    Exploring the combined anti-cancer effects of sodium butyrate and celastrol in glioblastoma cell lines: a novel therapeutic approach

    PubMed ID:38532150WOS:001191234900001Glioblastoma, a highly aggressive and lethal brain cancer, lacks effective treatment options and has a poor prognosis. In our study, we explored the potential anti-cancer effects of sodium butyrate (SB) and celastrol (CEL) in two glioblastoma cell lines. SB, a histone deacetylase inhibitor, and CEL, derived from the tripterygium wilfordii plant, act as mTOR and proteasome inhibitors. Both can cross the blood–brain barrier, and they exhibit chemo- and radiosensitive properties in various cancer models. GB cell lines LN-405 and T98G were treated with SB and CEL. Cell viability was assessed by MTT assay and IC50 values were obtained. Gene expression of DNA repair, apoptosis, and autophagy-related genes was analyzed by RT-PCR. Cell cycle distribution was determined using flow cytometry. Viability assays using MTT assay revealed IC50 values of 26 mM and 22.7 mM for SB and 6.77 μM, and 9.11 μM for CEL in LN-405 and T98G cells, respectively. Furthermore, we examined the expression levels of DNA repair genes (MGMT, MLH-1, MSH-2, MSH-6), apoptosis genes (caspase-3, caspase-8, caspase-9), and an autophagy gene (ATG-6) using real-time polymerase chain reaction. Additionally, flow cytometry analysis revealed alterations in cell cycle distribution following treatment with SB, CEL and their combination. These findings indicate that SB and CEL may act through multiple mechanisms, including DNA repair inhibition, apoptosis induction, and autophagy modulation, to exert their anti-cancer effects in glioblastoma cells.     Carpal tunnel syndrome prediction with machine learning algorithms using anthropometric and strength-based measurement

    PubMed ID:38630703Objectives Carpal tunnel syndrome (CTS) stands as the most prevalent upper extremity entrapment neuropathy, with a multifaceted etiology encompassing various risk factors. This study aimed to investigate whether anthropometric measurements of the hand, grip strength, and pinch strength could serve as predictive indicators for CTS through machine learning techniques. Methods Enrollment encompassed patients exhibiting CTS symptoms (n = 56) and asymptomatic healthy controls (n = 56), with confirmation via electrophysiological assessments. Anthropometric measurements of the hand were obtained using a digital caliper, grip strength was gauged via a digital handgrip dynamometer, and pinch strengths were assessed using a pinchmeter. A comprehensive analysis was conducted employing four most common and effective machine learning algorithms, integrating thorough parameter tuning and cross-validation procedures. Additionally, the outcomes of variable importance were presented. Results Among the diverse algorithms, Random Forests (accuracy of 89.474%, F1-score of 0.905, and kappa value of 0.789) and XGBoost (accuracy of 86.842%, F1-score of 0.878, and kappa value of 0.736) emerged as the top-performing choices based on distinct classification metrics. In addition, using variable importance calculations specific to these models, the most important variables were found to be wrist circumference, hand width, hand grip strength, tip pinch, key pinch, and middle finger length. Conclusion The findings of this study demonstrated that wrist circumference, hand width, hand grip strength, tip pinch, key pinch, and middle finger length can be utilized as reliable indicators of CTS. Also, the model developed herein, along with the identified crucial variables, could serve as an informative guide for healthcare professionals, enhancing precision and efficacy in CTS prediction. © 2024 Yetiş et al.     Çalışmada, Katılım Öncesi Mali Yardım Aracı kapsamında uygulanan projelerde nihai yararlanıcı olarak ifade edilen belediyelerin proje yönetim döngüsünde üstlendiği görev ve sorumluluklar, Niksar Entegre Su Tesisi Projesi örneği üzerinden incelenmiştir. Çalışmada, Niksar (Tokat) Belediyesi’nde görevli Proje Uygulama Birim Başkanıyla yüz yüze görüşme yapılmıştır. Yarı yapılandırılmış görüşme tekniğiyle elde edilen veriler, betimsel analize tabi tutularak yorumlanmış, temalarına göre gruplandırılarak incelenmiştir. Çalışmada, birincil hukuk kaynakları ile görüşme verilerinin bir sentezi yapılmıştır. Çalışmada, belediyelerin Katılım Öncesi Mali Yardım Aracı yönetiminde programlama, uygulama ve izleme, değerlendirme, raporlama olarak belirtilen aşamalarının hemen hepsinde direkt ya da dolaylı şekilde sorumluluklarının bulunduğu ortaya konulmuştur. Ayrıca belediyelerin proje yönetim sürecinde aracı makam rolü üstlendikleri; sektör sorumlusu kurumlar ile danışman firma arasında, danışman firma ile yüklenici firma arasında bir köprü görevi gördükleri belirtilmelidir. Bununla birlikte belediyelerin projeye ilgilerinin ve sahiplenme seviyelerinin, projenin başlamasından düzgün yürütülmesine ve sonuçlandırılmasına kadar tüm süreçlerde etkili olduğu bu araştırmanın temel çıkarımlarıdır.This study aims to elucidate the roles and responsibilities of municipalities, particularly as the final beneficiaries of projects executed under the Pre-Accession Assistance Instrument, by examining the Niksar Integrated Water Facility project as a case study. The research data were collected through a face-to-face interview with the Head of the Project Implementation Unit of the Niksar (Tokat) Municipality. Using semi-structured interview techniques, the data were subjected to descriptive analysis, interpreted, and thematically organized for a more profound understanding. The study integrates both primary sources and interview data to draw its conclusions. The research reveals that municipalities are intricately involved in nearly all stages of the Instrument for Pre-Accession Assistance management, including programming, implementation, monitoring, evaluation, and reporting. In the project management phases, municipalities serve as pivotal intermediary authorities, facilitating communication between sectoral institutions, consultancy firms, and contractors. Moreover, the study underscores that the level of interest and ownership demonstrated by municipalities significantly influences the success of the project, from its inception through to its effective execution and completion

