    A rapid and inexpensive RNA-extraction method for high-throughput virus detection in grapevine

    The extraction of RNA from grapevine tissue is a crucial step for virus diagnostics via multiplex reverse transcription-polymerase chain reaction (mRT-PCR). Conventional methods are either time-consuming or expensive when convenient extraction kits are used. Here we present an easy, but reliable extraction method that fulfills the requirements of epidemiological research (high sample throughput with maximum accuracy). A further advantage of the protocol beside the low costs is the absence of harmful chemicals like phenol or chloroform and the possibility to manage \u27high-throughput\u27 extractions and analyses

    Effects of soil warming and altered precipitation patterns on photosynthesis, biomass production and yield of barley

    Crop productivity and plant physiology are affected by rising temperatures and altered precipitation patterns due to climate change. We studied the impacts of an increase in soil temperature of 2.5 °C, a decrease in summer precipitation amount of 25%, a reduction in summer precipitation frequency of 50%, and their interactions on photosynthesis, biomass production, and yield of spring barley (Hordeum vulgare L. cv. RGT Planet) in a temperate agricultural ecosystem near Stuttgart (Germany). Leaf gas exchange of barley appeared to be affected mainly by drought in the form of reduced precipitation frequency or by a combination of changes in soil temperature and precipitation patterns. In contrast, biomass production and yield parameters were more affected under soil warming alone. In addition, biomass of roots increased under soil warming at stem elongation. Stable grain yield was observed under reduced precipitation amount and also under increased evaporation through soil warming. These findings provide additional evidence that barley is relatively drought tolerant, which should be taken into consideration in the context of appropriate crop selection under climate change

    Deciphering the complex interaction of microbial inoculants with plants

    Pflanzenwachstumsfördernde Mikroorganismen (PWFM) stellen eine effiziente und umweltfreundliche Alternative zu Agrochemikalien dar. PWFM interagieren mit Wurzeln, beeinflussen das Pflanzenwachstum und die Nährstoffversorgung. Die Kombination von kultivierungsabhängigen und -unabhängigen Methoden zur Überwachung von PWFM und Untersuchung der mikrobiellen Gemeinschaften in der Rhizosphäre führte zu einem besseren Verständnis der komplexen Interaktionen in der Rhizosphäre. Pseudomonas sp. RU47 ist ein Rhizosphären-kompetenter Stamm, der sowohl pflanzenwachstumsfördernde als auch Biokontroll-Aktivitäten besitzt. Die Untersuchung der Genomsequenz von RU47 zeigte, dass der Stamm zu dem taxonomischen Cluster P. koreensis gehört und möglicherweise eine neue Spezies repräsentiert. Es konnten verschiedene Gene identifiziert werden, die eine Rolle bei den pflanzenwachstumsfördernden und Biokontroll-Aktivitäten spielen. Basierend auf der RU47-Genomsequenz wurden Primer für vier verschiedene Gene, potentiell involviert in Pflanzen-Bakterien-Interaktionen, entworfen und für die Detektion und Quantifizierung von RU47 in Gesamt-DNA der Rhizosphäre von Tomate und Mais, die in Böden mit reduzierter P-Düngung angebaut wurden, getestet. RU47 wurde in der Rhizosphäre von Tomate und Mais sechs Wochen nach Pflanzung detektiert. Der Einfluss von vier PWFM (Trichoderma harzianum T-22, Pseudomonas sp. DSMZ 13134, Bacillus amyloliquefaciens FZB42 und Pseudomonas sp. RU47) auf das Wachstum von Tomate und Mais sowie auf die mikrobielle Gemeinschaft in der Rhizosphäre unter reduzierter P-Verfügbarkeit wurde untersucht. Es wurde eine hohe Rhizosphärenkompetenz von bakteriellen Inokula an Tomatenwurzeln beobachtet, die positiv mit Pflanzenwachstum und Akkumulation von Nährstoffen im Spross korrelierte. Des Weiteren stimulierten die Inokula das Pflanzenwachstum und führten zu Veränderungen in der mikrobiellen Gemeinschaft in der Rhizosphäre von Tomate. Alle inokulierten Stämme besiedelten die Wurzeln von Mais und veränderten die bakterielle Rhizosphärengemeinschaft, wodurch es zu einer Erhöhung der Maispflanzen-Biomasse kam. Diese Studie liefert einen wichtigen Beitrag hinsichtlich der Verwendung von RU47 im landwirtschaftlichen Bereich, was zur Reduzierung des Einsatzes von Pestiziden und mineralischem Dünger beitragen kann. Darüber hinaus zeigt diese Studie die Fähigkeit von RU47, die Rhizosphäre zu kolonisieren, was zu einer maßgeblichen Erhöhung des Pflanzenwachstums führte aber auch zu Veränderungen des Rhizosphären-Mikrobioms, die möglicherweise auch zu den positiven Effekten beigetragen haben. Auf Wunsch des Autors / der Autorin ist diese Dissertation nur als Druckausgabe verfügbar.Plant growth promoting microorganisms (PGPM) have been proven as an efficient and environmentally friendly alternative of agrochemicals. PGPM interact with roots, affect plant growth and nutrient acquisition. Combined cultivation-dependent and independent methods to monitor PGPM and to investigate the microbial communities in the rhizosphere led to better understanding of the complex interactions in the rhizosphere. Pseudomonas sp. RU47 is a rhizosphere-competent strain showing plant growth-promoting and biocontrol activities. The genome sequence of plant beneficial strain RU47 was analyzed and revealed that the strain belongs to the P. koreensis cluster and likely represents a novel distinct species. Several genes contributing to the plant growth-promoting and biocontrol activities were also identified. Based on the genome sequence data of RU47, primers targeting four different genes potentially involved in plant-bacteria interaction were designed and used for detection and quantification of RU47 in total community DNA from the rhizosphere of tomato and maize plants grown in soils with reduced P-fertilization. RU47 could be detected in the rhizosphere of tomato and maize six weeks after planting. The influence of four PGPM (Trichoderma harzianum T-22, Pseudomonas sp. DSMZ 13134, Bacillus amyloliquefaciens FZB42 and Pseudomonas sp. RU47) on the growth of tomato and maize plants as well as on the rhizosphere microbial community under reduced P supply conditions were tested. High rhizocompetence of bacterial inoculants was observed on tomato roots and positively correlated with plant growth and shoot nutrient accumulation. Furthermore, the bacterial inoculants stimulated the plant growth and overlapping rhizosphere community changes of tomato plants. All the inoculants colonized the maize roots and beneficially changed the rhizosphere bacterial community which led to increased maize plant mass. This study supports potential use of RU47 in agriculture, which might contribute to a reduced application of pesticide and mineral fertilizer. Moreover, this research showed the successful ability of PGPM to colonize the plant roots, leading to a massive increase in the plant growth which might be also due to the shifts in the rhizosphere microbiome detected.     PA contamination in medicinal plants: occurrence, limits and measures

