    Remarks on the Messinian evaporites of Zakynthos Island (Ionian Sea, Eastern Mediterranean)

    Detailed mapping of the Neogene deposits on Zakynthos Island shows that the Messinian primary evaporite basins, formed over Ionian basement, are delimited by the westernmost outcrop of the Triassic evaporitic diapirs, located west of the Kalamaki-Argasi Messinian gypsum unit. The post-Miocene external Ionian thrust is emplaced west of the Triassic diapirs. Planktonic foraminifera biostratigraphy indicates that primary evaporite accumulation took place probably during the first stage of the Messinian salinity crisis (5.96-5.60 Ma), in shallower parts of a foreland basin, formed over the Pre-Apulian and the Ionian zone basement. Establishment of these depositional environments, before the Ionian thrust emplacement, was probably due to the particularities of the foreland basin, which extended from the external Ionian to the internal Pre-Apulian zone. Field observations, borehole data and an onshore seismic profile show that the Neogene sediments over the Pre-Apulian basement correspond to the foredeep through forebulge domain of the foreland basin, as it is documented from their spatial thickness distribution. In contrast, the Neogene sediments over the Ionian basement correspond to the wedge top of the foreland basin, which was less subsiding, as it is deduced by their reduced thickness. This lower subsidence rate was the result of the concurrent diapiric movements of the Ionian Triassic evaporites. In Agios Sostis area, located over Pre-Apulian basement, the Neogene sequence is intercalated by decametre-thick resedimented blocks consisting of shallow water selenite. To the southeast, this mass-wasting Messinian gypsum passes to mainly gypsum turbidite. In Kalamaki-Argasi area, located over Ionian basement, the shallow water environment led to the deposition of the observed primary gypsum. Erosion of the primary gypsum of both forebulge and wedge top supplied the foreland basin’s depocenter with gypsum turbidites. Η λεπτομερής χαρτογράφηση των Νεογενών αποθέσεων της Ζακύνθου δείχνει ότι οι λεκάνες πρωτογενούς Μεσσήνιας εβαποριτικής απόθεσης, που σχηματίστηκαν πάνω σε Ιόνιο υπόβαθρο, οριοθετούνται από τη δυτικότερη εμφάνιση των Τριαδικών εβαποριτικών διάπειρων (δυτικά της Μεσσήνιας ενότητας γύψου Καλαμάκι-Αργάσι). Η μετα-Μειοκαινική εξωτερική Ιόνια επώθηση τοποθετείται δυτικά αυτών των Τριαδικών διάπειρων. Βιοστρωματογραφική ανάλυση των συναθροίσεων πλαγκτονικών τρηματοφόρων στα Νεογενή ιζήματα δείχνει ότι η συσσώρευση πρωτογενούς εβαπορίτη έλαβε χώρα πιθανά κατά το πρώτο στάδιο της κρίσης αλμυρότητας του Μεσσηνίου (5.96-5.60 Εκατ.χρ.), στα αβαθέστερα τμήματα της λεκάνης προχώρας, η οποία σχηματίστηκε πάνω στο Προ-Απούλιο και Ιόνιο αλπικό υπόβαθρο. Ο σχηματισμός των περιβαλλόντων ιζηματογένεσης, πριν από την Ιόνια επώθηση, πιθανά οφείλεται στις ιδιαιτερότητες της λεκάνης προχώρας, η οποία εκείνο το διάστημα εκτείνονταν από την εξωτερική Ιόνια ως την εσωτερική Προ-Απούλια ζώνη. Νέα δεδομένα υπαίθρου, σε συνδυασμό με τη βιοστρωματογραφία, τα στοιχεία γεωτρήσεων και μία διαθέσιμη σεισμική τομή δείχνουν ότι τα Νεογενή ιζήματα πάνω στο Προ-Απούλιο υπόβαθρο αντιστοιχούν στον τμήμα της λεκάνης προχώρας, από το προβύθισμα (foredeep) ως την πρόσθια ανύψωση (forebulge), όπως αυτό διαπιστώνεται από την χωρική κατανομή του πάχους τους. Αντίθετα, τα Νεογενή ιζήματα πάνω στο Ιόνιο υπόβαθρο αντιστοιχούν στο τμήμα της λεκάνης προχώρας που βρίσκεται πάνω στο μετωπικό πρίσμα της ορογένεσης (wedge top), το οποίο βυθίζονταν λιγότερο όπως συνάγεται από το ελαττωμένο πάχος των ιζημάτων. Ο μικρότερος ρυθμός βύθισης οφείλεται στις διαπειρικές κινήσεις των Τριαδικών εβαποριτών. Στη Νεογενή ακολουθία, που βρίσκεται πάνω σε Προ-Απούλιο υπόβαθρο (περιοχή Άγιος Σώστης), παρεμβάλλονται επανιζηματοποιημένα μπλοκ δεκαμετρικού πάχους αποτελούμενα από σεληνίτη μικρού βάθους απόθεσης. Προς νότο, αυτή η βαρυτικά μεταφερόμενη μάζα της Μεσσήνιας γύψου μεταπίπτει κυρίως σε τουρβιδιτική γύψο. Στην περιοχή Καλαμάκι-Αργάσι που βρίσκεται πάνω σε Ιόνιο υπόβαθρο, το αβαθές περιβάλλον οδήγησε στην απόθεση της παρατηρούμενης πρωτογενούς γύψου. Η διάβρωση της πρωτογενούς γύψου τόσο της πρόσθιας ανύψωσης όσο και του μετωπικού πρίσματος επώθησης τροφοδότησαν το προβύθισμα της λεκάνης προχώρας με τουρβιδίτες γύψου

