    Inhibitors of the Small Membrane (M) Protein Viroporin Prevent Zika Virus Infection - dataset

    Flaviviruses, including Zika virus (ZIKV), are a significant global health concern, yet no licensed antivirals exist to treat disease. The small Membrane (M) protein plays well-defined roles during viral egress and remains within virion membranes following release and maturation. However, it is unclear whether M plays a functional role in this setting. Here, we show that M forms oligomeric membrane-permeabilising channels in vitro, with increased activity at acidic pH and sensitivity to the prototypic channel-blocker, rimantadine. Accordingly, rimantadine blocked an early stage of ZIKV cell culture infection. Structure-based channel models, comprising hexameric arrangements of two trans-membrane domain protomers were shown to comprise more stable assemblages than other oligomers using molecular dynamics (MD) simulations. Models contained a predicted lumenal rimantadine binding site, as well as a second druggable target region on the membrane-exposed periphery. In silico screening enriched for repurposed drugs/compounds predicted to bind to either one site or the other. Hits displayed superior potency in vitro and in cell culture compared with rimantadine, with efficacy demonstrably linked to virion-resident channels. Finally, rimantadine effectively blocked ZIKV viraemia in preclinical models, supporting that M constitutes a physiologically relevant target. This could be explored by repurposing rimantadine, or development of new M-targeted-therapies

    Data for Design of experiments investigation into the production of all cellulose composites using regenerated cellulosic textiles

    Data for All cellulose composites (ACCs) can be produced from native and man-made cellulosic fibres; use of the latter provides an additional application for waste-derived regenerated fibers. ACCs were prepared using an ionic liquid dissolution method, utilizing a regenerated cellulose (Tencel) textile, with and without an interleaf cellulosic film. A design of experiments methodology was applied to explore process-property relationships; concentration of the ionic liquid and the processing time and temperature were investigated. It was found that the film remained in-between the textile layers, rather than penetrating the fiber assembly, in contrast to our previous work on cotton-based ACCs. This is due to the structural differences between Tencel and cotton fabric. A multi-response optimization was conducted through a central composite face centered strategy, which captured the film system more strongly. Optimized processing conditions were identified, yielding a Young's modulus and strain-to-failure of 5.3 GPa and 3.5% respectively, validated through in-lab samples. Mechanical Testing Tensile strength, Young's modulus and strain-to-failure were evaluated using an Instron 5584 universal tensile tester according to ASTM D1846. Test specimens were cut using a laser cutter to a gauge length of 30 mm and width of 5 mm and tested using a crosshead speed of 10 mm/min. Density measurement A gravimetric approach was used to determine the densities of the optimized ACCs. The dimensions of cut specimen samples were measured using an RS PRO digital caliper to obtain the specimen volume, and subsequently weighed. For each ACC, three specimens were measured and weighed to obtain an estimation of error. A reference value of 1.5 g/cm3 was used for the density of the cellulosic materials within the ACC, drawing from existing literature; the density of plant fibers is reported to be between 1.4 and 1.5 g/cm3 [44]. Cellulose II is reported to have an approximate density at 1.5 g/cm3, and the density of bulk amorphous cellulose is estimated to be in the range of 1.48 to 1.5 g/cm3. Optical microscopy Cross-sections of the prepared ACCs were observed using an Olympus BH2 microscope in reflection mode. Composite thickness measurements were then extracted from these images utilizing ImageJ software. To enhance image clarity, samples were embedded in epoxy resin using a silicon mold and subsequently polished. To ensure an accurate representation of each sample was presented in this paper, multiple images were obtained from which thickness measurements were taken. For each ACC sample, an average thickness value, along with the corresponding standard error, was calculated from six measurements

    Appropriating the ‘real’ is a practice-led investigation that researches the extent to which verbatim theatre techniques can be appropriated to create a fictional performance. Criticisms of verbatim theatre’s ‘truth claims’ – that it is a re-presentation of the ‘real’ – are both dramaturgical and ethical. Addressing these concerns, the practice presented here sets out to experiment with “indecidability” (Lehmann, 2006:101), heightening the audience’s perception of the gap between ‘truth’ and ‘fiction’, chiefly by drawing attention to the role of the theatre maker. The practice-led research is made up by this trilogy of performances: The Unsettling; Everybody Always Tells the Truth; and Is It Different Now? and an accompanying thesis

    This dataset is associated with a paper on phase coexistence and transitions of a synthetic antiferromagnetic multilayer, and contains imaging using a number of different complimentary microscopies to study the transition mechanisms of the synthetic antiferromagnet. Samples were measured in the bulk using SQUID-VSM, imaged on the surface using magnetic force microscopy around the transition point and XMCD-PEEM at fields near zero, and imaged in transmission using both Fresnel and DPC Lorentz transmission electron microscopy at the transition point. This data was used to build up a picture of the transition mechanism of the synthetic antiferromagnet and image chiral textures therein

    Porcine cysticercosis is an important public health problem in sub-Saharan Africa. We carried out a cross-sectional survey of 744 pigs in 6 abattoirs in Rwanda. Pig characteristics were recorded and infection status was assessed using a commercial antigen-ELISA. Whole carcass dissection was carried out on a subset of 67 pigs, in order to estimate the diagnostic accuracy of the ELISA and to confirm the species of tapeworm present. The data indicate a high prevalence of porcine cysticercosis in Rwandan abattoirs