    Seit mehreren Jahren wird eine mögliche Kontamination von Arzneipflanzen mit pyrrolizidinalkaloid-haltigen Beikräutern diskutiert. Die Erkenntnisse über ein solches Risiko hat die Anbauer und Lieferanten pflanzlicher Drogen und die Hersteller von pflanzlichen Arzneimitteln veranlasst, Ursachenforschung zu betreiben und Maßnahmen zur Reduktion und Vermeidung einer möglichen PA-Kontamination zu ergreifen, die u.a. zusammenfassend in einem Code of Practice beschrieben sind. Die Ergeb­nisse von Datensammlungen der Hersteller zeigen über die letzten Jahre eine deutliche Verminderung der PA-Belastung in pflanzlichen Drogen und Extrakten, die für die Herstellung pflanzlicher Arzneimittel eingesetzt werden. Es wird jedoch auch deutlich, dass das Problem einer PA-Kontamination in naher Zukunft nicht vollständig gelöst werden kann. Das Herbal Medicinal Products Committee der europäischen Zulassungsagentur EMA hat im Jahr 2016 für eine Geltungsdauer von 3 Jahren einen vorläufigen Grenzwert von 1,0 μg Pyrrolizidinalkaloiden pro Tag bezogen auf das Fer­tigarzneimittel empfohlen. Diese Frist ist im Januar 2019 um weitere zwei Jahre verlängert worden.Since 2013, a potential contamination of medicinal plants with pyrrolizidine alkaloid-containing weeds has been discussed. The knowledge about such a risk of contamination induced cultivators and suppliers of medicinal plants and manufacturers of herbal medicinal products to investigate the situation and to take action in order to reduce or avoid potential PA contamination as summarised e.g. in a Code of Practice. Over the past years, the results of data collections show a remark­able reduction of the PA burden in herbal drugs and herbal extracts used for the production of herbal medicinal products. However, it is obvious that the problem of contamination cannot be completely resolved in the near future. The Herbal Medicinal Products Committee at the European Medicines Agency recommended a transitional limit of 1.0 μg pyrrolizidine alkaloids per day related to the final product for three years which has recently been prolonged by further two years

    Nachdem Pyrrolizidinalkaloide (PA) in Tees, Kräutern und Arzneipflanzen als Kontaminanten nachgewiesen worden waren, setzten sich alle Akteure der Branche dafür ein, das Risiko der PA-Kontamination zu verringern. Auf der Basis von Risikoanalysen wurden mögliche Eintragswege der PA-haltigen Unkräuter identifiziert. In verschiedensten Veranstaltungen informierten sich Landwirte und Verarbeiter über Ursachen und Maßnahmen zur Risikominderung. Forschungsprojekte zur Bewertung des PA Potenzials von Unkräutern, zur Verbesserung der Unkrautbekämpfung und zur PA-Eliminierung wurden durchgeführt oder begonnen. Dabei arbeiten Landwirte, Verarbeiter und Forscher eng zusammen. Über Fachgremien wie den Deutschen Fachausschuss für Arznei-, Gewürz- und Aromapflanzen laufen die Informationen zusammen, die wiederum in Aktivitäten der Akteure entlang der Prozesskette münden. Die Anstren­gungen aller Branchenmitglieder haben bereits zu rückläufigen PA-Gehalten in den Rohstoffen geführt. Eine hundert­prozentige PA-Freiheit wird es nicht geben. Künftige toxikologische Daten werden eine differenziertere Bewertung der Kontamination mit den verschiedenen PA erlauben.As soon as pyrrolizidine alkaloids (PA) had been detected in herbal teas, spices and medicinal plants, all players of the sector committed themselves to reduce the risk of PA contamination for their products. Based on risk assessments, likely contamination pathways from PA containing weeds into the herbal products were identified. In numerous events farmers and herbs processing companies informed themselves about PA sources and measures to reduce the risk of contamination. Research projects were run or started to evaluate the PA potential of weeds, to improve weed control and to eliminate PA from herbal material. Farmers, processers and researchers collaborate closely to achieve these goals. Professional bodies like the German Committee on Medicinal and Aromatic Plants are platforms for discussion of recent PA information and for initiation of further activities along the process chain. The efforts of all players of the sector already led to reduced PA concentrations in herbal products. However, a perfect solution for zero contamination will not come true. Upcoming toxicity data will allow to individually evaluate contaminations with different PA substances
