Identification of Excretory – Secretory Sporozoite Antigens of Eimeria tenella with Monoclonal Antibody
The research done specifically to identified the. Eimeria tenella excretory-secretory sporozoit antigen by using the monoclonal antibody reaction instead. Eimeria tenella excretory-secretory sporozoit antigen. The 107 sporozoits were obtained from existation of 5x107 oocysts with 25 ml tripsin and sodium taurocholat. Excretion - secretion antigen were preparated by freezing and thawing and then be isolated with ice bath 20 second, 20 meter of amplitude as the further treatment. Determinating the excretion -secretion sporozoit protein by using BCA-test. While the visualization sporozoit protein antigen by SDS-PAGE. Monoclonal antibody were produced through hybridization between B BALB/c limphosit immunized with Eimeria tenella excretion-secretion sporozoite antigen and myeloma cells. The monoclonal antibody identification of excretion – secretion antigen were done through western blott. The visualization of protein molecule weight of Eimeria tenella sporozoit were resulted 20 protein fraction, included 14, 16, 17, 18, 20, 24, 26, 32, 34, 36, 39, 43, 47, 56, 65, 67, 76, 87, 94, and 96 KDa. There were 12 kinds of monoclonal antibody could be produced from these proteins. While the 5 hybridoma immunoblotting resulted specific reaction by the appearances of reaction ribbon obviously, there were MABset 1 which identified protein epitop with 14.7 KDa of molecule weight, MAbset 2 which identified protein epitop with 43 KDa of molecule weight, MAbset 3 which identified protein epitop with 42 KDa of molecule weight, MAbset 4 which identified protein epitop with 47 KDa of molecule weight and MAbset 5 which identified protein epitop with 90 KDa of molecule weight. As the conclusion, five of excretion-secretion sporozoit antigen were proper immunogen to stimulate the hospest immunity. Effect of Artificial Insemination Time on the Fertility of Layer Hens
A study on the effect of AI time on the fertility of layer hens was carried out. Semen was collected from eight kampung roosters, with age ranging from 12-18 months. Completely Randomized Design was used to analyze the data obtained. Every eight hens (n=8) was inseminated intravaginally at 0-2 hrs, >2-4 hrs, > 4-6 hrs and >6-8 hrs after oviposition. The results showed that insemination time did not affect (P>0.05) the percentage of fertility from days 2 to 20. However, the insemination time affected (P0.05) the mean duration of the fertile period. In conclusion, the time of insemination at 0 – 2 hours after oviposition gave the best fertility. It is recommended that for the best fertility flock inseminations should be performed at a time of 0-2 hours after an egg is laid every 9 days. Grassing as potential factor in Gastrointestinal Nematodiasis in Goat in Banyumas District
Goat system manajemen like housing, feeding dan grassing have been studied on the diseases association in gastrointestinal nematodiasis infection. The method applied was survey on 150 goat (Jawarandhu) faeces sample in Banyumas District. The Research was carried out from July to September 2004. The result showed that the nematode infection 1,54 times to goat if grassing and 88,62% infection in groups. Housing, Feeding have no risk and associated with disease infection. Genetic Relatedness Between Batur, Merino and Local Sheep Based on Random Amplyfied Polymorphism DNA Marker
RAPD analysis to determine the diversity, status and genetic close relatioship Batur with Merino, Garut, Thin Tail and Fat Tail sheep genomic DNA was used in 27 Batur, 15 Merino, 17 Garut, 15  Thin Tails and  Fat Tails animals. The process of RAPD-PCR used five primers of 10 nucleotides. PCR results were electrophoresed with 2% agarose gel. Identified similarities and differences between individual RAPD bands one and cross-sample of the population, genetic distance and closeness of genetic relationship. The process 89 sample  sheep RAPD-PCR generated of 4189 band from 100 to 1500 bp which consisted of 91 type. Batur sheep samples produced bands at most (1666 tape) and the lowest Fat Tails (493 bands). The number monomorfism of bands most of the Batur (891 bands) and the lowest Garut (170 bands), but the polymorphism  band's highest  of the Batur (775 bands) and the lowest Fat Tails (262 bands). Between individuals within populations have similar genetic Merino highest and the lowest Thin Tail. Cross-population genetic similarity is highest individuals in the population Batur and Merino, while the lowest Merino and Thin Tail. The closest genetic distance was Batur with Merino population and the most apart distant Merino with Thin Tail or Merino and Fat Tails. Batur sheep population has the closest genetic relationship with the Merino and the most apart distant is with Fat Tail. The Effect of Fish Meal Substitution with Squilla empusa on Reproductive Performance of Local duck
