Chapter 1 Background and Objectives 
Pickled vegetables selling at the wet market are common diets in Cambodia, and most of them are 
made using a Cambodian homemade-style. Many cases of food poisoning from homemade pickles 
happened in Cambodia. Unfortunately, there is no guideline of food safety due to insufficient 
information on how to make a safe food. It has been reported that homemade pickles selling at wet 
markets in Cambodia are contaminated with microbes due to the poor sanitation and personal 
hygiene of the food preparer and handler, which leads to a high risk of food spoilage and food 
poisoning. Therefore, heat treatment, such as hot pack filling and pasteurization, should be added in 
the processing to obtain a microbiologically safe product. 
This research aimed to examine the effect of the heating treatments (hot pack filling and 
pasteurization) on the preservative properties of vegetable pickles and to propose safe vegetable 
pickles in Cambodia. 
 
Chapter 2 Improvements in Preservation Properties of Homemade-Style Pickled Cucumber  
From Chapter 1, the pickles using homemade recipes are at a high risk of being spoiled and 
causing food poisoning, as there are no heating steps in the production process. Therefore, it is 
necessary to improve the hygienic conditions of the food supply for producing a safe pickle in 
Cambodia. 
Heat treatments using hot pack filling and pasteurization lead to microbiologically safe product 
and an extended shelf life. In this Chapter, the effect of heat treatments (hot pack filling and 
pasteurization) on preservative properties of pickled cucumbers is examined, and the acceptability of 
the pickled cucumbers prepared with heating process is evaluated.

Pickled cucumbers prepared with and without the heating process were made using a traditional 
Cambodian recipe and stored at 30 ºC for 7 days. In the heat treatment, cucumbers were packaged in 
a glass bottle with hot pack filling, and pasteurization was performed by placing the bottle in boiling 
water with a center temperature over than 75 ºC for 10 min. The presence of total viable bacteria, 
coliforms, Escherichia coli, yeasts, and molds in the pickled products were tested during the storage. 
To investigate the acceptability of the pickled cucumbers prepared with the heating process,sensory 
evaluation tests were performed.  
The samples without the heating process were contaminated with coliforms, Escherichia coli, 
yeasts, and molds and were in an initial putrefactive state because of the number of total viable 
bacteria of 107 CFU/g. On the other hand, coliforms, E. coli, yeasts, and molds were not detected in 
the samples prepared with the heating process within 7 days. This result indicated that the 
pasteurization treatment killed many spoilage microbes, including foodborne pathogens. From the 
results of the sensory evaluation and questionnaire survey, the pickled cucumbers prepared with the 
heating process made either in Japan or Cambodia were acceptable to the Cambodian panelists; 
however, further improvement in the softness of the pickles was commented upon. 
 
Chapter 3 Proposal of New Pickles to Improve Food Sanitation at Wet Markets  
From Chapter 2, the non-use of heat treatment is a possible factor leading to highly contaminated 
pickles with microbes, including the bacteria that cause food poisoning. The proposed heat treatment 
was effective in reducing the microbial content of pickles. In this Chapter, the applicability of the 
proposed heat treatment to young papaya is investigated, and the acceptability of the pickled young 
papaya is evaluated.  
Two kinds of pickled young papaya were made and stored at 30 ºC and 5 ºC for 14 days. The 
presence of total viable bacteria, coliforms, E. coli, yeasts and molds in the pickled products was 
tested during the storage. Sensory evaluations were conducted to assess the acceptability of new 
pickled young papaya among Cambodia and Asian panelists. 
The result showed no detection of total viable bacteria, coliforms, E. coli, yeast and mold during 
the storage at 30 ºC and at 5 ºC for 14 days. This ensures that the proposed heat treatment is 
applicable to young papayas. Pickled young papayas can be stored at room temperature (30 ºC) for 
14 days without microbes, indicating two-fold increases in storage duration at a room temperature of 
30 ºC compared to the pickled cucumber in Chapter 2. The sensory evaluation indicated that the 
pickled young papayas were acceptable to Cambodian and Asian people, but further improvement on 
lowering the salt content and increasing the sweetness and crunchiness of the salty sample was 
recommended.

Chapter 4 Summary and Recommendation 
Pickles made from agricultural products are popular ready-to-eat foods among Cambodians. Some 
of pickles selling at wet markets are made using a traditional Cambodian style. Usually, traditional 
Cambodian homemade pickles are produced without any heating steps in the production process. 
Food sanitation for the homemade pickles in Cambodia is poor. Importantly, there is a lack of 
knowledge relating to food sanitation and hygiene at the level of the producer, seller, consumer, and 
committee on food safety.  
In this study, a heat treatment was proposed and applied to make pickles using Cambodian 
homemade-style. It was found out that the proposed heat treatment was effective for producing a safe 
pickled cucumber. In addition, the proposed heat treatment was also applicable to pickled young 
papayas.  
The outcomes of this research can be recommended to the committee on food safety to create 
guidelines for food safety in Cambodia, and directly to the producer or manufacturer for making a 
safe pickle. The committee can educate the producers, manufacturers, sellers, consumers, and 
farmers accordingly to this guideline. Thus, the producers and sellers can produce and sell safe 
homemade pickled vegetables to consumers. The storage duration of the homemade pickles can be 
increased compared to the current state of 3 to 4 days. These lead to a decrease in food waste, an 
increase in income for producers and sellers, and a decrease in expenses for consumers. More 
importantly, farmers can effectively utilize agricultural products by decreasing agriculture waste, 
increasing income, and creating more job opportunities. Especially, this can help promote women's 
empowerment. It can be said that the outcome of this research contributes to a sustainable society by 
achieving the following the sustainable development goals (SDGs): (1) no poverty; (2) zero hunger; 
(3) good health and well-being; (9) Industry, innovation, and infrastructure; (12) responsible 
consumption and production. 
 
カンボジアにおける安全な野菜ピクルスのための加熱処理の適用 
カンボジアでは多くの種類の漬物が製造されており，漬物はウェットマーケットといわれ
る市場でも販売されている。ウェットマーケットでは冷蔵設備を備えていない店舗が多く，
さらに，ほとんどの漬物が外気にさらされた状況で販売されており，陳列時の交差汚染の危
険性も高く，食品衛生に関する意識が乏しいと感じられる。自家製漬物の販売業者は食品安
全や食品衛生に関する教育や情報が不足している。また，カンボジアにおいて，食品の安全
性に関するガイドラインは存在していない。そのため，残念ながら販売されている漬物が原
因物質となる食中毒が頻発している。微生物学的に安全な漬物製品を得るためには，製造工
程中の加熱処理工程が有効で，食品の安全性の改善・向上にはホットパック充填や低温殺菌

などの熱処理を製造工程に加える必要がある。本研究では，加熱処理（ホットパック充填，
および低温殺菌）が野菜ピクルスの保存性に及ぼす影響を検討すること，カンボジアにおけ
る安全な野菜ピクルスを提案することを目的とした。 
第 2 章では，カンボジアにおける自家製ピクルス（キュウリ）を試料として用い，加熱処
理（ホットパック充填，および低温殺菌）が保存性に及ぼす影響を検討すること，加熱処理
したピクルスの現地での受入度を調査することを目的とした。加熱処理した試料と加熱処理
していない試料を試作し，これらを 30 ℃で 7 日間保存した。保存期間中，試料の一般生菌
数，大腸菌群および大腸菌，酵母，カビの有無について検査した。加熱処理を含まない試料
は，大腸菌群や大腸菌，酵母，カビが混入しており，一般生菌数の結果から初期腐敗状態で
あった。一方，加熱処理を行った試料では，保存期間中において大腸菌群，大腸菌，酵母，
カビは検出されなかった。また，加熱処理した試料の一般生菌数は，保存期間において腐敗
とみなされるレベル以下（107 CFU/g 以下）であった。この結果は，加熱処理によって食中
毒菌を含む多くの腐敗微生物が死滅したことを示している。加熱処理したピクルスの受入度
を調査するために官能評価およびアンケート調査を行った。その結果，硬さなどテクスチャ
ーの改良が指摘されたが，カンボジア人パネリストの受入度は良好であった。 
第 3 章では，カンボジアにおいて農産物を有効かつ新規に利用するために，ピクルス原料
として用いられていない青パパイヤ（完熟していない，未熟な状態のパパイヤ）を原料とし
て用い，ピクルスを試作した。試料の製造工程では第 2 章と同じ加熱処理を適用した。ここ
では，2 種類の味付け（酸味，塩味）の青パパイヤピクルスを試作し，これらの受入度を調
査するとともに，保存性を検証することを目的とした。5 ℃および 30 ℃で 14 日間，試料
を保存して，試料中の一般生菌数，大腸菌群および大腸菌，酵母，カビの有無について検査
した結果，試作したピクルスは 30 ℃で 14 日間保存可能であることが示唆された。また，
官能評価結果から，試作した 2 種類のピクルスはカンボジアにおいて受け入れられる可能性
が示された。 
以上のように，本研究においては，カンボジアにおける野菜ピクルスに加熱処理は有効で，
加熱処理によりピクルスの保存性ならびに安全性を向上できることを示した。また，安全な
野菜ピクルスを開発・提案することができた。これらの成果は，カンボジアにおける食品衛
生の向上や農産物の有効利用，農業従事者の収入向上につながると思われる。 
 
