    From Revolutionary Pariah to Respected Pundit: Zinovi Peshkov and American Audiences, 1915-1917

    This article considers a pivotal moment in the life of Zinovy Peshkov: his 1916-1917 speaking tour of the United States. Adopted by famed author Maksim Gorki, Peshkov spent much of his youth among Russian revolutionaries. Yet, he turned his back on their path when he joined the French Foreign Legion during the First World War. Combat wounds led to the loss of an arm but also to a transformation once Peshkov was sent to the United States by a French government hoping to increase American support for the Allied War effort. The advent of the Russian Revolution made Peshkov an important pundit – one with access to American politicians, businessmen, and opinion-makers – and a heroic figure in the eyes of the public

    Oxidative stress-associated gene signature predicts the prognosis and therapy response in prostate adenocarcinoma

    Background: Oxidative stress contributes to the development of prostate adenocarcinoma (PRAD). However, the prognosis prediction and therapy response predicted based on oxidative stress-associated genes lacked comprehensive study. Herein, an integrated bioinformatics approach was adopted to identify the prognosis-associated oxidative stress genes for PRAD. Methods: From Gene-Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) databases, the transcriptomic data and clinical data were collected. Genes related to oxidative stress were retrieved from oxidative stress pathway "GOBP_RESPONSE_TO_OXIDATIVE_STRESS" in the MsigDB. A risk model was constructed based on the hub genes selected by both WGCNA analysis and LASSO analysis. GEO cohort was used for verifying the model robustness. By running CIBERSORT and ESTIMATE algorithm, immune cell infiltration was quantified. TIDE algorithm and Spearman correlation analysis were used for evaluating the immunotherapy response and drug sensitivity. RT-qPCR analysis and CCK-8 were carried out to validate the results. Results: We established oxidative stress-related gene signature (NUDT7, NTRK3, MAP3K12, DRD5, C3orf18, and B3GALT2) as an independent factor for the prognostic survival of PRAD. In virto experiments showed that MAP3K12 had a higher expression in PRAD cell lines, and knockdown of MAP3K12 inhibited PC3 cell viability. The risk score was positively linked with T_ cells_ regulatory_ Tregs and Macrophages_ M2 and negatively linked with Plasma_cells. High-risk patients showed higher expressions of PDCD1, CD274, CTLA4, LAG3, PDCDILG2, BTLA, HAVCR2, TIGIT, and higher myeloid-derived suppressor (MDSC) score. Docetaxel, Cisplatin, and Bicalutamide could benefit low-risk patients more. Calibration curves and DCA showed an accurate prediction by the nomogram. Conclusion: We established a novel and reliable prognostic model for PRAD patients

    Aberrant Immunohistochemical stains among 573 cases of Diffuse Mesothelioma

    Background: Diagnosis of malignant mesothelioma often requires differentiating it from other metastatic malignancies including breast cancer, lung cancer, ovarian cancer, colorectal cancer, etc. Immunohistochemical stains (IHC) are important means to assist in confirming mesothelioma and ruling out metastatic cancers. Although mesothelial specific markers have been used for diagnosis, aberrant immunostains were also observed in clinical practice which often confounded the diagnosis. In this study, we analyzed the positive rate of commonly used IHC markers in the diagnosis of mesothelioma among 573 patients. Design: 427 cases of epithelioid mesothelioma, 87 cases of biphasic mesothelioma and 59 cases with sarcomatoid mesothelioma were retrieved from the pathology consultation files between 2020-2023. The positive rates of over 50 IHC markers including over 30 aberrant IHC markers were analyzed. Results: The positive rates of mesothelial markers, such as calretinin, WT-1 and D2-40, and epithelial markers, such as cytokeratins, were highest in epithelioid type, intermediate in biphasic type and lowest in sarcomatoid type mesothelioma. The mesenchymal marker vimentin and additional marker GATA-3 were highly expressed in sarcomatoid mesothelioma. The highest loss rate of BAP-1 was in epithelioid type, while the highest loss rate of MTAP was in sarcomatoid type. The CDKN2A (p16) deletion was observed equally in both epithelioid and sarcomatoid/biphasic types. Over 30 aberrant markers were observed. For epithelioid type, commonly observed aberrant IHC markers included MOC-31, BerEP4, PAX-8, p63, CK20, NKX3.1 and ER. For biphasic type, significant aberrant IHC markers included MOC-31, Ber-EP4, PAX8, CK20 and p40. For sarcomatoid type, notable aberrant IHC markers included p63, SMA, ERG and CD31. Among these aberrant IHC markers, MOC-31, Ber-EP4 and p63 were more commonly observed in pleural epithelioid mesothelioma, whereas PAX-8 in peritoneal epithelioid mesothelioma. Conclusions: Our data demonstrated that aberrant immunostains were common in all of the histological types of mesothelioma. Therefore, the diagnosis of mesothelioma should be based on correlation of clinical presentation, radiological findings, and selective panel of immunostains, and should not be distracted by aberrant immunostains

