【緒言】 
内臓脂肪型肥満，糖尿病，高脂血症，高血圧は，長年にわたる悪質な生活習慣を一因とす
る代表的な疾患である。特に内臓脂肪型肥満と，残る 3 項目のうちの 2 項目以上を併発し
た状態はメタボリックシンドロームと呼ばれ，動脈硬化，心筋梗塞や脳梗塞を導く主たるリ
スクとなることが知られている。こうした「生活習慣病」に分類される疾患は以上の限りで
はない。日々の悪質な食生活，運動不足やストレスが，がん，骨粗鬆症，歯周病や腸疾患な
どの疾患にも関与することが益々明らかにされている。一方，生活習慣病の若年化とこれに
伴う医療費の増額が懸念されており，本疾患に対する効果的な治療もしくは予防素材の開発
が求められている。 
生活習慣病の医薬品による治療を考えたとき，海洋生物は候補化合物を探る天然物ソース
として有望である。これまで数多くの海洋天然物（もしくはその類縁体）が，医薬品として
上市されてきた。化合物ライブラリーを系統的な合成経路で効率的に多品種合成する
Combinatorial Chemistry に基づく新薬開発が発展する一方，生物間の生存競争というふる
いにかけられ残されてきた天然物の化学構造には人知を越えた知恵が隠されており，今後も
医薬品開発に大きく寄与することが期待される。実際，現在でも複数の海洋天然物由来化合
物が開発フェーズに進んでいる。 
生活習慣病に対する異なるアプローチとして，プロバイオティクス，プレバイオティクス
およびこれらの組み合わせであるシンバイオティクスもまた有望な素材である。プロバイオ
ティクスとは，生きて腸に届くことで宿主に便益をもたらす，乳酸菌やビフィズス菌を代表
とする細菌群である。一方，プレバイオティクスとは，有益な細菌に選択的に資化されるこ
とで健康効果を導く，イヌリンやガラクトオリゴ糖を代表とする化合物群である。プロバイ
オティクスとプレバイオティクスを上手く組み合わせることで両者の相乗効果が導かれる
ことが期待され，こうした素材はシンバイオティクスと呼ばれている。シンバイオティクス

は食品および医療の現場で腸疾患に対する効果や整腸作用が認められているものの，その組
み合わせ方の科学的根拠は薄く，プロバイオティクスとプレバイオティクスの相乗効果が必
ずしも認められないことが課題となっている。 
以上の背景から本研究では，生活習慣病に対する治療もしくは予防法を，二つのアプロー
チから検討することとした。第一のアプローチとして，海洋生物からの医薬品候補化合物の
探索をおこなった成果を，本論文第 1 章と第 2 章にて報告した。また第二のアプローチと
して，プロバイオティクスとプレバイオティクスの最適組み合わせからなる，高い健康効果
が期待されるシンバイオティクス素材の開発を試みた成果を，第 3 章にて報告した。なお，
実験動物の権利愛護に関する議論が活発化していることを鑑み，現段階では in vitro 試験系
にて可能な限り高いポテンシャルを有する素材を選定することとした。 
 
【研究内容】 
第 1章：ヒト子宮頸がん由来 HeLa細胞株を用いたスクリーニング系による，がん細胞に対
する細胞傷害活性を有する海洋生物由来化合物の探索 
本章では，東京大学水圏天然物化学研究室が保有する海洋無脊椎動物ライブラリーより，
がん細胞に対する細胞傷害活性を有する医薬品候補化合物の探索をおこなった。生理活性は，
ヒト子宮頸がん由来 HeLa 細胞株を用いた MTT 試験系により確認した。 
第一に，ライブラリーに属する無脊椎動物から調整した粗抽出画分をランダムスクリーニ
ングした結果，カイメン‐赤藻共生体 Ceratodictyon spongiosum/Haliclona cymaeformis
の脂溶性画分に強力な活性が認められたため，本サンプルより活性化合物の抽出を試みた。
続いて，前述の試験系における生理活性を指標として各種クロマトグラフィーによる分画を
進めた結果，HPLC にて単一のピークを示す 4 つの画分を得た。二次元 NMR と質量分析
計を用いた平面構造解析より，これらは新規 1-アルキル-グリセロールの一群であることが
判明したため，我々は本化合物群を Ceratodictyols と命名した。最後に，化合物中の不斉
点の立体構造を改良モッシャー法により解析し，Ceratodictyols に共通して存在するグリセ
ル基の不斉点が S 体構造を取ること，HPLC で得られた 4 画分のうち 2 画分は，アルキル
基に存在するアリルアルコール部位の不斉点が S 体と R 体である化合物の混合物（エピマ
ー混合物）であることを明らかにした。 
抽出された各 Ceratodictyol の生理活性を評価したところ，細胞傷害活性は極めて弱かっ
た（IC50 = 67 M）。分子構造も単純であることから，医薬品への応用は困難であると推察
された。

