The molecular mechanism of salivary dysfunction amelioration by calorie restriction
研究成果の概要(和文):マウスに35%のCR(カロリー制限食)を与えると唾液分泌量は増加し、若齢マウスを上回った。唾液腺の炎症の程度について検討を行うと、自由摂取群と比較してCRマウスでは、浸潤細胞は減少し、組織内で産生される炎症性サイトカインも低値を示し、アミノ酸代謝関連遺伝子にも変動が認められ、さらにCR群ではケトン体が経時的に上昇した。カロリー制限による様々な作用はヒストン脱アセチル化酵素のsirtuinが関与しており、ポリフェノール類のレスベラトロールがこれを活性化することが報告されていることから、レスベラトロールのNODマウスの唾液分泌亢進作用を有する事を確認したことから臨床応用の基盤となる成果が得られた。研究成果の概要(英文):Calorie restriction (CR) is known to decrease the risk of disease onset resulting in lifespan elongation. Xerostomia is accompanied with aging, however, the pathogenesis is unknown yet and effective treatment has not been established. We studied the effect of CR on xerostomia accompanied with aging.Method Twelve-month-old male C57BL/6 mice were divided into ad libitum-fed (AL) group and CR group. The amount of food intake in CR group was 35% less than in AL group. The saliva secretion was measured after three and six months of CR. At the end of experiment, salivary glands were extracted following histological and gene expression analysis. Results The amount of saliva secretion decreased with aging in AL group, whereas increased saliva secretion was seen from three months after CR. Elucidation of the microRNA-mediated control of the alveolar bone metabolism by PDL cells under each mechanical stresses
研究成果の概要(和文):矯正学的歯の移動でなぜ圧迫・伸展刺激で骨吸収・骨形成と異なる骨代謝反応が惹起されるかについて明らかとすべく、圧迫・伸展刺激を負荷された歯根膜線維芽細胞セルラインが発現するマイクロRNA (miRNA)について解析を行った。圧迫・伸展刺激では異なるmiRNA発現が誘導された。骨代謝制御に重要な因子であるOPGに着目し、歯根膜線維芽細胞における圧迫・伸展刺激での発現変動と、miRNA発現の関連について解析を行った。その中で、OPGを標的とするmiR-3198がOPG発現変動と逆の変動をし、圧迫・伸展刺激負荷による歯根膜線維芽細胞のOPG発現変動に重要な役割を果たしていることを明らかとした。研究成果の概要(英文):We aimed to elucidate the regulatory mechanism of the induction of the different bone metabolisms, bone resorption and bone formation, at the compression and tension side during orthodontic tooth movement. We focused on the microRNA (miRNA), and analysed the expression of miRNA in PDL cells under compression and tension. We discovered that different miRNAs were upregulated in PDL cells under compression and tension. Then we examined the relationship between the change of OPG expression by different mechanical stresses and the difference of miRNA induction. miRNAs which can target OPG were firstly selected, and we found that miR-3198 was the miRNA which exhibited different change of the expression between compression and tension. miR-3198 and OPG inversely changed by compression and tension. Elucidation of the role of oral-derived endotoxin as a common risk factor for periodontal disease and dementia
研究成果の概要(和文):ダウン症(DS)者30名と健常者38名の口腔衛生状態の評価、細菌学的検査、免疫学的検査を行い、DS者のアルツハイマー型認知症(AD)発症リスクとの関連を検討した。その結果、血中アミロイドβ(Aβ42)濃度、Candida albicansの分離頻度、唾液中IL-6、IL-8濃度、PCR、OHI、GI、GBIが健常者(対照群)に比べてDS群で高かった。歯周病菌には両群で有意差が見られなかった。DS者では、不十分な口腔清掃が原因と考えられる口腔内の炎症を示す臨床所見がみられた。DS者は、血中アミロイドβ(Aβ42)濃度が高いので、口腔の慢性炎症は健常者以上にAD発症のリスクになると考えられる。研究成果の概要(英文):Amyloid-β(Aβ42) concentration in the blood, oral hygiene, periodontopathic bacteria and immunological condition of 30 patients with Down's syndrome (DS) and 38 healthy subjects were examined. Based on the results, we estimated the association of DS with the risk of developing Alzheimer type dementia (AD). Aβ42 concentration in blood, salivary IL-6, IL-8 concentration, salivary Candida albicans isolation frequency, PCR, OHI, GI, GBI were higher in the DS group than in the control healthy group. There was no significant difference in periodontopathic bacteria between two groups. Clinical findings showing intraoral inflammation, which is thought to be caused by insufficient oral hygiene, were observed in DS patients. A Study of the Heian Period Kana Literature Based on philology
研究成果の概要(和文):平安時代の仮名文学作品は、今日様々に活字化され、読まれてきている。しかし活字化に際しての、底本選定に関する従来説には、不適切なものが多かった。結果、善本ではない古典籍が底本に採用されたり、同じ古典籍が何回も底本とされ続けたり、重要他伝本が忘れ去られてしまったり、という弊害が生じていた。本研究は、それら従来説を批判し、調査研究し直して、平安時代仮名文学の伝本や本文に関する新たな知見の提示を試みたものである。結果、古今集・発心和歌集・二十巻本類聚歌合・伊勢物語・源氏物語・栄花物語・枕草子ほかの伝本や本文に関し、重要資料を発掘・再発掘し、評価・再評価して、学術論文等に結実させていくことができた。研究成果の概要(英文):In the Heian period of Japan, literature written in pseudonyms was written. They are currently being read in print. However, when being reprinted, there was a mistake in previous studies on which classic nationality is to be based. As a result, materials of poor quality were adopted, the same material was used many times, and important material was forgotten. Therefore, in this research, we criticize the conventional theory on those materials and texts. It also tried to present survey and research again and to present. As a result, we were able to excavate and re-evaluate many important materials. Development of the virtual articulator combining dentition data and jaw movement data
研究成果の概要(和文):歯列データと下顎運動データを統合するバーチャル咬合器の開発を行い,バーチャル咬合器で表現される機能運動路(デジタルFGP)を製作した.また,従来法(FGP,FBI)と比較することで臨床応用の可能性について検討した.その結果,デジタルFGPは従来法に比較して機能運動時の情報が含まれ,より機能に即した補綴装置の製作が可能であることが示唆された.研究成果の概要(英文):We developed the virtual articulator which combined dentition data and jaw movement data. And compared the the functional guided path (digital FGP) of the virtual articulator with the traditional approach (FGP, FBI) and examined the possibility of the clinical application. As a result, digital FGP includes a lot of information of the functional movement than a traditional approach.
