Mycoplasmosis is one of the most important porcine
diseases. It has a great impact on the productivity and
the economic performance in swine industry world-
wide including Thailand where the pig production
system has been becoming more and more industrial-
ized. ‘Industrialized’ means ‘intensive management’
in which numerous pigs are reared in confined air
space. In these environments chronic diseases includ-
ing respiratory diseases, diarrhea and arthritis are
most common in every country. Recently catastrophic
disease called PRDC (porcine respiratory disease com-
plex) prevails all over the world. Among various eti-
ological agents which are claimed to be associated with
the disease Mycoplasma hyopneumoniae is considered
to play an essential role together with PRRS (porcine
reproductive and respiratory syndrome) virus. It not
only causes pneumonia (mycoplasmal pneumonia of
swine : MPS) by itself but also aggravates infections
caused by other agents especially PRRS virus infec-
tion, which escalates the economic losses induced by
PRDC. Mycoplasma hyosynoviae causes nonpurulent
polyarthritis in breeding and fattening pigs. The
disease is also prevailed all over the world. Little is
known on porcine mycoplasmas in Thailand because of
their difficulties of isolation and cultivation of the
organisms. The present investigations were carried
out to get information which will improve the diag-
nostic techniques of porcine mycoplasmosis and con-
tribute to the control of the diseases in that country.
In the first chapter antimicrobial susceptibilities ofM.
hyopneumoniae were investigated since it is an im-
portant etiological agent of not only MPS but also
PRDC. For isolates with resistance to macrolides, the
first choice drugs in chemotherapy for mycoplasmosis,
mechanisms of the resistance were examined by se-
quencing 23S rRNA of resistant isolates. In chapter 2
antimicrobial susceptibilities of M. hyosynoviae were
also investigated and in vitro development of macro-
lide and lincomycin resistance was examined to elu-
cidate the mechanisms of the resistance using type
strain S16 of this organism. Since early medication is
required for the effective chemotherapy of arthritis
due toM. hyosynoviae, rapid detection of this organism
was developed in chapter 3. Finally methods to see
the genetic diversity of field isolates ofM. hyosynoviae
in Thailand were evaluated from the perspective of
getting epidemiological information of the disease.
1. Antimicrobial susceptibilities ofMycoplasma
hyopneumoniae field isolates and occurrence of
enroflaxacin, macrolides and lincomycin resist-
ance
To assess the in vitro susceptibilities ofM. hyopneu-
moniae to antimicrobial agents is inevitable for the
chemotherapy of MPS. In this chapter a total of 159
Thai isolates of M. hyopneumoniae derived from

pneumonic lungs of pigs in 41 farms during 2006-2011
were subjected for susceptibility testing against 12
antimicrobial agents widely used in Thailand ; chlor-
tetracycline (CTC), oxytetracycline (OTC), doxycycline
(DOXY), lincomycin (LCM), josamycin (JM), kitasamy-
cin (KT), spiramycin (SPM), tylosin (TS), erythromy-
cin (EM), tiamulin (TM), florfenicol (FFC) and en-
rofloxacin (ERFX). The type strain J of M. hyopneu-
moniae was also included in the test as the control
strain. Obtained data were compared with those for
27 isolates collected during 1997-1998.
Tiamulin showed the lowest minimal inhibitory
concentrations (MICs) among drugs tested ranged
between 0.013 and 0.2mg/ml and its MIC90 (MIC at
which 90% of the isolates tested were inhibited) was
0.1mg/ml. Next to TM, TS and LCM showed high
activities with MIC90 of 0.39mg/ml, however, two iso-
lates were resistant to these drugs showing a MIC
value of higher than 12.5mg/ml. Three macrolides,
SPM, JM and KT also showed high potency with
MIC90 of 0.78, 0.78 and 1.56mg/ml, respectively. Eryth-
romycin showed exceptionally low activity among five
macrolides tested with MIC50 of 50mg/ml. Florfenicol
showed moderately high activity with MIC90 of 1.56
mg/ml. Both OTC and DOXY showed similar activities
with FFC and their MIC range distributed from 0.39
to 12.5mg/ml with MIC90 of 6.25mg/ml. On the other
hand, activity of CTC to Thai isolates of M. hyopneu-
moniaewas low with MIC range from 3.12 to 100mg/ml,
MIC90 of 50mg/ml and MIC50 of 25mg/ml. The MICs of
ERFX distributed in a broad range from 0.1 to 25mg/ml
and those for 76 of 159 (47.7%) Thai isolates exceeded 2
mg/ml which is the breakpoint of this drug.
The comparison of susceptibilities of the present 159
isolates with those of isolates obtained from 1997 to
1998 in Thailand revealed that susceptibilities of the
present isolates to CTC, OTC, JM and TS were de-
creased considerably. The MIC90 for previous isolates
were 6.25, 0.78, 0.2 and 0.1, whereas those for present
isolates were 50, 6.25, 0.78 and 0.39mg/ml, respectively,
showing 4- to 8-fold higher MIC90. No change was ob-
served in MIC90 of LCM and TM between the present
isolates and the previous ones. The sequences of do-
main V of 23S rRNA of the two macrolides and LCM
resistant isolates were compared with those of M.
hyopneumoniae strain J and strain ATCC 27719 (Gen-
bank accession number AE017243 and X68421, respec-
tively). The result revealed a point mutation at
A2058G (E. coli coordinates) in both macrolides and
LCM resistant isolates.
2. Antimicrobial susceptibilities ofMycoplasma
hyosynoviae field isolates in Thailand during
2008-2011 and in vitro development of resistance
to tylosin and lincomycin in type strain S16 ofM.
hyosynoviae
No information is available on antimicrobial suscep-
tibilities of field isolates of M. hyosynoviae in Thai-
land. In this chapter 41 Thai isolates of M. hyosyno-
viae derived from tonsils (29), lungs (10) and joint
fluids (2) of pigs from 9 farms during 2008-2011 were
investigated for their in vitro susceptibilities against
10 antimicrobial agents widely used in Thailand,
namely OTC, DOXY, LCM, JM, KT, SPM, TS, TM,
FFC and ERFX. The type strain S16 ofM. hyosynoviae
was also included in the test. All isolates and type
strain S16 were examined for 23S rRNA transitions at
domain II and V which relate to macrolide resistance
and compared with MIC levels of macrolides and
lincomycin for each strain. In vitro development of
macrolides and lincomycin resistance in type strain
S16 of M. hyosynoviae were also investigated to
analyze the mechanisms of macrolides and lincomycin
resistance.
Of 10 antimicrobials tested, TM showed the lowest
MICs of 0.016-0.125mg/ml against all M . hyosynoviae
field isolates. Next to TM, LCM and JM showed high
activities with MIC90 of 1mg/ml. Both KT and FFC
showed moderately high activities with MIC90 of 2-4
mg/ml, whereas SPM and TS showed lower activities
with MIC90 of 8mg/ml. Activities of tetracyclines to
Thai isolates ofM. hyosynoviae were low and MICs for
OTC ranged from 16 to 128mg/ml and those for DOXY
from 4 to 16mg/ml. The MIC range of ERFX was
between 0.25 to 2mg/ml. Of the Thai isolates of M.
hyosynoviae considerable portion was resistant to
some of the drugs ; 41 of 41 for OTC (break point ; ≧16
mg/ml) , 15 of 41 (36.6%) for ERFX (≧2mg/ml) and 13 of
41 (31.7%) for TS (≧4mg/ml).
An acquired G745A transition in domain II was

