Pharmacological inhibition reveals participation of the endocytic compartment in positive feedback IL-6 secretion in human skeletal myotubes
IL-6 is an important cytokine involved in metabolic, immunological, and cell-fate responses. It is released upon stimulation by skeletal muscle cells through partially characterized mechanisms. In some cell types, IL-6 has been reported to activate a positive feedback loop involving endocytic vesicles, but evidence is mostly based on transcription and signal transduction mechanisms and is very scarce in muscle cells. Our aim was to directly demonstrate the presence of positive feedback in the ATP-induced release of IL-6 into the supernatant of human skeletal muscle cultures. The total release (production) of IL-6 was reduced for higher volumes of supernatant, when the secreted IL-6 molecules are more diluted, and enhanced when the supernatant volume was lower. In addition, secretion was impaired both by tocilizumab, a blocker of human IL-6 receptors, and by the soluble form of the receptor. The secretion in response to ATP was also inhibited by treatment with the endocytosis inhibitor dynasore, and by disruption of the acidic gradient of the endocytic compartment using different methods (chloroquine, NH4Cl or monensin). IL-6 secretion was also impaired by NED-19, a specific inhibitor of the two pore channels receptor mediating Ca2+ release from the endolysosomal compartment. IL-6 and ATP increased IL-6 mRNA levels, an effect blocked by tocilizumab. Altogether, our results demonstrate that ATP-secreted IL-6 activates a positive loop based on IL-6 receptors, endocytosis, two pore channels and IL-6 transcription. Given the importance of muscle IL-6 as a systemic regulator and as an inflammatory mediator, our study can help to understand muscle pathophysiology
Predicción de glucosa mediante transformaciones del espectro al campo de visión, computación evolutiva y aprendizaje profundo
Programa de Doctorado en Tecnologías InformáticasLa Diabetes Mellitus es una enfermedad autoinmune que se caracteriza por unos niveles de glucosa en sangre elevados si no se actúa de manera adecuada. Existen dos tipos, la Diabetes Tipo 1 en la que el páncreas no produce la insulina necesaria para que las células puedan procesar la glucosa y la Diabetes Tipo 2, donde el páncreas sí produce insulina, pero las células receptoras son incapaces de procesarla correctamente para absorber la glucosa. Para las personas con diabetes es de vital importancia mantener los niveles de glucosa en lo que se conoce como rango saludable, no llegando a los extremos, tanto inferiores (Hipoglucemia) como superiores (Hiperglucemia). Los avances en los ´últimos años en el ´ámbito de la Inteligencia Artificial están consiguiendo resolver problemas complejos donde es necesario predecir situaciones y/o valores que hasta ahora no eran posible. Esto nos lleva a plantear en este trabajo de tesis la hipótesis de poder generar modelos que puedan predecir situaciones potencialmente peligrosas a personas con diabetes desde dos enfoques diferentes. Un primer enfoque, donde se busca la predicción del valor exacto de glucosa mediante el uso modelos basados en sistemas de lógica difusa, y un segundo enfoque, donde el objetivo es predecir situaciones de hipoglucemias e hiperglucemias futuras mediante el uso de transformadas de espectro al campo de visión y modelos de aprendizaje profundo. De esta forma, estos modelos permitirán informar con antelación a las personas con diabetes para que puedan tomar las acciones correctoras con el objetivo de evitar situaciones peligrosas..
Diabetes mellitus is an autoimmune disease characterised by elevated blood glucose levels if not treated properly. There are two types: Type 1 diabetes, in which the pancreas does not produce the insulin necessary for the cells to process glucose. cells to process glucose, and Type 2 diabetes, in which the pancreas does produce insulin but the receptor cells are unable to process it correctly to absorb glucose. For people with diabetes, it is vital to keep glucose levels within the so-called healthy range, avoiding the extremes of low (hypoglycaemia) and high (hyperglycaemia). The progress made in recent years in the field of artificial intelligence is capable of solving complex problems where it is necessary to predict situations and/or values that were not possible and/or values that were not possible until now. This leads us to propose in this thesis the hypothesis of generating models that can predict potentially dangerous situations for people with diabetes from two different approaches. Self-perception of young university students about their leisure time before, during and after covid-19. An approach to the spanish context
The aim of this research is to explore and analyse the self-perception of a group of young Spanish university students in relation to their leisure activities, in a temporal space defined in three moments: before the COVID-19 pandemic, during the confinement and after the confinement (return to normality). Information was collected with a self-administered questionnaire to 54 young Spaniards. The results show that before the pandemic, leisure was in person with activities shared with friends. During confinement, leisure was non-face-to-face, significantly increasing family and individual activities. After the confinement, the time that young people spent with their family was reduced, and the time with friends in person and outdoors with “post-COVID” concerns increased. It is concluded that young people have changed their guidelines, activities and with whom they spend their leisure time in comparison to their experience before and during confinement. The return to normality has also changed how young people spend their leisure time, and with whom, by adopting preventive behaviors.This publication is part of the research project entitled Development of Youth COVID 19 programmes – “Delve into the improvement of the lives of social service workers and their degree of motivation to help improve the quality of life of the most vulnerable families and groups, as well as to know and understand the needs of young people in the CAM for the year 2022”, funded by the Institute Department of Districts, Youth and Citizen Participation of the Autonomous City of Melilla (BOME-BX-2022-60).peerReviewe