    Standardized protocol for collecting community-level bee data

    A key component of assessing bee biodiversity patterns and supporting bee conservation is documenting bee communities. When integrated with additional ecological data, community-level data help reveal the relative impact of local- and landscape-scale factors on bee taxa. As such, these data can inform management decisions to support bee diversity and mitigate environmental drivers of decline. However, methods for sampling bee communities vary greatly across projects, making it difficult to compare existing datasets or design new, interoperable studies. Here, we provide a standardized protocol for collecting community-level bee biodiversity data and offer guidance on inventorying, surveying, and monitoring of bee communities. We also present case studies to illustrate how different components of the protocol could be implemented. Although we discuss the benefits of collecting physical specimens, we emphasize the importance of responsible collecting and highlight key strategies to minimize environmental impact while maximizing the value of the work in new projects. This protocol is part of a series developed in association with the U.S. National Native Bee Monitoring Network to standardize bee monitoring practices.

    Notes on the genera Ammobatoides and Melitturga in Europe (Hymenoptera: Apoidea)

    The bee genus Ammobatoides Radoszkowski (Apidae) is a species-poor Old World group that predominantly parasitises the pollen-collecting genus Melitturga Latreille (Andrenidae). Nine species of Ammobatoides are currently recognised, but due to the rarity of most species, four are only known from a single sex. Examination of material from Greece has yielded the previously unknown female of Ammobatoides okalii Kocourek, a species previously known only from its male holotype. The suspected host is Melitturga spinosa Morawitz. Revised identification keys for Ammobatoides and Melitturga in Europe are provided, along with a clarification of the statuses and distributions of Melitturga praestans Giraud and Melitturga syriaca Friese

    Estimating body size in the large carpenter bees (Xylocopa)

    Body size is a salient functional trait in bees, with implications for reproductive fitness, pollination ecology, and responses to environmental change. Methods for quantifying bee body size commonly rely on indirect estimates and vary widely across studies, particularly in studies of the large carpenter bees (Xylocopa Latreille) (Apidae: Xylocopinini). We evaluate the robustness of three common body size parameters (intertegular distance, head width, and costal vein length) as predictors of dry body mass within and among 11 species of Xylocopa (and 5 subspecies). We found that all three size measurements provide robust body size estimates, accounting for 92–93% of intraspecific variation in body mass. Within species, however, these measurements were considerably less predictive of body mass, explaining on average only 36.8% (intertegular distance), 57.4% (head width), and 38.8% (costal vein length) of the variation in body mass. We also highlight a novel application of photogrammetry and 3D modeling to estimate surface area and volume across species, and comment on the utility of these methods for body size estimates in Xylocopa and in insects more broadly. These findings provide practical guidelines for body size estimation methods within and among carpenter bee species.