found in 13 isolates by sequence analysis of domain II
and V of 23S rRNA of all Thai isolates and type strain
S16 of M. hyosynoviae. The MICs of TS were of 4-8
mg/ml for these 13 isolates and 0.5-2.0mg/ml for re-
maining 28 isolates, suggesting strongly that this
G745A transition inM. hyosynoviae may confer a mild
resistance to TS. MIC values of other macrolides and
LCM were not correlated to this transition.
Type strain S16 of M. hyosynoviae was highly sus-
ceptible to both TS and LCM with the MICs of 0.032
and 0.125mg/ml respectively. In vitro development of
resistance to TS and LCM were investigated by pas-
saging this strain in broth medium containing various
concentrations of TS or LCM. At the first and second
passage, the highest concentration of TS and LCM
that allowed the growth of S16 was the same as that
for parent strain. From the third passage, the strain
began to get resistance to TS or LCM rapidly and could
grow at about 4 to 32 times higher concentrations in
each passage. At the sixth passage the highest con-
centration of the drug that S16 mutant strain could
grow was 4,000 or 1,000 times higher for TS or LCM
respectively. In LCM selection, the A2058C/G or
A2059C transitions were obtained in S16 mutants.
This mutant was resistant not only to LCM but also to
all macrolide drugs tested. In TS selection the A2062G
transition was obtained which conferred the resist-
ance to macrolides but not to LCM.
3. Development of semi-nested PCR for detec-
tion of 16S rRNA gene of Mycoplasma hyosyno-
viae
Successful treatment of arthritis due toM. hyosyno-
viae in pigs is best achieved by injecting effective
antimicrobials as early as possible. It needs several
days, however, to diagnose the disease since the iso-
lation of mycoplasmas is time-consuming. In this
chapter semi-nested PCR to detect M. hyosynoviae
using three oligonucleotide primers was developed for
the rapid diagnosis of the disease. The primers were
designed based on 16S rRNA gene of M. hyosynoviae
accession number U26730 from the three positions.
The position of a forward primer was at 196nt to 219nt
whereas the positions of an outer reverse primer and
an inner reverse primer were at 825nt to 844nt and
468nt to 491nt, respectively. The expected sizes of the
PCR products from the first and the second round of
the semi-nested PCR were 649bp and 295bp, respec-
tively. Using the adequate concentration of the M.
hyosynoviae DNA, the first round of semi-nested PCR
could generate an amplified fragment about 649bp
followed by an amplified fragment about 295bp for the
second round of semi-nested PCR. The specificity of
the test was examined with porcine mycoplasmal and
bacterial species commonly encountered in pneumonic
and/or arthritic lesions in pigs. The organisms tested
were as follows : Mycoplasma hyopneumoniae, M. hy-
orhinis, M. flocculare, M. arginini, M. bovigenitalium,
Arcanobacterium pyogenes, Actinobacillus pleuropneu-
moniae, Bordetella bronchiseptica, Escherichia coli,
Haemophilus parasuis, Klebsiella pneumoniae Pas-
teurella multocida Staphylococcus aureus and Strep-
tococcus suis. None of the primer pairs yielded PCR
products or non-specific bands with DNA from those
mycoplasmal and bacterial species. The semi-nested
PCR developed here detected as little as 10−14g of
purified M. hyosynoviae DNA in a reaction and the
limit of detection for clinical materials was at least
103 CFU (colony forming unit) per gram of the lung
samples. To evaluate the applicability of this method
a total of 300 tonsillar samples were collected at a
slaughterhouse and compared the detection rate with
that by cultivation. As a result 45 of 300 (15%) sam-
ples were positive by semi-nested PCR while 17 of 300
(5.7%) were positive by cultivation. Thus the semi-
nested PCR was shown to be a useful tool for rapid
detection ofM. hyosynoviae in clinical materials in pig
herds.
4. Genetic diversity ofMycoplasma hyosynoviae
field isolates in Thailand
It is essential to know the epidemiology of the dis-
ease such as source of infection or mode of transmis-
sion for the establishment of control measures for
arthritis due to M. hyosynoviae. Recently genetic het-
erogeneity of causal organisms has been widely util-
ized to get epidemiological information in various in-
fectious diseases. In this chapter genetic charac-
terization of field isolates of M. hyosynoviae was in-
vestigated by pulsed-field gel electrophoresis (PFGE)

and random amplified polymorphic DNA (RAPD)
analyses. Mycoplasma hyosynoviae strains used were
the same as those in chapter 2. Well-separated DNA
fragments were obtained from 37 of 41 Thai isolates
and type strain S16 with PFGE. Four Thai isolates
did not yield clear bands. The banding patterns of the
typable strains consisted of 6-12 fragments in the size
range of 2-48kbp. Twenty-two different patterns were
detected among these strains. Among isolates derived
from the same farm, two to five different profiles were
also detected in addition to the same profiles. Though
different profiles were usually detected among isolates
derived from different farms, one common profile was
detected in two isolates from two different farms on an
occasion. These two farms were located in the same
province, suggesting ‘farm to farm’ transmission of
these isolates with the same profile. The PFGE tech-
nique had a high reproducibility with identical band-
ing patterns for replicate samples. Thus it revealed
that M. hyosynoviae had high genetic heterogeneity
both within a pig farm and among the farms by PFGE.
On the other hand it was suggested that PFGE might
be a useful tool for the epidemiological studies of M.
hyosynoviae infection from the fact that common pro-
file was detected in each isolate derived from different
farm in one occasion.
The RAPD analyses were carried out after various
preliminary experiments. The clear patterns were ob-
tained by using RAPD primer 6 and RAPD beads
(Ready-To-Go RAPD Analysis kit, GE healthcare, Life
Science, USA). The 41 Thai isolates and type strain
S16 were all typed with banding patterns consisting of
1-12 fragments in size range of 200-1,500bp. Thirty-
nine different patterns were detected among 42 strains
and two isolates with the same profile by PFGE
derived from different farms showed different pattern
by RAPD. On the other hand, the same banding pat-
terns were not necessarily obtained by repeated
analyses of the same strains. Due to this low repro-
ducibility RAPD analyses were not considered appro-
priate to obtain epidemiological information of the
disease despite its high discriminatory power.
The results on antimicrobial susceptibilities of M.
hyopneumoniae described in this work have great
importance for controlling not only MPS but also
PRDC in whichM. hyopneumoniae plays an important
role. Rapid detection of M. hyosynoviae enables the
rapid diagnosis and earlier medication of arthritis due
to M. hyosynoviae and improves the treatment to-
gether with the results of antimicrobial susceptibil-
ities. The elucidation of the mechanisms of macrolide
and LCM resistances ofM. hyosynoviae will contribute
to establish the prudent use of antimicrobials in ani-
mal husbandry. Lastly, PFGE analyses ofM. hyosyno-
viae isolates in Thailand will facilitate the molecular
epidemiological study of animal mycoplasmosis in that
country.

