    Variation of Interleukin-8 (IL-8) Receptor Gene Based on Single Strand Conformation Polymorphism (SSCP) in Indonesian Friesian Holstein Dairy Cattle

    Interleukin-8 (IL-8) receptor gene is response to bacterial invasion, endothelial and epithelial cells by release the chemokine interleukin-8 (IL-8). IL-8 mediates neutrophil function, allowing neutrophils to resolve bacterial infections by migrating through blood vessel walls and to the site of infection. IL-8 also impacts neutrophil killing and survival ability during the inflammatory response.  Genes that associated with neutrophil function also has potency as a genetic marker for mastitis disease, the migration of neutrophil from blood to infections side is essentially to detect the mastitis pathogen in general. It has been known that mastitis disease was the major problem in dairy cattle industry since influencing the milk quality, especially in bacterial content. The aim of this research is to detect the variation of IL-8 receptor gene in Indonesian Frisien Holstein (FH) dairy cattle which keep by farmer at Pujon District of Malang Regency.  DNA genom was extracted from whole blood cells by standart phenol-chloroform methods.  PCR reaction was done by using genomic DNA as a template while primer was the set of nucleotide that flanking IL-8 receptor gene. Polymorphism analysis of IL-8 receptor gene was based on number and frequency of SSCP (Single Strand Conformation Polymorphism) pattern of  PCR products.  From research result showed that was found five patterns of SCCP from IL-8 receptor gene of Indonesian FH which came from Pujon District, Malang Regency.  This is also illustrated that of IL-8 receptor gene of Indonesian FH dairy cattle was enough polymorphism.  The number pattern of SSCP-1 and SSCP-2 were found higher than others.  Also, those patterns less than 50 % of samples were suspected as sub clinical mastitis. It means that this preliminary study showed that SSCP-1 and SSCP-2 patterns could be as a candidate marker for mastitis resistance of Indonesian FH dairy cattle. While others patterns could be used in determination of IL-8 receptor gene polymorphism of Indonesian FH dairy cattle based on SCCP patterns.  This is important because we need this data for completing the genetic variation of Indonesian cattle data base.     The effect of Strain and Altitude on Broiler Performance

    The research aimed to study the effect of strain and altitude on broiler performance, and to find out suitable strains kept either on low and high altitude. Low altitude was in Purwonegoro North Purwokerto (80 m asl) and high altitude was in Munggangsari Baturaden (755 m asl). The research was carried out since September 5th to December 5th, 1996 using 2x3 Factorial based on Completely Randomized Design. Factor I were land altitude (d) : low (d1) and high (d2). Factor II were broiler strains (g) : CP-707 (g1), AS-101 (g2) and Lohmann (g3). Data obtained were analyzed using F test continued  to Least Significant Difference Test. The research indicated that interaction between altitude and strains significantly affected  feed convertion and income over feed  and doc cost, while strain had significant effect on feed convertion. AS-101 and Lohmann strains  kept on low altitude had the highest final weight. The low altitude gave better feed convertion and income over feed and doc cost, whereas CP-707 strain had the best feed convertion.     Effects of Lemuru Oil and ZnSO4 Supplementation in the Diet Consisted of Cacao Pod Silage and Urea on Zinc Absorption and Growth of Calves

