    .Sex determination in prepuberal jewelled lizards (Timon lepidus). clinical and conservational implications

    La lucertola ocellata (Timon lepidus) è il più grande sauro d’Europa, attualmente la specie risulta essere minacciata e la popolazione sul territorio italiano è in continua diminuzione. In letteratura, al momento, esistono pochi studi riguardanti la determinazione del sesso in esemplari immaturi del genere Timon e non esistono lavori riguardanti l’identificazione del sesso in T. lepidus. Il dimorfismo sessuale delle lucertole ocellate è evidente a circa un anno di età, difficilmente è possibile riconoscere fenotipicamente il sesso degli esemplari subadulti basandosi solamente sulle caratteristiche morfologiche del soggetto. Per questo motivo, in questo lavoro di tesi, abbiamo indagato la possibilità di utilizzare l’ecografia come tecnica diagnostica precoce in grado di visualizzare, misurare e valutare morfologicamente le gonadi in esemplari immaturi nati in cattività. Per la realizzazione di questo studio sono stati inclusi sette esemplari di Timon lepidus e sottoposti ad esame ecografico a due mesi di età (T0), quattro mesi di età (T1) e a sei mesi di età (T2). Le gonadi sono state identificate a livello di celoma caudale, caudalmente al fegato e cranialmente al polo craniale renale, adese alla parete dorsale della cavità celomatica, lateralmente all’aorta, medialmente e craniali ai corpi adiposi. A T0 e T1, non è stato possibile distinguere morfologicamente le ovaie dai testicoli. La prima differenza statisticamente significativa si è notata a T2 dove le gonadi sono state identificate come strutture bozzellate nelle femmine e lisce nei maschi. I risultati di questo studio preliminare risultano interessanti in quanto è stato possibile identificare delle differenze morfologiche nelle gonadi dei soggetti studiati mesi prima dello sviluppo dei caratteri sessuali secondari. Il sessaggio precoce ecografico è uno strumento non invasivo utile non solo a fini conservazionistici, ma anche commerciali. In questo modo è possibile riconoscere e gestire le coppie prevenendo conflitti tra esemplari del medesimo sesso e formare gruppi riproduttivi precocemente, prima dell’anno di età. I risultati preliminari di questo lavoro sono quindi essenziali al fine di una possibile reintroduzione in natura di esemplari nati in cattività, tutelando le popolazioni autoctone tramite reintroduzioni e contrastando il prelievo in natura a fini collezionistici.The Jewelled lizard (Timon lepidus) is the largest lizard in Europe. Currently, the species is threatened, and its population is continuously decreasing in Italy. In the literature, there are few studies on sex determination in immature specimens of the genus Timon, and no research exists on sex identification in T. lepidus. Sexual dimorphism in ocellated lizards becomes evident at about one year of age, and it is challenging to phenotypically identify the sex of subadult specimens based solely on their morphological characteristics. For this reason, in this thesis, we investigated the possibility of using ultrasound as an early diagnostic technique to visualize, measure, and morphologically assess the gonads in immature captive-born specimens. Seven Timon lepidus specimens were included in this study and underwent ultrasound examination at two months of age (T0), four months of age (T1), and six months of age (T2). The gonads were identified at the level of the caudal coelom, caudally to the liver and cranially to the cranial renal pole, adhering to the dorsal wall of the coelomic cavity, laterally to the aorta, and medially and cranially to the fat bodies. At T0 and T1, it was not possible to morphologically distinguish ovaries from testes. The first statistically significant difference was observed at T2, where gonads were identified as nodular structures in females and smooth in males. The results of this preliminary study are interesting because morphological differences in the gonads of the studied subjects were identified six months before the development of secondary sexual characteristics. Early ultrasound sexing is a non-invasive tool useful not only for conservation purposes but also for commercial ones. This method allows for the recognition and management of pairs, preventing conflicts between specimens of the same sex and forming breeding groups early, before reaching one year of age. Therefore, the preliminary results of this work are crucial for the potential reintroduction of captive-born specimens into the wild, protecting native populations through reintroductions and countering wild collection for commercial purposes

    Study of the electroencephalographic signal spectrum in relation to motor sequences learning based on Action Observation Training