マイコプラズマを原因とする肺炎や関節炎は養豚の生
産性を阻害する難治性疾病として重要である。本研究
は，肺炎起因菌であるMycoplasma hyopneumoniaeお
よび関節炎起因菌であるM. hyosynoviaeのタイ国分離
株には，マイコプラズマに対する第 1次選択薬であるマ
クロライド系薬剤のみならず，第 2次選択薬であるエン
ロフロキサシン耐性菌が増加していることを明らかにし
た。またマクロライド系薬剤に対する耐性機構は菌種に
より異なること，さらにM. hyosynoviaeでは抗菌剤の
存在により急激に耐性化することを明らかにした。これ
らの成績は，畜産領域における抗菌剤の慎重使用の確立
に貢献すると考えられる。またM. hyosynoviaeの迅速
検出法を考案したことは，本マイコプラズマによる関節
炎の早期診断と早期の薬剤投与を可能とし，薬剤感受性
成績とあいまって治療成績を向上させることができる。
さらにM. hyosynoviae分離株のパルスフィールド電気
泳動法による解析は，該菌の遺伝的多様性を明らかにす
るとともに，本方法が分子疫学調査に利用できることを
示した。
よって，審査員一同は博士（畜産学）の学位を授与す
る価値があると判断した。

1． 研究の背景と目的
卵殻の役割を人工容器に代替させた鳥類胚の培養は，
遺伝子導入の効率化，希少鳥類の保護・増殖に貢献する
ものであるが，未だ方法が確立されていない。鳥類の卵
は卵殻の中に栄養物質である卵黄や卵白を大量に含んで
おり，卵殻内において胚発生の大部分が進行する。卵殻
は主に炭酸カルシウム（98.43%），炭酸マグネシウム
（0.84%），リン酸カルシウム（0.73%）から構成され，
胚発生においてミネラル供給源，ガス調整機能および培
養容器としての 3つの機能を有している。このため，胚
の培養に用いる容器はこれらの機能を満たす必要があ
る。Perry（1988）の代理卵殻法は初めて本来の卵殻外
でのニワトリ胚の培養を可能にした。この培養法は，
System I（1細胞期から胚盤葉期），System II（胚盤葉
期から培養 72時間迄）および System III（培養 72時
間以降）の 3つの段階のうち，ガラス容器で培養する
System I以外は培養容器に加工した卵殻を用いており，
卵殻の持つ機能をそのまま利用している。培養容器に人
工の素材のみを用いた胚培養法は，実験動物として広く
用いられているニホンウズラを対象として検討されてお
り，Ono et al.（1994）は System I，Kawashima et al.
（2005）は System II，および Kamihira et al.（1998）
は System IIIにおける培養法をそれぞれ検討している
が，これらの報告は各 Systemに限定したものであり，
1細胞期または胚盤葉期から一貫して人工容器で培養し
た報告は見当たらない。
本研究では，Kamihira et al.（1998）を参考にポリテ
トラフルオロエチレン（PTFE）膜とポリプロピレン製
のコニカルチューブを培養容器として，ニワトリの水様
性卵白に乳酸カルシウムや卵殻粉末を添加したものを培
養液として用い，胚盤葉期から孵卵 60時間後までの 5
つの段階（胚盤葉期および孵卵 27・48・55・60時間後）
の胚を 38℃で培養することで，人工容器による培養環
境が胚に与える影響を調べ，胚盤葉期胚からの培養技術
における問題点を明らかにしようとした。
2． 培養容器における空気層の有無と培養開始時期の違
いが胚に与える影響
ウズラ胚を対象として人工容器の培養環境（培養中の
空気層の有無）が胚にどのような影響を与えるのか検討
するため，胚盤葉期および胚盤葉期から 60時間孵卵
（前孵卵）を行った後に人工容器を用いて培養を開始し
た。
培養には胚の上面に空気層を設けた区（空気層あり）
と，卵白で満たし空気層を排除した区（空気層なし）を
設定し，それぞれの実験区に対して胚盤葉期および前孵
卵後から人工容器で培養した。なお，通常孵卵した胚を
対照区とした。また，添加ミネラルとして乳酸カルシウ
ム 35mg を加えた。その結果，胚盤葉期から人工容器
で培養した場合，生存率は 13日目に空気層ありの 42%
に対し，空気層なしでは 19%と有意（p＜0.05）に低い
値を示した。前孵卵後に人工容器で培養した場合，生存
率は 12日目に空気層ありの 80%に対し，空気層なしで
は 40%と有意（p＜0.05）に低い値を示した。胚盤葉期
から人工容器で培養した胚は，前孵卵後に人工容器で培
養を行った胚より全期間を通じて低い生存率で推移し
た。また，羊膜を破り孵化に相当する段階まで発生した
胚は，空気層ありで胚盤葉期および前孵卵後から人工容
器で培養した場合に 10∼15%存在した。培養中に死亡
した胚の発生段階やサイズを比較した結果，人工容器で

