Localization of disulfide bonds in ribonuclease using low pH trypsin and LC-ESI-QTOF-MS
We evaluated a method to localize disulfide bonds in bovine pancreatic ribonuclease (RNase) by applying low pH trypsin protein digestion with a bottom-up LC-ESI-QTOF-MS approach. The goal was to minimize disulfide bond scrambling during sample preparation. By using N-ethylmaleimide (NEM) for alkylation of free cysteines, we achieved 92% sequence coverage and successfully identified the native disulfide bonds between specific cysteine residues. However, scrambled disulfide bonds were also observed. Our results indicate that while this low pH digestion method helps preserve native disulfide bonds, further refinement is needed to fully prevent disulfide bond rearrangement during sample preparation

Large language models in cybersecurity ::threats, exposure and mitigation
This open access book provides cybersecurity practitioners with the knowledge needed to understand the risks of the increased availability of powerful large language models (LLMs) and how they can be mitigated. It attempts to outrun the malicious attackers by anticipating what they could do. It also alerts LLM developers to understand their work's risks for cybersecurity and provides them with tools to mitigate those risks.
The book starts in Part I with a general introduction to LLMs and their main application areas. Part II collects a description of the most salient threats LLMs represent in cybersecurity, be they as tools for cybercriminals or as novel attack surfaces if integrated into existing software. Part III focuses on attempting to forecast the exposure and the development of technologies and science underpinning LLMs, as well as macro levers available to regulators to further cybersecurity in the age of LLMs. Eventually, in Part IV, mitigation techniques that should allow safe and secure development and deployment of LLMs are presented. The book concludes with two final chapters in Part V, one speculating what a secure design and integration of LLMs from first principles would look like and the other presenting a summary of the duality of LLMs in cyber-security.

Do we offer what they ask for? Comparing sustainable sonsumer demands with sustainability offers of international tour operators
Consumer behaviour in tourism is influenced by a growing concern for sustainability in many Western societies. How do today’s customers interpret sustainable tourism and how do international tour operators respond to cross-cultural sustainable consumer demands? The purpose of this study is to critically analyse the interpretation of sustainable tourism offerings in outbound tourism in order to improve the promotion and product development of international tour operators. Consumer demand is analysed using a mixed methods approach to combine the advantages of quantitative and qualitative research. The results confirm the different perceptions of sustainable tourism between industry professionals and tourists. The information on sustainability factors provided by tourism suppliers does not meet the needs of tourists to make informed choices. There is a need for customer-centered communication and fact-based information tailored to different customer segments
Stock market reaction to the voluntary adoption of nature-related financial disclosure ::an event study
This article investigates the stock market reaction to the voluntary adoption of nature-related financial disclosures (TNFD) Recommendations, which focus on nature-related dependencies, impacts, risks, and opportunities. Investors may view this adoption as either value-destroying or value-enhancing. Using an event-study methodology, we analyze a global sample of listed companies. The results reveal significant negative cumulative abnormal returns when companies announce their adoption of the TNFD Recommendations. This negative effect is stronger for firms adopting the Recommendations as of fiscal year 2025 (versus 2024) and for firms in the basic materials, real estate, and utilities industries
Comparison between the effects of guided and free museum visits on the physical demands and well-being of sedentary individuals over 50 ::an observational comparative study
Background : Cultural activities can be a valuable means of supporting individuals who are not receptive to physical activities. The objective was to assess the number of steps taken during the guided or free visits among sedentary individuals and compare physical capacities and well-being before and after visits for the entire group and between middle-aged and older age subgroups. Methods : The observational study included 80 sedentary participants over 50 years old. Of those, 40 participated in a guided visit and 40 visited the museum freely (one session 60–75 minutes) to the Ariana and Museum of Art and History in Geneva. Before the visit, balance, gait speed, quadriceps strength, fatigue, stress, pain, and well-being were assessed. During the visit, the participants had an activity tracker to measure the step numbers. In the end, all tests were repeated and satisfaction was assessed. Depending on the normality of the data, a parametric or nonparametric test was conducted for intergroup and intragroup comparisons. Results : Participants took 889 ± 381 steps during the free visit and 819 ± 244 steps during the guided visit (p = 0.195). Balance and gait speed in dual-task activities (free visit), stress, fatigue, and well-being showed improvement after the visits (p < 0.050). For older individuals (≥ 65 years), the guided visit led to more favorable well-being outcomes, whereas in younger individuals (< 65 years), the free visit led to increased physical demands. Conclusion : Both visit modalities exceed the threshold of 775 steps. A museum visit is a relevant intervention to physically stimulate sedentary individuals and improve their well-being
