論文内容の要旨 
 
The development of the mammalian cerebral cortex is a highly intricate and meticulously orchestrated series of events. 
Especially, accurate mitotic division and differentiation of neural stem/progenitor cells (NSPCs) are crucial for the 
coordinated generation of progenitors and neurons, and accurate positioning of the neurons is critical as a prerequisite 
for higher brain functions. In the nervous system, members of the Kinesin superfamily motor proteins (Kifs) are 
localized in both mitotic and post-mitotic cells. While the role of Kifs in neurons is well-established, their functions 
in the NSPCs during embryonic brain development remain less explored. Here, I investigated the pivotal role of two 
Kifs, i.e., Kif23 (N-Kif) and Kif2C (M-Kif), in the embryonic mouse NSPCs. I found that Kif23 and Kif2C were 
highly expressed in the mitotic NSPCs and exhibited dynamic changes in the localization during cell cycle. 
Knockdown (KD) of Kif23 led to precocious neurogenesis, possibly due to an accelerated cell cycle exit as a result of 
disrupted mitotic spindle orientation and impaired cytokinesis. Kif23-KD induced upregulation of the -H2AX-p53-
p21 signaling pathway, ultimately culminating in cytokinetic failure. In addition to precocious neuronal differentiation, 
Kif23 deficiency caused an increase in apoptosis of differentiated neurons. Furthermore, Kif23 depletion perturbed 
the apical surface structure of NSPCs and disrupted the proper localization of apical junctional proteins. To delve 
deeper into the neuropathology of microcephaly, I assessed the function of human KIF23 in the mouse Kif23-KD 
model. The data demonstrated the successful rescue of Kif23-KD-induced phenotypes by introducing wild-type human 
KIF23, but not with a microcephaly-associated variant of KIF23. To understand the function of another target Kif, 
Kif2C, a similar KD approach was used as for Kif23. Like Kif23, KD of Kif2C induced a reduction in NSPC 
proliferation and acceleration of cell cycle exit, eventually impaired maintenance of the NSPC pool. Kif2C-KD also 
caused mitotic defects and chromosomal segregation errors during mitosis, which in turn upregulated -H2AX-p53-
p21-induced cell cycle exit. Furthermore, Kif2C deficiency disrupted NSPC basal processes and impaired NSPC 
dependent neuronal migration. In conclusion, this study revealed novel roles of two mitotic Kifs, i.e., Kif23 and Kif2C, 
in the NSPCs in cortical development. These findings may provide new insights into the molecular mechanisms 
underlying congenital brain anomalies such as microcephaly.  
 
  
論文審査結果の要旨 
 
大脳皮質発達においては、神経幹/前駆細胞（NSPC）の正確な細胞分裂によるニューロン分化が必
須である。神経系では、モータータンパク質のキネシン(Kif)はニューロンで機能するが、胚発生中
の NSPC での機能はあまり理解されていない。 
Sharmin Naher 氏は、E14.5 マウス終脳の空間トランスクリプトームおよび単一細胞 RNA シーケン
シング解析により、Kif23 と Kif2C が増殖する NSPC に発現することを発見し、免疫染色によりこ
れらのタンパク質が細胞周期依存的な細胞内局在を示すことを明らかにした。 
NSPC における Kif23 の機能解明のため、子宮内電気穿孔法（IUE）による Kif23 のノックダウン
（Kif23-KD）を行ったところ、早期の神経分化亢進が認められた。紡錘体微小管に局在する Kif23
の KD により紡錘体形成の異常や細胞分裂の角度が変化した。Kif23-KD ニューロンでの凝縮核・二
重核の検出により、細胞質分裂が完了できないことが示唆された。加えて DNA 損傷応答マーカー
γ-H2AX と下流分子 p53、p21 の発現が上昇し、細胞周期停止と細胞死の経路の活性化が検出され
た。さらに Kif23-KD による NSPC 頂端部の表面構造の乱れが認められた。小頭症の病態理解のた
め、マウスに野生型のヒト KIF23 を導入したところ、Kif23-KD 表現型を回復できたが、小頭症関連
変異型 KIF23 では回復できなかった。以上より、Kif23 が神経発生を制御し、小頭症の病態発生に
関連することが考えられた。 
Kif2C においても、IUE による Kif2c-KD を行った。その結果、Kif2C-KD による早期の神経分化亢
進およびγ-H2AX-p53-p21 細胞周期停止経路の発現上昇による細胞周期からの早期離脱が認められ
た。さらに、Kif2C-KD により有糸分裂や染色体分離の異常が生じた。また Kif2C-KD ではニューロ
ンの放射方向への移動が傷害され、これは NSPC の放射状突起消失による可能性がある。以上よ
り、Kif2C は神経分化とニューロン移動の両側面を調節することが示唆された。 
上記の研究成果は、Sharmin Naher 氏が科学者として自律的な研究活動を行うに必要な高度な研究能
力と学識を有することを示しており、Sharmin Naher 氏が提出した論文は博士（生命科学）の博士論
文として合格と認める。

