    Peningkatan penduduk lansia akan berdampak peningkatan ketergantungan lansia karena kemunduran fisik, psikis, sosial dan kesehatan lansia. Dibutuhkan suatu program pembangunan kelanjutusiaan yang mampu meningkatkan Kualitas hidup. Posyandu lansia sebagai salah satu upaya kesehatan yang bersumber daya masyarakat di Puskesmas yang dibantu oleh kader. Kader Posyandu lansia berperan dalam melakukan screening dini kesehatan lansia dan pengukuran status gizi dan kesehatan secara rutin. Puskesmas Purwokerto Utara II terdapat posyandu lansia baru 44,4%, ada beberapa RW digabung jadi satu, belum dilaksankan secara rutin bimbingan teknis ke kader dan posyandu lansia, kurangnya pengetahuan lansia mengenai manfaat posyandu, akses untuk ke posyandu lansia yang belum merata, masih terbatasnya sarana dan prasarana posyandu lansia. Kegiatan dilakukan pelatihan, demonstrasi, pendampingan dan pembentukan forum posyandu lansia sebagai salah satu upaya kesehatan yang bersumber daya masyarakat untuk memberikan pelayanan kesehatan bagi lansia. Media modul gizi lansia dan vidio kesehatan lansia oleh tim pengabdi. Ada perbedaan pengetahuan sebelum dan sesudah diberikan pelatihan dengan nilai p 0,000< α (0,05).     Singkong (Manihot esculenta) merupakan sumber karbohidrat penting, telah diolah menjadi tepung mocaf melalui fermentasi, menghasilkan produk dengan karakteristik mirip tepung terigu dalam hal ini dapat digunakan dalam pembuatan mi sebagai alternatif yang lebih lokal. Pelatihan ini bertujuan meningkatkan pengetahuan dan keterampilan ibu-ibu PKK pengurus desa serta RT Desa Sokawera dalam pembuatan mi singkong, serta mempromosikan penggunaan bahan pangan lokal. Melalui penyuluhan dan praktik, peserta menunjukkan peningkatan signifikan dalam pengetahuan dan keterampilan. Tahapan dalam metode ini adalah pre-test, penyuluhan, praktik bersama, dan post-test. Alat-alat yang dibutuhkan dalam pelatihan adalah kertas kuesioner, pulpen, proyektor, laptop, dan pengeras suara. Program ini merupakan bagian dari upaya pengembangan infrastruktur pendidikan berbasis green school.     Government Policy in the Regulation of Nurse Practice in Hospitals

    Government policy in the regulation of nurse practice aims to protect the public and nurses through nurse accountability in practice. How nursing regulations are implemented in hospitals, the policies implemented and the strengths and weaknesses in their implementation need to be studied. The purpose of the study is to provide an overview of the implementation of government policies in the regulation of nurse practice in hospitals. The research uses an empirical/sociological juridical approach with descriptive research specifications. Research respondents were the Head of the Cilacap District Health Service, the Director of the Cilacap Regional Hospital, the Head of the Nursing Section, the Head of the Nursing Room and 59 nurses who served at the Cilacap Regional General Hospital. Data collection used in-depth interviews with data analysis using content analysis and qualitative analysis. The results of the research show that in implementing regulations, the roles of the parties involved are in accordance with their proper duties, but not all nurses understand the regulations. The form of practice is nursing care, but there are still weaknesses in supervising the tasks carried out. Suggestions are given to nurses to actively obtain information through print media, electronic media, the internet, seminars, coaching and under be active in organizations

    Isolation and Purification of Breast Milk Folate Binding Protein: Salting-Out and Chromatography Techniques

    ABSTRACT. Folate binding protein (FBP) is a protein in breast milk that plays a role in the regulation and bioavailability of folic acid. In contrast to cow's milk FBP, information about breast milk FBP is still limited. This research aims to determine the isolation and purification methods of breast milk FBP and the molecular weight of breast milk FBP. The sample in this study was 1000 mL of breast milk. Breast milk was prepared in several stages to yield whey. Isolation and purification of FBP from whey were carried out in stages, salting-out, ion exchange chromatography, and affinity chromatography. Whey salting-out with 95% saturation of ammonium sulfate could precipitate folate-binding proteins. This precipitate showed three peaks on DEAE chromatography. Peak II DEAE 95% was thought to be a negatively charged folate-binding protein. Peak II DEAE 95% also showed the presence of two peaks on affinity chromatography. It was believed that Peak II AF 95% was a pure folate-binding protein. Peak II AF 95% showed the presence of a single band on SDS-PAGE and western blot. This indicated that the folate-binding protein was 100% pure. FBP can be isolated from breast milk by the salting-out method using 95% ammonium sulfate, DEAE chromatography, and affinity chromatography. FBP from breast milk has a molecular weight of approximately 37 kDa. The final level of FBP isolated from breast milk is approximately 0.022 mg/mL. The successful isolation of FBP from breast milk provides an opportunity to use it to understand the clinical role of FBP in increasing folic acid levels in both breast milk and infant serum, as well as to develop methods for determining folic acid levels in these fluids.     Keywords: Breast milk, folate binding protein, isolation, purification, molecular weigh