培養した胚は通常孵卵した胚に比べ発生が遅延する傾向
が認められ，Stage45胚のサイズ（全長）も 35∼53%小さ
かった（p＜0.05）。また，胚盤葉期から人工容器で培養
した胚は，前孵卵後から人工容器で培養した胚よりサイ
ズ（全長）が 29%小さかった（p＜0.05）。
一方，胚盤葉期から人工容器培養を行った胚には 4日
目以降から約 20%の奇形が認められたのに対し，通常
孵卵した胚および前孵卵後から人工容器で培養した胚に
は奇形の発生が認められなかった。また，発生した奇形
の部位は，約 67%が眼に関するものであり，眼以外に
も頭蓋骨や胸骨の形成不全が認められた。
以上のことから，本実験での人工容器による胚培養で
は，これまでの前孵卵後に人工容器で培養した胚に関す
る報告と同様に成長が遅延または制限されることが確認
された。加えて，成長の遅延や制限の度合いは人工容器
での培養開始時期が早いと大きくなることが明らかと
なった。また，胚成長に必要な乳酸カルシウムを添加し
たにもかかわらず胚が小さったことから，添加した乳酸
カルシウムからのカルシウムの吸収が不十分であると思
われた。この現象は空気層が排除された場合に顕著であ
ることから，培養容器のガス調整機能が不十分である可
能性も考えられた。さらに，胚盤葉期から発生 60時間
後までの初期発生における培養環境が，胚の奇形発生に
関与することが示唆された。
3． 人工容器での培養開始時期が胚の外部計測値とカル
シウムおよびマグネシウムの含有量に与える影響
人工容器で培養した胚は通常孵卵した胚に比べ発生が
遅延し，胚のサイズも小さい。胚のサイズは骨の形成と
関係があるとされているが，人工容器による胚培養で
は，乳酸カルシウムを添加しても通常孵卵した胚に比べ
発生の遅延や胚のサイズが小さいといった現象が認めら
れた（実験 2）。このことから，人工容器による培養開
始時期（胚盤葉期または孵卵 60時間後）の違いがカル
シウム（Ca）とマグネシウム（Mg）の含有量，発生ス
テージおよび胚の外部計測値に与える影響を明らかにし
ようとした。なお，添加したミネラルは，Kamihira et
al.（1998）の報告と予備実験の結果から，乳酸カルシ
ウム 35mgまたは乳酸カルシウム 25mg＋卵殻粉末 10
mgとした。
その結果，培養初期（発生 60および 120時間後）に
おける外部計測値は，培養開始時期の違いにかかわらず
人工容器で培養した胚と通常孵卵した胚の間に差は認め
られなかった。しかしながら，培養中期以降（発生 240
および 360時間後）においては，人工容器で培養した胚
は通常孵卵した胚より乾重量で 54∼57%，湿重量で 32
∼54%，第 3趾長で 19∼35%，全嘴峯長で 21∼27%，お
よび全長で 26∼28% 小さかった（p＜0.05）。また，胚
盤葉期から人工容器で培養した胚は，前孵卵後から培養
した胚および通常孵卵した胚より眼直径が 11∼22%小
さかった（p＜0.05）。一方，人工容器で培養した胚の
CaとMgの含有量は，培養開始時期に関係なく通常孵
卵した胚に比べ低い値（Ca : 12∼88%，Mg : 28∼75%）
を示した。また，胚盤葉期から培養を開始した胚は，前
孵卵後から培養を開始した胚よりサイズ（全長）が
19∼25% 小さく，Ca および Mg の含有量がそれぞれ
55∼61%および 41∼42%低かった（p＜0.05）。
以上のことから，人工容器での培養開始時期に関係な
く，乳酸カルシウムや卵殻粉末を添加しても通常孵卵し
た胚に比べ胚成長に必要な CaおよびMgが十分に吸収
されないこと，培養開始時期が早いとサイズが小さくな
り Ca および Mg の吸収量が少なくることが明らかと
なった。
4． 培養開始時期の違いが奇形発生に及ぼす影響
実験 2において胚盤葉期から前孵卵 60時間までの間
に奇形が発生する要因が存在することが示唆されたこと
から，さらに詳細に奇形発生の要因が存在する時期を特
定するため，人工容器での培養を胚盤葉期，孵卵 27時
間後，48時間後，55時間後および 60時間後から開始
し，生存率の推移と奇形の発生率について調べた。な
お，培養容器内には空気層を設け，ニワトリの水様性卵
白に乳酸カルシウムを 35mg添加したものを培養液と
して用いた。
その結果，発生 7日目まではいずれの区も生存率に差
は認められなかったが，12日目以降は全ての培養区に
おいて急激な生存率の低下が認められ，15日目には全
ての胚が死亡した。奇形は前孵卵 55時間未満から人工
容器で培養した胚に 16%認められたが，55時間後およ
び 60時間後から培養を開始した胚には認められなかっ
た。また，55時間後から培養を開始した胚には孵化直
前である Stage 45まで到達した胚が 10%認められた。
なお，発生した奇形の 57%が眼に関するものであった。
胚を保護する羊膜の形成は孵卵 29時間後から胚の頭側
で始まり，孵卵 35時間後には眼が分化する予定の前脳
のほぼ全域が羊膜で覆われることが知られている。
以上のことから，奇形発生と羊膜形成の関係が強く示
唆され，孵卵 55時間未満の胚は，人工容器による培養
環境の影響を受けやすいことが明らかとなった。
本研究は，ニホンウズラ胚を胚盤葉期から孵化予定日

まで一貫して人工容器で培養し，人工容器による培養が
ニホンウズラ胚に及ぼす影響を初めて明らかにした。
本研究の結果から，人工容器による培養環境や培養開
始時期の違いが胚の体の大きさ，ミネラル含有量，生存
率および奇形の発生率に影響を及ぼすことが明らかと
なった。また，羊膜形成の段階と胚の奇形発生に関係性
が示唆されたことから，奇形発生の認められた孵卵 55
時間未満の培養環境を胚へのダメージが少なくなるよう
に改善する必要がある。
審査報告概要
卵殻を人工容器に代替させた鳥類胚の培養は，遺伝子
導入の効率化，希少鳥類の保護・増殖に貢献するもので
あるが，胚盤葉期から一貫して人工容器を用いた培養法
は確立されていない。本研究は，胚盤葉期から孵卵 60
時間後までの 5段階のニホンウズラ胚を，ポリテトラフ
ルオロエチレン膜とポリプロピレン製のコニカルチュー
ブを用いた人工容器により培養し，胚盤葉期胚に対する
人工容器培養技術の問題点を明らかにしようとした。そ
の結果，前孵卵 55時間未満の胚にのみ奇形が発生する
ことを初めて明らかにし，羊膜が未完成の胚は，人工容
器の培養環境による致命的な影響を受けやすいことを示
した。また，人工容器による培養では成長の遅延や胚の
サイズが小さくなることから，胚への CaおよびMgの
吸収量を測定し，添加したミネラルの胚への吸収が不十
分であることを明らかにした。さらに，空気層の排除が
胚の生存率を低下させることから，培養容器のガス交換
機能が不十分である可能性を示した。本研究は，胚盤葉
期胚からの人工容器による胚培養法について検証した初
めての研究であり，今後，同手法を改良し発展させる上
での貴重な知見を示すものである。
よって，審査員一同は博士（畜産学）の学位を授与す
る価値があると判断した。