    An experiment conducted to study the effect on Zinc, prostaglandin-E2 (PGE2) concentaration, alkaline phosphatase activities (APA) in the growth of bull calves fed cocoa pod silage diet supplemented with lemuru oil (LO) and ZnSO4. The animals were randomly assigned into three group of treatments, i.e.:supplementation of  0, 25, 50 and 75 mg kg -1 diet ZnSO4 and 0, 1.5 and 3% kg -1 diet LO. Basal diet consisted of cocoa pod silage and concentrate consists of wheat bran, cassava waste, soya bean meal in the ratio 1:3 which contained 70% Total Digestible Nitrient (TDN) and 16% CP as prescribed by NRC (1988). The diet was the supplemented  with 1% urea. LO and ZnSO4 supplementation in the diet was significantly increased Zinc absorption (P<0.01) as well as increased PGE2 concretation (P<0.14) in the growth of bull calves. Relationship of the rate of apparent Zinc Absorption (Va, mg.d-1) with the rate Zinc intake (Va, mg.d-1), PGE2 concentration (mg.L-1) and Vi interaction for the level of 0, 1,5 and 3% LO supplementation were: Va (0) = -5.69 + 0.976 Vi + 1.98 PGE2 – 0.0855 Vi PGE2;  Va (1.5) = -2.6 + 0.962 Vi – 14.3 PGE2 + 0.435 Vi PGE2 and Va (3) = -5.69 + 1.03 Vi  -  0.394 PGE2 + 0.125 Vi PGE2. Level of LO supplementation was significantly linier increased (P<0.01) in serum of APA. Relationships of Zinc absorption with APA for the level of 0, 1.5 and 3% LO supplementation was significantly (P<0.01) increased by the following equations: APA (0) = 3.90569 – 0.0204 Va;  APA (1.5) = 3.61119 – 0.0167 Va and APA (3) = 4.18421 – 0.0199 Va. The average daily gain of the animals (ADG, kg) could by state as ADG = 0.603 + 0.00025 Zn + 0.00004 Zn2 + 0.113 LO – 0.0275 LO2 + 0.00002 Zn LO – 0.00001 Zn2 LO2 (R2 = 0.653; Sb = 0.0926). It can be concluded that supplementation of LO or ZnSO4 in the diet can increased Zinc absorption in the growth of bull calves with their average daily gain was higher at 1.5% LO and 75 mg ZnSO4 kg-1 diet than other levels.     Syncronization Technology for The Reproductive Management in Sambar Deer (Rusa Unicolor Brookei)

    Research to study the synchronization technology to improve the reproductive of captive sambar deer (Rusa unicolor brookei) was conducted using 10 hinds and three stags aged 8 -10 years. Estrous induction was done using local intravaginal spon containing 450 mg Medroxy progesterone acetate (MPA), inserted the intravaginaly for 9 days. At 2 to 6 days post-sponge withdrawal, hinds were put together with stags in hard antler conditions. The results showed that retention rates of sponge reached 100% and no sign of irritation on the vagina. Two days post sponge withdrawal. Three hinds showed an estrous behavior and were mated, whereas four other hinds showed estrous behavior three days post sponge withdrawal. Overall 90% of hinds gave fawns at the end of pregnancy whit length of gestation 235,66 ± 3,60 days (SD). It can be concluded that synchronization using locally made sponge showed a positive response in sambar deer.     The Effect of Garlic (Allium sativum L) Supplementation on Production and Egg Cholesterol Level of Hysex Brown Laying Hens

    This aims of this study was to evaluate the effect of garlic supplementation on production and egg cholesterol level of Hysex Brown laying hens. This study was conducted based on Completely Randomized Design with four treatment of garlic containing 0; 2; 4 and 6% and five replicate.  In each replicate, there were four hens aged 38 weeks. The formulation diet of 2.900,01 ME kcal/kg, 16.5% of protein and drinking water were prepared in ad libitum during 4 weeks observation. The result showed that the effect of garlic supplementation 0, 2 and 6% respectively did not show any significant effect toward feed intake, egg production, egg cholesterol, egg weight, egg yolk and drinking water during 4 weeks of observation.  It was found that 4% garlic supplementation significantly (P<0.05) reduced on egg cholesterol by 14.23% and did not affect the eggs production.  It was concluded that garlic supplementation did not affect egg production, but garlic supplementation 4% decreased egg cholesterol of Hysex Brown laying hens.     Function Analysis of Milk Yield on Small Holder Dairy Cattle in Sumbang Banyumas Region

