    Alternative splicing in human cells exposed to ionizing radiation: a comprehensive review of ex vivo and in vivo studies

    Purpose: This study reviews how ionizing radiation (IR) induces alternative splicing (AS) in non-tumor and tumor cells under both ex vivo and in vivo irradiation conditions. The relevance and limitations of IR-induced AS in identifying potential biomarkers are highlighted for two main applications: biodosimetry and radiotherapy. Conclusions: Radiation promotes alterations in AS, which may differentially affect the response in both tumor and non-tumor cells. This response can occur in genes that change their overall expression as well as in those that remain unaltered in response to IR. Although cis-regulators modulate AS, trans-regulators like splicing factors are more involved in the IR response. Variants of key genes involved in the DNA damage response (DDR) are regulated in non-tumor cells while they are often deregulated in tumor cells favoring radioresistance. Identifying IR-induced AS variants could enhance the sensitivity of biodosimeters for dose estimation and biomarkers for radiosensitivity, offering potential strategies to personalize radiotherapy and improve outcomes.     Analyzing water uptake of apple trees using isotopic techniques in the Shandong Peninsula, China

    Study regionThe hilly area of Shandong Peninsula is a pivotal apple-producing region in China. However, the precise water sources utilized by the apple trees for transpiration remain poorly understood in this region.Study focusHere we quantify the water sources used by apple trees in this area using stable isotopic tracing methods. Through on-field studies in a representative apple orchard and subsequent isotopic assessments, the primary water sources tapped by the apple trees were identified in three plots with contrasting soil characteristics and through 5 days of sub daily sampling.New hydrological insights for the regionOur results show that apple trees have a marked preference for soil water centered at the 60 cm depth, with more deep water use at plots without weathered layers. Notably, the isotopic compositions of the xylem water leaned more towards signatures of soil water, rather than immediate irrigation water or groundwater. Given the irrigation water used to be the dominant water source recharging into soil, the weak contribution of irrigation water to plant would be attributed to the high soil evaporation rates during the growth phase, which strongly alter the isotopic composition of irrigation water in shallow soil layers. These insights boosted our comprehension of water sourcing mechanisms in the sloped orchard ecosystems in the Shandong Peninsula and lay the groundwork for deeper exploration into the irrigation ratio to rainwater utilized by apple trees in comparable regions.Fil: Pang, Tianze.     Adding anaerobic digestate to commercial farm fields increases soil organic carbon

    Integrating anaerobic digestion technology canhelp farms meet agronomic, environmental, and economic goals. The slurryby-product from anaerobic digestion – anaerobic digestate – can be applied to croplands as an organicamendment and fertilizer. However, digestate effects on soil organic carbon(SOC) and soil fertility are poorly understood, especially at the field scale.In this study, we analyzed data from a multi-fieldcommercial farm in Iowa, USA, which integrates agricultural production withanaerobic digestion. The digestate produced in this system was applied to14 crop fields over 5-12 years. To assess the digestate effects on SOC stocksand soil plant-available nutrients, we analyzed the digestate chemicalcomposition, digestate rates, and soil test results of 421 samples taken at0-15 cm soil depth. Most sampled points (86%) increasedSOC stock, with greater gains observed in soilswith lower initial SOC levels. The average SOC accrual rate was 0.8 Mg ha-1y-1 (confidence interval: 0.7-0.9 Mg ha-1 yr-1),and isotope analysis (13C and 15N)indicated that new soil organic matter is primarily derived from digestate. Assumingsimplified SOC dynamics, the SOC formation efficiencyfrom digestate was estimated at 18% (higher than the estimate for raw manure).Anaerobic digestate also increased soil test phosphorus (STP) and potassium,with STP values doubling over eight years, exceeding crop requirements.Integrating anaerobic digestion on farms can help reverse soil degradation andenhance agricultural sustainability, although STP should be monitored toprevent potential adverse environmental impacts.Fil: Villarino, Sebastián Horacio.     Phylogeny of the planthopper genus Megamelus (Hemiptera, Delphacidae), with the description of two new species from South America