1章． 緒 論
本研究は，熱帯亜熱帯沿岸域におけるマングローブ葉
生成タンニンの生態系内動態と溶存鉄供給にかかわる役
割を明らかにすることを目的とし，以下の仮説を立て，
その検証を行った。「マングローブに生息する植食性の
カニが樹種葉と林床土壌との接触機会を増加させる結
果，その葉中タンニンが林床土壌中の鉄を溶存化させ，
この溶存鉄が植物プランクトンの増殖に寄与する」その
研究成果は以下のように要約される。
2章． タンニン溶液による溶存鉄の土壌溶出
マングローブにおけるタンニンの役割を明らかにする
ため，マングローブ葉中のタンニン含有率と，異なる塩
分濃度のタンニン溶液をマングローブ土壌に添加した際
に溶出する溶存鉄量を検証した。これらの実験にはマン
グローブ樹種 6種と非マングローブ樹種 16種の葉及び，
マングローブ林床土，暗赤色土と黒ボク土を供試した。
得られた結果は以下の通りである。
1）タンニン酸溶液濃度を高める（∼100mM）と，マ
ングローブ土壌から溶出した溶存 Fe量（∼20mgg−1）
も増加した。
2）マングローブ土壌に 100mMタンニン酸溶液を添
加した際に溶出した溶存鉄量は，暗赤色土のそれよりも
5.5倍高かった。
3）10mMタンニン酸溶液湛水 1日目のマングローブ
土壌から溶出した溶存鉄量は暗赤色土のそれより高かっ
たが，湛水 2日目以降のそれらの増加度合に違いはな
かった。
4）10mMタンニン酸溶液中の塩分濃度を高めると，
土壌から溶出する溶存鉄量は有意に減少したが，塩分濃
度 35‰の同溶液添加時にマングローブ土壌から溶出し
た溶存鉄量は，同条件の暗赤色土から溶出したそれより
6.1倍多かった。
5）供試したマングローブ樹種（n＝6）及び非マング
ローブ樹種（n＝16）葉中のタンニン含有率（mean±
SE）は，それぞれ 99±16mgg−1及び 76±19mgg−1で
あった。
6）マングローブ土壌から溶出した溶存鉄量は，添加
した供試葉抽出液中のタンニン含有率と有意な正の相関
関係がみられた。
これらの結果は，マングローブ樹種葉中のタンニンが
塩水条件下においても，マングローブ土壌から溶存鉄溶
出を促進する可能性があることを示している。
3章． マングローブリター量に占める植食性カニによ
るリター持去り量の比率
マングローブ葉中タンニンが林床土壌と接触するまで
の動態を明らかにするた日本の亜熱帯域に位置する沖縄
地域の宮古島の島尻地区マングローブにおいて，リター
キャッチャーを用いて植食性カニによる落葉の巣穴への
持去り量を通年調査した。得られた結果は以下の通りで
ある。
1）リター総量は 1221gm−2y−1であり，そのうちカ
ニによる持去り率，林床への現存率，及び海域への持去
り率はそれぞれ 68.9，9.6及び 21.4%であった。
2）総リター量における各樹種（Bruguiera gymnor-
rhiza, Rhizophora stylosa 及び Derris trifoliata）の重
量割合はそれぞれ 61.4，31.1及び 7.5%であり，各樹種
葉のカニによる持去り率はそれぞれ 72.8，69.3及び
35.7%であった。
3）落葉及びカニに持去られた落葉中のタンニン含量
は，それぞれ 92.6及び 65.7gm−2y−1であった。本調査

地点におけるリターのカニによる持去り率は，熱帯域で
同様に行われた先行研究の値と同等であった。
これらの結果から，植食性カニがマングローブ葉を選
択的に摂食するという行為は，葉中タンニンと林床土壌
との接触機会を増加させ，溶存鉄溶出を促進させる可能
性が示唆された。
4章． 植食性カニの樹種葉摂食性向とその葉中タンニ
ンの動向
本章はマングローブ樹種葉中タンニンと土壌との接
触/反応における植食性カニの役割を明らかにすること
を目的とし，宮古島の島尻林帯に優占的に生息する植食
性カニである Neosarmatium smithiによるマングロー
ブ葉の摂食性向，同カニの糞中及び巣穴内壁土壌中のタ
ンニン含有率を調査した。最後に同カニ糞水抽出液を林
床土壌に添加した際に溶出する溶存鉄含量を定量した。
得られた結果は以下の通りである。
1） N. smithiによる Rhizophora stylosa, Bruguiera
gymnorrhiza 及び Kandelia candel 葉の摂食速度は，
供試したその他の樹種葉より有意に高かった。
2）N. smithi の糞，巣穴内壁黒色土，及び黄色土中
のタンニン含有率は，それぞれ 9.93±3.14，0.49±007，
及び 0.11±0.02であった。
3）N. smithi の糞，林床土，及びそれら混合物の水
抽出液中の溶存鉄含有率（mean±SE）は，それぞれ
0.08±0.003，0.15±0.015，及び 0.65±0.015mgg−1であっ
た。
これらの結果により，植食性のカニはタンニン含有率
の高いマングローブ葉を選択的に巣穴に持ち込んで摂食
し，その葉，もしくは糞中のタンニンは土壌と接触し，
土壌から溶存鉄溶出を促進する可能性があることが示唆
された。
5章． マングローブ植生内表流水中のタンニン含有率
と溶存鉄含有率との関係
本章では，マングローブ植生域表流水中の溶存鉄濃度
の実態及び，表流水中の溶存鉄とタンニン含有率との関
係性を検証するため，マングローブ植生域における表流
水中の水質を測定し，両者の関係を解析した。調査対象
地域は，マングローブ植生域として宮古島の島尻地区と
川満地区とし，その対照として非マングローブ植生域の
同島浦底地区とした。供試表流水は 2011年 8月から
2012年 3月まで月 1回，大潮の最干潮時に各地区の上
流域から沿岸海域まで計 16地点において採取した。得
られた結果は以下の通りである。
1）マングローブ植生域の表流水中溶存鉄含有率
（0.11∼0.27mgL−1）は非マングローブ水域の表流水中
溶存鉄含有率（0.09∼0.14mgL−1）よりも有意に高かっ
た。
2）表流水中の溶存鉄とタンニン含有率は，全ての調
査対象地区において，上下流域では低く，中流域では高
かった。
3）表流水中の溶存鉄とタンニン含有率との関係は，
マングロー植生域である島尻地区と川満地区ではそれぞ
れ P＜0.01と P＜0.05水準で有意な正の相関関係がみら
れたが，非マングローブ植生域である浦底地区では有意
な関係はみられなかった。
以上の結果から，宮古島のマングローブ水域表流水中
の溶存鉄含有率は高いことが明らかになり，その要因と
して上中流域の植物体中で生成されるタンニンが関連し
ている可能性が示唆された。
6章． タンニンが植物プランクトンの鉄吸収に及ぼす
影響
本章では，マングローブ生態系におけるタンニンの役
割を解明する端緒として，宮古島の島尻林帯河口域に生
息する珪藻の Cylindrotheca closteriumを培養し，試薬
タンニン酸と FeCl3を添加した際の cell数の挙動を調
査すると共に，海水中におけるタンニン錯体鉄の溶存率
を測定した。得られた結果は以下の通りである。
1）鉄除去培養液にタンニン酸と FeCl3を加えて C.
closteriumを 15日間培養すると，処理区（FeCl3+タン
ニン）の cell 数は，対照区（FeCl3）の 29.5倍多かっ
た。
2）塩分濃度を変えた鉄除去培養液（35‰，26‰，
18‰，9‰，0‰）にタンニン酸と FeCl3を加えて C.
closteriumを 15日間培養すると，塩分濃度 18‰以下の
試験区では cell数が増加しなかった。
3）鉄除去培養液に FeCl3と濃度別タンニン酸（4，
40，400mM）を加えて C. closteriumを 9日間培養する
と，4mM区の cell数は，分注時に比べ 47.8倍増加した
が，40mM，400mM タンニン酸水溶液を添加した培養
液中の cell数は培養期間を通して増加しなかった。
これら結果はマングローブ落葉中タンニンと林床土壌
中の鉄との反応により土壌溶出した鉄が，植物プランク
トンへの鉄供給源として重要である，との説を支持す
る。
以上の結果により，マングローブ林床に生息する植食
性カニが，マングローブ葉中タンニンと林床土壌との接
触機会を増加させる結果，マングローブ葉中タンニンと

