial properties were successfully transferred from the nanomaterial to the nano-enabled product. Thanks to these case studies workers, users, and environmental exposures were assessed. NEPs antibacterial efficacy and product durability were evaluated. The Safe and Sustainable by Design approach was applied for the synthesis of gold-platinum nanoparticles catalysts. The green chemistry principles were implemented on multiple levels, first for the safe and sustainable synthesis of the nanoparticles and then in their application for biomass valorization by converting biomass wastes into chemical building blocks. Two different gold-platinum nanoparticles structures were studied core-shell (gold core and a platinum external shell) and alloy. The correlation between the structure and the catalytic activity of these materials was studied thanks to an in-depth physicochemical characterization (TEM, XRD, ICP-OES, UV-Vis, DLS, ELS). The catalytic activity was evaluated in the model reaction of hydrogenation of nitrophenol to aminophenol. Synergistic effects were observed for the bi-metallic nanoparticles, thanks to which a reduced load of platinum allows excellent catalytic activity. The increasing contamination of hydric resources caused by human activities has highlighted the necessity to develop new technologies for wastewater remediation. From a circular economy perspective, natural adsorbent materials were selected for this purpose. Clays and hydrotalcites share a layered structure able to host counterions, the deriving ion exchange capability has been efficiently exploited for the adsorption of heavy metals and organic pollutants. The selected adsorbents were characterized by XRD, BET, and ELS, subsequently, they were tested in the adsorption of simulated pollutants, namely the cationic dye rhodamine B, anionic dye methyl orange, and copper (II) cation. Clays and hydrotalcites were embedded into renewable seaweed-derived biopolymeric scaffolds made of k-carrageenan, chitosan, or agarose to improve their handleability. The pollutants adsorption tests were replicated on the scaffolds and the resulting adsorption process kinetic followed the pseudo-second-order model

    Nel corso degli anni, sono stati sviluppati vari antibiotici con diversi meccanismi d’azione. Tuttavia, la maggior parte di questi sono soggetti ad antibiotico-resistenza, specialmente se il meccanismo d’azione è basato sull’inibizione enzimatica. Ad oggi è appurato che un meccanismo d’azione aspecifico, come ad esempio la distruzione fisica della membrana, sia meno suscettibile alla resistenza. Questo effetto si può ottenere tramite diverse molecole, inclusi i peptidi antimicrobici (AMPs). Nonostante il loro caratteristico meccanismo d’azione, l’attività antimicrobica dei peptidi antimicrobici e limitata, specialmente perché suscettibili all’azione proteolitica di enzimi endogeni o batterici. Per ovviare alla degradazione proteolitica e incrementare le conoscenze relative a questi agenti antibatterici, abbiamo pianificato la sintesi di una libreria di peptidomimetici antimicrobici che possano bypassare le criticità relative a questa classe di molecole, come la loro instabilità metabolica. Inoltre, il nostro obiettivo e lo sviluppo di composti che non solo mimino la struttura peptidica ma che posseggano anche proprietà membrano litiche simili a quelle degli AMPs, favorendo così la diretta distruzione della membrana batterica. La struttura molecolare di riferimento per la progettazione di queste molecole è rappresentata dalla Brilacidina, una molecola mimetica delle defensine. Tale molecola è risultata attiva contro S. aureus, Enterococcus faecium, E. Coli. Ispirandoci alla struttura della Brilacidina e alla significativa letteratura a riguardo, abbiamo identificato un ipotetico farmacoforo per la sintesi di analoghi strutturalmente correlati. La modifica delle braccia laterali è cruciale per ottenere la specifica conformazione richiesta per l’inserimento nel doppio strato fosfolipidico. Prima di continuare la sintesi di altri analoghi, abbiamo focalizzato la nostra attenzione in un’ampia caratterizzazione biologica delle molecole sintetizzate. Conseguentemente, le molecole sono state testate su un ampio numero di patogeni clinicamente rilevanti dimostrato incoraggianti risultati e analisi preliminari sul meccanismo d’azione hanno confermato l’effetto membranolitico ipotizzato.Over the years, many antibiotics with different mechanisms of action have been developed. However most of them succumb to antimicrobial resistance, especially when their mechanism of action depends on enzymatic inhibition. Nowadays it is recognized that a non-specific mechanism, such as physical disruption of the membrane, is less susceptible to resistance. This can be achieved through specific molecules, including antimicrobial peptides (AMPs). Despite their unique mode of action, the antimicrobial activity of AMPs is limited, primarily due to their high susceptibility to proteolytic degradation by endogenous or microbial enzymes. To counteract proteolytic degradation and to improve the knowledge around these antibacterial agents, we planned to synthetize a library of antimicrobial peptides mimetics overcoming specific weaknesses related to this class of molecules, such as their metabolic instability. Additionally, our goal is to develop compounds that not only mimic peptides but also possess membranolytic properties similar to AMPs, enabling direct disruption of the bacterial membrane. The molecular template for the development of our library of peptidomimetics compounds is represented by Brilacidin which is a defensin-mimetic molecule. This molecule was showed to have activity against S. aureus, Enterococcus faecium, E. Coli. Taking inspiration by the Brilacidin structure and relevant literature, we identified a hypothetical pharmacophore for synthesizing structurally related analogues. The modification of side arm lengths is crucial to achieving the specific conformation required for perpendicular insertion into phospholipid bilayers. Before synthesizing other analogues, we focused my interest on the comprehensive biological characterization of the molecules synthesized.     