旧東ドイツにおける難民・移民の受容と地域社会

 ドイツの難民政策の基本になっているのは、ドイツ連邦共和国基本法第 16a 条第１項
の「政治的に迫害されている者は、庇護権を有する。」という条文で、それが、ナチス
のユダヤ人迫害・虐殺に対する反省から生じたということはよく知られている1。 
 ユダヤ人迫害により、多くのユダヤ人は故郷を追われ、難民となった。その原因とな
ったのがドイツであったからである。しかし、ドイツ人自身にも難民となった記憶があ
る。第二次世界大戦まで、多くのドイツ人がドイツ国外のヨーロッパ中東部に暮らして
いた。彼らは、ナチス支配と共にその土地にやってきたわけではなく、何百年もの間、
その地に定住していたが、第二次世界大戦後、ナチスと同一視されて、断罪され、ドイ
ツ領内へ追放された。近年では、難民としてのドイツ人の記憶も取り上げられることが
多くなっている2。旧東ドイツのザクセン＝アンハルト州では、1945 年に住民の３分の
１近くが、被追放者だったとされる3。 
 こうした時代背景のもと、2015 年に「欧州難民危機」が生じた。中東・アフリカでの
政治的混乱の中、その地、特にシリアから多くの難民が、ヨーロッパを目指した、もち
ろん、本特集のクノスト氏の論考にあるように、この状況は突然起こったわけではない。
ドイツでは当時のメルケル首相が、人道的見地から移民を歓迎したこともあり、多くの
難民の目的地となった。しかし、グリュットナー氏の論考にあるように、欧州連合 EU
のダブリン規則では、難民は EU 域内で最初に入国した国で、難民申請をすることにな
っており、空路の入国以外では、ドイツがその申請国になることはあり得ない。ドイツ
の態度は、EU の境界国に負担を強い、不満を生じることとなった。また、難民自身も、
ドイツより貧しく、難民支援の環境が整っていない境界国に留まることをよしとしなか
った。 
 さて、この特集の日本の執筆者は、2017年度から 2020年度の科学研究費基盤研究（B）
「EU における難民の社会統合モデル―ドイツ・ハレ市の先進的試みの可能性と課題―」

と 2021 年度から 2024 年度の科学研究費基盤研究(B)「EU における移民・難民統合政策
の課題と展望―旧社会主義圏を中心に―」の助成を受け、ドイツの執筆者と共に共同研
究を遂行してきた。本特集の和英の翻訳もこの助成を受けている。また、これらの共同
研究は、2016 年に東北大学内のプロジェクトで、本特集執筆者の藤田氏らがハレを訪
問したことに始まった。 
 ハレを調査地として選んだことは偶然ともいえるが、前述のように、旧東ドイツから
多くの被追放民を受け入れたザクセン＝アンハルト州に位置していること、更に歴史を
遡ると、三十年戦争期にボヘミアからの避難民を受け入れた実績まであること、そして、
本特集の論考にあるように、難民・移民の受け入れに独自の試みを成していたことなど、
調査地にふさわしいことがわかった。 
 難民・移民排斥と極右の台頭で取り上げられることの多い旧東ドイツ、しかも州は違
うとはいえ、ライプツィヒと近いのにも関わらず、排斥は目立たない。難民・移民統合
のための行政と市民のネットワークがうまく機能しているようであった。 
 人口約 24 万人のハレは、ザクセン＝アンハルト州の州都マクデブルクをわずかに上
回り、ザクセン＝アンハルト州最大の人口をかかえる。もともと塩水泉が沸き、製塩業
で栄え、現在もその遺跡が残る。また、作曲家ヘンデルの生誕地としても知られ、「ヘ
ンデルの町」を名乗っている。また、本特集のドイツの執筆者が関係するマルティン・
ルター大学ハレ・ヴィッテンベルクを抱える大学町として、多くの学生が住んでいる。
17 世紀末からほぼ大学と同じ歴史を誇る学術機関フランケ財団の存在も、ハレの文化
的香りを高めている。難民・移民統合における大学や学生の役割については、クノスト
氏の論考に書かれているとおりである。 
 製塩業のほかのハレの産業としては、19 世紀にドイツ最古のチョコレート工場が建
設され、現在も盛業中であるが、社会主義期には二つの化学工場も建設され、ハレの代
表的産業となった。工場労働者用に建設された団地群からなるハレ＝ノイシュタット地
区は、現在では移民が集住する地域となっている。 
 本特集では、ハレ及び近隣のライプツィヒにおける８年間の研究成果の一部を公表す
る。

