    Biodiversity Assessment of Foxtail Millet (Setaria italica L.) Genotypes Based on RAPD Marker

    Foxtail millet (Setaria italica L.) is an important crop in areas where harsh environmental conditions limit crop productivity, including in high salinity and drought prone areas. In Indonesia millet is cultivated in certain areas; however, superior varieties are less developed in the country. The objective of this study was to analyze the genetic diversity among foxtail genotypes using RAPD markers. Genomic DNA of ten foxtail millet genotypes was amplified using 26 random primers through RAPD analysis. Of these primers, 22 produced reproducible amplicons and were polymorphic among the 10 foxtail millet genotypes. The number of polymorphic markers for each primer varied from 1 (primer E15) to 14 (primer M17). The amplified product size ranged from 120 to 2500 base pairs (bp). A dendrogram constructed based on the UPGMA clustering method put all genotypes in 5 distinct groups at 0.64 coefficient level. Diverse genotypes identified in this study can be used as potential parents in an efficient crop improvement program

    Can We Produce True Seed of Shallot (TSS) from Small Size Shallot Sets?

    The Indonesian government has been promoting the use of true seed of shallot (TSS) for shallot production due to its higher productivity potentials and greatly reduced risk of disease transfer. This research was conducted at Leuwikopo experimental station, Department of Agronomy and Horticulture, Bogor Agricultural University, West Java, Indonesia, in 2015. The aim of the research was to study TSS production from small size shallot sets “Bima Brebes”, an easy to flower shallot cultivar.  The trial was arranged in a completely randomized block design with two factors; BAP concentration i.e. 50, 100 150, 200 ppm as the first factor and the sizes of mother bulbs, i.e. small (3-4 g) and medium (5-10 g), as the second factor. The results showed that the vegetative growth of plants from the two bulb sizes were similar. TSS production and TSS quality from small size (3-4 g) bulbs was also comparable to that of medium size shallot set, except for 1000 seeds weight in which medium size mother bulb produced heavier 1000-seeds weight than that of small size shallot set. Therefore small size shallot set can potentially be useful for TSS production.

    Improvement of Shelf Life and Sensory Quality of Pummelos by Fruit Waxing and Wrapping

    The aims of this study were to evaluate the physical and chemical change of pummelo fruit during storage with waxing and wrapping, and to determine the effect of waxing and wrapping and their  its interaction in affecting shelf life of pummelo fruits. This research was arranged in a randomized complete block design with two factors and three replications. The first factor was waxing consisted of without waxing, 10% beeswax, 5% beeswax, and chitosan. The second factor was wrapping consisted of without wrapping and wrapping using either transparent yellow or white plastic wrap. Pummelo fruits experience weight loss, reduced total soluble solid (TSS), decrease in firmness and total acidity during 48 days of storage in room temperature. Waxing using beeswax or wrapping using plastic wraps significantly suppressed weight loss and maintained fruit firmness compared to those without waxing and wrapping. However, waxing and wrapping did not significantly affect the quality of pummelo fruits. Based on the fruit’s weight loss, firmness, and panelist preferences, 5% or 10% bees wax can be used as waxing. Both transparent yellow and white plastic wraps can be used to maintain fruit weight, firmness, and appearances during storage.

    The Potentials of Limau (Citrus amblycarpa Hassk. Ochse) as A Functional Food and Ornamental Mini Tree Based on Metabolomic and Morphological Approaches

    Limau (Citrus amblycarpa (Hassk.) Ochse) is one of the native Citrus species of West Java that is grown in the local backyard, thus is easily found in local cuisines. Limau has great potentials for use as a potted plant in home gardens for its aesthetic value and fragrance. The objective of this research was to describe the metabolites fingerprint and morphology of Limau as a potential functional and ornamental mini tree. Methanol leaf extracts from one-year-old Limau originated from Bogor were analyzed using untargeted Gas Chromatography Mass Spectrometry (GCMS). Methanol leaf extract was dominated by vitamin E and composed of other 18 metabolites including phytosterols, fatty acids and terpenes. The major portion of fruit extract consisted of 6-octadecenoic acid, palmitate and various fragrance compounds such as alpha sinensal, alpha limonene, beta citronellal, citronellol, and sabinene. Limau fruits are also a functional food due to its rich antioxidant and aromatic content in the fruits. The morphology of limau tree is suitable as a potted ornamentals or mini-fruits tree for its small crown, sparse branching with unifoliate aromatic leaves and small spines, and plentiful aromatics mini fruits with the diameter of around 2-4 cm. 