 
審 査 報 告 概 要 
 
カンボジアにおいて，ポピュラーな加工食品の一つである漬物が原因物質となる食中毒が
頻発している。本研究では，カンボジアの生鮮市場で販売されている自家製漬物を対象とし

て，加熱処理が野菜漬物の保存性に及ぼす影響を明らかにすること，安全な野菜漬物を提
案・開発することを主目的とした。その結果，加熱処理工程を適用して試作した漬物は常温
で 7 日間程度保存可能であることが示され，本研究において提案した加熱処理により漬物の
保存性ならびに安全性を向上できることが明らかとなった。また，官能評価の結果，加熱処
理を施した漬物の受入度は良好であり，安全な野菜漬物を開発・提案することができた。こ
れらの成果は，カンボジアにおける食品衛生の向上や農産物の有効利用，農業従事者の収入
向上などにもつながり，また提案した加熱処理はカンボジア以外でも広く適用可能な基礎的
技術として高く評価できると考えられる。これらの研究成果等を詳細に検討した結果，審査
委員一同は博士(農学)の学位を授与する価値があると判断した。

Climate change is dramatically affecting the availability of water and soil resources, 
resulting in reduced rainfall in some parts of the world; soil moisture will eventually be 
reduced due to increased water evaporation, leading to crop drought and reduced yields. 
Khao Dawk Mali 105 (KDML105) is the most important rice in the Thai economy due to 
its ease of preparation and characteristics (e.g. Stickiness, softness and pleasant aroma), 
which have made KDML105 rice popular with consumers. However, KDML105 can only 
be planted once a year during the rainy season due to cultivation constraints, including 
the seasonal change in day length. KDML105 is not a known drought-tolerant variety, 
but its adaptability in a rainfed lowland ecosystem has been attributed to root system 
plasticity under abiotic stress. In Thailand, most rice-growing areas in rainfed regions 
have relatively limited water availability, which affects plant growth and reduces rice 
production. Drought is a serious factor limiting rice production and quality. Liu et al. 
(2006) reported that lack of water can cause a reduction in the number of panicles and 
developed seeds. Thawatchai (1992) showed that when rice plants are exposed to 
drought during the growing season, the yield is reduced by about 17% and by a further 
30% during the reproductive stage when young grains are formed.  
 
1. Preliminary assessment of selected rice varieties under different irrigation 
conditions. 
Drought is a major factor limiting rice production and quality. In recent years, 
drought damage in rainfed lowland rice areas has become a serious problem in Thailand. 
The study aimed to clarify the growth and yield-related characteristics of Thai Jasmine

rice compared to three other rice varieties under paddy and upland conditions. The 
effects of different water supply levels corresponding to paddy and upland conditions on 
four rice cultivars, i.e. KDML105, Dro1-IR64, Kinandang Patong (KP) and IR64, were 
studied. All four rice cultivars showed decreased growth (plant height and number of 
tillers per plant), photosynthesis rate, transpiration rate, stomatal conductance, 
number of panicles per plant, percentage of filled grains, number of spikelets per panicle, 
1000 grain weight and number of spikelets per plant under upland conditions, while the 
50% days to heading (50% DTH) increased. The 50% DTH of KDML105 indicated that 
this variety flowered later even under paddy conditions and the longer days (218.5 days) 
under drought stress conditions. Compared to paddy conditions, the 50% DTH under 
upland conditions increased by 10.91% for KDML105, 19.51% for Dro1-IR64, 18.91% for 
KP and 17.81% for IR64. For yield related traits, the percentage of filled grains in 
upland conditions was reduced by 3.77% in KP, 3.97% in IR64, 9.10% in Dro1-IR64 and 
12.42% in KDML105 compared to paddy conditions. From the result of this study, it can 
be concluded that the water deficit conditions resulted in reduced growth and yield 
components. Moreover, under drought conditions, KDML105 was less drought tolerant 
than other rice varieties. In contrast, Dro1 had the best drought tolerance, followed by 
IR64 and KP. However, the long growing season of KDML105, being a photosensitive 
variety, exposes it to the risk of drought for a prolonged period of time, affecting rice 
production in the long run. To avoid this unexpected feature, improving 
non-photosensitive varieties is a challenge that should be addressed against climate 
risks in the near future. 
 
2. Investigation of the breeding method using IR24 of the Thai jasmine rice variety 
KDML105 for early heading based on combined marker-assisted and phenotypic 
selection. 
The Thai rice variety Khao Dawk Mali 105 (KDML105) is the most important rice 
variety grown in Thailand. However, this variety can only be planted once a year during 
the rainy season due to some cultivation constraints. Flowering time (or heading date) is 
one of the most important agronomic traits in crops. The response of plants to flowering 
time is considered to be an aspect of their adaptation to climate change, which affects 
the yield potential of plants. We conducted this study to (i) improve Thai rice varieties 
by giving them the ability to escape dehydration and (ii) contribute to a database and 
future rice breeding programmes under unpredictable conditions. Regarding the

expression of heading traits in F2 hybrid plants, we observed that under natural day, 
both rice hybrids had 50% days to heading (DTH): at 105-125 days after sowing (DAS) in 
KDML105/IR24 and 110-125 DAS in IR24/KDML105. For the F3 phenotyping analysis, 
it was found that KDML105/IR24 on lines 27, 36 and 73 and IR24/KDML105 on lines 7, 
70 and 77 had low variation in DTH. These six lines are considered to be homozygous 
strains with a fixed heading period. In the genotyping of F3, we observed that 
KDML/IR24 on lines 27, 69, 73 and 87, including the combination of IR24/KDML105 on 
line 7, had the same expressions of Hd1and Hd3a as IR24 (the parent plant) and did not 
express DTH8. Based on these data, these five lines can be used in future breeding 
programmes. 
 
3. Investigating the breeding method using Dro1-IR24 with Thai jasmine rice variety 
KDML105 for a deep rooting trait based on combined marker-assisted and phenotypic 
selection. 
  Drought is a serious factor limiting rice production and quality. Many drought 
tolerance quantitative trait loci (DT-QTLs) identified in the rice genome were located on 
different chromosomes. DT-QTLs were located on chromosomes 1, 3, 4, 8 and 9, 
especially on chromosome 9, characterising deep rooting 1 (Dro1); deep rooting may help 
plants to avoid drought-induced stress by extracting water from deep soil layers. The 
objectives of this study were (i) to improve Thai rice varieties by giving them the ability 
to tolerate drought and (ii) to contribute to a database and future rice breeding 
programmes under unpredictable conditions. Regarding the root growth trait expression 
of F2 hybrid plants, the selection of rice with dominant root depth characteristics, it was 
found that rice plants with root lengths greater than 1500 cm and Ratio of Deeper 
Rooting (RDR) greater than 0.40 were selected for further variety selection studies. F3 
populations, the relationship between plant growth and root growth was found that 
tiller/plant was highly positively correlated with lower root length, RDR, lower root DW 
and total root DW. For F3 plants, there was a relationship between the number of tillers 
per plant and bottom root length. It can be observed that the plants can be divided into 3 
groups according to the bottom root length. The first group has a total root length of 100 
to 500 cm (KDML105, line 82, 97, 104 and 110). The second group was Dro1 IR64, lines 
6, 50 and 85. In this group a total root length is greater than 500 but not greater than 
1000 cm. The last group has a total root length than 1000 cm (more than in the parent 
plants), which are lines 29 and 38. In the genotyping of F3, we observed that

KDML105/Dro1 IR64 on lines 29, 38 and 50 had the same expression of Dro1 as Dro1 
IR64 (the parent plant). Based on these data, these three lines can be used in future 
breeding programs. 
 
General discussions 
Drought is a major factor limiting rice production and quality. In recent years, 
drought damage has become a serious problem in lowland rainfed rice areas in Thailand. 
We sought to determine the growth and yield characteristics of Thai jasmine rice under 
paddy and upland conditions. The results showed that KDML105 was less drought 
tolerant than the other varieties and yield-related traits were not significantly affected 
under water stress conditions. However, KDML105's long growing season (as a 
photosensitive variety) exposes it to drought risk for an extended period, affecting rice 
production in the long term. To avoid this unexpected feature, improving non-sensitive 
types is a challenge that should be against climate risks in the near future. Plant 
response to flowering time can be considered as an aspect of plant adaptation to climate 
change that affects plant yield potential. The results of this study confirm that 
expressing the same Hd1 and Hd3a as IR24 and not expressing DTH8 is effective for the 
early flowering of KDML105.  
Deep rooting can help plants avoid drought stress by extracting water from the 
deeper layers of the soil. KDML105-Dro1 has an improved root system, especially since 
the deep root length was significantly higher than that of KDML105. This is probably 
because this variety resulted from a crossing of thick and deep rooting from Dro1-IR64; 
therefore, the gene associated with deep rooting from Dro1-IR64 has been successfully 
introgressed into KDML105 by rice plants with dominant deep rooting traits expressed 
the same Dro1-gene as Dro1-IR64.  
KDML105 is known worldwide as jasmine rice, the most important rice in the Thai 
economy. The results of this study will contribute to the use of climate-smart jasmine 
rice breeding programs to prepare for the future. 
 
