    The current state of the population of the golden potato cyst nematode Globodera rostochiensis (Nematoda, Heteroderidae) in the northwest of Ukraine

    The golden potato cyst nematode (GPCN) Globodera rostochiensis (Wollenweber, 1923) Skarbilovich, 1959 is a highly specific parasite of the roots of the nightshade plants (Solanaceae). Thus, the state of the pest population demands constant monitoring and control of distribution and numbers. The distribution of G. rostochiensis in Volyn region of Ukraine was studied in 2008–2018 using the data of the state institution the Volyn Regional Phytosanitary Laboratory. The present article gives the analysis of the study results. The disease foci were detected by visual above-ground inspection of potato plantings, also by manually collecting soil samples before planting the potatoes and after harvesting, and consequently analyzing theme. The initial (pre-planting) and conclusive (after harvest) population density of GPCN in the soil was determined by the number of cysts and the mean number of larvae and eggs in cysts obtained from 100 cm3 of soil. Cysts were isolated from soil samples by the standard funnel flotation method. The dead and living larvae were identified visually by the shape of the body and the state of internal organs. According to the results of assessing pre-planting and post-harvest nematode numbers in soil, the reproduction coefficient Рf/Рі was calculated. In Volyn region, G. rostochiensis was first observed in 1968 on farmland and since then, the parasite has spread. Pest foci were recorded in 15 districts of the region in 303 settlements on the area of 946.123 hectares. It was however found that during the latest decade, the area of soils affected by G. rostochiensis in Volyn region decreased by 147.647 hectares. The largest infected areas (over 100 ha) were located in Kovel, Rozhyshche and Manevistky districts, the least infected area was observed in Ivanychi district of the region. The pest was not found in Lutsk district. The highest infection rate was recorded in Rozhyshche district. The highest ratio of viable cysts was observed in the soils of Volodymyr-Volynsky and Rozhyshche districts. The soils of the southern districts (located in the natural zone of forests and steppe) of the region demonstrated 1.5 times higher infection rates compared to soils of the northern districts (in the natural zone of mixed forests). The soils of the southern districts also harboured stable and strong pest populations. The pre-planting soil infection rates proved to directly depend on the reproduction coefficient of GPCN. If the values of Рі, initial infection rate, were lower than 1,000 larvae and eggs per 100 cm3, the reproduction coefficient was 1.18. Increase in the pre-planting infection rate to 2,000 eggs and larvae per 100 cm3 did not affect the reproduction coefficient. At approximately 5,000 eggs and larvae per 100 cm3 the reproduction coefficient exceeded 2, which should be considered in developing the pest control measures

    The impact of temporal patterns of temperature and precipitation on silver Prussian carp (Carassius gibelio) spawning events