    The Interaction of the Chemotherapeutic Drug Chlorambucil with Human Glutathione Transferase A1-1: Kinetic and Structural Analysis

    Glutathione transferases (GSTs) are enzymes that contribute to cellular detoxification by catalysing the nucleophilic attack of glutathione (GSH) on the electrophilic centre of a number of xenobiotic compounds, including several chemotherapeutic drugs. In the present work we investigated the interaction of the chemotherapeutic drug chlorambucil (CBL) with human GSTA1-1 (hGSTA1-1) using kinetic analysis, protein crystallography and molecular dynamics. In the presence of GSH, CBL behaves as an efficient substrate for hGSTA1-1. The rate-limiting step of the catalytic reaction between CBL and GSH is viscosity-dependent and kinetic data suggest that product release is rate-limiting. The crystal structure of the hGSTA1-1/ CBL-GSH complex was solved at 2.1 A° resolution by molecular replacement. CBL is bound at the H-site attached to the thiol group of GSH, is partially ordered and exposed to the solvent, making specific interactions with the enzyme. Molecular dynamics simulations based on the crystal structure indicated high mobility of the CBL moiety and stabilization of the Cterminal helix due to the presence of the adduct. In the absence of GSH, CBL is shown to be an alkylating irreversible inhibitor for hGSTA1-1. Inactivation of the enzyme by CBL followed a biphasic pseudo-first-order saturation kinetics with approximately 1 mol of CBL per mol of dimeric enzyme being incorporated. Structural analysis suggested that the modifying residue is Cys112 which is located at the entrance of the H-site. The results are indicative of a structural communication between the subunits on the basis of mutually exclusive modification of Cys112, indicating that the two enzyme active sites are presumably coordinated

    The Use of Highly Specific GSTs towards the Development of Stress Tolerant Transgenic Plants (pp. 263-274)

    Glutathione transferases (GSTs) belong to a super-family of multifunctional proteins. GSTs play a key role in cellular detoxification from xenobiotic substances like herbicides, secondary metabolites and toxic degradation products resulting from oxidative stress and cellular metabolism. Furthermore, environmental conditions generate oxidative stress, the products of which have to be detoxified by plants. It is anticipated that environmental stresses will worsen over the following years due to climate change. Hence, plants must adapt rapidly to the new environmental conditions in order to both survive and satisfy the constantly increasing human demand for agricultural products. Genetic engineering has been successfully used to develop plants resistant to stresses and, having taken all the necessary precautions, could offer a solution as it can help to develop plants with desirable traits in a short period of time. We present here the use of GST isoenzymes in the development of transgenic plants. Although transgenic plants over-expressing various GSTs have been used for “in planta” evaluation of the enzymes used in response to different stresses, the results show that GSTs could be of great value for generating stress tolerant plants. However, the literature is limited and more studies should be performed in order to exploit their full potential