 
審 査 報 告 概 要 
 
タイのジャスミンライスとして知られるカオドマリ 105（KDML105）の干ばつ耐性の遺
伝的改良を目的に，干ばつに対する逃避性を付与し，KDML105/IR24 の交雑 F2雑種集団お

よび F3後代系統の出穂期の表現型と遺伝子型を解析した。遺伝子型解析では，Hd1 および
Hd3a を発現し，DTH8 を発現しないことで，非日長反応性になることが確認された。次に，
乾燥ストレスを回避する深根性を付与し， KDML105/Dro1 IR64 の交雑 F2雑種集団および
F3後代系統の形質発現および表現型と遺伝子型を解析した結果，深根性を持つイネの分げつ
数は下根長，総根長，下根乾物重，総根乾物重と形質相関があることがわかった。さらに，
総根長が 1000cm より大きい系統が，深根性遺伝子 Dro1 を発現していることが明らかにな
った。本研究の結果から作出された育種系統および形質相関の知見は，今後の気候変動に対
応したジャスミンライスの育種改良に貢献すると考えられる。よって審査員一同は博士（国
際農業開発学）の学位を授与する価値があると判断した。

【背景・目的】 
 シアノバクテリアは酸素発生型光合成を行う唯一の原核生物であり，増殖に外部からの有
機物を必要とせず，太陽光と CO2 ，微量のミネラルのみで生育する。さらに光エネルギー
をバイオマスに変換するエネルギー効率が陸上植物と比較して高い（植物：0.5%以下，シア
ノバクテリア：3~9%）。これらのことから，シアノバクテリアは CO2 ニュートラルに有用
化合物を生産するホストとして期待されている。しかし，シアノバクテリアは大腸菌などの
従属栄養生物に比べて生育が遅く，生産性において劣ることが多い点や，シアノバクテリア
に対する遺伝子工学ツールの不足が課題である。今後シアノバクテリアを用いた物質生産を
産業レベルに発展させるためには，さらなる遺伝子工学ツールの拡充やシアノバクテリアに
特徴的な細胞内環境，あるいは特異的代謝経路を活かした有用物質生産が重要になってくる。
本研究ではシアノバクテリアの遺伝子工学ツールの拡充と新規有用物質の生産系構築を目
的とし，幅広いシアノバクテリア種で利用できるベクターを開発した。さらに新規構築した
ベクターを用いて，植物由来有用テルペノイドの生産に取り組んだ。テルペノイドはステロ
ールやホルモンなどの生体成分や，精油，原薬，天然ゴムなどの有用二次代謝産物を含み，
現在利用されているテルペノイドの大部分は植物から抽出したものである。植物の葉緑体の
祖先生物であるシアノバクテリアはテルペノイドの基礎骨格を作る代謝経路が植物と同一
という点に着目し，シアノバクテリアを利用した異種テルペノイド生産系（1,8-シネオール，
ストリゴラクトン）の構築に取り組んだ。 
 
【結果】1. シアノバクテリアにおいて高い複製活性を持つ自律複製領域の探索と過剰発現
プラスミドの構築 
 自己複製プラスミドは，プラスミドにコードされた複製開始因子（Rep タンパク質など）

を用いて，ホストの複製機構を利用して自律的に複製される。自律複製配列は，複製開始因 
子をコードする遺伝子とその結合配列からなり，複製起点となる。これまでにシアノバクテ
リア内在性プラスミドを利用したベクターが開発されているが，その多くが単一のシアノバ
クテリア種のみで自律複製され，広宿主ベクターとして報告されているものはわずかである。
また RSF1010 ベクターはホストレンジが広く複数のシアノバクテリアで維持されるが，コ
ピー数が染色体と同程度であるため，遺伝子の過剰発現には適していない。シアノバクテリ
アの自律複製領域に関する新たな知見を得るため，ライブラリスクリーニングとシーケンシ
ングを組み合わせた新しい手法，Autonomous Replication Sequencing（AR-seq）を確立した。
スクリーニングのゲノムソースとして，染色体の他に 7 つのプラスミドを持ち，少なくと
も 7 つのレプリコンが共存できる Synechocystis sp. PCC 6803 (S. 6803) を選択した。S. 6803 
から構築されたゲノムライブラリーを形質転換するホストとして，極めて高い形質転換能を
有し，内在性プラスミドを一つしか持たない Synechococcus elongatus PCC 7942 (S. 7942) を
利用した。まず S. 6803 ゲノムライブラリー（ライブラリーA）を S. 7942 へ導入し，得ら
れたコロニーより再び DNA を抽出しライブラリーB とした。S. 7942 染色体へインテグレ
ーションせずに自律複製しているプラスミドを得るために，S. 7942 形質転換体ライブラリ
ーから抽出したDNA を E. coli へ再び導入しコロニーを取得した（ライブラリーC）。次に，
各ライブラリーに含まれるゲノム領域を明らかにするため，網羅的なシーケンス解析を行っ
た。ライブラリーB，C において配列リードを S. 6803 ゲノムにマッピングしたところ，S. 
6803 に含まれるプラスミドである pCC5.2 の ORF B（CyRepA2）と周辺領域が含まれてい
た。S. 7942 における CyRepA2 の複製活性を評価するために，CyRepA2 遺伝子領域を含む
プラスミドベクターに，レポーター遺伝子として  gfpmut2 を配置した発現ベクター
pYS1C-GFP を構築し S. 7942 へと導入した。 gfp 上流には IPTG による制御が可能なプロ
モーターを配置しており，IPTG 依存的な GFP の発現が期待できる。蛍光顕微鏡やウェス
タンブロッティングにより解析したところ，pYS1C-GFP を持つ S. 7942 では IPTG 依存的
に GFP の発現が観察された。染色体に導入した発現系と比較したところ，pYS1C-GFP を
導入した細胞の GFP 発現量は約 10 倍以上であり，CyRepA2 を保有する pYS ベクターを
用いた発現系が過剰発現に適していることが示された。続いて，pYS ベクターのホストレ
ンジを評価するために，系統的に異なるモデルシアノバクテリア S. 6803，Synechococcus sp. 
PCC 7002 ，Anabaena sp. PCC 7120 への導入を試みた。これら 3 種においても S. 7942 と
同様に GFP の蛍光が確認され，pYS ベクターが幅広いシアノバクテリア種で機能する過
剰発現ベクターとして利用できることが示された。 
 
2. pYS ベクターを利用したシアノバクテリアにおける 1,8-シネオールの生産 
 pYS ベクターが有用物質生産に利用可能か検証した。最初のターゲットとして 1,8-シネオ

ールを選択した。1,8-シネオール（C10H18O）は揮発性モノテルペンであり，その心地よい香
りと味から食品添加物，香料，化粧品，洗口液として使用されている。1,8-シネオールは，
主に植物のユーカリなど精油から抽出されており，これらの植物では，ゲラニルピロリン酸
（GPP）からテルピネオール合成酵素（TER）とシネオール合成酵素（CIN）の 2 つの酵素
によって 1,8-シネオールが生成される。近年，Streptomyces clavuligerus ATCC 27064 から発
見されたシネオール合成酵素（CnsA）は，GPP から直接 1,8-シネオールを合成することが
可能であるため，シアノバクテリアの 1,8-シネオール生産に cnsA 遺伝子を用いることとし
た。cnsA 遺伝子を S. 7942 に対してコドンを最適化し，pYS ベクターに導入することで 
pYS1C-CnsA を構築した。pYS1C-CnsA を S. 7942 へ導入し IPTG の存在下または非存在
下で細胞増殖を比較した結果，IPTG 存在下では pYS1C-CnsA 株の増殖は著しく遅延し，
CnsA の過剰発現による細胞毒性が示唆された。次に，培養液のヘッドスペースの揮発性成
分を回収し，CnsA 発現株における 1,8-シネオールの収量を解析した。SPME ファイバーを
用いて揮発性成分を回収しガスクロマトグラフィーで分析したところ CnsA 発現株は，3
時間で菌体湿重量当たり 105.6 μg の 1,8-シネオールが得られた。 
 
3. シアノバクテリアと酵母を利用したストリゴラクトン生産 
 シアノバクテリアにおける物質生産のさらなるターゲットとしてストリゴラクトン（SL）
を選定した。植物ホルモンである SL はアフリカで農業生産上，最も問題となっている根
寄生植物 Striga の発芽刺激物質であり，圃場試験から Striga の駆除に高い効果が認められ
ている。しかし植物からの分泌量は微量であるため，SL を安価かつ大量に生産する系の確
立が求められている。SL は β-カロテンを初発物質として D27，CCD7，CCD8 に触媒され
る反応を経て SL 前駆体カーラクトン（CL）まで葉緑体で合成された後，小胞体において 
P450 酵素である MAX1 ファミリーにより多様な SL アナログが合成される。まず CL 合
成反応を触媒する 3 遺伝子を pYS1C にクローニングし（pYS1C-CL），これを S. 7942 に
導入した（CL 生産株）。CL 生産を LC/MS/MS 解析した結果，CL 生産株は同湿重量にお
ける植物体の根と比較して 500 倍以上の CL を生産しており，CL は培地中に排出されて
いることが確認された。S. 7942 によって合成された CL の生理活性を確認するため，新た
に根寄生植物 Orobanche minor（O. minor）を用いた簡易的なアッセイ系を構築した。CL を
培地に排出されることを利用してシアノバクテリア培養液を寒天で固め，培養液寒天の上に 
O. minor の種子を播種した。その結果 CL 生産株の培養液寒天上でのみ種子の芽が観察さ
れ，S. 7942 由来 CL が生理活性を有することを確認した。CL 生産株において pYS 空ベ
クター導入株と生育を比較した際，IPTG 添加時にわずかな生育遅延が確認された。CL 生
産時にカロテノイドの蓄積量が減少していることを予想し，カロテノイド蓄積量を HPLC 
により解析したところ β-カロテンの下流にあるゼアキサンチンの蓄積量がわずかに減少し