    Evidenze sempre crescenti in ambito neuroscientifico suggeriscono come integrare l’osservazione di azioni all’interno di pratiche di apprendimento motorio possa essere un elemento decisivo nell’accelerare e migliorare la prestazione. All’interno di un ampio studio volto a indagare i correlati neurali e cinematici dell’apprendimento motorio integrato attraverso Action Observation Training (AOT), sono state condotte registrazioni elettroencefalografiche (EEG) su soggetti sani, prima e dopo un periodo di due settimane di training, mentre gli stessi osservavano le sequenze motorie da apprendere, con l’obiettivo di identificare i correlati EEG dell’apprendimento tramite AOT. Il contributo fornito da questo elaborato di tesi riguarda l’acquisizione e analisi di dati elettroencefalografici, svolta su 25 soggetti, finalizzata a valutare se vi sono specificità nella modulazione del segnale EEG in relazione all’esperienza motoria acquisita mediante il metodo AOT, confrontando questi dati con quanto riscontrato in relazione all’apprendimento tramite esercizio motorio e in relazione ad atti motori all’interno del repertorio motorio. I risultati dello studio mostrano come l’esperienza motoria acquisita durante l’apprendimento impatta significativamente sul segnale EEG in diverse bande di frequenza: la banda alfa mostra una modulazione legata all’esperienza motoria indipendente dalla tipologia di training, mentre la sincronizzazione della banda theta nella regione parietale destra, associata al metodo AOT e al follow-up, suggerisce un possibile vantaggio dell’AOT nella generalizzazione a nuove sequenze. I risultati provenienti da questa analisi evidenziano una specificità del metodo AOT sui correlati EEG dell’apprendimento motorio. Sebbene questi risultati debbano ancora essere integrati i dati di cinematica articolare per valutare le relazioni reciproche di questi segnali, essi aprono alla possibilità di monitorare indirettamente i percorsi di apprendimento motorio tramite valutazioni integrate del comportamento motorio e della reattività dei rispettivi segnali neurali.Increasing evidence in the field of neuroscience suggests that integrating action observation into motor learning practices can be a crucial factor in accelerating and enhancing performance. Within a comprehensive study aimed at investigating the neural and kinematic correlates of integrated motor learning through Action Observation Training (AOT), electroencephalographic (EEG) recordings were conducted on healthy subjects before and after a two-week training period, during which they observed motor sequences to be learned. The objective was to identify EEG correlates of learning through AOT. The contribution of this thesis lies in the acquisition and analysis of electroencephalographic data from 25 subjects, aimed at evaluating whether there are specific modulations in the EEG signal related to motor experience acquired through the AOT method, compared with findings related to learning through motor exercise and motor acts within the motor repertoire. The study results demonstrate that motor experience acquired during learning significantly impacts EEG signals across different frequency bands. Specifically, the alpha band shows modulation linked to motor experience independent of the type of training, while synchronization of the theta band in the right parietal region, associated with the AOT method and follow-up, suggests a potential advantage of AOT in generalizing to new sequences. These findings highlight a specificity of the AOT method in influencing EEG correlates of motor learning. While further integration with assessments of joint kinematics is necessary to evaluate the mutual relationships of these signals, they offer the possibility of indirectly monitoring motor learning pathways through integrated assessments of motor behavior and the reactivity of respective neural signals

    Evaluation of intestinal microbioma before and after therapy in "Inflammatory Bowel Disease" affected horses

