    Effects of pesticides on sperm quality in honey bee (Apis mellifera L.) drones

    In this study, the effects of pesticides, which are extensively used in Turkish agriculture, on drone semen were investigated. Some of the most used pesticides in Turkey; abamectin, cypermethrin, glyphosate, and spinosad were selected. The study consisted of four pesticide groups, the solvent for these pesticides (methyl alcohol), and a control group, a total of six groups. There was a total of 36 colonies used in the experiment, with six colonies in each group. Before beginning the experiment, the colonies were homogenized, and sister queens were admitted in all colonies. When drone eggs began to form, pesticides were applied to the colonies. Sperm was taken 17 days after the drones emerged reached maturity. Semen volume and sperm count measured by spectrophotometer were recorded, and sperm concentration was calculated based on these two parameters. In addition, sperm length and viability were also assessed. When sperm concentration was examined, the lowest concentration was found in drones collected from abamectin-treated colonies (P 0.05). In the study conducted to determine the stress caused by pesticides in the larval period, it was found that abamectin and cypermethrin caused stress at the applied dose in the larval period (P < 0.05)

    Molecular Detection and Phylogenetic Analysis of Orf Virus From Dermatological Lesions in the Teats of Goats

    BackgroundThe orf virus (ORFV) is a viral pathogen that primarily causes contagious ecthyma in humans and different ruminants. The infection, which is common worldwide, causes large-scale economic losses to animal breeders.Objective and MethodsIn this study, tissue samples collected from eight randomly selected goats with dermatological lesions on the teats were examined in different goat herds. B2L gene-specific primer pairs (PP1, PP3 and PP4) were used to reveal the presence of ORFV by molecular methods and for phylogenetic analysis.ResultsViral DNA was detected in four of eight tissues using the semi-nested PCR method. In addition, the data obtained by performing sequence analyses of the amplicons with positive results were compared with the information of different ORFV isolates registered in the GenBank database. Based on the sequence analysis of the field isolates obtained in our study, it was found that the nucleotide similarities among these isolates and those from Asian countries were 100%. Furthermore, ORFV isolates collected from different species and produced in T & uuml;rkiye over various periods exhibited homologous nucleotide sequences with similarities ranging from 98.1% to 98.8%. In the phylogenetic tree drawn based on the B2L genomic region, it was observed that our field isolates were classified in Group I together with other Turkish and Asian strains.ConclusionAs a result, while other pathogenic agents are considered the cause of disease in goats with dermatological lesions on their mammary tissue, the ORFV should also be evaluated, and protection and control programs should be prepared accordingly

    Hatay İli Reyhanlı İlçesi Zeytin Ağaçlarının Yaprak ve Toprak Örnekleri ile Beslenme Durumunun Belirlenmesi

    Çalışma, Hatay ili Reyhanlı ilçesini kapsayacak şekilde belirlenen zeytin bahçelerinin bitki besin elementlerinin durumunu belirlemek amacıyla yapılmıştır. Bu amaç için 15 farklı zeytin bahçesinden 0-30 ve 30-60 cm olmak üzere iki farklı derinlikten 30 adet toprak ve 15 adet yaprak örneği alındı. Toprak ve yaprak örneklerinde bazı makro-mikro besin element analizleri ile topraklarda ek olarak bünye, kireç, pH, tuz, KDK, organik madde analizleri yapılmıştır. Elde edilen bulgulara göre, araştırma alanı topraklarının; tamamı killi tın bünyeli, hafif alkalin reaksiyonlu, genelde organik maddece fakir, tuzsuz, kireçli ve çok kireçli sınıfında yer aldığı belirlenmiştir. Çalışmada, toprakta bulunan K, Mg, Fe ve Cu içerikleri ile ve yaprakta bulunan K, Mg, Fe ve Cu içerikleri uyumlu olarak belirlenmiştir. Fakat, toprak örneklerde yapılan analizlerde N (%26.67) ve P (%6.67) noksan olarak bulunmasına rağmen, yaprak örneklerinin tümünün N ve P içeriklerinin yeterli bulunmuştur. Ayrıca, toprakların Zn ve Mn içeriklerinin büyük bir kısmının yeter sınırın altında olmalarına rağmen, yaprak örneklerinde Zn içeriği tüm örneklerde, Mn içeriklerinin ise büyük çoğunluğunun (%93.33) yeterli olarak bulunmuştur. Araştırma alanından elde edilen verilere göre, Hatay ili Reyhanlı ilçesi zeytin bahçelerinde bazı bitki besin elementlerinin toprak ve bitki analizleri neticesinde belirlenen ve yukarıda belirtilen dengesizlikler, bitkilerin beslenmelerinde bazı sorunların olduğunu, bölgede olan zeytin bahçelerinde analize dayanmayan gübreleme yapıldığı belirlenmiştir. Bu sebep ile zeytin bahçelerinde beslenme sorunları net olarak belirlendikten sonra, yaprak ve toprak analizlerinin ve bu analizlere dayalı gübreleme programları oluşturmanın ne kadar önemli olduğu hakkında çiftçiler bilinçlendirilmelidir.The study was carried out to determine the nutritional status of olive orchards selected to represent Reyhanlı district of Hatay province. For this purpose, 30 soil and 15 leaf samples were taken from 15 different olive gardens from two different depths: 0-30 and 30-60 cm. Some macro-micro nutrient element analyzes were performed on soil and leaf samples, and additional texture, lime, pH, salt, KDK and organic matter analyzes were performed on soils. According to the findings, the soils of the research area; It has been determined that it has a clay loam texture, slightly alkaline reaction, is generally poor in organic matter, in the salt-free, calcareous, and very calcareous class. In the study, K, Mg, Fe and Cu contents were determined to be compatible with soil and leaf contents. However, although 26.67% of the N content and 6.67 % of the P content of the soil samples were found to be deficient, the N and P contents of all leaf samples were found to be sufficient. In addition, although most of the Zn and Mn contents of the soil were below the sufficient limit, it was determined that all the leaf Zn contents, and most of the Mn contents (93.33 %) were sufficient. As a result, the findings obtained reveal that the imbalance of some plant nutrients determined in the soil and plant analysis in the olive groves of Reyhanlı district of Hatay province has some problems in the nutrition of the plants, and the existence of plant nutrition programs that are not based on soil and plant analysis in the olive orchards in the region. For this reason, after the problem is clearly identified by increasing olive nutrition studies, in addition to the necessity of appropriate fertilizer studies and determination of doses, producers need to give due importance to soil, and leaf analyzes and raise awareness about nutrition based on scientific foundations