    Megamelus is a genus of Delphacidae widely distributed and mostly associated with plants in freshwater environments. Despite various taxonomic revisions and thorough research, the delimitation of the genus, its diversity, and its evolutionary history need to be further explored. Moreover, features originally considered distinctive of the genus exhibit variation and should be reassessed. Here, the genus Megamelus in South America was examined, describing two new species, Megamelus delticus Remes Lenicov & Mariani, sp. nov. and Megamelus serpentinus Mariani & Remes Lenicov, sp. nov., and providing information on their host plants and geographical distribution. The distribution and host range knowledge of Megamelus iphigeniae and Megamelus timehri are also expanded, the male brachypter of M. timehri described for the first time, and a key to distinguish the species, based on male and female genitalia and their external morphology, is provided. Moreover, the first phylogenetic analysis of the genus is presented, based on the mitochondrial COI gene to clarify the interspecific relationships among its members. Our combined findings support the monophyly of the genus and refine diagnostic features, including the importance of the pygofer’s lobed appearance. This comprehensive revision highlights the need for further multidisciplinary approaches to fully understand the evolutionary history of Megamelus and its interactions with host plants and environments.Fil: Salinas, Nicolas Alejandro.     Agroforestry systems can protect yerba mate crops from extreme environmental conditions without altering their secondary metabolites and productivity

    Agroforestry systems (AFS) offer sustainable alternatives to monoculture for crops such as yerba mate (Ilex paraguariensis). This study assessed the impact of AFS and monoculture on yield, secondary metabolites, and physiological responses of yerba mate plants, alongside seasonal variations in microclimatic conditions and herbivory attacks. The AFS, featuring Toona ciliata intercropped with yerba mate, provided approximately 45% shade, reducing extreme temperatures and thermal fluctuations. While yerba mate in monoculture exhibited higher photosynthetic capacity, plants in AFS developed morphological adaptations, such as thinner leaves, enhancing performance under shade. Herbivory was initially higher in AFS during spring but equalized between systems later in the season. Secondary metabolite profiles were similar across both systems, indicating no compromise in product quality. Despite distinct microclimatic conditions, crop yields were comparable between systems. These results highlight the capacity of AFS to buffer environmental stress, enhance plant resilience, and maintain productivity, reinforcing its potential as a sustainable cultivation strategy in a changing climate.     The use of molecular and cell biology scientific news to facilitate learning and scientific thinking

    Molecular Cell Biology (MCB) should be taught according to the scientific practices, avoiding cumulative and memory knowledge construction, but favoring scientific thinking. A way to achieve this goal is to apply activities involving scientific news, which construct knowledge through significant learning and the development of critical thinking. The study aimed to evaluatethe implementation of learning activities involving scientific news in the MCB course at the undergraduate level. The perspective of design-based research was applied, whereas the cognitive and interpersonal aspects were evaluated by means of the professors´ narration, class registration, questionnaires answered by the students, and the evaluation of the activities carried out bythe students. Results showed that the activity involving scientific news reinforces the construction and integration of new knowledge with that previously acquired and consolidates the acquisition of scientific thinking. Even though the completion of the activity involved a complex process, according to the professor and students´ opinion, the students observed positive aspects such asthe application of biological and scientific language and the motivation to search for related information. Regarding the cooperative learning strategy, students perceived that it is a methodology that facilitates their learning. In summary, incorporating scientific news into MCB courses will enhance professors´ effectiveness in achieving didactic goals while also fostering scientific thinking in students, equipping them for future roles as biologists and professorsin biological sciences.Fil: Giojalas, Laura Cecilia.     Failure of doramectin and ivermectin in preventing Cochliomyia hominivorax myiasis in a subtropical region: A pharmacokinetic-pharmacodynamic study