    The equine gastrointestinal (GI) tract functions through the support of a complex and diverse population of microorganisms known as the intestinal microbiota. Microbiota plays an essential role in maintaining animal health, contributing to digestion, metabolism, and protection against pathogens, thus directly and indirectly influencing the host's development and growth. Typically, microbiome assessments in horses rely on fecal samples avoiding invasive procedures that involves intestinal samples. While fecal samples provide a good reflection of microbial communities in the large and small colon, they are less indicative of the proximal intestinal regions. Despite the known importance of gut microbial communities, there is limited knowledge about the differences in microorganisms' relative abundances between a “normal” and “abnormal” microbiome. Moreover, define “normal” or “healthy” microbiome in horses is challenging due to its complexity and the variable nature of the GI environment. Dysbiosis (or microbial imbalance) is linked to conditions such as acidosis, diarrhea, colitis, colic, laminitis, equine metabolic syndrome, and equine grass sickness. However, it remains unclear whether dysbiosis is a cause or consequence of disease, making it difficult to establish clear relationships. Chronic inflammatory bowel diseases (IBD) are recurrent GI tract disorders observed in various animal species, including horses. IBD is characterized by persistent inflammation, with immune reactions triggered by unidentified antigens like bacteria, viruses, fungi, parasites, or dietary elements. Despite extensive research, the exact pathogenesis of IBD in horses remains unclear. However, it is well understood that decreased microbial diversity and growth of pathogenic bacteria are associated with GI illnesses. However, the cause-and-effect relationship between microbiota changes and IBD in horses is still poorly understood, with conflicting data in current literature. Probiotics and prebiotics have been suggested as potential therapeutic aids in managing colitis and other GI disorders in animals including IBD. However, the scientific evidence supporting the efficacy of commercial prebiotic formulations in horses remains limited. While in vitro studies may show promising results, in vivo evidence is harder to obtain. This study aims to assess the therapeutic effects of a commercial prebiotic product, FiberPlex (Scandinavia s.r.l.), to treat microbiota dysbiosis caused by IBD in horses. The prospective clinical trial included 14 horses with a presumptive diagnosis of IBD. Fecal samples were collected at three time points (T0, T1, and T2) to evaluate the microbiome before (T0) and after treatment (T1, T2). At T0, analysis revealed mild dysbiosis, with reduced bacterial diversity and the presence of pathogenic bacteria. Despite some bacterial families maintain crucial roles in digestion, an imbalance persisted. At T1, following treatment and prebiotic administration, notable reductions in pathogenic bacteria from the phyla Proteobacteria and Actinobacteria were observed. However, beneficial bacteria from the phylum Firmicutes were also reduced, indicating persistent dysbiosis. A persistent treatment (T2) allowed to a microbiome overall adequacy with an improved Shannon index from T1, indicating an initial restoration in diversity. However, Shannon Index remain lower than T0 and lower than healthy horses index suggesting persistent imbalances. In fact, the absence of beneficial bacteria like Akkermansia muciniphila and Fecalibacterium prausnitzi remained, suggesting that full microbiome recovery may require further intervention or longer prebiotic administration. In summary, while the use of prebiotics showed some promising results in restoring balance to the gut microbiome in horses with IBD, significant imbalances remained after treatment, highlighting the complexity of equine GI health and the need for further research.Il corretto funzionamento dell’apparato gastrointestinale (GI) degli equini è garantito dal supporto di una popolazione complessa e diversificata di microrganismi nota come microbiota intestinale. Il microbiota svolge un ruolo essenziale nel mantenimento della salute animale, contribuendo alla digestione, al metabolismo e alla protezione contro i patogeni, influenzando così direttamente e indirettamente lo sviluppo e la crescita dell'ospite. Tipicamente, le valutazioni del microbioma nei cavalli si basano su campioni fecali per evitare procedure invasive che comportano il prelievo di campioni intestinali. Sebbene i campioni fecali forniscano una buona percezione delle comunità microbiche presenti nel grosso e piccolo colon, sono meno indicativi delle regioni intestinali prossimali. Nonostante l'importanza riconosciuta delle comunità microbiche intestinali, esistono poche conoscenze sulle differenze nelle abbondanze relative dei microrganismi tra un microbioma "normale" e uno "anormale" . Inoltre, definire un microbioma "normale" o "sano" nei cavalli è ad oggi una sfida, data la complessità e la natura variabile dell'ambiente GI. La disbiosi, o squilibrio microbico, è legata a condizioni come acidosi, diarrea, colite, colica, laminite, sindrome metabolica equina e malattia dell'erba equina. Tuttavia, non è chiaro se la disbiosi sia una causa o una conseguenza della malattia, il che rende difficile stabilire relazioni chiare. Le malattie infiammatorie croniche intestinali (IBD) sono disturbi ricorrenti del tratto GI osservati in varie specie animali, inclusi i cavalli. L'IBD è caratterizzata da un'infiammazione persistente, con reazioni immunitarie innescate da antigeni non identificati come batteri, virus, funghi, parassiti o elementi dietetici. Nonostante numerose ricerche, la patogenesi esatta dell'IBD nei cavalli rimane poco chiara. Tuttavia, è noto che una diminuzione della diversità microbica e la crescita di batteri patogeni siano associati a malattie GI. La relazione causa-effetto tra le variazioni del microbiota e l'IBD nei cavalli è ancora poco compresa, con dati contrastanti nella letteratura attuale. Probiotici e prebiotici sono stati suggeriti come potenziali coadiuvanti terapeutici nella gestione della colite e di altri disturbi gastrointestinali negli animali, tra cui l’IBD. Tuttavia, le prove scientifiche a supporto dell'efficacia delle formulazioni commerciali di prebiotici nei cavalli sono ancora limitate. Sebbene gli studi in vitro possano mostrare risultati promettenti, le prove in vivo sono più difficili da ottenere. Questo studio ha l'obiettivo di valutare gli effetti terapeutici sul microbiota intestinale di un prodotto prebiotico commerciale, FiberPlex (Scandinavia s.r.l.), per il trattamento delle disbiosi intestinali associate all’IBD . Lo studio clinico prospettico ha incluso 14 cavalli con diagnosi presunta di IBD. I campioni fecali sono stati raccolti in tre momenti (T0, T1 e T2) per valutare il microbioma prima (T0) e dopo il trattamento (T1, T2). A T0, l'analisi ha rivelato una lieve disbiosi, con ridotta diversità batterica e la presenza di batteri patogeni. Nonostante fossero presenti alcune delle famiglie batteriche che hanno ruoli cruciali nella digestione, persisteva uno squilibrio. A T1, dopo il trattamento, sono state osservate riduzioni significative dei batteri patogeni appartenenti ai Phyla Proteobacteria e Actinobacteria. Tuttavia, anche i batteri benefici del phylum Firmicutes erano presenti in percentuale ridotta, indicando persistente disbiosi. Un trattamento prolungato (T2) ha portato a un'adeguatezza complessiva del microbioma con un indice di Shannon migliorato rispetto a T1, indicando un iniziale ripristino della diversità microbica. Tuttavia, l'indice di Shannon rimane inferiore a T0 e al valore di riferimento dei cavalli sani, suggerendo squilibri persistenti. Infatti, l'assenza di batteri benefici come Akkermansia muciniphila e Fecalibacterium prausnitzii persiste, suggerendo che il pieno recupero del microbioma potrebbe richiedere ulteriori interventi o una somministrazione prolungata del prebiotico