A study has been conducted to know the effect of fishmeal substitution with Squilla empusa on reproductive performance of local duck. Randomized Completely Design was applied in this experimental. Ninety-six grower local ducks were divided into six treatments ration. Each treatment was replicated by four replications and consists of four local ducks each. The treatment was P0 = 0 % Squilla empusa and 10 % fish meal, P1 = 2 % Squilla empusa and 8 % fish meal, P2 = 4 % Squilla empusa and 6 % fish meal, P3 = 6 % Squilla empusa and 4 % fish meal, P4 = 8 % Squilla empusa and 2 % fish meal, and P5 = 10 % Squilla empusa and 0 % fishmeal. Parameters observed was reproductive performance consisted of sexual maturity, average egg production and egg weight during 3 months of production period. The result showed that fishmeal and Squilla empusa had very significant by effect (P<0,01) on feed consumption, sexual maturity, egg production and egg weight. It was significant by (P< 0,05) on egg production and weight. It was concluded that substitution up to 4.96 % of Squilla empusa give the latest of sexual maturity (5.8 month); the lowers egg production was at 6 % Squilla empusa; at 10 % Squilla empusa egg weight tended to increase. Digestibility and Egg Yolk Pigment Intensity of Local Ducks Fed Shrimps Head Meal, Leucaena Leaf Meal and L-Carnitine Supplementation
The experiment was conducted to determine the nutrient digestibility and egg yolk pigment intensity of local ducks which fed shrimps head meal, leucaena leaf meal and L-Carnitine supplement. The experimental method used was a Completely Randomized Design (CRD) with four treatments i.e : R0 (feed with leucaena leaf meal), R1 (feed with 25 ppm L-Carnitine supplement), R2 (feed with 2% shrimps head meal) and R3 (feed with 4% shrimps head meal). A total of 0,20% FeSO4/kg feed were supplemented to all treatments. The results showed that L-Carnitine supplements and fed shrimps head meal (2% and 4%) have no significant effects (P>0,05) on crude fat and crude protein digestibility but have significant (P<0.05) effects on yolk pigment intensity. L-Carnitine supplements resulted in 86,69% crude fat and 90,78% crude protein digestibility). Ducks fed with 4% shrimps head meal (R3) produced eggs with highest yolk pigment intensity (10,55). Identification of Body Size and Body Shape of Garut Sheep Fighting Type and Meat Type and Garut Cross Based on Principal Component Analysis
This research was done to identify the body size and shape  among three types of local sheeps of Garut fighting type, Garut meat type and their crossbred fighting and meat type in Garut district (Margawati, Sukawening and Wanaraja) and Bogor district (Ciomas and Cinagara). The total number of sheep from Garut district used in this study were 520 head consisting of crossbred sheep from Margawati (71 head), fighting sheep from Wanaraja (79 head), meat sheep from Wanaraja (84 head), fighting sheep from Sukawening (87 head) and meat sheep from Sukawening (62 head). Whereas the total number of sheep from Bogor district were 137 head consisting of those of fighting sheep from Ciomas (66 head) and from Cinagara (70 head). Some body measurenments measured in this study were body weight, body length, wither height, chest width, chest circumfrence, cranium, tail length and tail width. Analysis of all those sizes were based on principal component analysis (PCA) then visualized into the group of crowded diagram. Different group of crowed builded on the base of scoring in body size and body shape derivated from covarian matrix were able to identifed morphologycal penotypic differences among garut sheep studied.  