たが，β-カロテン蓄積量およびカロテノイドの総量に変化はなかった。 
 シアノバクテリアを用いた SL 生産系にむけて，ストライガへの高い発芽活性が知られ
ている 4-デオキシオロバンコール（4DO）の合成酵素である MAX1 ホモログ（OsCYP711A2）
の発現および機能化を試みた。CL 生産株への OsCYP711A2 の導入をはじめとして，様々
なエンジニアリングを行ったが，現状ではシアノバクテリア細胞内での OsCYP711A2 の機
能化は成功していない。原核生物と真核生物の P450 は構造が大きく異なるため，真核生物
由来 P450 を原核生物内で機能させることは困難であり，大腸菌やシアノバクテリア内での
成功例はわずかである。そこで単一生物内での合成にこだわらず，OsCYP711A2 による CL 
から 4DO の変換を別の生物で行うことを考えた。真核生物由来の P450 研究は酵母を利用
することが多く，MAX1 ホモログの活性評価にも用いられている。また CL 生産株は CL 
を培地中に排出している。そのため抽出の工程を経ずにシアノバクテリアと酵母の複合培養
により SL を生産できるか検討した。CL 生産株の培養液を OsCYP711A2 発現酵母の培養
液に添加したところ，酵母は細胞内に CL を取り込み，4DO を合成した。生産量は菌体湿
重量 1 g あたり 160.7±18.0 ng であり，イネと比較すると 10,000 倍以上の 4DO 生産量で
あった。さらに本システムで生産した SL が生理活性を有することを，アフリカで実際に
農業被害を与えている種である Striga hermonthica の種子を用いた発芽試験により確認し
た。 
 
【総合討論】 
 本研究ではシアノバクテリアの有用物質生産の実用化に向けて幅広いホストで利用可能
な発現ベクターの開発と新規の有用物質生産を行った。S. 6803 ゲノムライブラリーを用い
た AR-seq 解析により，CyRepA2 が S. 7942 において高コピーで保持されることを明らか
にした。この成果は CyRepA2 領域が異種シアノバクテリアにおいても自律複製活性を有す
ることを示した最初の報告である。さらに今回の結果から，いくつかのシアノバクテリアに
おいて，pYS ベクターが遺伝子過剰発現に利用できることが明らかになった。これらの結
果は，本研究で構築した pYS ベクターが優れた遺伝子工学ツールであることを強く示唆す
るものである。本実験ではモデルシアノバクテリア 4 種について評価を行なってきたが，
pYS の広宿主性を利用することで，今後，非モデルのシアノバクテリアの遺伝子工学研究
にも利用できると期待している。 
 さらに，pYS ベクターを用いて S. 7942 をホストとした 2 種の植物由来テルペノイド
（1,8-シネオール，CL）生産系を構築し，さらに酵母との共培養により高い効率で 4DO を
生産することに成功した。植物由来の生理活性物質には医薬品や香料，染料など有用な物質
が見出されているが，その多くは植物体から回収できる量が限られる。さらに地理的条件や
気候などの環境要因に依存することを考慮すると，シアノバクテリアを用いた生産系は，

CO2 ニュートラルかつ持続的にこれらのテルペノイドを生産するための有用な手段である。
本研究成果によって得られた植物由来テルペノイドの生産量は，まだ実用化レベルに達して
いない。しかし，本研究において構築した 1,8-シネオールの生産，検出系はテルペノイド代
謝フローの評価系として有用であり，テルペノイド代謝の複雑な制御の理解や，CL や 4DO 
の生産量を強化する際の代謝改変にも利用できると考える。また，CL 生産株は依然として
カロテノイドを高蓄積しているため，CL 生産増強の余地が残されている。今後の課題とし
て β-カロテンから CL 生産に関与する酵素を最適化することで，本来のカロテノイド代
謝経路から CL 生産経路への基質の流量を増加させることができると考えている。シアノ
バクテリアと従属栄養生物の共培養研究はすでに報告があるがシアノバクテリアはあくま
でグルコースの供給源に過ぎず，共培養系を植物の生理活性物質生産に応用した研究は未だ
前例がない。また，微生物によって根寄生植物発芽活性を有する CL，SL（4DO）を生産し
た世界で初めての例である。さらに本 SL 生産システムは酵母に導入する MAX1 ホモログ
を変更することで，4DO 以外の SL を生産することも可能である。本研究によって見出さ
れたシアノバクテリア・酵母共培養による植物由来の生理活性物質生産の方法論は，SL 生
産をフラッグシップモデルとして，さらなる構造が複雑な化合物の生産にも応用可能である。 
 
 
審 査 報 告 概 要 
 
本研究では，酸素発生型の光合成を行い CO2 ニュートラルな有用物質生産の宿主として
期待されているシアノバクテリアの遺伝子工学技術をさらに発展させるため，まずシアノバ
クテリア Synechocystis sp. PCC 6803 の内在性プラスミドが有する CyRepA2 が，幅広いシア
ノバクテリア種において自律複製活性を有することを明らかにし，これを利用した広宿主域
発現ベクターpYS を開発した。次に，pYS を用いて Synechococcus elongatus PCC 7942 を宿
主としたテルペノイド生産を試み，香料に利用されている揮発性モノテルペン 1,8-cineole
の生産に成功した。さらに，希少植物ホルモンであるストリゴラクトン（SL）の前駆体カ
ーラクトン（CL）の生産系を構築し，CL 生産株と SL 合成酵素を導入した酵母を共培養す
ることで，植物の 10,000 倍以上の効率で SL 生産することに成功した。これらの CL およ
び SL は根寄生植物に対する生理活性を有しており，本研究はシアノバクテリア物質生産系
の有用性を示した点で高く評価される。これらの研究成果等を詳細に検討した結果，審査員
一同は，学位請求者に博士（農学）の学位を授与する価値があると判断した。

本論文は，世界人口の増加と乾燥地の拡大という食料安全保障において相反する問題が進
展するなか，乾燥地の緑化や農業開発に対する深層地下水利用の可能性を検証することを目
的に，極乾燥地のジブチ共和国を対象に，主に，1)同国における深層地下水の化学的特徴と
分類・分布， 2)涵養域と年代，3)同国沿岸に生育するマングローブの体内水の起源，につ
いて調べた結果をまとめたものである。 
深層地下水の化学的特徴と分類・分布については，同国の全域的な地点において約 50～
200m 深から採取した地下水を対象に測定した各種イオン濃度等を用い，多変量ならびに階
層クラスター解析を行うとともに，GIS マッピングツールを用いた流域解析を行った結果，
内陸部深層地下水の大半は火山岩由来の無機成分を主な組成とし，飲用や農業利用に適して
いることを明らかにした。 
深層地下水の涵養域はδ2H とδ18O，また年代は 3H と 14C 濃度を用い推定した結果，複
数の涵養域に由来する水が混合して深層地下水が形成されており，その年代は，若い地下水
（約 70 年未満）と古い地下水（約 70～3000 年前）が，同一の地下水流域内においても混
在していることを明らかにした。 
ムシャ島とジブチ市沿岸に生育する 2 種のマングローブ（Rhizophora mucronata と
Avicennia marina）体内水の起源を，δ2H とδ18O により推定した結果，ワジが形成され
ていないムシャ島においてもマングローブが，何らかの淡水を利用しており，その給源は深
層地下水である可能性が示された。 
以上のように，ジブチ共和国のように極端な乾燥地であっても，一定のフラックスをもつ
深層地下水が存在すれば，その量に応じ，緑化や農業開発をはじめとする民生の向上に有効
であると考えられた。

審 査 報 告 概 要 
 
本論文は，降水量がきわめて少なく乾燥した地域の緑化や農業開発において，深層淡水地
下水を利用することの可能性を，ジブチ共和国での調査・研究結果をもとに論考したもので
ある。その調査・研究において主に，1）同国深層地下水を無機成分組成，パイパーダイヤ
グラム，ヘキサダイヤグラム，クラスター分析および主成分分析により分類し，その流域別
分布を明らかにし，2）水安定同位体自然存在比，3H および 14C 分析により深層地下水の由
来と年代を推定したとともに，３）降水量が極めて僅少な地域に生育するマングローブ植物
が利用する淡水の起源について論考し，結論的に，深層地下水の利用可能性を評価するため
の一連解析手法の有効性を示すとともに，乾燥地における深層地下水の硝酸態窒素に対する
脆弱性と，乾燥地に生育するマングローブ植物が淡水地下水の存在を示す指標となる可能性
を示した。以上の研究成果等を詳細に検討した結果，審査委員一同は，博士（国際農業開発
学）の学位を授与するに値すると判断した。