    Optimization of Chitosan-Based Edible Film with the Addition of Velvet Bean Aqueous Extract and Cinnamon Essential Oil for Antibacterial Packaging

    ABSTRACT. The current global warming issue has encouraged a lot of research on edible films. The use of edible films has become an international trend and is now essential for our products to compete in the global market. Bioactive compounds from plants can be incorporated into edible films to enhance their biological activity, including their antibacterial properties. This study developed a chitosan-based edible film with the addition of velvet bean aqueous extract (VAE) and cinnamon essential oil (CEO), aiming for high antibacterial activity. The objectives of this study were to isolate and identify the chemical components of CEO, test its antibacterial activity, find the most optimum formulation of a chitosan-based edible film with VAE and CEO in terms of antibacterial activity (Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 25923), and analyze the physical and morphological properties of the resulting edible film. The research methods included a literature review and laboratory experiments, with procedures involving the isolation and GC-MS-based identification of CEO chemical components, antibacterial activity testing of CEO, optimization of a chitosan-based edible film formulation with the addition of VAE and CEO based on antibacterial activity test against E. coli ATCC 25922 and S. aureus ATCC 25923 (well diffusion method), and characterization of the best film’s physical properties (thickness, tensile strength, elongation, solubility, WVP, and WVTR) and morphological analysis (SEM method). The results of this study were as follows: (a) CEO contained cinnamaldehyde (52.86%) and 2- methoxycinnamaldehyde (47.06%); (b) CEO had very strong antibacterial activity against E. coli ATCC 25922 and S. aureus ATCC 25923, with inhibition zones of 46.28 mm and 47.95 mm, respectively; (c) the most optimal edible film formulation consisted of chitosan with 30.0% VAE and 4.0% CEO, yielding strong antibacterial activity against E. coli ATCC 25922 (15.50 mm inhibition) and S. aureus ATCC 25923 (16.71 mm inhibition), with a thickness of 0.15 mm, tensile strength of 0.32 Mpa, elongation of 0.32%, solubility of 4.89%, WVP of 8.82 g/m2.h), and WVTR of 3.92 g/m.h.     Response Surface Methodology to Optimize Laccase Produced from Pleurotus ostreatus Using Corn Cob and Rice Bran as Substrate

    ABSTRACT. Laccase is an industrially significant enzyme capable of oxidizing a broad range of phenolic compounds, making it valuable for applications such as bioremediation, textile dye decolorization, and pulp bleaching. Therefore, this study aimed to optimize laccase production from Pleurotus ostreatus using agricultural waste substrates corn cob (BJ), rice bran (DP), and their 1:1 mixture (BJ:DP) through Response Surface Methodology (RSM). Extracellular laccase was obtained by centrifuging the cultured medium, followed by ammonium sulfate precipitation and dialysis. The enzyme activity was quantified, and the fraction with the highest specific activity was analyzed using SDS-PAGE. Furthermore, Central Composite Design (CCD) was used to assess the effects of three independent variables, namely ABTS concentration (0.01–0.1 mM), incubation temperature (20–30 °C), and reaction time (20–30 minutes). The results showed that for BJ substrate, the highest enzyme activity (13.7 ± 0.05 U/mL) was observed in the 0–20% ammonium sulfate fraction, with specific activities of 76.25 ± 0.09 U/mg and 90.28 ± 0.03 U/mg in the 0–20% and 20–40% fractions, respectively. Conversely, DP substrate achieved a maximum specific activity of 209.67 ± 0.028 U/mg in the 20–40% fraction. The crude extract from BJ:DP mixture showed a high protein content (0.636 ± 0.006 mg/mL) but the specific activity was substantially lower (19.33 ± 0.003 U/mg). Based on RSM analysis, the optimal conditions were ABTS concentration of 0.05 mM, incubation temperature of 22.21 °C, and reaction time of 28.64 minutes, resulting in a predicted laccase activity of 13.99 U/mL.   Keywords: Corn cob, Laccase, Pleurotus ostreatus, Response Surface Methodology, Rice bra