林床土壌中の鉄とが反応し，林床土壌から溶存鉄を大量
にマングローブ及び沿岸生態系に供給している可能性が
示され，そのようにして溶出する溶存鉄は植物プランク
トンに吸収可能であることが明らかとなったと言える。
そこで，本研究で得られた実験結果を基に，世界のマ
ングローブから落葉由来タンニンによって土壌溶出され
る溶存鉄量を概算すると，4560万 km2の海域における
鉄の散布量に当たり，これは HNLC海域と言われる南
極海の面積 2033万 km2の 2.24倍に相当する。
このように，マングローブの落葉由来タンニンから土
壌溶出される年間溶存鉄量は世界の海洋生産性に大きな
影響を与えるだけのポテンシャルを秘めている。本研究
で得られた成果により，マングローブの価値は益々見直
されるだろう。

タンニン含有率と有意な正の相関が見られた。水中の溶
存 Feの機能を確認するために珪藻を培養し，試薬タン
ニン酸と FeCl3を用いた実験から「マングローブの葉中
タンニンと林床土壌中の Feとの反応で土壌より溶出し
た Feが，植物プランクトンへの Fe供給として重要で
ある」との説を支持する結果を得た。これらのことから
マングローブの存在が植物プランクトンを含む沿岸域の
生態系を形成している機構を明らかにした。これらの成
果は，世界のマングローブの溶存 Fe の推計やマング
ローブの環境保全機能の再評価に寄与するものである。
よって，審査員一同は博士（環境共生学）の学位を授
与する価値があると判断した。

論文内容の要旨
筋活動時においては，伸張性収縮の繰り返しにより骨
格筋損傷が生じるが，その詳細なメカニズムや回復・治
癒過程に関する知見は未だ乏しい。近年，一酸化窒素
（NO）と組織の炎症との関連性が報告されている。そ
の役割は酸化ストレスを増大させる一方で，その殺菌力
により炎症を縮小するという善悪両面の作用を持つこと
が知られている。骨格筋損傷の炎症と治癒に NOが関
与していることが考えられるが，その詳細は不明であ
る。そこで本研究では動物モデルを用い，筋活動に伴い
筋組織中における NO の発生と，損傷・治癒との関連
を明らかにすることを目的とし，5つの研究を遂行し
た。
1． メカニカルな筋損傷モデルにおける NO生成
ラビットの脚部に機械的な伸張性筋損傷モデルを作成
した。筋損傷作成後数日遅れて組織内に増加する NO
は誘導性 NO 合成酵素（inducible Nitric oxide syn-
thase : iNOS）由来であり，損傷後 4時間および 2日
後に二峰性の上昇を示すことを見出した。メカニカルな
伸張性筋損傷後に引き起こされる NO生成は，筋組織
の中に存在する iNOSによって損傷受傷とはタイムラ
グをおいて発生することが示唆された。
2． 一過性の走運動刺激による NO生成
機械的に損傷を作成したモデルと実際の筋運動におけ
る NO生成の差異を調べるために，短時間の急性高強
度ランニングを負荷したラットの下肢骨格筋を観察し
た。NO生成および iNOS mRNA発現は，走運動 3時
間後にピークを迎えることを明らかにした。走運動にお
いて，筋活動と NO生成には機械的に作成したメカニ
カルな損傷モデル同様にタイムラグが生じていることが
示唆された。
3． 過重力負荷による筋損傷と NO生成
普段の筋運動と異なるメカニカルストレスである過重
力が，骨格筋に及ぼす影響について検討した。短時間の
過重力環境に暴露された下肢筋内には機械的損傷モデル
と同レベルの NO生成やインターロイキン等の損傷様
物質が発生するが，組織学的な損傷を確認することはで
きなかった。短時間の過重力環境による筋内 NO 生成
は 1G環境下の筋損傷とは異なることが示唆された。
4． 筋損傷治癒と NO生成との関連
機械的に作成したメカニカルな筋損傷モデルでは，タ
ンパク分解を促進するプロテアーゼである Calpainや
筋損傷部位の再構築に働く酵素であるMyo-Dが，筋内
NO 生成と同様に損傷 3日以降に上昇した。薬理的に
NO生成をブロックすることで，この現象が消滅し治癒
が遅れることも明らかになり，損傷後の NO生成がト
リガーとなり筋再生が促進されることが示唆された。
5． 筋損傷治癒と遅発性筋痛の関連
機械的に作成したメカニカルな筋損傷モデルにおい
て，筋内における発痛物質やその受容体の変化を調べ
た。筋損傷治癒時における発痛関連タンパクや遺伝子の
発現は，NO生成がピークとなる損傷 3日以降に上昇す
ることが明らかになった。NOが発痛物質や情報伝達に
おける受容体の発現調節を行うことにより，遅発性筋痛
の発生に関与している可能性が示唆された。
これらの検討により，伸張性収縮モデル，高強度走運
動，過重力負荷といった筋活動により骨格筋損傷が発生
すると，数日のタイムラグをおいて NO生成が増大し，