    A survey on smallholder dairy cow had conducted in Banyumas region from November 8th 1999 up to February 8th 2000. Forty six respondents were involved in this study (as samples). Cobb - Douglas analysis was applied in this survey. From the study can be concluded that a small holder owned 3.02 heads of dairy cow (2.2  ST), 1.44 heads of lactating cow with 9.77 L/day milk production on average. The benefit of the backyard farming was RP.70.603,225 per month on average. Feeding was the biggest influence on benefit because feed cost was 65.81 percent from the production cost. The milk production function , by the equation: Y = 0,8958 X1â°Â·â°Â³Â³Â² X2 0.049 X30.168 X40.608 The four variables can explain 92.4 percent (R²=0,924)on the milk  production variation .It can be suggested that to improve milk production on dairy cows can be done by intensifying labour, and improving the forage feeding.     Program pengembangan dan perbaikan sistim produksi peternakan dapat diawali dengan penilaian terhadap potensi suatu bangsa ternak melalui serangkaian proses pencatatan, evaluasi on-farm, dan monitoring. Tujuan kajian ini adalah (1) mengetahui penampilan reproduksi dan kematian cempe prasapih Kambing Peranakan Etawah pada sistim produksi di kelompok tani ternak kambing Gumelar Banyumas, dan (2) mengetahui faktor-faktor non-genetik yang berpengaruh terhadap penampilan reproduksi dan kematian cempe prasapih.  Digunakan kompilasi data penampilan reproduksi dan kematian cempe prasapih hasil penelitian lapang melibatkan 562 cempe dan 344 ekor induk kambing. Uji Chi-Square dan prosedur General Linear Model (GLM) diterapkan untuk menguji faktor-fator non-genetik (jenis kelamin, tipe kelahiran, paritas) yang berpengaruh  terhadap jumlah anak sekelahiran, kematian cempe prasapih, dan jarak beranak. Hasil penelitian menunjukkan rataan kematian cempe prasapih sebesar 5,9%. Kematian cempe prasapih betina (6,3%) nyata lebih tinggi  daripada jantan (5,4%). Kejadian kematian cempe prasapih paling sering dijumpai pada kelahiran kembar tiga (16,7%), sedangkan pada kelahiran kembar dua dan tunggal masing-masing sebesar 5,6% dan  2,9%.  Kematian cempe prasapih dipengaruhi oleh paritas induk, dan kecenderungan menurun dengan peningkatan paritas.  Rataan jumlah anak sekelahiran sebesar 1,64 ekor, dan dipengaruhi sangat nyata oleh paritas induk. Induk pada paritas 1, 2, 3, 4, 5, 6, dan 7 menghasilkan jumlah anak sekelahiran berturut-turut 1,45; 1,71; 1,73; 1.95; 1,76; 1,83; dan 2,13 ekor.  Rataan jarak beranak 285 hari dan nyata dipengaruhi oleh faktor paritas induk dan tipe kelahiran.  Jarak beranak nyata lebih pendek dengan peningkatan paritas induk (1-7) berturut-turut adalah 319, 271, 261, 234, 236, 230, dan 228 hari.     Growth Of Female New Zealand Crossbreed Rabbit On Variation Of Feed Protein Level

    The Growth of Female New Zealand White Crossbreed Rabbit on Variation of Feed Protein Level have studied on Experimental Farm Animal Husbandry Faculty, University of Jenderal Soedirman Purwokerto.   On – Station Research by Factorial Pattern based on Two-Way Classification Design, were involved 42 heads rabbit two months of age. Factors examine consist  of : kinds of feed as a first factors (a1 = without fish meal, a2 = fish meal 2%, a3 = fish meal 12%, a2 = fish meal 22%), and body weight as a second factors (b1 = 601-700 gram,b2 = 701-800 gram, b3 = 801-900 gram). Variance analysis showed that there were significance (P< 0.01) interaction between kinds of feed and body weight on daily gain and feed consumption. Increasing feed protein level (2, 12 and 22% of fish meal) clearly increase (P<0.01) of daily gain and feed consumption on overall of body weight conditions  (b1,b2 and b3).     Comparative Study on Fat and Cholesterol Contents of Shredded Beef and Bufallo Meat

    Penelitian ini bertujuan untuk membandingkan kandungan lemak dan kolesterol abon daging sapi dan kerbau antar umur ternak. Sampel daging diambil pada bagian paha belakang (rump) dari sapi jantan, kerbau jantan dan kerbau betina yang dipotong di RPH Pemalang. Ternak dibedakan atas umur muda dan tua dengan 4 (empat) kali ulangan. Peubah yang diamati adalah kandungan lemak dan kolesterol. Data yang terkumpul dianalisis ragam dengan pola tersarang (Nested Classification), sebagai grup adalah jenis ternak (sapi jantan, kerbau jantan dan kerbau betina). Sub grup adalah antar umur dalam jenis ternak (muda dan tua). Anlisis ragam menunjukkan kandungan lemak antar jenis ternak menunjukkan perbedaan nyata (P<0,05) sedangkan antar umur ternak menunjukkan perbedaan sangat nyata (P<0,01). Pada kandungan kolesterol, menunjukkan antar jenis ternak tidak berbeda nyata. Hasil penelitian dapat disimpulkan bahwa kandungan lemak dan kolesterol abon daging sapi dan kerbau tidak menunjukkan perbedaan sehingga daging kerbau dapat digunakan sebagai alternatif penyediaan abon daging.