Yams (Dioscorea spp.), annual tuberous crops, are an important staple for millions of people in 
tropical and subtropical regions (Raphael et al., 2015). Yams also serve as a major source of 
production in West Africa (Fu et al., 2011; Verter and Becvarova, 2015). However, yam cultivation 
requires high levels of chemical fertilization to maintain productivity, resulting in high production 
costs (Carsky et al., 2010). Biofertilizers (also known as bioinoculants, biocontrol agents, and 
biopesticides) are gaining interest as a sustainable solution to environmental problems caused by the 
inappropriate use of chemical compounds. The most widely used microorganisms are plant 
growth-promoting bacteria (PGPB), which promote plant performance directly and/or indirectly via 
three primary modes of action: increased nutrient accessibility, production of plant growth regulator, 
and increased plant host systemic resistant induction/antagonistic activity (Mohammadi and Sohrabi, 
2012; Expósito et al., 2015; Sathya et al., 2017). However, previous studies on bacterial inoculation 
used as a biofertilizer seem to face similar problems in terms of biofertilizer development, with many 
studies as well as in yam studies reporting significant plant growth-promoting effects under in vitro 
conditions and others reporting deleterious effects or much fewer effects on plant growth and yield 
under field conditions. Therefore, to further address the problems associated with biofertilizer 
development, this study evaluated the effects of bacterial inoculation of water yam with a synthetic 
bacterial community consisting of five dominant PGPB on their growth and native bacterial 
communities using high-throughput 16S rRNA amplicon sequencing. This study also estimated the 
effects of inoculation techniques (soil application and root dipping) and the degrees of interaction 
between four water yam varieties and their native bacterial communities.

1. Growth of Water Yam (Dioscorea alata L.) as Affected by Bacterial Inoculation under 
Growth Chamber Conditions 
Previous studies on water yam plants revealed that the endophytic nitrogen-fixing bacteria 
associated with water yam plants, which 38% of nitrogen were contributed by these bacteria. 
Therefore, this study aimed to evaluate the effects of bacterial inoculation on the growth and source 
of nitrogen in water yam cv. A-19 under growth chamber conditions. The plants were inoculated with 
two different inoculant concentrations (104 and 106 CFU.ml-1) of Agrobacterium sp. strain 343, 
which was previously isolated from the root of water yam cv. A-19, along with a non-inoculated as 
the control treatment. In addition, this bacterial strain was previously evaluated for the plant 
growth-promoting traits related to nitrogenase activity, phosphorus solubilization and production of 
phytohormone (IAA) and siderophores which all were positive. The results showed that most of the 
growth parameters (i.e., the number of leaves, leaf areas, stem length, and dry weights of leaf, stem, 
and tuber) at 90 days after inoculation (DAI) were significantly affected by the inoculation. In 
addition, the nitrogen content in leaves was significantly increased by the inoculation in comparison 
with the control at 60 and 90 DAI, which the natural abundance of stable nitrogen isotopes (δ15N) 
results suggested that 33 to 51% nitrogen in inoculated plants could be derived from the atmosphere 
as the result of the biologically fixed nitrogen provided by the inoculated bacteria. These findings 
suggest the potential of isolated bacterial inoculation on yam production. 
 
2. Replacement of Water Yam (Dioscorea alata L.) Indigenous Root Endophytes and 
Rhizosphere Bacterial Communities via Inoculation with a Synthetic Bacterial Community of 
Dominant Plant-Growth-Promoting Bacteria 
Biofertilizers containing high-density plant-growth-promoting bacteria are gaining interest as a 
sustainable solution to environmental problems caused by eutrophication. However, owing to the 
limitations of current investigative techniques, the selected microorganisms are not always preferred 
by the host plant, preventing recruitment into the native microbiota or failing to induce plant 
growth-promoting effects. To further understand this, five plant-growth-promoting bacteria 
previously isolated from water yam (Dioscorea alata L.) plants and showing a dominant abundance 
of 1% or more in the water yam microbiota were selected for analysis of their 
plant-growth-promoting activities when used as a synthetic bacterial inoculant. The synthetic 
bacterial community consisting of bacterial strains S-60 (Neorhizobium), S-64 (Rhizobium), S-163 
(Enterobacter), S-392 (Stenotrophomonas), and 1615 (Ralstonia) was used for the inoculation. Water 
yam cv. A-19 plants were inoculated twice at 10 and 12 weeks after planting under greenhouse 
conditions. Bacterial communities in the root, rhizosphere and bulk soil samples were characterized

using high-throughput 16S rRNA amplicon sequencing. Compared with non-inoculated plants, all 
bacterial communities were significantly altered by inoculation, mainly at the genus level. The 
inoculation effects were apparently found in the root bacterial communities at 16 weeks after 
planting, with all inoculated genera showing dominance (in the top 35 genera) compared with the 
control samples. However, no significant differences in growth parameters or nitrogen contents were 
observed between treatments. At 20 weeks after planting, the dominance of Stenotrophomonas in the 
inoculated roots decreased, indicating a decline in the inoculation effects. Interestingly, only the 
Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium clade was dominant (>1% relative 
abundance) across all samples, suggesting that bacteria related to this clade are essential core bacteria 
for water yam growth. This study is the first report to show that native bacterial communities in water 
yam plants can be replaced by a synthetic bacterial community, with growth remaining similar 
between the control and inoculated plants. 
 
3. Different Growth and Bacterial Communities of Water Yam (Dioscorea alata L.) as Affected 
by Two Different Inoculation Methods Using a Synthetic Essential Core Bacterial Community 
 
The inoculation techniques are considered as one of the factors that affect the colonization and 
plant growth–promoting effects of inoculants. Therefore, this study aimed to compare the effects of 
two inoculation techniques (soil application and root dipping) using a synthetic bacterial community 
on water yam cv. A-19 growth and its native bacterial communities under growth chamber conditions. 
Five plant growth–promoting bacteria (i.e., S-12, S-17, S-60, S-84, and 3E-17) related to 
Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium clade that were previously identified as 
essential core bacteria for water yam growth were used for synthetic bacterial inoculation. Compared 
with the root dipping technique, inoculation using the soil application technique significantly 
improved plant growth in terms of leaf number, area, and dry weight as well as SPAD value, 
suggesting that the soil application technique is an efficient method for bacterial inoculation in yam 
plants. The metagenomic analysis revealed that the root bacterial communities across all control 
plants were dominated by Actinobacteria followed by Proteobacteria and Firmicutes. However, the 
relative abundance of these phyla was different between inoculation techniques which the relative 
abundance of Actinobacteria, Proteobacteria and Firmicutes were 87.45%, 9.71% and 1.15% in 
control soil application plants, respectively, and 42.94%, 27.01% and 3.66% in control root dipping 
plants, respectively. The inoculation using both techniques significantly altered root bacterial 
communities with decreases in the relative abundance of Actinobacteria and increases in 
Proteobacteria. Higher growth parameters of plants under soil application treatments compared with

plants under root dipping treatments suggest that the soil application can deliver or induce more 
beneficial bacteria into the water yam bacterial communities. Therefore, soil application should be 
used as an inoculation method for yam plants in further study. 
 
4. Effects of a Synthetic Core Essential Bacterial Inoculation on Growth and Native Bacterial 
Community of Four Water Yam (Dioscorea alata L.) Varieties 
To estimate the degrees of interaction between water yam plants and their native bacterial 
communities, four water yam varieties (namely, A-19, A-62, A-64, and A-133) were used in this 
study. Growth and the bacterial communities were observed 90 days after inoculation with a 
synthetic essential core bacterial community consisting of five plant growth-promoting bacteria 
related to Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium clade (i.e., S-12, S-17, S-60, S-84, 
and 3E-17). Growth of four yam varieties responded differently to the inoculation which only 
inoculated A-62 plants had significantly higher growth parameters (i.e., number of leaves, leaf area 
and dry weight of leaf) compared with the control plants. On the other hand, deleterious effects were 
observed other varieties which SPAD values and tuber dry weight decreased in inoculated A-64 and 
A-133 plants, respectively. Bacterial communities of all varieties were identified using a 
high-throughput 16S rRNA amplicon sequencing. Across all non-inoculated plants, root bacterial 
communities were predominated by phyla Actinobacteria (51.85-87.45%), Proteobacteria 
(9.71-34.05%), Firmicutes (1.15-2.66%), and Bacteroidota (0.19-9.64%). The root bacterial 
communities of all varieties were apparently affected by the inoculation, which increases in 
Proteobacteria (from 9.71% to 74.43% in A-19; from 12.37% to 65.00% in A-62; from 34.05% to 
66.06% in A-64; from 27.60% to 56.72% in A-133) and decreases in Actinobacteria (from 87.45% to 
17.91% in A-19; from 83.78% to 18.59% in A-62; from 51.85% to 26.84% in A-64; from 66.49% to 
31.30% in A-133) were observed. Based on Venn diagram analysis, the smallest number of taxa in 
the native bacterial communities were found in A-62 plants (65) followed by A-19 (131), A-133 
(156), and A-64 (214), suggesting the control A-62 plants may have the lowest association with their 
native plant growth-promoting bacteria. The number of taxa in A-62 increased to 244 as well as 
common taxa across all varieties (from 42 to 80) following inoculation. Together, these findings 
suggest that the inoculation increased bacterial diversity and united bacterial communities across all 
varieties. Moreover, the positive effects were found in A-62 plants may be due to a low association 
with their native bacterial communities. 
 