    Life Cycle Assessment of a Steel-Framed Residential Building

    One of the most widely acknowledged policies, which is also strongly promoted by legislation and government officials globally, is sustainable development. Since the introduction of the term and the development of its content, the movement for sustainable development has been accepted by all business sectors as a set of principles that have to be incorporated into standard practice. Particularly in the case of business sectors such as construction that have been identified as the largest consumers of raw materials and energy there has been considerable pressure to optimize processes in terms of sustainability, with particular emphasis on the environmental impact caused. Steel structures constitute a construction technology which holds significant potential in terms of sustainability. The purpose of the current research is to quantify this potential by calculating the environmental impact caused throughout the life cycle of a steel-framed residential building. A life cycle assessment is conducted, taking into account issues such as raw material acquisition, construction and waste management. The results obtained are used to draw conclusions regarding the application of the life cycle assessment methodology to steel buildings and the environmental data required. Furthermore, observations regarding the quantification of the environmental impact caused by the steel-framed residential building and the identification of the most environmentally damaging processes in regard to the life cycle of the building are also made

    Ultrasonic energy input influence on the production of sub-micron o/w emulsions containing whey protein and common stabilizers

    Ultrasonication may be a cost-effective emulsion formation technique, but its impact on emulsion final structure and droplet size needs to be further investigated. Olive oil emulsions (20 wt%) were formulated (pH 7) using whey protein (3 wt%), three kinds of hydrocolloids (0.1–0.5 wt%) and two different emulsification energy inputs (single- and two-stage, methods A and B, respectively). Formula and energy input effects on emulsion performance are discussed. Emulsions stability was evaluated over a 10-day storage period at 5 C recording the turbidity profiles of the emulsions. Optical micrographs, droplet size and viscosity values were also obtained. A differential scanning calorimetric (DSC) multiple cool–heat cyclic method (40 to 40 C) was performed to examine stability via crystallization phenomena of the dispersed phase. Ultrasonication energy input duplication from 11 kJ to 25 kJ (method B) resulted in stable emulsions production (reduction of back scattering values, dBS 1% after 10 days of storage) at 0.5 wt% concentration of any of the stabilizers used. At lower gum amount samples became unstable due to depletion flocculation phenomena, regardless of emulsification energy input used. High energy input during ultrasonic emulsification also resulted in sub-micron oil-droplets emulsions (D50 = 0.615 lm compared to D50 = 1.3 lm using method A) with narrower particle size distribution and in viscosity reduction. DSC experiments revealed no presence of bulk oil formation, suggesting stability for XG 0.5 wt% emulsions prepared by both methods. Reduced enthalpy values found when method B was applied suggesting structural modifications produced by extensive ultrasonication. Change of ultrasonication conditions results in significant changes of oil droplet size and stability of the produced emulsions

    The aim of the present study is to document undergraduate students’ understanding of aspects and properties of the notion of periodicity when confronted with various tasks mostly taken from their secondary school textbooks. In order to accomplish the above goals we designed and contacted three separate research activities (case studies). This was considered necessary in order to study different aspects of periodicity within a broad range of participants, as well as to integrate this kind of research activities in researcher’s teaching experiences as in the case of Act 1

    3D Facial landmark detection under large yaw and expression variations

    A 3D landmark detection method for 3D facial scans is presented and thoroughly evaluated. The main contribution of the presented method is the automatic and pose-invariant detection of landmarks on 3D facial scans under large yaw variations (that often result in missing facial data), and its robustness against large facial expressions. Three-dimensional information is exploited by using 3D local shape descriptors to extract candidate landmark points. The shape descriptors include the shape index, a continuous map of principal curvature values of a 3D object’s surface, and spin images, local descriptors of the object’s 3D point distribution. The candidate landmarks are identified and labeled by matching them with a Facial Landmark Model (FLM) of facial anatomical landmarks. The presented method is extensively evaluated against a variety of 3D facial databases and achieves state-of-the-art accuracy (4.5-6.3 mm mean landmark localization error), considerably outperforming previous methods, even when tested with the most challenging data