損傷治癒や筋痛の発生に関与することが明らかになっ た。
審査報告概要
筋活動により骨格筋内では炎症とタンパクの崩壊にと
もなう損傷を生じ，その後その炎症，損傷および筋痛は
治癒に向かうことが知られている。また，筋活動時には
骨格筋内の一酸化窒素の生成が増大することも報告され
ている。本研究では，筋活動により生じる骨格筋の損傷
と治癒に対する一酸化窒素の果たす役割について明らか
にした論文である。その結果，骨格筋に負荷される筋活
動として考えられる高強度走運動，過重力負荷およびメ
カニカルな伸張性筋収縮モデルを用いた場合，骨格筋損
傷が発生し，その後タイムラグで一酸化窒素の生成が増
大し，それが損傷治癒や筋痛の発生に関与していること
が明らかになった。また，メカニカルな伸張性収縮によ
る筋損傷モデルにおいて，筋損傷と治癒過程では筋内の
発痛物質や情報伝達における受容体の発現調節を行うこ
とが，遅発性筋痛の発生に関与している可能性が示唆さ
れた。本研究は，筋活動にともなう損傷から治癒に至る
生理的メカニズムについて，一酸化窒素の生成増大が深
く関与するという新知見が得られた。
よって，審査員一同は博士（運動生理学）の学位を授
与する価値があると判断した。

Vietnam has reformed its economy that known as
“Doi Moi” since 1986 and enhanced a reasonable level
of macro-economic stability over the past twenty-five
years. It is now focusing on issues of competitiveness
and productivity through micro-economic reform
programs. Although reforming the economy following
the trend of reducing density of agriculture in Gross
Domestic Product (GDP), Vietnam is still the country
depending on agriculture heavily. However, invest-
ment on agriculture is not adequate to its contribution
to GDP and creating jobs. Consequently, agricultural
development has not shown sustainable and complete
progress with small scale and low productivity.
This dissertation is considered a contribution to
improving rural investment climate and promoting
growth of agro-enterprises. Firstly, it assesses the
situation of rural investment climate in Northern area
of Vietnam, and then, examines the impacts of rural
investment climate factors on business activities of
agro-enterprise.
Objectives of the dissertation are (1) to collect evi-
dences of rural investment climate factors, indicators
of firm’s business activities, and relationship between
rural investment climate and business activities of
enterprise, (2) to describe the situation of rural in-
vestment climate in Northern area of Vietnam and
analyze the
Government’s efforts to create an enabling rural
investment climate, (3) to examine impact of rural
investment climate factors on business activities of
agro-enterprise for both indicators of total factor
productivity and investment decisions, and (4) to
suggest implications that can contribute to improve
rural investment climate and promote growth of agro-
enterprises.
In terms of structure, the dissertation has seven
chapters. The dissertation begins by defining the re-
search problem with research questions, objectives
and providing the originality and significance of the
research study. Chapter one also reviews the research
background, and presents summary of method, con-
ceptual framework, scope and limitation of the study.
Chapter two reviews extant literature related to re-
lationship between rural investment climate and busi-
ness activities of enterprises. Theoretical framework
of the research study is also developed in this chapter.
Chapter three provides a description of characteristics
of Northern area of Vietnam and a summary statistics
of the observations.
Data analysis and findings are presented in chap-
ters four, five and six. Chapter four presents the situ-
ation of rural investment climate in Northern area of
Vietnam by understanding the constraints of rural
investment climate and discussing on the Govern-
ment’s efforts to remove these constraints. Chapter
five examines the relationship between rural invest-
ment climate and agro-enterprise performance in
Northern area of Vietnam. This examination empha-

sizes the impact of rural investment climate factors on
total factor productivity of agro-enterprise. Chapter
six presents descriptive findings of the research study
related to measuring the influence of rural investment
climate factors on agro-enterprise’s investment deci-
sions which include decision to invest and level of
investment made. Finally, the dissertation ends with
chapter seven where conclusions drawn from data
analysis and implications of the research study are
respectively summarized and presented.
The dissertation provides descriptive findings of the
situation of rural investment climate that are some-
what reflected by identification of rural investment
climate constraints and how to remove these con-
straints. In addition, the dissertation demonstrates
the impact of rural investment climate factors on
business activities of agro-enterprise which are
indicated by total factor productivity and investment
decisions.
As a consequence of the research findings, this re-
search study provides many implications including
the policy implications and implications for the further
research. Firstly, implications are mentioned in policy
recommendations for the Government to remove con-
straints towards enabling rural investment climate
and for the agro-enterprises to improve their capabil-
ity and building strategies. The implications for the
further research are suggested.

地方に立地する農企業にとって，農村の投資環境が企
業の事業成果を大きく左右することは広く認められてい
る。本研究は，ベトナム北部 4州の農村で事業を展開し
ている農企業を調査対象にして，農村投資環境と農企業
の事業活動の関係について実証的に明らかにし，農企業
が事業成果を向上させるために，農村の投資環境をどの
ように整備していくべきかを政策提言するという，すぐ
れて実践的な内容である。本研究では，①近年政府が農
村投資環境の整備にあたって強化してきた事項，②農企
業の事業成果に対する農村投資環境のインパクト評価
（全要素生産性の計測による），③農企業の投資決定に対
する農村投資環境の影響について，計量モデルを用いて
明らかにした。その結果，農村投資環境の制約要因とし
て，土地と金融へのアクセス，行政上の諸手続き，農村
インフラ，税制，労働力人材の熟練と教育が導出され
た。しかしながら，現実には，土地へのアクセスが複雑
であること，金融へのアクセスもまた困難であることが
指摘された。とはいえ，農村投資環境の整備は，農企業
の成長と発展にとって不可避であることから，政府への
いくつかの政策提言がなされた。本研究は，依然として
農業を主体とするベトナム経済の発展にとって重要な核
心をなす農村投資環境の整備に焦点をあてたものであ
り，その成果はきわめて斬新であり，政策的な意義が大
きい。
よって，審査員一同は博士（会計学）の学位を授与す
る価値があると判断した。