Conclusion 
The finding of this study suggests that the effects of bacterial inoculation in water yam plants can

be affected by growth conditions, plant growth stages, inoculation technique, plant genotype, as well 
as interaction levels between host plants and their native bacterial communities. Among four yam 
varieties, A-62 is a good genotype model for studying the inoculation effects on growth and bacterial 
communities. To further understand and confirm the findings in this study, the inoculation effects of 
a synthetic bacterial community consisting of dominant plant-growth-promoting bacteria should be 
expanded to greenhouse or field conditions with different fertilizations (chemical or organic 
fertilizer). 
 
 
審 査 報 告 概 要 
 
植物生育促進細菌（PGPB）は窒素固定，リンの可溶化，植物ホルモンの産生などにより植
物の生育促進や植物病原菌への拮抗作用を持つ細菌である。本研究はヤムイモの一種である
ダイジョに PGPB を接種し，生育に及ぼす影響並びにメタ 16S 解析による細菌叢の共生状態
について検討した。閉鎖環境においては接種細菌がダイジョの生育を促進したが，温室内で
栽培したダイジョ A-19 系統に接種したところ生育の促進効果は見られなかった。しかし，
接種植物体の細菌叢をメタ 16S 解析で確認したところ接種した細菌が共生することが確認
された。PGPBの混合溶液をダイジョ A-19系統の根に直接浸す方法と土壌に浸透させる方法
で生育を比較し土壌浸透法の効果を明らかにした。また多数のダイジョ系統における PGPB
の接種試験では，生育が促進される A-62 系統を明らかにした。これらの接種細菌とヤムイ
モとの共生メカニズムを明らかにすることが化学肥料に頼らない品種の開発につながると
考えられた。審査員一同は，これらの知見と発見は学術的に価値の高いものと認め，学位請
求者に博士（農学）の学位を授与するに値すると判断した。

論 文 内 容 の 要 旨 
 
コレステロール依存性細胞溶解素（CDC）は，一部のグラム陽性菌が産生する強力な細菌
毒素である。CDC は，典型的なⅡ型シグナルペプチドを有する 50-70 kDa の水溶性単量体と
して細菌細胞から分泌される。この毒素がコレステロールを含む真核細胞膜にさらされると，
毒素分子は膜表面に最大 30～50 個のモノマーを含む大きな円形のホモオリゴマー複合体を
形成し，最終的に脂質二重膜に孔を形成する。 
 Clostridium botulinum は，自然界最強の毒性を示すボツリヌス神経毒素（BoNT）を産生す
る。BoNT の血清学的特異性に基づいて，この細菌は血清学的に 7 つ（A～G 型，BoNT/A～
BoNT/G）に分類される。また，C. botulinum は，その生理学的・生化学的特徴から血清型
とは別に，4 つの生理学的グループ（I-Ⅳ）に分類される。これらの生理学的グループのう
ち，グループⅢ菌株は BoNT/C あるいは BoNT/D を産生し，鳥類や家畜に対してボツリヌ
ス症を引き起こす。 
 C. botulinum が産生するボツリノリジン（BLY）は，CDC に属するチオール活性化溶血毒
である。BLY は，赤血球や培養細胞に対して毒性を示すことが報告されている。しかし，
極めて強い毒性を示す BoNTと比較して，BLYについての研究はほどんど行われておらず，
その病原性発現機構の詳細は不明である。最近，C. botulinum D 型株 4947 株（D-4947）のゲ
ノムに 2 つのボツリノリシン遺伝子（bly1 および bly2）が存在することが報告された。bly1
と bly2 遺伝子の配列類似度は，ヌクレオチドレベルで 78.3％，アミノ酸レベルで 74.6％で
あった。CDC 産生細菌のゲノムを解析したところそのほとんどが，一つの CDC 遺伝子を保
有する。一方，C. botulinum グループⅢに加え，近縁種である C. novyi（ガス壊疽の原因菌）
や C. haemolyticum（細菌性血色素尿症の原因菌）に CDC 遺伝子の重複が生じていた。しか
し，これらの CDC の機能については不明のままである。 
本論文では，Clostridium 属における CDC 遺伝子の重複について着目し，各 CDC の特性

を in silico 解析および組換えタンパク質を使用した機能解析から明らかにすることを目的と
した。本論文において，第 1 章ではグラム陽性菌がもつ CDC のアミノ酸配列を用いて系統
解析を行い，C. botulinum グループⅢとその近縁種由来の CDC の特徴を解析した。第 2 章で
は，C. botulinum グループⅢ由来の 2 つの CDC（BLY1 および BLY2）の細胞毒性について
検証した。第 3 章では，C. novyi が持つ 2 つの CDC ファミリータンパク質（NLY1 および
NLY2）の細胞毒性について検証した。 
 
第 1章 CDC のアミノ酸配列に基づく系統解析 
 Clostridium，Bacillus，Streptococcus などの病原性グラム陽性菌は，組織や細胞への侵入を
補助するために CDC を産生する。ゲノムデータベースを用いた in silico 解析により多くの，
CDC 産生グラム陽性菌は，そのゲノム上に 1 種の CDC 遺伝子をコードしているが，C およ
び D 型 C. botulinum やその近縁種において，CDC の遺伝子重複が生じていることが明らか
にされた。遺伝学的研究により，グループⅢ C. botulinum は，他グループの C. botulinum よ
りも C. novyi および C. haemolyticum に近縁であることが知られている。この章では，CDC
のアミノ酸配列に基づいた系統解析を行い C. botulinum グループⅢと C. novyi および C. 
haemolyticum 由来の CDC について解析した。 
 その結果，C. botulinum グループⅢとその近縁種は，そのゲノム上にそれぞれが遺伝子重
複によって生じたと考えられる 2 つの CDC 遺伝子を有していた。また，C. botulinum グル
ープ III とその近縁種が持つ CDC タンパク質のアミノ酸配列を用いた系統解析により，少な
くとも 4 つのクラスター（α〜δ）に分類された。さらに，それぞれのクラスターは，アミノ
酸配列から予測されるシグナルペプチドの種類や等電点に差異が見られた。このクラスター
を基にすると，C. botulinum グループⅢとその近縁種は，α と δ あるいは β と γ の組み合わ
せで 2 つの CDC 遺伝子を保有していることが明らかとなった。 
 
第 2章 C. botulinum グループⅢ由来 CDC タンパク質 BLY1 および BLY2 の機能解析 
本章では，C. botulinum グループⅢに属する D 型菌 4947 株の CDC タンパク質 BLY1（ク
ラスターα に属する）および BLY2（クラスターδ に属する）細胞溶解活性について検証し
た。予測されたシグナルペプチドを含まない bly1 および bly2 遺伝子をタンパク質発現用プ
ラスミド pET28a にクローニングし，組換えタンパク質発現用大腸菌 BL21（DE3）を用いて
誘導・発現した。組換え BLY（rBLY1 および rBLY2）は，それぞれアフィニティーカラム
およびゲルろ過カラムクロマトグラフィーを用いて SDS-PAGE 上でほぼ単一バンドとして
精製された。ウマ赤血球に精製 rBLY1 および rBLY2 標品を供すると，いずれも溶血活性を
示した。rBLY1 および rBLY2 の HD50（50 % 溶血を与える毒素量）は，それぞれ 0.096 nM
および 0.412 nM であり，rBLY1（α）の方が高い活性を示した。さらに，両者の溶血活性は，

システイン濃度の用量依存的に増加した。したがって，これらのデータは，BLY1 および
BLY2 が共に CDC であることを示している。一方，rBLY2(δ)は，培養細胞（HeLa，IEC-6
および NRK-52E）において，rBLY1（α）よりも有意に低い細胞溶解活性を示した。rBLY1
と rBLY2をアミノ酸配列アラインメント解析から，細胞結合モチーフに変異が認められた。
2 つの CDC の毒性の違いは，その構造の違いによるものと推察された。 
 
第 3章 C. novyi 由来の CDC タンパク質 NLY1 および NLY2 の機能解析 
 C. novyi は，グラム陽性偏性嫌気性菌であり外傷に侵入することで，豚などの家畜および
ヒトにガス壊疽を起こす原因菌である。C. novyi A 型菌は，C. botulinum グループⅢと近縁種
であり，第 1 章の結果からクラスターβ と γ に分類される 2 種の CDC 遺伝子を保有する。
本章では，C. novyi A 型菌 KZ671 株から 2 種の CDC タンパク質 NLY1（β）および NLY２
（γ）をクローニングし，それぞれの組換え体を作製しその毒性を調べた。 
 大腸菌を用いた組換えタンパク質発現系により誘導・発現した組換え NLY（rNLY1 およ
び rNLY2）を精製した結果，rNLY1 は高純度に精製された。rNLY2 は N 末端側の約 10 kDa
が切断されていたがウマ赤血球に対して溶血活性を示した。また，システイン添加によって
rNLY1と rNLY2の溶血活性は共に活性化されたことからCDCに属するチオール活性化溶血
素であることが示唆された。rNLY1 および rNLY2 の溶血活性を比較すると rNLY1（β）が
高い溶血活性を示した。rNLY2（γ）は，培養細胞（HeLa，IEC-6 および NRK-52E）におい
て，rNLY1（β）よりも細胞溶解活性が有意に低かった。rNLY1 と rNLY2 をアミノ酸配列ア
ラインメント解析から，細胞結合モチーフに変異が認められた。2 つの CDC の毒性の違い
は，その構造の違いによるものと推察された。 
 