    The Use of Trellis and Mulch Increased Fruit Production of Spaghetti Squash (Cucurbita pepo L.)

    Spaghetti squash is squash of American origin that has spaghetti-like characteristic of fruit flesh, unlike any other squash varieties. This study aims to investigate the use of trellis and mulch to increase the spaghetti squash production. The experiment was conducted at the Experimental Station of Centre of Tropical Horticulture, Bogor from April to November 2016. The experiment used a completely randomized block design with four replicates. Two factors were tested, i.e. the use of trellis and mulch as the main plots, and accessions of the spaghetti squash as a sub-plot. The treatments were the use of trellis and mulch, mulch without the trellis, the trellis without mulch, and without the trellis and mulch. The spaghetti squash accessions used in this experiment were accessions from Taiwan (F1) and from America. Each experimental unit consists of five plants with a total a total of 160 plants. The results of this experiment showed that the growing squash using mulch and trellis had the greatest number of fruits per plant, more marketable fruits and had the largest fruit weight as compared with the other treatments. Taiwan accession had more fruits per plant and total marketable fruits, the largest fruit weight, the longest and widest fruits with large fruit diameter. Taiwan accession produced fruits at lower nodes of the plants, and fruits can be harvested earlier than the American accession. Correlation analysis indicated that the more fruits per plant, the more number of fruits that are marketable, the greater fruit weight and more fruits can be harvested from the higher nodes of the plants. From agronomical point of view, Taiwan spaghetti squash would be preferable to obtain the high fruit production provided the crops were mulched and grown with trellis.Keywords: Cucurbitacea, trellis, mulch, plant cultur

    Evaluation of Growth, Flowering and Seed Morphology of Batflower, Tacca chantrieri Andre

    Tacca chantrieri is a perennial plant belongs to Taccaceae family that has the potential to be marketed as an ornamental plant due to its unique shape and dark color inflorescence. To date, very limited publication on T. chantrieri are available. The aim of this research was to examine plant growth pattern and flower phenology of T. chantrieri from West Borneo, Indonesia, and from Queensland, Australia in relation to their potential development as ornamental pot plants. The study was extended to examine T. chantrieri seed morphology with an inclusion of an accession from Thailand.  The field experiment was conducted at the Leuwikopo experimental station in Darmaga, Bogor, Indonesia. Seed morphology examination was conducted at the Histology Laboratory of Department of Agronomy and Horticulture, Faculty of Agriculture, Bogor Agricultural University. T. chantrieri from West Borneo blooms six times a year whereas the Queensland accession twice a year. West Borneo accession had smaller bractea, fewer numbers of flowers per inflorescence and shorter flower stalk than Queensland accession. In addition, T. chantrieri Queensland grew faster than Borneo with the one leaf formed every two to three weeks, whereas it was at least four weeks in the Borneo accession. Both accessions were susceptible to leaf brown spot disease caused by Cercospora taccae. T. chantrieri Queensland seeds are oval dark brown, whereas West Borneo and Thailand accessions were shaped like kidneys (reniform) with a lighter brown color. This study demonstrated that there is great diversity in T. chantrieri including plant morphology, seed shape and color, which are important for species identification and for development of T. chantrieri as ornamental pot plant

    Nodulation and Nitrogen Fixation of Pongamia pinnata

    Nitrogen is one of the most important nutrients required by plants as a major component of all nucleic acids and proteins such as enzymes which control and enable their growth and reproduction. While much research has been conducted on the legume tree Pongamia (a candidate source for renewable biofuel), there is only a handful of studies on the mechanisms and regulation of nitrogen fixation, which is considered as one of the most important domestication traits that needs to be investigated.  Steps to optimize the symbiotic nitrogen fixation of Pongamia is, firstly, to select the best rhizobial isolates as inoculum among the naturally-occurring pool of bacteria in soils across Queensland. There have been reports on rhizobia nodulating Pongamia isolated from Western Australia and India but not in Queensland, Australia. This study is the first to report such rhizobia isolates that nodulated Pongamia.  Secondly, is to establish efficient nodulation by studying the factors such as nitrate and salinity. The published literature has provided extensive details on the effects of these factors in nodulation and their mechanisms in various legumes. However, only one preliminary study was published from our laboratory; the present study is the in-depth continuation of that effort. Lastly, nitrogen fixation in Pongamia must be assessed to determine if fixed nitrogen is sufficient to support its growth and reproduction. Acetylene reduction assay is the simplest and most common method of assessing fixed nitrogen but in this research, different methods were explored in order to compare both qualitative and quantitative results.     Adaptation of Wetland Rice to Extreme Weather