Análisis transversal del medio ambiente en la Agenda 2030: diplomacia pública en las páginas web de municipios de Portugal
This article delves into local public diplomacy from the perspective of climate action. A cross-sectional analysis is made of the climate perspective in the international 2030 Sustainable Development Agenda, considering the social, political, and economic challenges implied by human alteration of the environment. The websites of local municipal corporations in Portugal are analysed to identify the information referring to Agenda 2030 and its convergence in climate action. To this end, a content analysis method was designed based on the relationship of the Agenda’s sustainable development goals (SDGs) with their associated climate change goals. Attention is paid primarily to the websites’ fixed sections, and secondarily to their informational content and external portals. The method is applied to the websites of seven of Portugal’s municipalities. Conclusions are drawn concerning whether or not they provide information on climate change so that the effort directed by these local governments towards climate action can be determined. A comparison is made with the international case. This tool contributes to localizing the SDGs and prioritizing environmentally important components.peerReviewe
Spatial and temporal analysis of water resources in the olive-growing areas of Extremadura, Southwestern Spain
The increasing variability of precipitation, higher temperatures, and recurring droughts in the semi-arid regions due to climate change are leading to increased aridity, resulting in scarcer water resources for crops. The present study aimed to analyse the spatial distribution of climate variables related to water resources in the olive-growing areas throughout Extremadura, southwestern Spain. To perform this task, three climate variables were used: the potential evapotranspiration of the crop, the FAO aridity index, and the annual water requirement. Considering data from 58 weather stations located throughout Extremadura and 17 along boundaries with at least a 30-year length (within the 1991–2021 period), each variable was computed at each station. After calculating some descriptive statistics, a multivariate geostatistical (regression-kriging) algorithm, incorporating secondary information on elevation and latitude, was used to accurately map each climate variable. Later, temporal trends and their magnitude were analysed using the Mann–Kendall test and the Sen’s estimator, respectively. The highest evapotranspiration and water requirements are located in the southern part of the region, which has large areas dedicated to olive cultivation. In the northern part of the region, there is greater spatial variability in evapotranspiration and, consequently, in water requirements for olive groves due to the more rugged topography. Similarly, the olive-growing areas with the highest aridity are also in the south of Extremadura. In most areas of Extremadura, olive cultivation requires appropriate irrigation for optimal productivity. According to evapotranspiration trends, the water requirements will become greater in the future. However, it is not guaranteed that the water supply will be sufficient in olive-growing areas where aridity is higher and water resources are scarce. Problems related to the criminal sanction of vote-buying within the Spanish election law
«Si me votas, te daré trabajo». «Si me votas, te bajaré los impuestos». «Si me votas, construiré un puente que le vendrá muy bien a tu negocio». ¿Corrupción de electores o democracia? El legislador español ha confiado parte de la respuesta a tan trascendental cuestión al artículo 146 de la Ley Orgánica del Régimen Electoral General. Empero, el análisis de la norma revela su pésima factura, su obsolescencia fruto del olvido y una aplicación por los tribunales no solo escasa sino también poco atenta. De hecho, el precepto no contempla sanción alguna para el elector que vende su voto, lo que sugiere una interpretación restrictiva que deja un peligroso espacio de impunidad para la libre compraventa de votos. La laguna parece poderse llenar por lo menos en parte mediante una lectura distinta de la relación entre compra de votos y cohecho que coloca al intérprete en el debate relativo al espacio que tiene en España la sanción de la concusión.‘If you vote for me, I'll give you a job. ‘If you vote for me, I will lower your taxes. ‘If you vote for me, I will build a bridge that will do your business a lot of good’. Corruption of voters or democracy? The Spanish legislator has entrusted part of the answer to such a transcendental question to article 146 of the Organic Law on the General Electoral System (Ley Orgánica del Régimen Electoral General). However, an analysis of the law reveals that it has been poorly drafted, its obsolescence is the result of forgetfulness, and its application by the courts is not only scarce but also not very attentive. In fact, the provision does not contemplate any sanction for voters who sell their votes, which suggests a restrictive interpretation that leaves a dangerous space of impunity for the free buying and selling of votes. The gap seems to be filled at least in part by a different reading of the relationship between vote buying and bribery, which places the interpreter in the debate regarding the space that the sanction of extortion has in Spain.peerReviewe
