The Development of Prototype Polyclonal Antibody of Receptor Advanced Glycation of Endproducts (RAGE)
Abstract
Background
Receptor for advanced glycation endproducts (RAGE) is a transmembrane protein that belongs to the immunoglobulin superfamily. As its name implicates, it can bind to advanced glycation endproducts, the resulting product of non-enzymatic glycosylation, and it also has the ability to interact with multiple ligands having common motifs as a so-called multi-ligand receptor. The ligands include high-mobility group protein (B)1 (HMGB1), S-100 calcium-binding protein, amyloid-β-protein, Mac-1, and phosphatidylserine. Interaction between RAGE and its ligands activates various cellular processes, including inflammation, proliferation, apoptosis, autophagy, and migration.
Methods
The proccess of isolation protein of RAGE was initiated with extraction protein and purification of RAGE protein. After that, the immunization of Rats was be done to produce Anti-RAGE. The confirmation of Anti RAGE was be done by SDS PAGE and Immunobloting.
Results
The production of Anti RAGE was enough pure compared by Anti RAGE commercial. Anti RAGE was protein that have molecular weight around 35 kD.
Conclusion
The methods that used in this study effective to develop production of anti-RAGE.

The Risk Factors of Anorectal Malformation Patients Mortality in Palembang
Anorectal malformations are serious anal and rectal disorders ranging from stenosis to anorectal agenesis. The incidence of mortality in cases of anorectal malformations is influenced by many factors, including the type of anorectal malformation, associated congenital anomalies, sepsis, and prematurity as investigated in this study. The aim of this study was to identify the risk factor of mortality on anorectal malformation patients. This study is an observational analytic study with cross sectional study design. Secondary data collection was obtained from eighty-two medical records of anorectal malformation patients in Dr. Mohammad Hoesin Hospital Palembang from January 2015 to June 2017 that meets the inclusion criteria. From eighty-two cases of anorectal malformations found nineteen cases of mortality (23.2%). There were four risk factors identified, anorectal malformation type, associated congenital anomalies, sepsis, and prematurity. Two risk factors found significantly affect the mortality on anorectal malformation patients, anorectal malformation type (p value = 0.036) and sepsis (p value = 0.049) from four risk factors studied. Type of anorectal malformation and sepsis have a significant effect on the mortality of patients with anorectal malformations, but there is no effect on the mortality of the patients with assciated congenital anomalies and prematurity.

The Recommended Aerobic Gymnastics Has Better Effects on Improving Cognitive and Motoric Ability in Children
Abstract
Background Cognitive and motoric ability has important role in children’s development, whereas exercise has benefit effects on those abilities. Knowledge materials mostly have dominant role in primary school curriculum. Physical exercise lesson is only complementary lesson without a target to improve children’s cognitive ability. We conducted a study to give evidence that 3x/week structured exercise program has better effect compared to 1 x/week exercise on cognitive and motoric skill ability in children. Materials and methods Children aged 6-8 years old were divided into treatment (n=34) and control group (n=33). Treatment group were performing fun aerobic gymnastics guided by trained instructor, 45 minutes each, 3 times a week for 8 weeks with intensity target. Control group were also performing the same gymnastic activity for only once a week with no target. Cognitive and motoric ability were assessed before and after intervention. Results A recommended regular exercise has better effect on executive function, reaction speed, coordination, flexibility and agility, compared to control. There were no differences on memory and balance ability. Both groups have shown better result for all indicators after exercise. Exercise in both groups has positive effect on cognitive and motoric ability, but a recommended 3 x/week regular exercise has better effects compared to 1 x/week exercise. Conclusions School program should give more portions for exercise activity in their curriculum.