    This thesis investigates the nature and functions of visual behavioural patterns exhibited by domestic dogs (Canis lupus familiaris), with a specific focus on displacement behaviours and facial expressions. The first part of the work scrutinizes existing studies on domestic dogs' displacement behaviours and facial expressions, elucidating their association with physiological indicators of stress, their role as indicators of positive and negative emotions, and their communicative function during social interactions. The experimental inquiries explore domestic dogs' behavioural and hormonal responses to experimentally induced states of positive anticipation and frustration, with the presence or absence of an intra and interspecific social partner (audience effect). Cortisol levels associated with some displacement behaviours hint at a connection with negative valence arousal, while the presence of an audience generally modulated the behavioural patterns, suggesting a communicative function. Furthermore, different audience types (conspecifics or human partners) influenced the characteristics of the displays exhibited, confirming that this putative communicative signals could be shaped by natural selection based on the characteristics of the receivers. Further investigation evaluates the appeasement signals hypothesis of displacement behaviours, examining their role in mitigating potentially aggressive encounters. Findings suggest that putative appeasement signals are associated with a non-aggressive attitude in the sender but are not exclusive of a conflict ridden context. Finally, the research also explores the use of video stimuli in studying intraspecific dog communication, demonstrating dogs' ability to respond with socially appropriate behaviours to videos of conspecifics performing different communicative signals. Displacement behaviours are accentuated in response to neutral conspecifics, underscoring their link with potentially ambiguous social contexts rather than overt threat. In conclusion, this research enhances our understanding of domestic dogs' communication by elucidating the intricate interplay between displacement behaviours, facial expressions, and emotional states. The findings underscore the important role of social context and audience presence in shaping communicative signals, providing valuable insights into the complex dynamics of canine communication. This comprehensive exploration opens avenues for further research into the evolutionary underpinnings and functional significance of canine behavioural displays in diverse social contexts