    It was revealed that Gibel carp, Carassius gibelio (Bloch, 1782) spawning phenology is determined by regular patterns of variability in temperature and precipitation over the period of time after spawning in the previous year. The sensitivity to temperature of water as a stimulus to spawning was shown as being dependent on the characteristics of the environment and fish development trajectories over the previous year. Information about fish spawning was collected in the Dnipro-Orylskiy Nature Reserve in the following locations: Nikolayev system of water bodies, River Protoch system and the Obukhov floodplain, the channel of the River Dnipro, water bodies of the Taromske ledge. The materials that formed the basis of this research were collected from the waters of the reserve in the years 1997–2018. There are dependencies between the air and water temperatures that have their own characteristics depending on the type of water body. These dependencies are described by a logistic equation. The temperature regime in water bodies of the Taromske ledge is very similar to the temperature regime of Nikolayev system of water bodies. The difference is that the spring period of the temperature increase in water bodies of the Taromske ledge comes earlier – in late February – early March. The air temperature dynamics over the year is described by the fourth degree polynomial. The residuals of the polynomial regression for each year were subjected to principal component analysis. The principal component analysis allowed us to identify 8 principal components of the temperature regimes, which together describe 59.2% of the variation of the residual of the polynomial trend. Deviations from the linear trend of the precipitation are systematic throughout the year, reflecting the uneven nature of rainfall during the year. The residuals of linear trend regression models of cumulative precipitation can be used as indicators of the dynamics characteristic of precipitation during the year. The residuals were used for principal component analysis, which revealed that the dynamics of the precipitation can be divided into five orthogonal principal components. The first five principal components explain 82.6% of the feature space variation. The timing of the onset of C. gibelio spawning in 95% of cases occurs on the 113–139th days of the year (on average, it takes place on the 125th day). Regression analysis revealed that the climate principal components and types of the lake systems can explain 79% of the variation in the timing of the onset of C. gibelio spawning. The water temperature at the time of C. gibelio spawning was exposed to natural dynamics in year aspect. The local minimum of the onset of spawning temperatures occured in the mid 2000s. Total of 69% of the water temperature variation at which spawning begins can be described by climatic predictors and water biotope types. The variation explained by some temperature principal components may be increased considerably after including as covariates such variables as the precipitation principal component and the biotope type. This principal component which represents the high frequency variation of the air temperature regime with a characteristic period 20 and 45 days has the highest explanation ability of all the components and is a constant explanatory predictor for all considered spawning characteristics

    Morphological and essential oil variations among Iranian populations of Salvia chloroleuca (Lamiaceae)

    Salvia chloroleuca Rech. f. & Aell. is one of the perennial species of the Lamiaceae family in Iran, which has antimicrobial and antitumoural properties. In the current research, the morphological variables and hydrodistilled essential oils of two populations, from Polor and Neyshabur, of this species were evaluated. We studied 10 and 40 individuals for morphometrics and essential oils, respectively. Morphological data were analyzed using SPSS. Furthermore, the extracted oils were analysed using GC and GC/MS. The qualitative morphological characteristics were stable between the populations, while the quantitative ones highly varied. The one-sample variance test revealed significant differences for most of the studied features. The yielded oil highly differed between these populations. The first and second main groups of compounds were the same for both populations, but with different percentages. However, the third and fourth main groups of compounds were not similar. Furthermore, the main compounds of essential oil differed between the studied populations. The major compounds of the Polor population were: β-pinene (22.7%), α-pinene (18.3%), germacrene D (7.7%) and sabinene (6.6%). However, spathulenol (19.8%), bicyclogermacrene (11.4%), p-cymene (10.8%) and β-pinene (10.4%) constituted the major compounds of the Nyshabur population. In total, the Neyshabur population had higher amounts of sesquiterpene and oxygenated compounds than the other population. The results indicated that in this species the quantitative morphological characters and essential oils were strongly affected by environmental factors

    The influence of Cameraria ohridella (Lepidoptera, Gracillariidae) on the activity of the enzymatic antioxidant system of protection of the assimilating organs of Aesculus hippocastanum in an urbogenic environment