The results from canonical analyses showed that body lenght, chest girth,chest width, and wither height were the most discriminant variables had impact to the diferences score size between  types of Garut sheep. However, the tail length and tail width were variables had impact to diferences score shape. The  crowed diagram showed that an intersection among three types (fighting, meat and crossbred) of sheeps suspected as resulted from the same genetic resource, namely Garut sheep. Genetic flow was suspected from crossbred in Margawati to meat sheep in Cinagara as well as the sheeps from Sukawening and Wanaraja to fighting sheep in Ciomas. Close genetic distances that were found among fighting and meat sheeps in Sukaweining as well as fighting and meat sheeps in Wanaraja could be considered that these two groups of sheeps as the genetic resource of fighting sheep in Ciomas. Effectiveness of Temulawak (Curcuma xanthoriza) and Kunyit (Curcumae domestica) Extracts to Enhance Productivity and as Immunostimulator of Avian Influenza in Broiler
The objective of the experiment was to investigate the effectiveness of treating broiler with temulawak (Curcuma xanthoriza) and kunyit (Curcumae domestica) extracts to enhance productivity and as imunostimulator of avian influenza. Broilers were given either temulawak, kunyit or temulawak+kunyit extracts. The treatments, including a control, were arranged in a factorial design. Variables measured were production index and immune titter with haemaglutination inhibition (HI) test at 35 days of age. Results showed that control, temulawak-, kunyit- and temulawak+kunyit-treated chicken have production indexes of 302.80, 382.30, 327.71, and 358.30, respectively. HI test results were all negative. It can be concluded that neither temulawak, kunyit nor temulawak+kunyit extracts is effective imunostimulator of avian influenza in broiler. Brahman Cross Development in Village Breeding Centre of the Sarjana Membangun Desa: Pitfall and A Lesson Learned
This study was conducted to evaluate the cattle development regarding the pitfall to be lesson learned on cow-calf operation of Brahman Cross (BX) of farmer groups in Village Breeding Centre (VBC) in Central Java province. Data on productivity of BX were compiled from 547 Brahman cows of 43 VBC of The Sarjana Membangun Desa (SMD) program years 2008-2009 distributed in 10 regencies. Dynamic population, calf-crop, calf and dam mortality, rate of second calving, and service per conception (S/C) were recorded. Quantitative and qualitative descriptive analyses  were applied in this study. Cow-calf operation of Brahman Cross in 43 VBC was unsuccessful in terms of low reproductive rate for the second pregnancy and calving as well as a high rate of calf and dam mortality. The rate of the second calving was 2.89%. Calf and dam mortality were 17.27 and 12.23% for the SMD year 2009, meanwhile 25.67 and 7.08%  for the SMD year 2008, respectively. Cow-calf development of BX in 43 VBC with the goal of increasing cattle populations was not significant. Some experiences on unsuccessful cow-calf development of BX must be taken into account and become a lesson learned for policy and program formulation in terms of cattle development through cow-calf operation of BX. The real actions through synergism and collaboration among parties (stakeholders) to solve the problems in BX development should be implemented in order to increase farmers’ income, and to achieve the main goal in the acceleration of self-sufficiency in beef production. The Effect of Utilization Bentonite and Zeolite in the Broiler Diets on Performance and Fecal Characteristics
Ninety six commercial strain were conducted from 14 to 42 days of age to evaluate the affect of utilization zeolite and on performance of broiler and fecal characteristics. Birds were divided into four  treatment diets (4 replicates of 7 birds each): K (control), KB (K+ 1% bentonite),  KZ (K+ 1% zeolite), KBZ (K+ 1% Z+ 1% B). The birds were fed initially for two weeks a commercial starter ration and followed by treatment diets 15 to 42 days of age. All the diets were formulated to have 20% crude protein and 3000 kcal. ME and 0.91% Ca and 0.69% P for control and 0.34% treatments. Body weights and feed intakes were measured on a weekly basis. The manure from each group was sampled on weekly basis and the moisture content, pH and ammonia production was determined. The results showed the utilization zeolite and bentonite or both significant differences (P<0.05) on pH and (P<0.01) fecal ammonia content, but not on feces water content, final body weight, feed consumption, feed conversion ratio, and carcass.