    Bitcoin price prediction using deep learning and econometric model: lstm and arima

    Dünyada en çok rağbet gören kripto para birimi olması nedeniyle Bitcoin (BTC), yatırımcılar ve araştırmacılar için son yıllarda dikkat çekici hale gelmiştir. Merkezi bir para birimi olmaması ve spekülasyonlara açık olması BTC fiyatında yüksek oynaklığa sebep olmaktadır. BTC fiyatının oynaklığının dikkate alınarak tahminlenmesi özellikle yatırımcılar için büyük önem taşımaktadır. Son yıllarda Makine Öğrenmesi (ML) yöntemlerinin gelişmesiyle birlikte birçok finansal alanda olduğu gibi kripto paraların fiyat tahminlemesinde sıklıkla ML yöntemlerine başvurulmaktadır. ML yöntemleri geleneksel ekonometrik yöntemlerin aksine veri setinde meydana gelen dalgalanmaları herhangi bir varsayıma ihtiyaç duymadan dikkate almakta ve çoğu zaman daha iyi sonuçlar vermektedirler. Bu çalışmada, 01.01.2018 ile 21.12.2023 tarihleri arasında BTC fiyatı geleneksel ekonometrik yöntem olan ARIMA ile ML yöntemi olan LSTM kullanılarak tahminlenmeye çalışılmıştır. Yöntemler karşılaştırılırken performans kriterleri olarak RMSE, MAE ve MAPE kriterleri kullanılmıştır. Çalışmanın sonuçlarına göre LSTM yöntemi en düşük RMSE ve MAPE değerlerine sahip olmuştur.As the most popular cryptocurrency in the world, Bitcoin (BTC) has attracted the attention of investors and researchers in recent years. Its decentralized nature and the exposure to speculation lead to high volatility in the BTC price. Predicting the BTC price taking into account its volatility is of great importance, especially for investors. In recent years, with the development of Machine Learning (ML) methods, ML methods are frequently used in price predicting of cryptocurrencies as in many other financial areas. In contrast to traditional econometric methods, ML methods take into account the fluctuations in the data set without the making any assumptions and often give better results. In this study, the price of BTC between 01.01.2018 and 21.12.2023 is predicted using the traditional econometric method ARIMA and ML method LSTM. While comparing the methods, RMSE, MAE and MAPE criteria were used as performance criteria. According to the results of the study, LSTM method has the lowest RMSE and MAPE values