    In the last two decades, the horse chestnut (Aesculus hippocastanum L.), introduced into the steppe zone of Ukraine, has been severely affected by the horse chestnut leaf miner Camereraria ohridella Deschka & Dimič, 1986, which results in damage to the assimilating organs, premature leaf defoliation and, as a consequence, a significant reduction in the reserve substances required for normal life of the plant. In recent studies, the main focus has been placed on the study of the pest’s effects on the non-enzymatic antioxidant protection system of the representatives of the genus Aesculus, while the enzymatic system of horse chestnut protection from the active forms of oxygen under stress is still poorly understood. The purpose of this study was to evaluate the reaction of catalase and two peroxidases of A. hippocastanum leaves, which differ in the level of damage by C. ohridella. The intensity of damage to A. hippocastanum leaves by the horse chestnut leaf miner in the park zones and botanical gardens of Dnipro city was determined, the activity and isoenzyme composition of benzidine-peroxidase, activity of guaiacol-peroxidase and catalase were measured. The lowest average benzidine-peroxidase activity was found in the group of trees with low level of leaf blight and the highest activity – in the group with high level. The opposite dependence was shown by catalase, the activity of which significantly decreases with increasing level of damage inflicted by the phytophage on the chestnut’s assimilating organs. Based on the determination of the variation coefficients, it has been shown that benzidine-peroxidase activity has a higher level of variability than that of catalase and guaiacol-peroxidase. It is established that under the influence of the leaf miner, activity of guaiacol-peroxidase was significantly higher by 87.1% and 75.6%, respectively, for medium and high levels of damage caused to the leaf by this phytophage as compared to that for low levels of damage. The increased level of leaf damage caused by the phytophage is reflected in the change in the isozyme profile of benzidine-peroxidase. The high activity of benzidine-peroxidase in the leaves of A. hippocastanum is due to the presence of several molecular forms that exhibit maximum activity in the narrow pH range (4.15–4.69). Quantitative redistribution of activity between the different molecular forms of benzidine peroxidase can be considered as the main regularity of changes in the expression of benzidine-peroxidase caused by different levels of leaf damage. The results showed that only one benzidine-peroxidase isoform with an isoelectric point of 4.15 shows a significant increase in activity (on average by 2.1 times) in A. hippocastanum leaves with medium and high levels of damage by C. ohridella. Significant reduction in activity is reported for dominant isoperoxidase with an isoelectric point of 4.25 revealing medium pest damage, and for high damage only a decreasing tendency is shown. The data obtained show that horse chestnut trees can specifically respond to mechanical damage by C. ohridella to leaves due to the changes in the activity of individual molecular forms of peroxidase. Further studies of oxidative metabolism are needed to understand the formation of resistance of representatives of the Aesculus genus to damage caused by this moth species based on a wider range of redox enzymes

    Population density and structure of birds breeding in an urban habitat dominated by large baobabs (Adansonia digitata), Northern Namibia

    Large baobabs are regarded as key plant species in the savannah biome. In this study their role in shaping the avian community has been evaluated. The territory mapping method has been employed to quantify the avian breeding community in the town Outapi (ca. 130 ha), Northern Namibia, well-endowed with large baobabs. A total of 29 breeding bird species were recorded. The alien House Sparrow was by far the most numerous species comprising 48.4% of all breeding birds. The Blue Waxbill and African Palm Swift were also classified as dominant species, comprising together 17.4%. Granivores were by far the most numerous feeding guild, comprising 77.2% of all birds breeding, while the insectivores comprised only 11.8%. Birds nesting in/on buildings comprised 52.7%, those nesting on trees/shrubs – 42.0%. The population densities of many bird species (e.g. doves from the genus Stepropelia, bulbuls, weavers) were comparatively low. Such a situation could have been caused by the exceptionally high population density of the Pied Crow. In the study area, there were 15 breeding pairs and about 100–150 non-breeding individuals. The baobabs may provide them with a feeding resource in the form of the sweet fruits. The crows, as nest predators, feed also on the eggs and nestlings of other birds. Doves seem to be especially prone to such predation, as their nests are easily to detect and destroy. As a result, they have to breed in low density. This in turn, may release other dominant granivores in the study area, namely sparrows. They are not vulnerable to crow nest predation, as their nests are usually well-concealed under eaves and in holes of buildings

    Parasitofauna of pike Esox lucius of the Lower Tobol (Russia)

