    FROM TRADITIONAL SOLAR CHIMNEY POWER PLANTS TO NEW APPROACHES

    Solar energy holds great potential for sustainable energy. Solar energy, harnessed in various ways, can be used directly to generate electricity or indirectly to generate both thermal and electricity. One system that allows for indirect electricity generation from the sun is the solar chimney power plant (SCPP). With its simple structure consisting of a collector, chimney, and turbine, the SCPP uses the solar energy it absorbs to heat the air. The heated air is accelerated upwards by the density difference and the vacuum effect of the tall chimney at the center of the system. The kinetic energy of the accelerated air is converted into electricity by the turbine. This study evaluates the evolution of traditional SCPP to innovative approaches and its potential for generating clean water. Airflow over a seawater pool located at the center of a hybrid system, built with integrated photovoltaic modules, increases the relative humidity of the system air. The water is then condensed from the moist air discharged through the chimney, producing pure water. The system's potential and the potential of SCPP for generating clean water are evaluated.&nbsp;</p

    The combined effect of the gene copy number and chaperone overexpression on the recombinant bovine chymosin production in Pichia pastoris, with mutant ADH2 promoter

    Chymosin is an enzyme used to coagulate milk, in the cheese industry. This study aimed to increase recombinant production of the chymosin in Pichia pastoris by determining the optimum copy number and overproduction of a Protein Disulfide Isomerase (PpPDI) chaperon protein. Bos taurus chymosin was expressed under the control of a mutant ADH2 promoter. The clones containing 1-4 gene copy numbers of the chymosin were constructed using the in vitro cloning method, and the effect of chaperone protein on chymosin secretion was investigated. The enzyme production levels are 4, 6.3, 4.5, and 3 IMCU/mL for 1, 2, 3, and 4-copy clones. The secreted chymosin levels increased up to two copies, and increasing the number of copies decreased the secretion level. Therefore, PpPDI was over-expressed in the clones regulated with the ADH2 promoter. The over-expression of PDI gene increased chymosin secretion in clones compared to the counterpart host. However, the highest chymosin level was obtained with C2 (2-copy chymosin containing clone; 6.3 IMCU/mL) and C2P2 (2-copy chymosin/2copy PDI containing clone; 8.2 IMCU/mL). The maximum production was 39 IMCU/mL with the clone C2P2 in the fermenter scale production. The enzyme activity increased approximately 2-fold by adding two copies of the chaperone protein. The combined effect of gene copy number and chaperone overexpression on chymosin production was investigated. Two copies of the chymosin and PpPDI genes were the optimum among the tested clones

    Tarihte davranışları, ilkeleri ve eylemleri ile dünyaya damgasını vurmuş önemli kişilikler bulunmaktadır. Bunlardan biri de mutasavvıf, âlim ve İslam filozofu olarak tanınan Hacı Bektaş Veli’dir. Verdiği değerli eserlerin yanı sıra bir “Toplum Hocası” kimliğiyle insanlara doğru yolu göstermeye hayatını adamış olan Hacı Bektaş Veli, nasihatlerini ilettiği özlü sözleriyle de yaygın biçimde tanınmaktadır. Bu özdeyişler içeriği ve yapısal özellikleri açısından dilbilimsel bir incelemeye değer görülmektedir. Bu çalışmanın amacı, Hacı Bektaş Veli’nin özdeyişlerini dilbilgisel ve anlambilimsel düzlemde inceleyerek sistematik olarak tekrarlanan dilbilgisel yapıların ve anlambilimsel düzeneklerin işlevlerini ortaya çıkarmak ve elde edilen bulguları Bektaşi geleneği bağlamında yorumlamaktır. Çalışmanın derlemi Nevşehir Hacı Bektaş Veli Üniversitesi, Hacı Bektaş Veli Araştırma ve Uygulama Enstitüsü’nün resmi sitesindeki “Hacı Bektaş Veli Öğretileri” sayfasında yer alan, toplam 58 tümceyi kapsayan 31 özdeyişten oluşmaktadır. Çalışmanın kuramsal çerçevesinini oluşturan Söylem Çözümlemesi uyarınca derlem büyük ölçekli ve küçük ölçekli metinsel düzeyde incelenerek özdeyişlerin bağlamsal, dilbilgisel ve anlambilimsel özellikleri nitel araştırma yöntemiyle saptanmıştır. Elde edilen bulgular Hacı Bektaş Veli’nin bilge kimliğinin yanı sıra dil bilincine sahip olduğunu da ortaya koymakta ve bu yönüyle onun eğitimci kimliğini desteklemektedir.There are important figures in history who have left their mark on the world with their behavior, principles and actions. One of them is Hacı Bektaş Veli, known as a Sufi, scholar and Islamic philosopher. In addition to his valuable works, Hacı Bektaş Veli, who devoted his life to showing the right path to people as a “Community Teacher”, is also widely known for his aphorisms in which he conveyed his advice. These aphorisms are worthy of a linguistic analysis in terms of their content and structural features. The aim of this study is to analyze Hacı Bektaş Veli's aphorisms at the grammatical and semantic level, to reveal the functions of the systematically repeated grammatical structures and semantic devices fulfill, and to interpret the findings in the context of the Bektshi tradition. The corpus of the study consists of 31 aphorisms covering a total of 58 sentences on the page “Hacı Bektaş Veli’s Teachings” on the official website of Hacı Bektaş Veli Research and Application Institute of Nevşehir Hacı Bektaş Veli University. In accordance with Discourse Analysis, which constitutes the theoretical framework of the study, the corpus was analyzed at large-scale and small-scale textual level and its contextual, grammatical and semantic features were determined by qualitative research method. The findings support Hacı Bektaş Veli's identity as an educator by revealing his language awareness as well as his wise personality