総 括 
著者は，本研究において，C. botulinum グループⅢやその近縁種が持つ CDC は，少なくと
も 4 つのクラスター（α〜δ）に分類されることを示した。それぞれのクラスターに分類され
る CDC タンパク質を組換えタンパク質として作製しその毒性を比較した結果，C. botulinum
グループⅢおよびその近縁種は，異なる細胞溶解活性を示す 2 種の CDC 遺伝子を有するこ
とを示した。Clostridium 属は，家畜ボツリヌス症やガス壊疽など家畜における感染症等の原
因となることがあり，これらの菌が産生する毒素の性状解明は，それらの感染症の予防や治
療につながる。さらに，細胞溶解素は害虫への殺虫剤として，またガン細胞など特定の細胞
を除去する薬剤として活用する研究が行われており，本研究の成果は，農業や医療へと応用
できる重要な知見であると期待される。

審 査 報 告 概 要 
 
 ボツリヌス菌は，自然界で最も高い毒性を示すボツリヌス神経毒素（BoNT）以外に複数の
毒素タンパク質を産生するが，BoNT以外の毒素については，実験室レベルで毒性が検証さ
れているのみで，その生理的意義，構造や機能についてはほぼ不明のままである。本研究は，
それらの毒素のうち，コレステロール依存性細胞溶解素（CDC）を対象としており，特にボ
ツリヌス菌を含む一部の病原性クロストリジウム属だけが2種のCDC遺伝子を有することに
着目した研究を行なっている。細谷氏は，これらの CDCタンパク質が，そのアミノ酸配列か
ら少なくとも 4つのクラスターに分類されることを示し，それぞれが，異なるシグナルペプ
チドを有すること，等電点などのタンパク質化学的性質が異なることを明らかにした。また，
それぞれの菌は，毒性の強い CDCと弱い CDCをコードする遺伝子を有していることを明らか
にした。ボツリヌス菌を含む病原性クロストリジウム属は，複数の毒素を産生しており，家
畜やペットなどの動物において疾患を引き起こす。本研究の成果は，これらの疾患の発生メ
カニズムの解明に貢献するものであると考えられ，これらの研究成果等を総合的に評価した
結果，審査委員一同は博士（生物産業学）の学位を授与する価値があると判断した。

第１章 序論 
メタボリックシンドローム（以下，MetS）の有病率増加は世界的な問題となっており，日
本でも40～74歳の3人に1人がMetSが強く疑われる者または予備群と推定されている。MetS
は糖尿病や心血管疾患などの重大な疾患に繋がるため予防が重要であり，容易に日常生活に
取り入れることができる簡便な予防方法の提示が望まれる。 
MetS を予防するためのアプローチの 1 つとして食生活の改善が挙げられる。健康な食生
活において，ビタミンやミネラル，食物繊維，ファイトケミカルなどを豊富に含む野菜の摂
取は欠かせない要素の 1つである。これまでに複数の疫学研究によって，野菜と果物が豊富
な食事の摂取が MetS のリスクと負の相関を示すことが報告されている。 
トマトは世界での生産量が多い野菜の 1つであり，生で食される他，調理して食されるこ
とも多く，地中海食といった伝統料理を通しても古くから摂取されてきた。また，飲料，調
味料，サプリメントなどの加工品を通しても摂取されており，世界的に最も多く摂取される
野菜の 1つである。トマトに多く含まれる赤色のカロテノイド，リコペンは，強い一重項酸
素消去能を持つことから，体内で発生する活性酸素や酸化ストレスの消去に働くことが期待
され，ヒトの健康に与える影響を調査した一次研究が数多く報告されている。 
近年，トマトやリコペンの摂取が MetS の予防に有効であるかを検証したシステマティッ
ク・レビュー（以下，SR）が複数報告されており，MetS の診断基準に用いられるバイオマ
ーカーである血中中性脂肪濃度，血中 HDL-コレステロール（以下，HDL-c）濃度，収縮期血
圧，拡張期血圧，空腹時血糖値に与える影響について評価した SR も報告されている。しか
し，これらの先行研究は，文献検索を実施する際の言語が英語，その他のゲルマン語/ロマ
ンス語に限定されているものがほとんどであることに加えて，文献検索に使用された文献デ
ータベースの数が 1～4つと少なく，文献検索の網羅性に課題があると考えられた。

そこで，本研究では，リコペンの経口摂取が MetS 改善作用を持つか否かを明らかにする
ために，リコペンの経口摂取が MetS の診断基準に用いられるバイオマーカーに与える影響
を，先行研究と比較してより網羅的な文献検索（言語による制限を行わず 15 のデータベー
スを使用した文献検索）を行った上で，メタアナリシスを含む SRによって厳格に評価した。 
 
第２章 ウエスト周囲径に対する有効性 
目的：ウエスト周囲径は，内臓脂肪面積 100 cm2以上相当を目安に設定されており，MetS
か否かを診断する上での第一段階の基準であることから最初に着目すべき項目である。本章
では，リコペンの摂取がウエスト周囲径に与える影響についてメタアナリシスを含む SR で
評価した。 
方法：本研究のプロトコールは International prospective register of systematic 
reviews への事前登録を行った（登録番号 CRD42018104595）。研究の適格基準は，参加者：
18 歳以上の者，介入：リコペンを含む試験食品の摂取，比較対照：リコペンを含まない，
リコペンが低濃度の試験食品の摂取，または何も介入を行わない場合，評価項目：ウエスト
周囲径，研究デザイン：ランダム化，準ランダム化，非ランダム化の並行群間比較試験およ
びクロスオーバー試験とした。15 のデータベースを用いて言語による制限を行わずに文献
検索を実施し，独立した 2名以上で適格基準に合致する文献の抽出，構造化抄録の作成，デ
ータの抽出，文献の質評価を行った。統計分析ソフトウェア「Review Manager（Windows 版，
Ver. 5.3.5）」を用いてメタアナリシスを実施した。各研究における効果の大きさのばらつ
きを考慮した変量効果モデルを用いて標準化平均差（以下，SMD）を統合した。メタアナリ
シスは事前に 6つの感度分析（①研究デザインがランダム化並行群間比較試験かそれ以外か
で分けた解析，②サンプルサイズが極端に大きい研究を除いた解析，③試験食品の形態がサ
プリメントタイプかそれ以外かで分けた解析，④試験食品の摂取期間の長短で分割した解析，
⑤リコペン摂取用量の多寡で分割した解析，⑥試験参加者が健康な人か疾病を有する人かで
分けた解析）を設定したが，②⑤は実施しなかった。また，ファンネルプロットを作成し出
版バイアスの有無を判断した。 
結果：適格基準に合致する文献が 4編（5 trial arms）抽出され，全て中程度のバイアス
リスクと判断された。これらの文献でメタアナリシスを行った結果，試験参加者人数の合計
は 134名で，対照群と比較してリコペン摂取群でウエスト周囲径が有意に低下していた（SMD 
= -0.36 [95% CI: -0.70, -0.01], p = 0.04）。ファンネルプロットから出版バイアスは小
さいことが示唆された。感度分析では，試験食品の形態がサプリメントタイプの 2編（リコ
ペン摂取用量が 10 mg/日，15 mg/日），試験食品の摂取期間が 4週間より長い 4 trial arms
（摂取期間はいずれも 8週間）で実施したメタアナリシスで，対照群と比較してリコペン摂
取群でウエスト周囲径が有意に低下していた。

結論：リコペンの摂取がウエスト周囲径の低下に有効であることが示唆された。また，リ
コペンを 10～15 mg/日，8 週間摂取することでウエスト周囲径の低下が期待できると考えら
れた。 
 