    One of the predators of the ichthyofauna of the Lower Tobol is Esox lucius (Linnaeus, 1758) (Esociformes, Esocidae). The purpose of this work is to study the current state of the E. lucius parasite fauna in the lower reaches of the Tobol River. In the present study 47 individuals of the northern pike of different sex aged from 2+ to 8+ years were examined by the method of complete parasitological dissection. As a result of the research, 23 types of parasites were found in the pike. The largest number of species of parasitic organisms – 20 – were found in May, 12 were found in December and 11 in September. Of these, 10 species are specific to northern pike: Haemogregarina esoci (Nawrotzky, 1914), Chloromyxum esocinum (Dogiel, 1934), Myxidium lieberkuhni (Biitschli, 1882), Myxosoma anurum (Cohn, 1895), Henneguya psorospermica (Thelohan, 1995), Tetraonchus monenteron (Wagener, 1857), Gyrodactylus lucii (Kulakowskaja, 1951), Triaenophorus nodulosus (Pallas, 1781), Azygia lucii (Miiller, 1776), Raphidascaris acus (Bloch, 1779). For the first time an intraerythrocyte parasite – H. esoci – was found in pike in the Lower Tobol, the extensiveness of invasion was 18.7% in December, and 6.2% in May. Epistylis sp. and Trichodinella epizzotica parasitized on the surface of the body of fish only in May, whereas members of the genus Trichodina were found in winter and spring. In all seasons, parasitization by M. anurum and H. psorospermica was established on the gills. The greatest occurrence of spores of M. lieberkuhni and C. esocinum was observed in the spring. Myxosporidia parasitized in the kidneys and T. monenteron parasitized on the gills during all periods of the study (the latter with an extensivity of invasion of 100.0%). All examined fish are infected with T. nodulosus 100.0% of cases. In May, other types of cestodes were found: P. esocis and Dibothriocephalus latus. Three pike were infected with proteocephalis. The larval stage of diphyllobothriid was found in one fish in the liver and gonads in an amount of 139 individuals. In the pikes’ stomach, during all periods of the study, A. lucii was found in all the fish examined. In the autumn period of the study immature R. campanula were found in in the intestine of 4 fish specimens. The nematode R. acus was found in pike only in spring. This nematode was found in fish aged 4+–8+. In May, glochidia with a high intensity of invasion were found on fins, gill covers and gills of fish. E. sieboldi copepods were found on the gills; in May, one specimen was found in one pike and in September in 6 with AI of 0.7. Tetraonchus monenteron was a dominant species, in May its degree of dominance decreased with the greatest uniformity of species in this study period. Having considered the age dynamics of pike infection by various types of parasites, it was found that in the age group 4+–5+ the number of species of parasites was greatest (20), while in groups 2+–3+ and 6+–8+ it was 14. In the pike, the core parasitic fauna were M. anurum, H. psorospermica, T. monenteron, T. nodulosus, A. lucii. Seasonality has virtually no effect on the degree of infection with specific parasites. The age of fish largely determines the qualitative and quantitative composition of the parasite fauna

    Nematocidial activity of aqueous solutions of plants of the families Cupressaceae, Rosaceae, Asteraceae, Fabaceae, Cannabaceae and Apiaceae