論文内容の要旨
本研究では，ニーズに対応した切り花生産・流通・消
費のために，①切り花の輸送方式の違いによる品質保持
効果の定量的評価，②花持ち試験における品質の定量的
評価を目指し，目視による評価が中心となっている流通
過程・生産現場において，実用的であり簡易的な切り花
の定量的品質評価方法を検討した。
平成 21年現在，日本における切り花の生産および需
要量は 55億本以上で，キクが約 20億本と最も多く，次
いでバラ，カーネーション，トルコギキョウ，ユリと
なっている。また，世界的に取引きされる商品としても
バラ，カーネーション，キクが上位を占めており，本研
究では，主要切り花として流通しているバラを試料とし
て取り上げた。
各種の消費者アンケートによると，切り花を購入する
際のポイントに花持ちのよいことが重要視され，これま
で切り花の品質保持および評価法に関する研究も多くな
されているが，そのほとんどが植物の生理学に基づく栽
培法，光合成，蒸散，糖質に関するものとなっている。
切り花を採花したままの状態，いわゆる非破壊検査でな
く，花弁や茎葉を切り取り，物理的に破壊，搾汁して調
査するなどの方法が主流であった。このため，高品質な
切り花と判明しても消費者に渡すことができないことが
問題であった。そこで，花弁糖度を光センサーで測定，
評価する非破壊検査が試みられているが，果物のように
検査対象物の形状が同一でなく，花弁は蕾から開花に至
る段階で形状が変化することが，実用化の課題となって
いる。また，バラ切り花は水ストレスに弱く，ベント
ネックが起こりやすいと言われており，水ストレスを与
えずに流通させることが望まれているが，特にバラに関
しては非破壊で品質評価する方法は見あたらないのが現
状である。
このように，世界の主要切り花の中では，バラは日持
ちが短く，非破壊で検査する方法がない。そこで，非破
壊による品質評価法を確立することが急務と考え，物理
的に安定している茎に着目し，茎の曲げ特性と水分との
関係について検討した。
1． 力学特性による水分測定の可能性の検討
バラ切り花の茎を測定部位として力学特性に着目し，
品質変化の要因の一つである萎れの定量的な評価を行う
ため，まず，花首付近から切り取った茎を用いて破壊測
定による曲げ試験を行い，茎の力学特性値と水分の関係
を検討した。曲げ試験は，花首の下から長さ 8cmの茎
を切り取り，両端支持の単純梁モデルを設定し，スパン
中央に垂直に曲げ荷重を加えた。
茎の力学特性値として曲げ荷重，曲げ弾性係数，曲げ
剛性，茎のたわみに対する曲げ荷重の最大変化率
（DPmax）などに着目して水分との関係を検討した。そ
の結果，茎の水分が減少し萎れることにより曲げ荷重，
曲げ剛性の値が小さくなった。それらの関係を対数近似
式で求め，曲げ荷重と曲げ剛性それぞれから水分を推測
することができた。さらに，DPmaxが水分の減少で小さ
くなることが分かった。DPmaxによる水分推定式は曲げ
荷重や曲げ剛性よりも小さい平均二乗誤差 RMSDでバ
ラ切り花の茎の水分を推定できた。
2． 推定式の汎用化の検討
汎用的に使える推定式を検討するため，栽培時期の異
なる試料を用いて作成した推定式の適合性について検討
した。この結果，単一栽培時期の試料だけで作成した推

定式では，他の時期の試料の水分を推定することができ
ないことが確認できた。そのため新たに 3種類の異なる
栽培時期の試料を合わせて推定式を作成することで，単
一栽培時期の試料による推定式と比べて汎用性のある推
定式を作成することができた。そして，説明変数に平均
茎径を加えた重回帰分析により推定式を作ることで，
DPmaxだけを用いたときより汎用的な水分推定式を作成
することができた。
3． 非破壊水分測定式の検討
茎を切取り，曲げ荷重を測定し，DPmaxを用いること
で水分を推定できる可能性が示唆されたため，その結果
をもとに非破壊による水分推定式を検討した。曲げ試験
は，バラ切り花に花，葉が付いた状態で，花首の下の部
分の茎を測定対象部位とし，両端支持の単純梁モデルを
設定し，スパン中央に水平に曲げ荷重を加えた。
その結果，説明変数として曲げ荷重と平均茎径を用い
た重回帰式により，非破壊で茎の水分を推定することが
できた。さらに，説明変数に DPmaxと平均茎径を用い
ることで推定値の RMSDを小さくすることができ，非
破壊での水分推定が可能であることが分かった。
また，作成した水分推定式を用いて同一試料の茎の水
分の経時変化を推定することができた。
4． 結 論
バラ切り花の流通過程における品質評価技術として，
従来の目視を中心とした評価，特別な測定機器を用いた
評価にかわる新たな評価方法として，実用的であり簡易
的で定量的品質評価方法を検討した。
その結果，バラ切り花の茎の曲げ荷重と茎径をもとに
非破壊での水分推定式を得ることができた。さらに，こ
の水分推定式を用いて非破壊でバラ切り花の水分を測定
することができた。
以上のことより，定量的な非破壊品質評価方法がない
バラ切り花について，茎の曲げ特性による非破壊水分測
定法を流通過程における新たな簡易的品質評価方法とし
て提案することができた。
審査報告概要

とんどが植物の生理に関する内容で，現場で実用的に要
求される非破壊法については見当たらない。そこで，本

は“しおれ”に関係した水分を取り上げ，実用性を重視
した簡易な非破壊測定法について検討した。その結果，
成長過程や個体差による影響が小さく，比較的安定して
測定可能な茎の曲げ試験を採用した。そして，曲げ荷重
曲線の最大変化率の対数値と茎径の 2つの説明変数によ
る重回帰式により，収穫時期の異なる切り取られた茎の
水分推定式を作成できた。さらに，非破壊状態の切り花
の茎の水分推定式を同様に作成して，同一試料の茎の水
分の経時変化を推定した。本研究は，世界的に主要な切
り花であり，日持ちの短いバラについて，力学的な手法
によって初めて実用的かつ簡易的な非破壊品質評価法を
提案した
よって，審査員一同は博士（農業工学）の学位を授与
する価値があると判断した。