The Relationship Between Soil Transmitted Helminthes (STH) Infection and Nutritional Status in Students of State Elementary School Number (SDN) 200 Palembang Indonesia
Infection of Soil Transmitted Helminthes (STH) is caused by intestinal nematodes where as in its life cycle, soil is needed as the media for the eggs or larvae to mature into effective forms, most commonly happen to children of school age. This infection is still one of the main problems in public health, including Indonesia. STH infection are widely distributed in tropical and subtropical areas. Lack of personal hygiene, poor environmental sanitation and low socioeconomic status are some factors that plays role in increasing the occurrence of the infection. This infection is also one of the causes responsible for malnutrition in children by decreasing appetite and food intake thus ensued adverse consequences such as declining growth pace, impairment of physical health, and weakening cognitive function. This study was conducted to analyze the association of STH infection with nutritional status of SDN 200 students in Kertapati District, Palembang. This study was an analytic observational research with a cross sectional research design. Samples consist of 107 students chosen using proportional stratified random sampling technique. Data was collected by direct interview using questionnaires, measuring body weight and height to obtain nutritional status which then classified using CDC 2000 growth curve while fecal contamination was examined using Kato Katz and modified Harada Mori methode in the Laboratory of Parasitology Medical Faculty of Universitas Sriwijaya. Data then analyzed using Chi-square test. From 107 students, 27.1% infection of STH was found on 29 students with 6 students (20.7%) infected by A. lumbricoides and 23 students (79.3%) infected by T. trichiura. Proportion of malnutrition status was found at 43.9%. Statistical test showed a significant association between STH infection and nutritional status (p=0.036; OR=3.167; Cl 95%: 1.163-15.237). There was a significant association between STH infection and nutritional status in students of SDN 200 Kelurahan Kemas Rindo Kertapati District Palembang City
Identification of IL-10 Gene Polymorphisms in Leprosy
Background: Leprosy is a chronic disease caused by an infection of Mycobacterium leprae, which affects skin and nerve lesions, which can result in disability. The prevalence of leprosy continues to increase, especially in Indonesia, which is one of the endemic areas of leprosy. The unique immune response of each individual not only determines the susceptibility of individuals to leprosy, but also determines the type of leprosy that will manifest. One of the immune responses that plays an important role in the pathogenesis of leprosy is interleukin-10 (IL-10). Increased regulation of IL-10 can have an effect on decreasing macrophage activity in killing bacteria. Production of IL-10 is regulated by genes that encode the cytokine. Gen IL-10 is located on chromosome 1q32. Polymorphism in this gene can cause variations in the function of IL-10 protein, which can then influence the process of microbial elimination in the development of leprosy.Objective: This study aimed to identify IL-10 -819 C / T gene polymorphisms which were cut with the MslI enzyme in lepers.Method: This study is a laboratory study, which is also an observational descriptive study with a cross sectional study approach to 40 subjects with leprosy. Identification of IL-10 -819 C / T gene polymorphism was carried out by PCR amplification and RFLP (restriction fragment length polymorphism) technique using the MslI enzyme.
Results: Distribution of TT, CT, and CC genotypes in the study subjects were 0 (0%), 14 (35%), and 26 (65%). Based on age, the distribution was 0 (0%), 6 (15%), 19 (47.5%) for those under 50 years old, and 0 (0%), 8 (20%), 7 (17.5 %) for those over 50 years old. Based on gender, the distribution is 0 (0%), 5 (12.5%), 16 (40%) in men, and 0 (0%), 9 (22.5%), 10 (25%) in women. Based on the classification of diseases, the distribution was 0 (0%), 3 (7.5%), 7 (17.5%) for papalacillary, and 0 (0%), 11 (27.5%), 19 (47.5 %) for multibasilers.
Conclusion: More wild type genotypes are found in lepers (65%).

Sensitivity and Spesificity of Interleukin 18 as Biomarker for Acute Kidney Injury
Background : Incidence of AKI in intensive care unit patients reach 60-70%, and the mortality rate is about 60%. IL-18 is a pro-inflammatory cytokine which increased in endogenous inflammation process. Studies in human showed that IL-18 concentration increased prior to AKI.
Objective : The aims of this research is to determine the sensitivity and specificity of Interleukin-18 urine examination as an early biomarker for acute kidney injury.
Methods : There’re total of 66 subjects who met the inclusion criteria. All the subject were checked for the IL-18 urine level with Cloud Clone kit and creatinine serum were also checked 48 hours after admission.
Results :The results showed in the cut-off point of 411.25 Pg/mL , urine IL-18 has a sensitivity of 54.3 % and specificity 83.9%, positive predictive value 79.17%, negative predictive value 61.9% and accuracy of 68.18%
Conclusions : Our result suggest that IL-18 urine holds a promise as an early biomarker of AKI and more sensitive and specific as an early biomarker for AKI compared to creatinine serum.

