    Effects of flavonoids dietary supplementation on egg yolk antioxidant capacity and cholesterol level

    An experiment was conducted to examine the effects of supplementing laying hen feed with different levels of hesperidin or naringin, bioflavonoids that are abundant and inexpensive by-products of citrus cultivation, on the yolk antioxidant capacity and cholesterol level. Seventy-two laying hens, approximately twelve months old, were assigned into 6 experimental groups of twelve hens each. One of the groups served as control (C) and was given a commercial basal diet, without bioflavonoid supplementation, whereas the other five groups were given the same diet further supplemented with hesperidin at low (750mg/kg of feed) (H1) or high (1500mg/kg) (H2) concentration or naringin at low (750mg/kg) (N1) or high (1500mg/kg) (N2) concentration or α-tocopheryl acetate (200mg/kg) (E). Measurements of yolk antioxidant capacity were performed on 8 eggs from each dietary group, at 0, 4, 7, 28 and 63 days after the beginning of the experiment. Yolk cholesterol level was determined on the final day (63th) of the experimental period. Oxidative stability of egg yolk, expressed as ng MDA/g yolk, was significantly improved in the hesperidin and naringin groups even from the first four days of the supplementation period (P<0.001). However, no flavonoids effect on yolk cholesterol level (mg/g) was observed. Antioxidant properties of flavonoids seem to be a promising natural agent for improving the health status and the shelf life of laying hens’ egg

    The MDR-involved human GSTA1-1, an important isoenzyme overexpressed in several tumors leading to chemotherapeutic-resistant tumour cells, has been targeted by 2,2′-dihydroxybenzophenones and some of their carbonyl N-analogues, as its potential inhibitors. A structure-based library of the latter was built-up by a nucleophilic cleavage of suitably substituted xanthones to 2,2′-dihydroxy-benzophenones (5–9) and subsequent formation of their N-derivatives (oximes 11–13 and N-acyl hydrazones 14–16). Screening against hGSTA1-1 led to benzophenones 6 and 8, and hydrazones 14 and 16, having the highest inhibition potency (IC50 values in the range 0.18 ± 0.02 to 1.77 ± 0.10 μM). Enzyme inhibition kinetics, molecular modeling and docking studies showed that they interact primarily at the CDNB-binding catalytic site of the enzyme. In addition, the results from cytotoxicity studies with human colon adenocarcinoma cells showed low LC50 values for benzophenone 6 and its N-acyl hydrazone analogue 14 (31.4 ± 0.4 μM and 87 ± 1.9 μM, respectively), in addition to the strong enzyme inhibition profile (IC50(6) = 1,77 ± 0.10 μM; IC50(14) = 0.33 ± 0.05 μM). These structures may serve as leads for the design of new potent mono- and bi-functional inhibitors and pro-drugs against human GTSs

    Within the last few years numerous polysaccharide extracts have been proposed as emulsion stabilizing agents. This increased interest arises from the fact that commonly used food polysaccharides like guar gum are used in non-food applications, mainly in petroleum refining and pharmaceuticals(Vaughnaetal). Along with the lower global production this has resulted in price fluctuations, consequently severe price increase and supply shortage(Bahamdanetal, Baratietal, Anonetal). From a dietary point of view, the viscous property of fenugreek gum (Trigonella foenum graecum L.)has been proved to reduce in vitro the absorption of glucose and the plasma levels of triglycerides and cholesterol in vivo and could be used when designing low - at emulsified products. Ultrasonic emulsification is a cost effective technique and the interest for scale- up is increasing,as it is considered a “Green Processing” technology for the manufacture of nanoemulsions

    Estimating the spatiotemporal evolution characteristics of diffusive hazards using wireless sensor networks

    There is a fast growing interest in exploiting Wireless Sensor Networks (WSNs) for tracking the boundaries and predicting the evolution properties of diffusive hazardous phenomena (e.g. wildfires, oil slicks etc.) often modeled as “continuous objects”. We present a novel distributed algorithm for estimating and tracking the local evolution characteristics of continuous objects. The hazard’s front line is approximated as a set of line segments, and the spatiotemporal evolution of each segment is modeled by a small number of parameters (orientation, direction and speed of motion). As the hazard approaches, these parameters are re-estimated using adhoc clusters (triplets) of collaborating sensor nodes. Parameters updating is based on algebraic closed-form expressions resulting from the analytical solution of a Bayesian estimation problem. Therefore, it can be implemented by microprocessors of the WSN nodes, while respecting their limited processing capabilities and strict energy constraints. Extensive computer simulations demonstrate the ability of the proposed distributed algorithm to estimate accurately the evolution characteristics of complex hazard fronts under different conditions by using reasonably dense WSNs. The proposed in-network processing scheme does not require sensor node clocks synchronization and is shown to be robust to sensor node failures and communication link failures, which are expected in harsh environments

    Targeted determination of more than 1500 micropollutants & transformation products in wastewater samples by liquid chromatography quadrupole-time-of-flight mass spectrometry with an accurate-mass database

    High resolution mass spectrometry has dramatically improved the possibilities of the environmental analysis. The present study describes the development of an analytical method, based on liquid chromatography quadrupole-time-of-flight mass spectrometry (LC–QToF-MS) for the target determination of more than 1500 contaminants of emerging concern (CECs) and transformation products (TPs) including, among others, pharmaceuticals, illicit drugs, personal care products, pesticides, industrial chemicals, and sweeteners in wastewater. Analytes were extracted from wastewater samples by mixed mode solid-phase extraction, and data were acquired through broad-band Collision Induced Dissociation (bbCID) mode, providing MS and MS/MS spectra, simultaneously, in both positive and negative ionization mode (two separate runs). The in-house mass spectral database was built by injection of standard solution of the analytes and it includes information of the retention time, parent ions and adducts, as well as fragment ions. The raw data were analyzed with Bruker Target Analysis 1.3 software. Retention time, accurate mass of the precursor ion and adducts, isotopic pattern, in combination with absence of the peak in the procedural blank were the parameters used for confirmation of the target compounds. Experimental fragment ions were also considered, along with the ion ratio, intensity and isotopic pattern. Furthermore, semi-quantitation of these contaminants was possible. The method herein presented, in addition of providing accurate information about the presence of a large number of relevant substances, has the advantage that the data generated can be further processed for suspect and non-target screening, expanding the information on the samples.