第３章 血中中性脂肪濃度，血中 HDL-c濃度に対する有効性 
目的：血中中性脂肪濃度が高い状態と血中 HDL-c 濃度が低い状態は，MetS のリスク因子
であるだけでなく，心血管疾患のリスク因子でもある。本章では，リコペンの摂取が血中中
性脂肪濃度，血中 HDL-c濃度に与える影響についてメタアナリシスを含む SRで評価した。 
方法：本研究のプロトコールは International prospective register of systematic 
reviews への事前登録を行った（登録番号 CRD42018104595）。研究の適格基準は，評価項目
を血中中性脂肪濃度，血中 HDL-c濃度とする以外は，第 2章と同様とした。また，文献検索，
文献の抽出，構造化抄録の作成，データの抽出，文献の質評価，メタアナリシス，出版バイ
アスの判断も第 2章と同様に実施した。 
結果：適格基準に合致する文献が 22 編抽出され，19 編が中程度の，2 編が低い，1 編が
高いバイアスリスクと判断された。データが入手できなかった 6 編を除き，16 編をメタア
ナリシスに使用した。血中中性脂肪濃度は，15編（17 trial arms）でメタアナリシスを行
った結果，試験参加者人数の合計は 996名で，対照群とリコペン摂取群とで有意差は見られ
なかった（SMD = -0.01 [95% CI: -0.14, 0.12], p = 0.86）。ファンネルプロットから出版
バイアスは小さいことが示唆された。また，いずれの感度分析においても有意差は見られな
かった。血中 HDL-c 濃度は，15編（18 trial arms）でメタアナリシスを行った結果，試験
参加者人数の合計は 923名で，対照群と比較してリコペン摂取群で血中 HDL-c濃度が有意に
上昇していた（SMD = 0.22 [95% CI: 0.03, 0.41], p = 0.03）。ファンネルプロットから出
版バイアスは小さいことが示唆された。感度分析では，研究デザインがランダム化並行群間
比較試験以外の 5編，試験参加者の属性が疾病を有する人の 12 trial arms で実施したメタ
アナリシスで，対照群と比較してリコペン摂取群で血中 HDL-c濃度が有意に上昇していた。 
結論：リコペン摂取が血中中性脂肪濃度に与える影響は小さいと考えられた。リコペンの
摂取が血中 HDL-c濃度の上昇に有効であることが示唆された。また，血中 HDL-c濃度が低い
人に対しての方がリコペン摂取の効果が見られやすいことが示唆された。 
 
第４章 収縮期血圧，拡張期血圧に対する有効性 
目的：血圧が高い状態は，MetS のリスク因子であるだけでなく，冠動脈疾患や脳卒中の
リスク因子でもある。本章では，リコペンの摂取が収縮期血圧，拡張期血圧に与える影響に
ついてメタアナリシスを含む SRで評価した。 
方法：本研究のプロトコールは International prospective register of systematic

reviews への事前登録を行った（登録番号 CRD42018104595）。研究の適格基準は，評価項目
を収縮期血圧，拡張期血圧とする以外は，第 2章と同様とした。また，文献検索，文献の抽
出，構造化抄録の作成，データの抽出，文献の質評価，メタアナリシス，出版バイアスの判
断も第 2章と同様に実施した。 
結果：適格基準に合致する文献が 11 編抽出され，9編が中程度の，2編が低いバイアスリ
スクと判断された。データが入手できなかった 3編を除き，8編をメタアナリシスに使用し
た。収縮期血圧は，8編（9 trial arms）でメタアナリシスを行った結果，試験参加者人数
の合計は 612名で，対照群とリコペン摂取群とで有意差は見られなかった（SMD = -0.26 [95% 
CI: -0.56, 0.03], p = 0.08）。ファンネルプロットから出版バイアスは小さいことが示唆
された。感度分析では，研究デザインがランダム化並行群間比較試験以外の 3 編，試験参加
者の属性が疾病を有する人の 5 trial arms，事後に設定した試験参加者の属性が高血圧者
の 3 trial arms で実施したメタアナリシスで，対照群と比較してリコペン摂取群で収縮期
血圧が有意に低下していた。拡張期血圧は，7編（8 trial arms）でメタアナリシスを行っ
た結果，試験参加者人数の合計は 496名で，対照群とリコペン摂取群とで有意差は見られな
かった（SMD = 0.01 [95% CI: -0.24, 0.25], p = 0.94）。ファンネルプロットから出版バ
イアスは小さいことが示唆された。感度分析では，事後に設定した試験参加者の属性が高血
圧者の 3 trial arms で実施したメタアナリシスで，対照群と比較してリコペン摂取群で拡
張期血圧が有意に低下していた。 
結論：血圧が高い人に対して，リコペン摂取が収縮期血圧，拡張期血圧の低下に有効であ
ることが示唆された。 
 
第５章 空腹時血糖値に対する有効性 
目的：空腹時血糖値が高い状態は，MetS のリスク因子であると同時に MetS の下流にある
2 型糖尿病の特徴でもある。本章では，リコペンの摂取が空腹時血糖値に与える影響につい
てメタアナリシスを含む SRで評価した。 
方法：本研究のプロトコールは International prospective register of systematic 
reviews への事前登録を行った（登録番号 CRD42018104595）。研究の適格基準は，評価項目
を空腹時血糖値とする以外は，第 2章と同様とした。また，文献検索，文献の抽出，構造化
抄録の作成，データの抽出，文献の質評価，メタアナリシス，出版バイアスの判断も第 2
章と同様に実施した。 
結果：適格基準に合致する文献が 15 編抽出され，13 編が中程度の，1 編が低い，1 編が
高いバイアスリスクと判断された。データが入手できなかった 5編を除き，10編（11 trial 
arms）をメタアナリシスに使用した。これらの文献でメタアナリシスを行った結果，試験参
加者人数の合計は 750名で，対照群と比較してリコペン摂取群で空腹時血糖値が低下する傾

向が見られた（SMD = -0.15 [95% CI: -0.31, 0.00], p = 0.05）。ファンネルプロットから
出版バイアスは小さいことが示唆された。感度分析では，研究デザインがランダム化並行群
間比較試験の 8 trial arms，事後に設定した試験参加者の属性が 2 型糖尿病者の 2 編で実
施したメタアナリシスで，対照群と比較してリコペン摂取群で空腹時血糖値が有意に低下し
ていた。 
結論：リコペンの摂取が空腹時血糖値の低下に有効である可能性が示唆された。特に，2
型糖尿病者など血糖値のコントロールが悪い人に対して，リコペン摂取が空腹時血糖値の低
下に有効であることが示唆された。 
 
第６章 リコペン摂取の安全性 
リコペンの安全性については，国立研究開発法人医薬基盤・健康・栄養研究所，アメリカ
国立衛生研究所などに情報が纏められている。これらによると，普通の食品や食事に含まれ
る量であれば妊娠中・授乳中を含めて安全であると思われること，サプリメントで摂取した
場合には 15～45 mg 日の用量で 6ヵ月間は安全に使用されているが，妊娠中・授乳中につい
ては安全性に関して信頼できる十分な情報が見当たらないことが提示されている。また，本
研究で適格基準に合致した文献の範囲では，リコペン摂取によって重篤な有害事象が発現し
たとの報告は無かった。 
本研究でメタアナリシスに含めた文献におけるリコペン摂取量は 6～50 mg/日の範囲であ
り，特に 7～30 mg/日の文献が多かった。リコペン摂取量が報告されていない文献がありリ
コペンの多寡での感度分析を行えていないため，効果を発揮するのに必要なリコペン摂取量
を推定することが難しいが，7～30 mg/日の文献が多かったことから，30 mg/日以下の量で
上記のバイオマーカーの改善効果が期待できるものと推察される。例えば，市販のトマトジ
ュースには 15～60 mg/200mL 程度の，生トマトには数 mg/100g 程度のリコペンが含まれる。
そのため，通常の食品から十分に摂取できる量で効果を期待できると考えられるが，サプリ
メントから摂取する場合には過剰摂取にならないように注意する必要があると思われる。 
 
第７章 終章 
MetS は放置すると重大な疾患に繋がっていくため予防が重要であり，日常生活の中に容
易に取り入れることができる簡便な予防方法が望まれる。本研究では，メタアナリシスを含
む SRによって，リコペンの摂取が MetS の診断基準に用いられるバイオマーカーに与える影
響を評価した。言語での制限をせずに 15 の文献データベースを用いて網羅的な文献検索を
行ったところ，先行研究で見落とされていた文献を抽出することができ，リコペン摂取によ
るウエスト周囲径の低下，血中 HDL-c濃度の上昇，空腹時血糖値の低下など新規の知見を得
ることができた。従って，リコペンの経口摂取には一定の MetS 改善作用が期待できること，

特に，バイオマーカーの数値の悪い人に対してより効果が見られやすいことを明らかにした。
MetS の予防方法の 1 つとして適切な食事が挙げられるが，トマトのように世界的に生産量
が多く簡便に入手できる野菜の摂取が MetS 改善に繋がる可能性を示した本研究成果は，
MetS予防推進の一助になると考える。 
本研究では明らかにすることができなかったが，どの程度の用量を摂取すると効果が発揮
されるか，どのような摂取方法がより有効であるか，といった点が明確にできると，より分
かりやすく役に立つ MetS 予防方法にできると考える。 
 
 
審 査 報 告 概 要 
 
メタボリックシンドローム（MetS）の有病率増加は世界的な問題となっている。本研究は，
トマトに豊富に含まれるリコペンの経口摂取が MetS の診断基準に用いられるバイオマーカ
ーに与える影響を，メタアナリシスを含むシステマティック・レビュー（SR）によって厳格
に評価した。リコペンがウエスト周囲径の低下，血中 HDLコレステロール濃度の上昇，空腹
時血糖値の低下に有効であることを初めて明らかにした。また，収縮期血圧，拡張期血圧，
空腹時血糖値に対しては，数値が悪い人においてリコペンによる改善効果が高いことの科学
的根拠も明らかにした。トマトは世界的に生産量が多く，入手しやすい野菜の 1 つであり，
生鮮トマトやその加工品を通したリコペン摂取は日常生活の中で比較的容易にできる MetS
対策になりうることを示した。 
これらの研究成果等を詳細に検討した結果，審査員一同は博士（環境共生学）の学位を授
与する価値があると判断した。