The The Effect of Active Gambir (Uncaria gambir) Fraction on TNF-α Protein Expression and Lesion Size in White Rats Gastritis Model
Background: Gambir (Uncaria gambir) is empirically used to treat abdominal pain and vomittus caused by gastritis because of its anti-inflammatory effects, especially flavonoid. This study aims to determine the effect of active gambir fraction on TNF-α protein expression and wound size in white rats gastritis model.
Methods: The research method used experimental study design in vivo with post test with control group design. Rats were divided randomly in 11 groups and were induced to be gastritis for 1 day. Group 1 (negative control) was given aquadest of 5 mL, group 2 (positive control) was administered ranitidine 10 mg/kgBW, groups 3, 4, and 5 were given n-hexane fraction, groups 6, 7 and 8 were given a water fraction, and groups of 9, 10, and 11 were given ethyl fractions with each group receives dose of 20, 40, and 80 mg/kgBW and all groups were treated for 3 days. Rats were dissected on 5th day for examination of gastric mucosal lesion size and performed ELISA expression of TNF-α expression of gastric mucosal tissue. The results of this study were assayed by SPSS 18.
Results: The result of the research using Kruskal-Wallis test showed that there were significant differences (p <0.05) of the lesions size between the sample groups where control positive, ethyl fraction 20, 40, 80 mg/kgBW, and water fraction 20, 40 mg/kgBB had the gastric mucosal lesion size differed significantly with the negative control group, while the TNF-α protein expression test using Kruskal-Wallis showed that there was a significant difference (p <0.05) TNF-α levels of all groups against the negative control.
Conclusion: Active gambir fraction had a potention to reduce size of mucose gaster lesion and reduce expression of TNF-α protein
The Efficacy of Seluang Fish Oil (Rasbora agrotynea) Related Body Weight, Lipid Profile, Adiponectin and Leptin in Wistar Rats-Induced High Fat Diet
Background
Rasbora agrotynea (local name : Seluang ) is a fish native to Sumatera, Malaya and Borneo. This fish has a potential as a local wisdom for supplementation of omega 3 and omega 6. In Sumatera, Seluang fish was used as traditional food.
Aim of Study
Aim of this study to explore the efficacy of seluang fish oil in body weight, lipid profile, adiponectin and leptin level in Wistar Rats-Induced High Fat Diet
Methods
This study was an experimental study , pre-post test with control group design. The sample in this study was 30 male rats, 8 weeks old, weight 150-200 gram. Rats were given high fat diet and seluang fish oil at dose of 0,1 , 0,2 and 0,4 mL/200 gr BW/day for 2 weeks. Seluang fish oil was extracted by distilation methods. The results of this study were assayed by SPSS 18.
Results
Seluang fish oil 0,4 mL/200 gr BW was more potent to reduce body weight gain, triglyseride level, leptin and increase adiponectin level than seluang fish oil 0,1 mL/200 gr BW, 0,2 mL/200 gr BW, negative control and positive control.
Conclusion
Seluang fish oil had a potention to reduce body weight, triglyceride, leptin and increase adiponectin level.

The Efficacy of Combination Extract Andrographis peniculata and Syzygium polyanthum on Glucose Uptake in Skletal Muscle in Diabetic Rats
Insulin resistance is impaired insulin signaling cascade in target cells to respond normal or elevated circulating insulin to the final cellular effect, such as translocation of vesicles containing GLUT4 glucose transporters, which is the major mediator of glucose removal from the circulation and a key regulator of whole-body glucose homeostasis. Among the several plants, Andrographis peniculata (sambiloto) and Syzygium polyanthum (Daun salam). In the present study, a combination was made, Andrographis peniculata (sambiloto) and Syzygium polyanthum (Daun salam), and to determine the effects of combination on tretament insulin resistance. Male Wistar rats (weight, 200-300 g) were randomized into five groups (6 rats/group). Group 1: negative group. Group 2: positive group (metformin 63 mg/kgBW ). Group 3,4 and 5: treatment with extract combination, each group 250 mg/kgBW, 500 mg/kgBW and 1000 mg/kgBW. Rats were induced by high fat diet-glucocorticoid for insulin resistance. Insulin and GLUT-4 were assayed by ELISA. Treatment with extract combination (250, 500 and 1000 mg/kgBW) and metformin for 2 weeks showed a sigificant decrease in fasting plasma insulin compare with the negative control rats with a reduction of 11,2%, 33,6%, 20% and 19,4%, respectively. Two weeks treatment either extract combination or metformin in diabetic rats, significantly increased GLUT 4 level (p<0,05) with a percentage increase of 6,68%, 15,21%, 12,76% and 1,77%
Granulocyte Colony Stimulating Factor (G-CSF) Quantitative Analysis In Plasmodium Vivax Infected Malaria Patients Experiencing Trombocytopenia
Abstract
Introduction : P. Vivax has been refered as pathological factor underlying increasing prevalences of haematological abnormality including anemia and thrombocytopenia. Through the day, exact mechanism of thrombocytopenia in malaria infection has yet come to a conclusion, several hypothesis are still in considered, phagocytosis and platelets aggregation remain the major disscussion topics. G-CSF, cytocine with elevated serum quantity in P. Vivax infections, were responsible in increasing phagoctytosis and conducting direct effect on platelets aggregation using adequate ADP. Increasing number of ADP in malaria cases were correlated with erythrocyte haemolitic, leading to increasing platelets aggregation. Although numerous hypothesis has been compelled, only a-few research publication has been made corresponding to G-CSF serum level on malaria P. Vivax infection and its correlations to thrombocytopenic events.
Aim of study : To analyze the relations between G-CSF serum levels and parasite numbers towards platelets profile in infected malaria P. Vivax patients.
Methods : Study design using Prospective analysis. Thirty six patients with single infection of Malaria Vivax in Puskesmas Sukamaju (Primary Heath Care Centre) and Puskesmas Kota Karang were assessed for G-CSF plasma levels, platelet counts, and MPV. Data analysis were conducted using Spearman correlation methods with SPSS.
Results : Study results showing significant correlation between G-CSF serum levels towards Platelet conts (R = -0,397(p = 0,016)), without significant correlation between G-CSF and MPV value (p = 0,874)
Conclusion : G-CSF serum levels were related with thrombocytopenia, with no correlation towards MPV.