    In natural ecosystems of animals, introduction of larvae of parasitic nematodes into the litter layer from excrement facilitates their migration and search of new host vertebrate animals. In such conditions they are constantly affected by biologically active substances of the abundant species of plants which grow in pastures. Currently, the influence of substances present in the above-ground part of plants on the vitality of larvae of helminths in the environment remains unstudied. In this article, we present the results of our research on the nematocidial activity in vitro in the aqueous solutions of 21 species of plants distributed in the territory of Steppe Ukraine: Sanguisorba officinalis L., Rosa canina L., Crataegus sanguinea Pall., Crataegus pentagyna Waldst. & Kit. ex Willd., Armeniaca vulgaris Lam., Taraxacum officinale F. H. Wigg., Iva xanthiifolia Nutt., Artemisia campestris L., Arctium minus (Hill) Bernh., Ambrosia artemisiifolia L., Cannabis sativa L., Humulus lupulus L., Melilotus officinalis (L.) Pall., Vicia cracca L., Lotus ucrainicus Klok., Onobrychis arenaria (Kit.) DC., Foeniculum vulgare Mill., Eryngium planum L., Conium maculatum L., Juniperus communis L., Thuja occidentalis L. The level of vitality of nematode larvae of the Strongylida (Haemonchus contortus (Rudolphi, 1803)) and Rhabditida (Strongyloides papillosus (Wedl, 1856)) orders varied depending on the species of plant, and also five experimental concentrations in aqueous solutions tested in seven replications. The most notable nematocidial effect was exerted by Taraxacum officinale F. H. Wigg. – we observed death of larvae of third stage development L3 H. contortus and larvae of first-third stages L1–3 S. papillosus at 24 h exposure to 3% aqueous solution. Three percent aqueous solutions of S. officinalis and A. artemisiifolia displayed nematocidial properties only against S. papillosus: death of L1–3 S. papillosus was observed. Aqueous solutions of R. canina, A. vulgaris, A. minus, H. lupulus, V. cracca, L. ucrainicus, O. arenaria, E. planum, C. maculatum, J. communis, Th. occidentalis had lethal effect only on non-invasive larvae (larvae of the first and second stage L1–2) of S. papillosus. They displayed no nematocidial properties towards invasive larvae of H. contortus and S. papillosus. At exposure to aqueous solutions of the rest of the studied species of plants, over 50% of L3 H. contortus and L1–3 S. papillosus larvae remained alive. The determined patterns allow us to state that while living in the litter and soil in the root zone of plants nematode larvae undergoa negative influence caused by some plant species

    Bactericidal, protistocidal, nematodicidal properties and chemical composition of ethanol extract of Punica granatum peel

    We have studied the chemical composition and antibacterial profile of ethanolic extract of Punica granatum L. (Lythraceae) on strains of microorganisms in vitro. Analysis using GC-MS showed 5-hydroxymethylfurfural (36.6%), D-sucrose (23.2%), sorbitol (6.7%), palmitic acid β-monoglyceride (5.6%), 2-furancarboxaldehyde (3.5%) and β-D-glucopyranose (3.3%) as the major components of the title extract. The experiment revealed a positive antibacterial effect of extracts obtained from P. granatum on 14 strains specifically Enterobacteriaceae microorganisms (Escherichia coli, Enterobacter aegorenеs, Proteus vulgaris, Serratia marcescens, Klebsiella pneumonia), Listeriaceae (Listeria ivanovi, L. іnnocua, L. monocytogenes) and yeasts from the family Saccharomycetaceae (Candida albicans). Our study showed that in many cases these extracts more intensively affect multi-resistant strains of microorganisms than macrolide antibiotic azithromycin and is therefore a source of molecules to be exploited in medicine or by the pharmaceutical industry. The investigated extracts of P. granatum can be recommended for further in-depth research against poly-resistant strains of the above-mentioned microorganisms. Effective drugs perform a leading role in providing stable veterinary well-being of livestock and healthcare of the population. The present study showed that the studied plant species more intensively affects multi-resistant strains of microorganisms than sodium salt of azithromycin. Lethal concentration (LC50) of ethanol extract from pomegranate for Paramecium caudatum Ehr. equaled 0.3%. Death of 100% of nematode larvae of Strongyloides papillosus (Ihle) was recorded during 24 h exposition in 20% extract of P. granatum peel