    Behavioural strategies in response to frustration in the first year of life : the Barrier Task test in a cohort of the Life-MILCH project

    Il presente studio si propone di indagare l'impatto di variabili quali sesso, frequenza dell’asilo nido, uso del ciuccio, allattamento al seno e temperamento percepito dai genitori rispetto all’output comportamentale in un compito di induzione della frustrazione. Il paradigma utilizzato è quello del Barrier Task test, applicato a 29 bambini a 12 mesi di età, facenti parte del progetto europeo Life MILCH. I risultati suggeriscono un’influenza della condizione sperimentale sui diversi comportamenti analizzati, evidenziando una peculiarità per i bambini che non utilizzano il ciuccio e quelli allattati al seno, caratterizzati da una maggior durata dei comportamenti comunicativi all’interno della fase di induzione della frustrazione. Sembrerebbe esservi inoltre la tendenza per i bambini allattati al seno, di adottare comportamenti di etero-regolazione di durata minore, nonostante le differenze con i bambini non allattati non siano significative. I risultati ottenuti dal presente studio contribuiscono a una comprensione preliminare dei comportamenti regolatori e socio-emozionali dei bambini in una fase precoce dello sviluppo, suggerendo come alcune variabili potrebbero giocare un ruolo nell’influenza dei comportamenti in situazioni di frustrazione.The present study aims to investigate the impact of variables such as sex, nursery attendance, use of the pacifier, breastfeeding and the temperament perceived by parents in relation to the behavioral output in a task that induces frustration. The paradigm used is the Barrier Task test, applied to 29 children aged 12 months, who are part of the European project Life MILCH. Results suggest an influence of the experimental condition on the different behaviors analysed, highlighting a peculiarity for children which do not use the pacifier and those breastfed, characterized by a longer duration of communicative behaviors within the frustration induction phase.There would also seem to be a tendency for breast-fed children to adopt behaviors of hetero-regulation of shorter duration, although the differences with non-breastfed children are not significant. The results obtained from this study contribute to a preliminary understanding of the regulatory and socio-emotional behaviors of children in a early stage of development, suggesting how some variables might play a role in the influence of behaviour in situations of frustration

    Multicomponent non-pharmacological approaches to managing subjective cognitive decline (SCD) and mild cognitive impairment (MCI) : integrated interventions for mental health and physical well-being

    L'incremento della prevalenza di patologie neurodegenerative ha suscitato un interesse crescente per gli approcci terapeutici multicomponente destinati a soggetti con Subjective Cognitive Impairment (SCD) e con Mild Cognitive Impairment (MCI). La revisione bibliografica presentata in questo elaborato analizza e sintetizza le evidenze attuali sugli interventi multicomponente (MNPI), che spaziano dall'esercizio fisico alla stimolazione cognitiva e alla gestione del benessere psicologico, e sono finalizzati a migliorare le funzioni cognitive e il benessere psicologico in questi gruppi di pazienti. I risultati indicano che gli approcci multicomponente possono favorire miglioramenti significativi nel funzionamento cognitivo generale, nella memoria, nell’attenzione, nelle funzioni esecutive e nella qualità della vita, sebbene siano necessarie ulteriori ricerche per determinarne l'efficacia a lungo termine. La revisione rappresenta la base empirica del progetto SACS (Salute, Autonomia, Connessione Sociale). Un MNPI che integra la riabilitazione cognitiva, l’esercizio fisico e le attività sociali per rallentare la progressione dei sintomi in soggetti anziani cognitivamente fragili. A tal proposito, sarà presentato un caso clinico, attraverso il quale si evincerà l'importanza di un intervento personalizzato e olistico nella gestione della compromissione cognitiva.The increasing prevalence of neurodegenerative diseases has sparked growing interest in multicomponent therapeutic approaches for individuals with Subjective Cognitive Impairment (SCD) and Mild Cognitive Impairment (MCI). This review analyzes and synthesizes current evidence on multicomponent non-pharmacological interventions (MNPI), which range from physical exercise to cognitive stimulation and psychological well-being management. These interventions aim to improve cognitive functions and psychological well-being in these patient groups. The findings indicate that multicomponent approaches can lead to significant improvements in overall cognitive functioning, memory, attention, executive functions, and quality of life, although further research is needed to determine their long-term efficacy. The review serves as the empirical foundation for the SACS project (Health, Autonomy, Social Connection), an MNPI that integrates cognitive rehabilitation, physical exercise, and social activities to slow the progression of symptoms in cognitively frail elderly individuals. In this context, a clinical case will be presented to highlight the importance of a personalized and holistic intervention in managing cognitive impairment