Approximately 15% of PD patients have a family history of the disease in one or more first-degree relatives, and 5-10% of PD cases exhibit a classical Mendelian inheritance pattern. In 1997, the heritable transmission of PD was first documented. Recent studies have found 90 independent genome-wide signals at 78 loci that may be associated with PD. The identification of genes linked to PD and their functions has uncovered novel biological pathways and treatment options that play a role in the development of PD.     CGI applications in advertising: an examination of brands'CGI advertising applications

    Küreselleşen dünyada artan rekabet ve değişen tüketici beklentileri, markaların başvurmuş oldukları araç, yöntem ve stratejilerde değişimi kaçınılmaz hâle getirmektedir. Bu değişimle birlikte reklamcılık adına tüm aktör ve yapıların, dijitalleşmenin dinamiklerini anlaması ve stratejilerini bu dinamikler çerçevesinde geliştirmesi önem kazanmaktadır. Bu doğrultuda günümüzde CGI teknolojisi, markaların reklamlarını daha yaratıcı ve etkili hâle getirmek için kullandıkları yeni bir yöntem olarak öne çıkmaktadır. Bu araştırmada, markaların dijital platformlarda yayınlamış oldukları CGI reklam uygulamalarının, görsel ve içeriksel açıdan incelenmesi ve markaların CGI reklam kullanım pratiklerinin ortaya konması amaçlanmaktadır. Bu amaç doğrultusunda araştırmada, 20 farklı markanın 27 adet CGI reklam uygulaması içerik analizine ve betimsel analize tabi tutulmuştur. Araştırma sonucunda CGI reklam uygulamalarının büyük bir bölümünün ev eşyası ve elektronik grubunda bulunan ürünlere yönelik olduğu ve reklamlarda bina, sokak ve cadde gibi mekânların kullanımının çoğunlukta olduğu sonucuna ulaşılmıştır. Bununla birlikte reklamların yarısından fazlasında rasyonel çekicilik türüne başvurulduğu ve bu doğrultuda ürün ve hizmete yönelik rasyonel özelliklerin daha çok öne çıkarıldığı gözlemlenmiştir. Reklamlarda görsel etkinin sağlanması için ise ağırlıklı olarak gerçeklik unsuruna başvurulmuştur. Son olarak CGI reklam uygulamalarının içeriksel açıdan zengin ve etkileyici bir yapıda olduğu, bu yönüyle de reklamcılıkta giderek artan bir kullanım alanına sahip olduğu söylenebilir.In the globalizing world, increasing competition and evolving consumer expectations make change inevitable in the tools, methods, and strategies brands resort to. Alongside this change, it becomes crucial for all actors and structures in advertising to understand the dynamics of digitalization and develop their strategies within this framework. In this regard, CGI technology emerges as a new method used by brands to make their advertisements more creative and effective. This research aims to examine the CGI advertising applications published by brands on digital platforms from visual and content perspectives and to reveal brands' CGI advertising usage practices. In accordance with this purpose, 27 CGI advertisements from 20 different brands were examined using content analysis and descriptive analysis techniques in the research. As a result of the research, it was found that a significant portion of CGI advertisements are targeted toward household and electronic products, and the use of settings such as buildings, streets, and avenues is predominant in the advertisements. Moreover, it was observed that more than half of the advertisements resort to rational appeal, emphasizing rational features of the product or service accordingly. Realism is predominantly employed to achieve visual effectiveness in advertisements.     Total diz artroplastisi sonrası hastalara adduktor ve IPACK blok uygulamasının ameliyat sonrası iyileşme ve uyku kalitesi üzerine etkisi: randomize, kontrollü bir çalışma”