    The aim of this work is to present a case study where the failure of IVM 3.15 % and DRM 1 % to prevent natural infestations of C. hominivorax larvae in Argentina is investigated based on field efficacy tests and a pharmacokinetic and pharmacodynamic analysis. Thirty male crossbred Braford calves were randomly assigned to three experimental groups (n = 10), the IVM 3.15 % group (subcutaneously at 630 µg/kg), the DRM 1 % group (subcutaneously at 200 µg/kg) and the control group (saline solution subcutaneously). All treatments were performed at the time of castration surgery through a scrotal incision, and the wounds were exposed to natural infestations of C. hominivorax. Wound inspections were carried out on days 3, 6, and 13 post-treatments. Jugular blood samples were taken from experimental animals at 3- and 6-days post-treatment. In presence of C. hominivorax larvae, samples of both the larvae and wound secretions were collected in plastic vials on days 3 and 6 to measure concentrations of both drugs by high-performance liquid chromatography. On day 3 posttreatment, active myiasis was observed in 9 animals from the control group, 5 from the IVM 3.15 % group, and 6 from the DRM 1 % group. On day 6 post-treatment, 5 and 3 new myiasis were detected in the IVM 3.15 % and DRM 1 % group, respectively. No larvae were observed in the wounds on day 13 post-treatment. Interestingly, DRM concentrations tended to be higher in larvae compared to IVM on day 3 post-treatment (p = 0.051), and IVM plasma concentrations tended to be higher than those measured for DRM on day 6 (p = 0.087). There was a very strong correlation between DRM and IVM concentrations in plasma and wound secretions and larvae. The uptake of DRM from wound secretions to larvae was 83 % greater than that of IVM (p = 0.03). The results of this trial suggest the presence of C. hominivorax resistance to DRM and highlight concern about the lack of effectiveness of IVM 3.15 % in preventing C. hominivorax infestations.Fil: Muchiut, Sebastián Manuel.     Patterns and Determinants of Imported Malaria near the Argentina–Bolivia Border, 1977–2009

    In the Americas, the number of confirmed malaria cases decreased by 65.4% between 2000 and 2023, and malaria elimination is now in sight for many countries. Argentina is currently free of autochthonous malaria transmission. Until 2011, cases of malaria were concentrated in Northwestern Argentina, near the border with Bolivia, a country that continues to have malaria transmission. The Orán department (Salta province, Northwestern Argentina) had particularly high transmission near a main road that is a pathway for migration from Bolivia. The purpose of this study was to identify which factors best explain the extent and timing of changes in the proportion of malaria cases in this area that were locally transmitted versus acquired in another country. Combining information from routinely collected case investigations, epidemiological surveillance data, and satellite imagery, we used a logistic model and a multi-level model of change to identify how demographic and place-level variables influence the proportion of malaria cases that were imported over time. The findings showed that the proportion of cases that were imported varied significantly over time, with a clear trend from predominantly autochthonous cases at the beginning of the study in 1977 (94.52%) to a majority of imported cases in 1992 (53.33%), a pattern that continued and intensified, reaching 76% imported cases by the end of the series in 2009. Nationality and place of work were key demographic factors influencing this shift. In particular, there was a change in transmission patterns after a cross-border intervention was launched in 1996. As Argentina has obtained certification of malaria elimination, these results may inform focal strategies for preventing re-introduction.Fil: Berlin, Erica.     Enzymes of the outer mitochondrial membrane regulating cholesterol and fatty acid metabolism in cancer

    Mitochondria are major sites of ATP production, also serving as metabolic and biosynthetic hubs. The structure of mitochondria comprises a matrix enclosed by an inner membrane which is separated from the outer mitochondrial membrane (OMM) by the intermembrane space. The OMM is a lipid bilayer that forms an interphase between mitochondria and the surrounding cytosol. While its primary function is to act as a selective barrier, controlling the exchange of molecules between these two cellular compartments, the OMM also plays a crucial role in various metabolic and regulatory processes. It is home to 114 distinct proteins, including transporters, signaling molecules, and structural components. Among these, approximately 30 are enzymes that actively participate in the regulation of lipid metabolism, amino acid processing, calcium homeostasis, and heme biosynthesis. These enzymatic functions highlight the OMM’s significance beyond its structural role, positioning it as a key player in cellular energy balance, apoptosis, and intracellular signaling pathways. Here, we focus on OMM proteins involved in the synthesis and utilization of cholesterol and fatty acids. We describe the mechanisms of action, effects, regulation, association with cancer progression, and their potential as pharmacological targets of the steroid acute regulatory protein (StAR), translocator protein (TSPO), acetyl-CoA carboxylase β (ACCβ), acyl-CoA synthetases long chain family member 1 and 6 (ACSL1 and ACSL6), and carnitine palmitoyl transferases 1A and 1B (CPT1A and CPT1B). Overall, we provide a comprehensive view of these OMM enzymes in non-cancerous and cancer cells as well as their potential as targets for developing novel chemotherapies.Fil: Zhang, Leilei.