    Some geometric estimates for fractional Poincaré inequalities

    This thesis is devoted to study the following two problems: giving geometric lower bounds for the first eigenvalue of the fractional Dirichlet-Laplacian of order ss; determining the sharp constant for the fractional Hardy inequality. The two problems are tightly connected: indeed, for an open set having finite inradius, every lower bound on the sharp Hardy constant immediately translates into a lower bound for the first eigenvalue. \par For the first problem, we prove a geometric lower bound in terms of the {\it inradius} of the set, in the case of open planar sets having nontrivial topology. This is valid for 1/2<s<11/2<s<1 only, and we show that this condition is sharp. Moreover, by constructing suitable counter-examples, we prove that our lower bound is optimal, in many respects. The result is obtained through some non-trivial adaptions to the fractional case of techniques employed by Hayman and Taylor, to handle the case of the usual Laplacian operator. In adapting these techniques, we will develop some technical tools, which are interesting in themselves, in the context of fractional Sobolev spaces. \par For the second problem, we determine the sharp constant in the fractional Hardy inequality for open convex sets in every dimension. This is done by constructing suitable local weak supersolutions with geometric content, to the relevant Euler-Lagrange equation. The latter is a weighted eigenvalue--type equation, containing a negative power of the distance from the boundary. For 1/2s<11/2\le s<1, such supersolution is given by a suitable power of the distance function. The case 0<s<1/20<s<1/2 is much more difficult, since such a construction fails. In this case, we employ a directional decomposition method, which permits to reduce the problem to dimension 11. This technique is due to Loss and Sloane, in the fractional setting. In particular, we can compute the sharp constant in the whole range 0<s<10<s<1. This completes a result which was left open in the literature.In questa tesi studiamo i due seguenti problemi: dare stime geometriche dal basso per il primo autovalore del Laplaciano frazionario di ordine ss con condizioni di Dirichlet; determinare la costante ottima per la disuguaglianza di Hardy frazionaria. Tali problemi sono strettamente connessi: infatti, per un insieme aperto con inradius finito, ogni stima dal basso per la costante ottima di Hardy si può convertire in modo immediato in una stima dal basso per il primo autovalore. Per il primo problema, proviamo una stima geometrica dal basso in termini dell'{\it inradius} dell'insieme, nel caso di aperti del piano con topologia non banale. È essenziale seguire le linee guida standard quando si conducono studi sull'incidenza delle infezioni del sito chirurgico per ottenere dati coerenti tra gli studi. È altresì importante conoscere i fattori di rischio che possono predisporre all'insorgenza di un'infezione del sito chirurgico (SSI). Questa conoscenza consente di agire per prevenire o arginare l'insorgenza di SSI, che è cruciale per migliorare l'esito dei pazienti chirurgici. L'identificazione dei pazienti ad alto rischio di contrarre SSI, l'applicazione di regole asettiche corrette, la stretta sorveglianza dell'incidenza di SSI nella clinica e l'uso moderato di antimicrobici sono la base della corretta gestione del paziente chirurgico. È importante tenere in considerazione che non tutte le infezioni del sito chirurgico richiedono una terapia antimicrobica. Ideally, antimicrobial therapy should only be considered after conducting culture and sensitivity tests to minimize the unjustified use of antimicrobials. The unregulated use of antimicrobials has contributed to the rise in antimicrobial resistance. Therefore, it's ethically crucial for all veterinarians to use antimicrobials more consciously to combat this global issue