    Climate change is believed to increase the intensity and the frequency of extreme weather events in reference to strong winds and heavy precipitations. The extreme event is defined as strong wind at speed of 50 km.h-1 and rain fall intensity 10 to 20 mm.h-1 or more .  This condition is detrimental to rice production as this may lead to lodging and flooding which normally occurs during the grain filling stage to harvesting resulting in lower yield and grain quality.  The. Simultaneous extreme events and critical rice growth occured more frequently due to increasing cropping season within a year in Indonesia. Therefore, it is important to mitigate and develop adaptation strategies in order to sustain rice production. Efforts to adapt to these extreme environmental conditions are mostly based on genetics and agro ecological approaches. Genetically, rice with strong hills, high aerodynamic with low water retention is desired. Agro-ecological manipulation is conducted through wind break application, planting arrangement to facilitate better sunshine penetration, to manage water level and planting calendar.     Optimation of In vitro Lateral Shoots Multiplication of Papaya (Carica papaya L.) “Callina” with BAP and NAA

    Papaya is a popular fruit and is grown commercially in many subtropical and tropical countries. Papayas are generally grown from seeds; therefore the offsprings are not true-to-type and could come in three sexes, female, male, and hermaphrodite. Clonal propagation is required to obtain to grow true-to-type hermaphrodite papayas. In this research, we developed an in vitro protocol for shoot multiplication from lateral shoots from in vitro germinated papaya seedlings. The in vitro propagated plant materials could potentially be used as a source of papaya micro cuttings, or as scion for papaya grafting. The experiment was set up as a factorial experiment with NAA at 0, 0.1 and 0.5 mg.L-1, and BAP at 0, 0.1, 0.5, and 1.0 mg.L-1 in a completely randomized block design. BAP interacted with NAA in affecting the shoot production per explant. The optimum BAP and NAA concentration to produce lateral shoots was 0.54 mg. L-1 and 0.1 mg.L-1, respectively. Media without NAA reduced the number of lateral shoots and number of leaf per explant at any BAP concentration.Keywords: hermaphrodite, seeds, true to type, clonal propagation, micro cuttings

    Growth and Development of Potted Ornamental Chilli (Capsicum annuum L.) with Different Types of Fertilizer and Growing Media

    Chilli has great potential to be marketed as ornamental potted plants due to their diversity in colours and shape of the fruits. The purpose of this research was to determine the optimum growing media and types of fertilizer to improve the performance of three varieties of potted ornamental chilli, ”Syakira”, “Lembayung”, and “Ayesha”. This research was conducted in Cikabayan greenhouse, Bogor Agricultural University, from December 2016 until May 2017. The experiment was arranged in a split plot design with fertilizer as the main factor (without fertilizer, P0; AB mix; P1, and NPK + Gandasil D, P2) and growing media as sub plots, i.e. charcoal husk : soil : manure (M1), perlite: soil : manure (M1), and cocopeat : soil: manure (M3) with proportion of 2:1:1 (v/v). NPK fertilizer and foliar fertilizer Gandasil D showed the maximum plant height, number of branches, number of flowers and fruits. “Ayesha” treated with AB mix fertilizer had the largest fruit diameter whereas “Syakira” and “Ayesha” had the longest fruit under the same treatment. Ornamental chilli grown on charcoal husk and cocopeat had the maximum number of branches and flowers per plant. “Syakira” grown on charcoal husk with AB mix and NPK + Gandasil D, “Lembayung” using cocopeat with AB mix fertilizer, and “Ayesha” varieties using charcoal husk with AB mix fertilizer were the most preferred by the consumers
