Geographical Data Science and Spatial Data Analysis: Data for Chapter 7
This dataset contains "lsoa" a spatial dataset / layer of census areas for Liverpool from the UK Data Service and census attributes for these areas from Nomisweb (ONS); "oa" - the same source data as "lsoa" but at a different spatial resolution (LSOAs hold 1500 people and OAs hold 300); "properties" - this is house price data obtained from the Nestoria API. UK Census data can be downloaded from the Office of National Statistics (https://www.nomisweb.co.uk) and spatial layers of Census areas from the UK Data Service (https://borders.ukdataservice.ac.uk). The unemployment and age data were from the 2011 UK population census (https://www.nomisweb.co.uk ) and the greenspace proportions were extracted from the Ordnance Survey Open Greenspace layer (https://www.ordnancesurvey.co.uk/opendatadownload/products.html). The spatial frameworks were from the EDINA data library (https://borders.ukdataservice.ac.uk). The OAs and LSOAs are both projected to the OSGB projection (EPSG 27700). The OA layer also has a geo-demographic class label (OAC) from the OAC (see Gale et al. (2016)) of 8 classes as introduced in earlier chapters. Liquid water is known as the “universal” solvent, capable of dissolving a wide variety of different solutes. While much is now understood about the impact of solutes on water structure in binary solutions, it is much more challenging to deconvolute the potentially competing effects of more complex solutions. Here, we present a correlative NMR and neutron diffraction study to examine the solute induced perturbation of water structure and dynamics in a tertiary solution containing the naturally occurring osmolyte trimethylamine N-oxide (TMAO) and magnesium perchlorate (Mg(ClO4)2). We show that while TMAO and Mg(ClO4)2 perturb water structure in an opposing manner, the two solutes slow water dynamics in an additive manner. We quantify the relative ability of each solute to perturb water by introducing a weighting parameter, and show that TMAO is 1.54 times more effective at perturbing water structure and dynamics than Mg(ClO4)2. The combination of NMR, neutron diffraction and computational modelling offers unprecedented access to the structure and dynamics of more complex aqueous solutions, permitting deconvolution of solute specific perturbation of water. Such insight provides a new route to understand this universal solvent in the context of important and relevant aqueous environments
Supplementary Data - Regional policies targeting residential solid fuel and agricultural emissions can improve air quality and public health in the Greater Bay Area and across China
Air pollution exposure is a leading public health problem in China. The majority of the air pollution disease burden is from fine particulate matter (PM2.5) exposure, with smaller contributions from ozone (O3) exposure. Recent emission reductions have reduced PM2.5 exposure. However, levels of exposure and the associated risk remain high, some pollutant emissions have increased, and some sectors lack effective emission control measures. We quantified the potential impacts of policy scenarios on ambient air quality and public health across China. We show that PM2.5 exposure inside the Greater Bay Area (GBA) is strongly controlled by emissions outside the GBA. We find that reductions in residential solid fuel use and agricultural fertiliser emissions result in the greatest reductions in PM2.5 exposure and the largest public health benefits. A 50% transition from residential solid fuel use to liquefied petroleum gas outside the GBA reduced PM2.5 exposure by 15% in China and by 3% within the GBA, and avoided 191,400 premature deaths each year across China. Reducing agricultural fertiliser emissions of ammonia by 30% outside the GBA reduced PM2.5 exposure by 4% in China and by 3% in the GBA, avoiding 56,500 annual premature deaths across China. Our simulations suggest that reducing residential solid fuel or industrial emissions will reduce both PM2.5 and O3 exposure, whereas other policies may increase O3 exposure. Improving particulate air quality inside the GBA will require consideration of residential solid fuel and agricultural sectors, which currently lack targeted policies, and regional cooperation both inside and outside the GBA
Inter-domain dynamics in the chaperone SurA and multi-site binding to its outer membrane protein clients - dataset
SurA is the conserved major chaperone of outer membrane protein (OMP) biogenesis in the periplasm of Gram-negative bacteria, and plays a key role in cell envelope homeostasis and virulence. In E. coli, SurA comprises three domains: a core domain and two peptidyl prolyl isomerase (PPIase) domains (P1 and P2). The functional roles of these domains in chaperone activity, including their inter-subunit dynamics and interactions with substrates remain unclear. Here we have used chemical cross-linking, HDX-MS, oxidative labelling, single-molecule FRET and molecular dynamics simulations, to investigate the conformational dynamics of SurA in its apo state and to map the binding site(s) for its clients. We demonstrate that SurA predominantly samples conformations in solution in which the P2 domain is closer to the core and P1 domains than suggested by the extended conformation in its crystal structure. Using cross-linking experiments we show that SurA has multiple, yet specific, binding sites for OmpX located primarily in the core domain, and that residues spanning the whole OmpX sequence bind in these regions. HDX experiments additionally reveal that binding of OmpX, and the larger client OmpF, lead to inter-domain conformational changes between the core and P1 domains. Combining these data we generate a model for SurA OMP recognition in which unfolded OMP substrates bind in a cradle formed between the SurA domains, in a manner reminiscent of other ATP-independent chaperones. Structural flexibility between domains is likely a key feature of the SurA functional cycle, assisting in OMP recognition, binding and release of its broad range of OMP substrates for folding into the outer membrane