    The adductor canal block (ACB) and IPACK (Infiltration of local anesthetic between the popliteal artery and capsule of the knee) block are motor protective blocks that provide effective analgesia and allow early ambulation. The aim of this study was to investigate the effect on postoperative recovery and sleep quality of patients applied with ACB and IPACK for total knee arthroplasty (TKA). Material and Method: This prospective, double-blinded, randomized, controlled study included 80 patients who underwent unilateral TKA under spinal anesthesia. The patients were separated as those applied with ACB+IPACK (Group ACB+IPACK, n=40) and a control group (Group C, n=40). The primary outcome was the postoperative first-day quality of recovery scale (QoR-15). Secondary outcomes were postoperative first and second-day Timed-Up-and-Go (TUG) test and range of motion (ROM), the total amount of rescue opioid drugs required, pain scales at different time points in the first 48 hours postoperatively, sleep evaluation on the postoperative and 6 following nights, and evaluation of sleep quality for 1 month using the Pittsburgh Sleep Quality Index (PSQI). Results: The QoR on postoperative days 1 and 7 were better in the ACB+ IPACK group than in the control group (p= 0.001, p= 0.002, respectively). On postoperative days 1 and 2, the TUG (p= 0.035, p= 0.019, respectively) and ROM (p=0.003, p=0.000) values were higher in the ACB+ IPCAK group. Postoperative opioid consumption was lower in the ACB+IPACK group (p= 0.012). The PSQI values at 1 month postoperatively were similar in both groups (p =0.095). Conclusion: The study results demonstrated that ACB+IPACK applied with ultrasound for TKA postoperative analgesia provided effective analgesia, higher QoR and physical performance, and reduced postoperative opioid consumption. However, there was no effect on postoperative sleep quality

    In this study, a new bodipy/pillar[5]arene functionalized magnetic MS-Sp-P[5]-bodipy microcapsule sensor was prepared based on the use of environmentally friendly for the selective and sensitive detection of Cu(II) and Hg (II) ions in aqueous media. SEM results used in the characterization process of the materials synthesized at each stage confirmed the structural and morphological changes in the pore structure, while other characterization results (FT-IR and XRD) elucidated the role of pillar[5]arene compound and bodipy dye in the synthesis of magnetic microcapsule sensors. The colloidal solution of MS-Sp-P[5]-bodipy (water/ethanol)) showed two fluorescence bands centered at 402 and 540 nm. The detection limits of MS-Sp-P[5]-bodipy for Hg(II) and Cu(II) were calculated to be 0.06 mu M and 2.27 mu M, respectively (at 540 nm). The linear range of the magnetic sensor for Hg(II) and Cu(II) was found to be in the range of 1-150 mu M and 10-150 mu M, respectively.     The prediction and evaluation of recycled polypropylene fiber and aggregate incorporated foam concrete using Artificial Neural Networks

    Reinforcement with recycled fiber is widely investigated to improve the mechanical behavior of foam concrete. In addition, the use of recycled aggregate in concrete provides benefits in terms of sustainability as it reduces the use of raw materials. In this research, 6 mm-long fibers and aggregates obtained from waste polypropylene were utilized in foam concrete production. This study also presents an evaluation of the recycled polypropylene fiber (PppF) and aggregate (PppA) incorporated foam concretes using ANOVA and ANNs. The ANN model was developed to estimate the compressive and flexural strengths, dry density, and thermal conductivity. The results indicated that the use of recycled polypropylene fiber increased the compressive and flexural strengths, however, polypropene aggregates affected the strengths negatively. And mixtures with higher levels of RppA and RppF have lower thermal conductivities. The slump of fresh concrete had an apparent reduction with the increase in RppA quantity.     Parents’ attitudes towards childhood vaccines, and related factors

    Purpose: This study was aimed at determining parents' attitudes towards childhood vaccines and related factors. Methods: This cross-sectional study was conducted with 462 parents who had children aged 0-59 months and presented to the paediatric outpatient clinic of a hospital located in the central Anatolian region of Turkey between October 2020 and March 2021. The study data were collected using a questionnaire including the Personal Information Form and the Parents Attitudes about Childhood Vaccines scale. Findings: Of the parents participating in the study, 81.6% stated that they considered it necessary to vaccinate their children, 83.1% had their children get all the vaccines recommended by the Ministry of Health, 76.9% stated that they missed some of the recommended vaccinations due to the side effects of vaccines, and 25.7% were hesitant to have their children vaccinated. Conclusions: While determining parents' attitudes towards and beliefs about vaccination, and factors affecting their attitudes and beliefs is of critical importance for the success of vaccination programs, parents' lacking knowledge about vaccines and having vaccine hesitancy may adversely affect vaccination rates in the community. Therefore, we recommend that parents should be supported through trainings on the importance of vaccination, and that parents and the society should be informed about vaccination through public service announcements using mass media. © 2024, Medcom Limited.