    Küresel Popüler Kültür Metası: K-Pop ve K-Drama Türk Fanları Üzerinden Analiz

    Küreselleşme, dijitalleşme ve medya teknolojilerindeki gelişmeler, kültürel ürünlerin uluslararası dolaşımını hızlandırarak popüler kültürün küresel ölçekte etkisini artırmıştır. Bu bağlamda Güney Kore’nin kültürel diplomasi ve yumuşak güç stratejilerinin bir sonucu olarak ortaya çıkan Kore Dalgası (Hallyu), kısa sürede dünya çapında yayılmış ve özellikle gençler arasında güçlü bir etkileşim yaratmıştır. Bu çalışmanın amacı, Kore popüler kültürünün Türkiye’deki gençler üzerindeki etkilerini incelemek; kimlik, aidiyet, tüketim alışkanlıkları ve günlük yaşam pratikleri üzerindeki yansımalarını ortaya koymaktır.Araştırmada nicel yöntem benimsenmiş ve 526 katılımcıya anket uygulanmıştır. Veriler korelasyon, ANOVA ve t-testi gibi istatistiksel yöntemlerle analiz edilmiştir. Bulgular, katılımcıların %84,8’inin düzenli olarak K- Drama, %63,7’sinin K-Pop takip ettiğini; %47’sinin Kore mutfağına, %45,2’sinin Korece’ye, %44,5’inin ise Kore kozmetik ürünlerine ilgi duyduğunu göstermektedir. Katılımcıların yarısı her gün Kore içerikleriyle ilgilenmekte, %58,8’i Korece öğrenmek istemekte ve %60,3’ü Kore menşeli ürünlere yönelmektedir. Ayrıca katılımcıların yarısından fazlası Kore Dalgasının Türkiye’de yeni bir kültürel kimlik oluşumuna katkı sağladığını ifade etmiştir. Sonuç olarak, Kore Dalgası Türkiye’de yalnızca popüler bir eğlence fenomeni değil, aynı zamanda gençlerin kültürel aidiyet duygusu, kimlik inşası ve tüketim alışkanlıklarını etkileyen önemli bir olgu haline gelmiştir. Çalışma, Güney Kore’nin kültürel diplomasisinin başarısını ve Türkiye’deki gençlerin kültürel tercihlerindeki dönüşümü somut verilerle ortaya koymaktadır.</p

    Molecular characterization and population structure analysis of luffa (Luffa aegyptiaca Mill.) germplasm using inter‑Primer Binding Site (iPBS) retrotransposon markers

    Sponge gourd (Luffa aegyptiaca), an economically significant member of the Cucurbitaceae family, is widely cultivated in tropical and subtropical regions for its applications as a vegetable, medicinal plant, and fiber crop. This study aimed to genetically characterize 32 Luffa aegyptiaca genotypes using Inter-Primer Binding Site (iPBS) retrotransposon markers and evaluate their efficiency in detecting genetic diversity. A total of 25 iPBS primers were assessed, with an average polymorphism rate of 84.1% and a polymorphic information content (PIC) of 0.54, thereby indicating moderate-to-high genetic differentiation within the population. Primers such as iPBS-2231 and iPBS-2256 showed exceptional performance, achieving 100% polymorphism rates and high PIC values, while primers like iPBS-2253 exhibited limited effectiveness. The unweighted pair group method with arithmetic mean (UPGMA) dendrogram and Principal Coordinate Analysis (PCoA) revealed three main genetic clusters and unique outliers, with Genotype 17 identified as the most genetically distinct due to its lowest similarity rate (0.54). Additionally, genetic similarity was generally higher among genotypes from the same geographic region, although exceptions were noted. The findings emphasize the critical role of primer selection in genetic analyses and provide a foundational framework for breeding programs and conservation efforts. The study highlights the potential of genetically unique genotypes for enhancing diversity and advancing agricultural sustainability in Luffa cultivation