    Identifikasi Lalat Buah Tangkapan dengan Umpan Buah Manis dan Buah Masam

    Lalat buah dari genus Drosophila sp. memiliki peran yang sangat penting dalam perkembangan penelitian genetika. Serangga ini banyak dimanfaatkan sebagai organisme model di laboratorium karena memiliki berbagai keunggulan unik, seperti siklus hidup yang singkat dan kemudahan perawatan, sehingga mendukung studi genetika yang lebih efisien. Keunggulan ini membuat Drosophila sp. menjadi salah satu subjek penelitian favorit dalam bidang biologi dan genetika modern. Penelitian ini bertujuan untuk mengidentifikasi jenis lalat buah (Drosophila sp.) yang tertangkap menggunakan umpan berupa buah manis (pisang) dan buah masam (jeruk nipis) di Perumahan Ridena, Mendalo, Jambi. Metode penelitian ini menggunakan observasi alami dengan memasukkan buah ke dalam botol perangkap dan mengamati spesies yang tertangkap menggunakan mikroskop stereo serta kunci determinasi. Hasil penelitian menunjukkan bahwa lalat buah yang tertangkap hanya terdiri dari satu spesies, yaitu Drosophila melanogaster, dengan jumlah individu yang berbeda pada setiap jenis buah: 8 ekor pada pisang dan 7 ekor pada jeruk nipis. Kesimpulan menunjukkan bahwa perbedaan rasa buah mempengaruhi daya tarik lalat buah, namun tidak mengubah spesies yang tertangkap.     The In Silico Study of Bioactive Compounds from Some Tropical Trees as Inhibitors of Papain-Like Protease SARS-COV-2

    ABSTRACT. Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) is the causal virus of the global COVID-19 pandemic. Furthermore, it is known to encode 4 structural and 16 non-structural proteins, one of which is Papain-Like protease (PLpro). This protein is an essential enzyme that plays a role in the maturation of viral polyproteins, the formation of replicate-transcriptase, and the signaling of the host's innate immune response to viral infections. To prevent the activities of PLpro, several compounds with inhibitory effects have been developed from natural sources. Flavonoid compounds from bay leaf (Syzygium polyanthum) and mahogany bark (Swietenia macrophylla), as well as phenolics from white teak bark (Gmelina arborea Roxb.), have been reported to have antiviral activities, but there are no studies on their potential as PLpro SARS-CoV-2 inhibitors. Therefore, this study aimed to investigate the PLpro SARS-CoV-2 inhibitory effects of 11 bay leaf, 3 mahogany bark, and 6 white teak bark test ligands within silico method using YASARA and PLANTS. The best test ligands obtained were epigallocatechin-3-gallate (-98.93 kcal/mol), catechin (-80.10 kcal/mol), epicatechin (-80.12 kcal/mol), and balanophonin (-83.36 kcal/mol) based on energy binding values surpassing the reference ligands ribavirin (-74.06 kcal/mol) and benserazide (-90.93 kcal/mol). Additionally, these four ligands satisfied physicochemical, pharmacokinetic, and toxicity predictions, in contrast to the natural ligand VIR251, which carries the risk of inhibiting hERG, potentially resulting in fatal outcomes. Based on the results, the compounds could be developed as effective alternative drugs in inhibiting PLpro SARS CoV-2.   Keywords: Covid 19, molecular docking, Papain-Like protease, SARS CoV

    Development and Characterization of Edible Films from Sodium Alginate and Arrowroot Starch Plasticized with Sorbitol

    ABSTRACT. The demand for eco-friendly packaging is growing in industrial applications. This study aimed to develop edible films using sorbitol, sodium alginate (SA), and arrowroot starch (ARS). Sorbitol, serving as a plasticizer, was evaluated for optimal concentration, while various SA:ARS ratios were tested for film formation. Physicochemical properties, including thickness, color, tensile strength, elongation, moisture content, opacity, water solubility, water vapor transmission rate, biodegradability, FTIR, and SEM morphology were analyzed. Results indicated that 0.5% sorbitol produced films with ideal thickness, color, tensile strength, and elongation. SA:ARS ratios significantly influenced film properties, with the 0.9:0.1 ratio yielding a thin, smooth, and highly biodegradable film.
