    Fut9 Deficiency Causes Abnormal Neural Development in the Mouse Cerebral Cortex and Retina.

    pdfα1,3-Fucosyltransferase 9 (Fut9) is responsible for the synthesis of Lewis X [LeX, Galβ1-4(Fucα1-3)GlcNAc] carbohydrate epitope, a marker for pluripotent or multipotent tissue-specific stem cells. Although Fut9-deficient mice show anxiety-related behaviors, structural and cellular abnormalities in the brain remain to be investigated. In this study, using in situ hybridization and immunohistochemical techniques in combination, we clarified the spatiotemporal expression of Fut9, together with LeX, in the brain and retina. We found that Fut9-expressing cells are positive for Ctip2, a marker of neurons residing in layer V/VI, and TLE4, a marker of corticothalamic projection neurons (CThPNs) in layer VI, of the cortex. A birthdating analysis using 5-ethynyl-2'-deoxyuridine at embryonic day (E)11.5, 5-bromo-2'-deoxyuridine at E12.5, and in utero electroporation of a GFP expression plasmid at E14.5 revealed a reduction in the percentage of neurons produced at E11.5 in layer VI/subplate of the cortex and in the ganglion cell layer of the retina in P0 Fut9-/- mice. Furthermore, this reduction in layer VI/subplate neurons persisted into adulthood, leading to a reduction in the number of Ctip2strong/Satb2- excitatory neurons in layer V/VI of the adult Fut9-/- cortex. These results suggest that Fut9 plays significant roles in the differentiation, migration, and maturation of neural precursor cells in the cortex and retina.Journal Articlejournal articl

    Occupational therapy using a robotic-assisted glove ameliorates finger dexterity and modulates functional connectivity in amyotrophic lateral sclerosis.

    application/pdfIntroduction: Although rehabilitation is recommended for amyotrophic lateral sclerosis (ALS), improvement of functional decline has hardly been achieved. We investigated the effect of occupational therapy that uses a robotic-assisted glove (RAG) on hand dexterity and the functional connectivities found in the brain of ALS patients.Method: Ten patients diagnosed with ALS and admitted to the Shiga University of Medical Science (SUMS) Hospital from December 2018 to December 2021 participated in the study. These participants chose the hand side to wear RAG and exercised for two weeks. A sham movement was performed on the other side. We administered several functional assessments, including the Simple Test for Evaluating Hand Function (STEF), grip strength, pinch meter for grip strength, Canadian occupational performance measure (COPM), as well as nerve conduction study (NCS) before and after the exercise, and evaluated the results. We also analyzed six patients' resting-state functional magnetic resonance imaging (rs-fMRI).Results: Two-week robotic rehabilitation improved the STEF, grip strength, and COPM scores when compared with those of the other side. However, no significant effect was observed in the pinch meter and the NCS results. The rs-fMRI data analysis revealed that the robotic rehabilitation augmented two functional connectivities between the left pallidum-right supplementary motor cortex and right insular cortex-right sensorimotor network among the patients, which had beneficial effects.Conclusion: The occupational therapy using RAG displayed improved hand dexterity. The enhanced functional connectivities around the sensorimotor network might be associated with the improvement in hand dexterity because of the RAG.Journal Articlejournal articl

    Limited social support is associated with depression, anxiety, and insomnia in a Japanese working population

    application/pdfBackground: Lack of social support is associated with depression, anxiety, and insomnia. This study aimed to determine the source of support related to depression, anxiety, and insomnia among Japanese workers.Results: A total of 1,852 Japanese employees (38.4% male, 45.9 ± 12.9 years) participated in the survey, with 15.5, 10.8, and 8.2% of the participants having depressive symptoms (PHQ-9 ≥ 10), anxiety symptoms (GAD-7 ≥ 10), and insomnia (ISI ≥ 15), respectively. The logistic regression analysis suggested that job stressors were associated with depressive symptoms (p < 0.001), anxiety symptoms (p < 0.001), and insomnia (p = 0.009). In contrast, support from co-workers (p = 0.016) and family members (p = 0.001) was associated with decreased depressive symptoms. Support from family members was associated with decreased insomnia (p = 0.005).Conclusion: Social support from co-workers and family may be associated with reduced depressive symptoms, and family support may be associated with reduced insomnia in the Japanese working population.Clinical trial registration: https://clinicaltrials.gov/ct2/show/NCT03276585.Journal Articlejournal articl

    The prevalence of depression in isolated/idiopathic rapid eye movement sleep behavior disorder: A systematic review and meta-analysis.

    pdftiftiffThis systematic review and meta-analysis aimed to investigate depression prevalence and depression-related symptoms among patients with isolated/idiopathic rapid eye movement sleep behavior disorder (iRBD). We systematically searched online databases (PubMed and Scopus), performed meta-analyses of psychiatric symptoms prevalence using a random-effects model, and calculated 95% prediction intervals (PIs) and I2 values to evaluate the degree of heterogeneity. We conducted a meta-regression analysis to assess the relationship between psychiatric symptom severity, age at diagnosis, and disease duration from onset of iRBD. We analyzed 31 studies which included 3,576 patients (2,871 men, 80.3%; mean age, 66.6 ± 8.6 years). The pooled depression prevalence was 28.8% (95% CI 23.1-35.2, 95% PI 8.1-65.1, and I2 = 83.9%). We found a significantly negative correlation between depression-scale scores and disease duration in iRBD (p = 0.012, β = -0.36, R2 analog = 0.33). Pooled prevalence of apathy and anxiety was 38.4% (95% CI 27.7-50.4, 95% PI 0.02-99.9, and I2 = 57.8%) and 21.3% (95% CI 15.5-28.5, 95% PI 4.2-62.6, and I2 = 47.1%), respectively. Few articles on alexithymia were available for meta-analysis. This study confirmed high prevalence of depression, apathy, and anxiety in patients with iRBD.Journal Articlejournal articl

    Self-injury as a predominant challenging behavior in epilepsy : A study in a residential facility for profoundly disabled patients.

    pdfBackground: The association between challenging behavior (CB) and epilepsy in people with intellectual disability (ID) remains largely controversial.Aim: To clarify the correlation between CB and epilepsy, we investigated the clinical characteristics of CB in both people with and without epilepsy among individuals with ID hospitalized in our residential facility.Methods and procedures: A total of 63 individuals with CB was retrospectively investigated using the Behavior Problems Inventory, and the following items were collected from the medical records: sex, age, hospitalization period, etiology and risk factors, level of ID, type of CB, administration of psychotropic drugs, presence or absence of epilepsy and clinical features of epilepsy.Outcomes and results: Almost all individuals with CB showed profound ID. There was no significant difference in the rate of CB between people with and without epilepsy. A type analysis of CB revealed that self-injurious behavior was observed more frequently in epilepsy cases (66.7 %) than in cases without epilepsy (36.4 %) (p = 0.015), and self-injurious behavior was the predominant clinical form in people with epilepsy among the three subgroups of CB (self-injurious, aggressive and self-injurious and aggressive behavior).Conclusions and implications: The high incidence of self-injurious behavior in epilepsy with profound disabilities may imply the presence of common pathological basis of self-injurious behavior and epilepsy.Journal Articlejournal articl