    Antimicrobial activity of 50 plant extracts

    Antibacterial activity of plants is a subject of interest in the search for new antibiotics and fungicidal preparations. This article analyzes the effectiveness of the action of extracts of plants on microorganisms: six species of bacteria (Salmonella typhimurium, Listeria monocytogenes, Escherichia coli, Klebsiella pneumoniae, Corynebacterium xerosis, Proteus vulgaris) and one fungus (Candida albicans). For the assay, we used herbal material of 50 species (seeds, grass, shoots, leaves, compound fruit, peel) obtained at different periods of the growing season. We studied Levisticum officinale, Petroselinum crispum, Nerium oleander, Vinca minor, Eleutherococcus senticosus, E. sieboldianus, Yucca filamentosa, Artemisia annua, Echinacea purpurea, Matricaria recutita, Tanacetum vulgare, Betula pendula, Corylus avellana, Buxus sempervirens, Humulus lupulus, Crassula ovata, Bryophyllum daigremontianum, Juniperus communis, Platycladus orientalis, Cycas revoluta, Calluna vulgaris, Rhododendron ferrugineum, Ceratonia siliqua, Trigonella foenum-graecum, Ribes nigrum, Phellinus tuberculosus, Lavandula angustifolia, Melissa officinalis, Monarda fistulosa, Origanum vulgare, Salvia sclarea, Laurus nobilis, Punica granatum, Hibiscus rosa-sinensis, Menispermum dauricum, Ficus benjamina, Morus alba, Paeonia suffruticosa, Picea abies, Adonis vernalis, Amelanchier ovalis, Prunus armeniaca, Crataegus monogyna, Citrus sinensis, Salix babylonica, Bergenia crassifolia, Schisandra chinensis, Taxus baccata and Xanthoria parietina. The alcohol tincture was filtered with sterile multi-layer gauze disc filters. Before the discs were put on the surface of agar with inoculation of the corresponding culture, they were dried in a sterile laminar box under ultraviolet rays. Antibacterial activity of various tinctures was determined by the disk diffusion method in agar with the measurement of the diameter of the growth suppression zone of the culture using a template ruler. Maximum inhibiting effect was achieved for Punica granatum on K. рneumonia, L. monocytogenes, S. typhimurium, P. vulgaris, C. хerosis and E. сoli, Lavandula angustifolia – on P. vulgaris, K. рneumonia and S. typhimurium, Echinacea purpurea – on C. аlbicans, E. сoli, P. vulgaris, K. рneumonia, Bergenia crassifolia – on P. vulgaris, K. pneumonia and S. typhimurium

    Investigation of vegetation as a precondition for improving the management of a national nature park (on the example of Dzharylhatskyi NP)

    The protected area of Dzharylhatskyi National Park is only 3% of the total area. This is not enough to preserve dynamic ecosystems that are very vulnerable to natural factors and almost impossible to restore following damage by anthropogenic factors. In the investigated area there are territories with high potential for conservation, which are characterized by the presence of species from the Red Data Book of Ukraine and International Red Lists, plant communities from the Green Data Book of Ukraine and biotopes of global significance. The proposed ten sites should receive protected status, which will bring the protected area up to the necessary minimum to preserve the rare ecosystems of the park from tourism and economic pressure, which have intensified in recent years. For the period of research in 2014–2018, 14 types of sozophytes were identified and confirmed, two of them are the highly localised species endemic to Dzharylhach Island: Molinia euxina Pobed. and Poacynum russanovii (Pobed.) Mavrodiev, A. Laktionov et Y. Alexeev. The syntaxonomic structure of the sozologically valuable coenoses is represented by two basal communities, two subassociations and 8 associations belonging to 9 alliances, 9 orders and 9 classes. Out of them, we provisionally propose the new following groups: ass. Apero maritimi-Chrysopogonetum grylli nom. prov., subass. Apero maritimi-Chrysopogonetum grylli, Stipetum borysthenicae nom. prov., subass. Cladietum marisci, Caricetum extensae nom. prov., BC Molinia euxina [Molinion caeruleae] nom. prov. The proposed sites represent 8 biotopes from Annex I of the Habitat Directive, which imposes obligations for their conservation at the world level. We carried out an analysis of the Ukrainian legislative acts, their correlation with international requirements and the zoning of the NPP “Dzharylhatskyi” in relation to these requirements and recommendations. Thus, the urgent need to expand the boundaries of the protected area of the Dzharylhatskyi National Nature Park by including distinguished protected tracts has been confirmed