Acceptance of Refugees and Immigrants in Former 
East Germany and Its Local Communities: 
Introduction

   The basis of Germany's refugee policy is Article 16a, Section 1 of the Basic 
Law of the Federal Republic of Germany, which states, "Persons persecuted on 
political grounds shall have the right of asylum." It is well known that this 
article arose from a reflection on the Nazi persecution and genocide of the 
Jews.1 
   Due to the persecution of Jews, many Jews were forced to leave their 
homeland and became refugees. Germany was the cause of the persecution of 
Jews. However, Germans themselves also have memories of becoming refugees. 
Until World War II, many Germans lived outside of Germany namely in Central 
and Eastern Europe. They did not come to the land with the Nazi occupation, 
but settled there for hundreds of years. But after World War II, they were 
identified with the Nazis, condemned, and deported to German territory. In 
recent years, the memory of Germans as refugees has also been increasingly 
discussed.2  In the state of Saxony-Anhalt, nearly one-third of the residents 
in 1945 were deportees.3  
   Following this historical background, the "European refugee crisis" arose 
in 2015. Amid political turmoil in the Middle East and Africa, many refugees 
from that region, especially from Syria, set out for Europe, and of course, as 
Knost discusses in this special issue, this situation did not happen suddenly. 
Germany became a destination for many refugees, because then-Chancellor

Merkel welcomed refugees from a humanitarian standpoint. However, as 
Grüttner argues, under the Dublin Regulation of the EU, refugees are supposed 
to apply for refugee status in the first country they enter within the EU, and 
Germany cannot be that country of application, except for entry by air. 
Germany's attitude has placed a burden on the EU's bordering countries and 
has caused their dissatisfaction. The refugees themselves did not want to 
remain in bordering countries that were poorer than Germany and less 
conducive to refugee assistance. 
   The Japanese authors of this special issue have conducted joint research 
with the German authors, funded by Grant-in-Aid for Scientific Research (B) 
"Social Integration Models for Refugees in the EU: Possibilities and 
Challenges of Advanced Attempts in Halle, Germany," from FY2017 to FY2020, 
and Grant-in-Aid for Scientific Research (B) "Migrant and Refugee Integration 
Policies in the EU: Challenges and Prospects: Focusing on the Former Socialist 
Bloc" for the fiscal years 2021 to 2024. The Japanese-English translation of 
this special issue has also been supported by this grant. In addition, these joint 
research projects began in 2016 when the authors of this special issue, Prof. 
Fujita and her colleague, visited Halle as part of a project within Tohoku 
University. 
   The choice of Halle as the research site can be seen as a coincidence, but 
as mentioned above, it is located in Saxony-Anhalt in former East Germany, 
which received many deportees, and further back in history, it even received 
refugees from Bohemia during the Thirty Years' War. The site was found to be 
a suitable location for the study, as it had made its own attempts to receive 
refugees and immigrants. 
   Located in the former East Germany, which is often featured for its 
exclusion of refugees and immigrants and the rise of the far right, and despite 
its proximity to Leipzig, albeit in a different state (Sachsen), exclusion was not 
noticeable in Halle. The network of city government and citizens for 
integration of refugees and immigrants seemed to be functioning well. 
   With a population of approximately 240,000, Halle has the largest 
population in Saxony-Anhalt, slightly larger than that of Magdeburg, the 
capital of Saxony-Anhalt. Halle originally prospered from saltwater springs 
and the salt manufacturing industry, the remains of which can still be seen

today. It is also known as the birthplace of the composer Händel, and calls 
itself "Händel's Town. The presence of the Franke Foundation, an academic 
institution with a history almost identical to that of the university since the end 
of the 17th century, also adds to Halle's cultural flavor. The role of universities 
and students in refugee and immigrant integration is described in Knost's 
discussion. 
   In addition to the salt industry, the oldest chocolate factory in Germany 
was built in the 19th century and is still thriving, and two chemical factories 
were built during the socialist period and became representative industries in 
Halle. The Halle-Neustadt area, which consists of apartment complexes built 
for factory workers, is now home to many immigrants. 
   In this special issue, we publish some of the results of eight years of our 
research in Halle and nearby Leipzig.