Gli antibiotici sono stati scoperti negli anni ‘30 del 1900 e, a partire da questi anni è aumentato esponenzialmente l’utilizzo di queste molecole farmaceutiche per la terapia di malattie infettive. Sono stati scoperte nuove molecole antibiotiche e ora vi sono diverse classi antibiotiche disponibili sul mercato (anche se i nuovi farmaci risultano insufficienti rispetto alle esigenze mondiali) . La principale conseguenza dell’uso massivo degli antibiotici è stato che i batteri hanno sviluppato e diffuso delle caratteristiche genetiche di resistenza agli antibiotici; infatti, al giorno d’oggi l’antibiotico resistenza rappresenta una delle maggiori problematiche sanitarie e veterinarie a livello globale rappresenta quella che potremmo definire la vera “pandemia” di questo secolo. La resistenza antibiotica nei microrganismi può avere due origini: resistenza intrinseca, ossia il batterio è resistente naturalmente alle molecole farmaceutiche; oppure resistenza acquisita, ovvero, il batterio recepisce la caratteristica di resistenza agli antibiotici attraverso l’inglobamento di nuovo materiale genetico, o grazie a mutazioni cromosomiche proprie del batterio. Le modalità con cui i microrganismi possono scambiare materiale genetico sono principalmente tre: trasformazione, trasduzione e coniugazione. A livello mondiale esiste un monitoraggio continuo dei batteri antibiotico resistenti e, l’Istituto Superiore di Sanità, in Italia, gestisce il programma di sorveglianza dell’Antibiotico Resistenza AR-ISS. In questo documento sono stati identificati e classificati i batteri patogeni antibiotico resistenti presenti a livello ospedaliero, i quali appartengono alle seguenti 8 specie: Staphylococcus aureus, Streptococcus pneumoniae, Enterococcus faecalis, Enterococcus faecium, Escherichia coli, Klebsiella pneumoniae complex, Pseuodomonas aeruginosa, Acinetobacter species. Il tema dell’antibiotico resistenza è molto sentito anche nell’ambito veterinario, ed in particolare, negli allevamenti zootecnici, nei quali gli antibiotici vengono ampiamente sfruttati, a volte anche per prevenire malattie infettive negli animali. È fondamentale, in questo settore, che si inizino a seguire delle procedure e delle misure di igiene e biosicurezza tali da poter diminuire la somministrazione di farmaci agli animali. Infine, anche l’ambiente e gli ecosistemi risentono dell’influenza degli antibiotici, poiché a causa dello spargimento dei reflui animali o dello scarico delle acque delle industrie farmaceutiche, gli antibiotici raggiungono l’ambiente, influendo sulla proliferazione e la crescita della flora. In questa cornice interdisciplinare, a livello italiano e a livello mondiale, sono stati sviluppati dei programmi di controllo e uso responsabile degli antibiotici (PNCAR e Antimicrobial Stewardship) che coinvolgono tutti gli operatori dei settori connessi alla problematica e hanno lo scopo di minimizzare l’utilizzo degli antibiotici. La prospettiva da mettere in campo corrisponde senza alcun dubbio all’approccio One Health, ossia dobbiamo considerare la salute come un equilibrio tra salute umana, animale e ambientale che sono strettamente interconnessi e si influenzano vicendevolmente.Antibiotics were discovered around the 1930s, and since then, their use for the treatment of infectious diseases has increased exponentially. New antibiotics have been discovered, and today, there are various antibiotic classes available on the market. The main consequence of the widespread use of antibiotics is that bacteria developed and disseminated genetic traits of antibiotic resistance; indeed, nowadays, antibiotic resistance represents one of the most significant human health and veterinary challenges on a global scale. Antibiotic resistance in microorganisms can have two origins: intrinsic resistance, where the bacterium is naturally resistant to pharmaceutical compounds, or acquired resistance, where the bacterium acquires resistance traits through the uptake of new genetic material or through its own chromosomal mutations. The mechanisms by which microorganisms can exchange genetic material are mainly three: transformation, transduction, and conjugation. Globally, there is continuous monitoring of antibiotic-resistant bacteria. In Italy, the Istituto Superiore di Sanità manages the Antibiotic Resistance surveillance program AR-ISS. This program has identified and classified antibiotic-resistant pathogenic bacteria found in hospitals, belonging to the following eight species: Staphylococcus aureus, Streptococcus pneumoniae, Enterococcus faecalis, Enterococcus faecium, Escherichia coli, Klebsiella pneumoniae complex, Pseudomonas aeruginosa, and Acinetobacter species. The issue of antibiotic resistance is also highly relevant in the veterinary field, particularly in livestock farming, where antibiotics are widely used, sometimes even to prevent infectious diseases in animals. It is crucial in this sector to begin following hygiene and biosecurity procedures to reduce the administration of drugs to animals. Moreover, the environment and ecosystems are affected by antibiotics due to the spread of manure or the discharge of wastewater from pharmaceutical industries. These antibiotics reach the environment, impacting the proliferation and growth of flora. In this interdisciplinary context, both in Italy and worldwide, control programs and responsible use of antibiotics (such as PNCAR and Antimicrobial Stewardship) have been developed, involving all stakeholders related to this issue. These programs aim to minimize the use of antibiotics. The approach to be adopted is the One Health perspective, which considers health as a balance between human, animal, and environmental health, all of which are closely interconnected and influence each other

In partcular we are interested in the energy of models such as Peridynamic, Micromagnetism, Elasticicty and many other. It is well known that Calculus of Variations is a key tool in order to understand these models. Our aim is to show some examples of how classical results of Calculus of Variations and Functional Analysis such as the Direct Method of Calculus of Variations and Gamma-convergence can be used to study these energies.In questa tesi studiamo le proprietà di alcuni funzionali relativi alla Scienza dei Materiali. In particolare siamo interessati dall'energia di modelli come la Peridinamica, il Micromagnetismo, l'Elasticità e molti altri. E' risaputo che il Calcolo delle Variazioni è uno strumento fondamentale per lo studio di questi modelli.
