マウスを用いた長期にわたる学習能力に関する研究

　本研究の目的は，加齢に伴う脳の変異が，長期
にわたる行動に，どうのような変化となって現れ
るに現れるのかを調べる事である。実験動物を用
いた加齢に関する研究においては，運動，飼育環
境，餌等さまざまな条件実験群を比較して，その
影響を調べることが多い。本研究では，同一個体
の加齢による変化を長期（約 18 ヶ月）に渉って
調べる。本研究は長期にわたり行う実験のため，
継続して実施することが必要である。このような
実験の成果は，マウスを用いた認知・行動に関わ
る基礎的なデータであり，加齢の脳神経疾患の動
物モデルを作ることを可能とするものである。
　長期の行動データを収集するためには，実験装
置およびデータ収集システムが重要な役割を果た
す。行動実験装置は，従来から 2 段式オペラン
ト実験箱を用いている。これは通常用いられるマ
ウス用飼育箱の上に，オペラント箱を重ねたもの
で，今回は更に回転籠をつけ，活動量の測定を行っ
た。この装置の特徴は，実験動物の行動を連続的
に観察できるようになっていることである。 上
段に置かれたオペラント箱と下段の飼育箱の間は
自由に行き来できるようにしてある。オペラント
箱は，他のマウスと隣り合っているため，効果的
に遮音するため。防音素材を箱間に入れている。
それぞれのオペラント箱内部には，ルームランプ
を付け，10：14 時間の明暗サイクルで点灯して
いる。このオペラント箱を用いることにより，被
験体への接触を出来る限り少なくして，長期の実
験を可能としている。実験の度に動物に接触し，
移動し，異なった環境に置くといった操作や，あ
る一定の時間のみの行動を観察するという状況を
回避し，その影響を最小にすることができるので
ある。
　本年度の実験では，雄マウス（C57BL/jcl）12
匹を用い，24 時間連続のオペラント学習を行っ
た。30 週齢の加齢マウスと，10 週齢の若年マウ
スを比較している。2 つのレバーに対する反応，
および回転籠における活動量において，加齢マウ
スは若齢マウスより減少するが，昼夜の運動パ
ターン，反応の位置嗜好などには差が認められな
い。今後は，課題に対する正反応率の変化を中心
に，認知能力の変化を見ることとしている。