第 2 章：マウス由来 ST-13 前駆脂肪細胞株を用いたスクリーニング系による，前駆脂肪細
胞から成熟脂肪細胞への分化を誘導する海洋生物由来化合物の探索 
正常な脂肪細胞は，血中に過剰量存在する糖質や脂質の倉庫としての役割を果たす。加え
て，抗炎症サイトカインであるアディポネクチンを産生し，主にインスリン抵抗性を改善さ
せることで糖尿病の予防および治療に寄与する。一方，過度に肥大した脂肪細胞は TNF
をはじめとする炎症性サイトカインを産生し，メタボリックシンドローム全般の悪化を引き
起こす。前駆脂肪細胞を脂肪細胞へと誘導する化合物の多くは，正常な脂肪細胞の数を増加
させる一方，肥大した脂肪細胞のアポトーシスを誘導し，結果として抗糖尿病効果を導くこ
とが期待されている。 
そこで本章では，東京大学水圏天然物化学研究室が保有する海洋無脊椎動物ライブラリー
より，前駆脂肪細胞を成熟脂肪細胞へと分化誘導する活性を有する化合物を探索した。生理
活性は，マウス由来 ST-13 前駆脂肪細胞株を用いたアッセイにより評価した。播種後 8 日
間の培養によりコンフルエントに達した細胞に被験物質を加え，4 日後における細胞内への
脂肪滴の存在を Oil Red O にて染色することで分化誘導の有無を確認した。本アッセイで認
められた生理活性を指標として目的化合物を探索した結果，カイメン Xestospongia 
testudinaria より 5 つの新規化合物を含む計 16 種の臭素化アセチレン酸の一群を単離およ
び平面構造決定した。このうち，特徴的なテトラヒドロフラン環を有していた化合物
（Testufuran A と命名）の立体構造は，円二色性分散計による CD スペクトル分析と二次
元 NMR による NOE スペクトル分析を組み合わせることで確定した。 
単離された化合物の多くは，2‐130 M オーダーの濃度にて前駆脂肪細胞を脂肪細胞へ
と分化誘導した。各化合物の構造と活性の比較より，活性にはカルボキシル基の存在が不可
欠であることが示唆された。分化誘導後の細胞からはアディポネクチン産生が認められ，本
効果は特に Testufuran A で顕著であった。Testufuran A のモチーフを利用した医薬品素材
の開発が期待されるが，より低濃度で効果が導かれるよう，更なる構造活性相関の検証が求
められる。 
 
第 3章：In vitro試験系の組み合わせによる最適なシンバイオティクス素材の探索 
本章では，最適なシンバイオティクス素材の組み合わせを，複数の in vitro 試験系を組み
合わせることで探索した。本素材に期待する効果としては，腸管上皮細胞内に誘導される「小
胞体ストレス（後述）」に対する抑制効果を求めることとした。なお，プレバイオティクス
として用いる素材は，株式会社ヤクルト本社より入手可能なガラクトオリゴ糖（以下，GOS）
に固定した。 
研究は 2 段階に分けて実施した（以下 A および B）。ステップ A では，小胞体ストレス抑
制効果を導くプロバイオティクスとして乳酸菌とビフィズス菌のいずれに焦点を当てるべ

きかを検討した。ステップ B では，ステップ A にて焦点を当てた菌属のうち，どのような
菌株が GOS を効率的に利用するかを検証した。 
＜ステップ A＞ 
ヒト腸上皮由来 Caco-2 単層モデルを用いた，腸管上皮細胞内に誘導される小胞体ストレス
に対する乳酸菌およびビフィズス菌の効果検証 
小胞体ストレスは，心理ストレスをはじめとする悪質な生活習慣を一因とし，細胞小器官
である小胞体内に折り畳み不全のタンパク質が蓄積されることで誘導される。過度の小胞体
ストレスが誘導されると，本器官が担当する重要な細胞機能であるタンパク質の合成や分泌
が阻害され，最終的に細胞死が誘導されることが知られている。特に腸管上皮細胞内に誘導
される本ストレスは，炎症性腸疾患をはじめとする種々の腸炎への関与が注目されている。 
そこで本ステップでは，ヒト腸上皮由来 Caco-2 単層モデルを用い，乳酸菌とビフィズス
菌の各 4 菌種が腸管上皮細胞内に誘導される小胞体ストレスに及ぼす効果を検証した。
Caco-2 単層を各試験菌種の加熱死菌体と共培養後，小胞体ストレス誘導物質であるツニカ
マイシン刺激を加え，①腸管上皮バリアの強度（経上皮電気抵抗値），②細胞膜の頑強性（ト
リパンブルー染色），および ③小胞体ストレスマーカーの発現（RT-qPCR，ウェスタンブ
ロット）を比較した（括弧内には評価に用いた手法を示した）。その結果，ツニカマイシン
刺激に起因する腸管上皮の破綻と小胞体ストレスマーカーの誘導は，ビフィズス菌の存在に
より抑制された一方，殆どの乳酸菌では効果が認められなかった。以上より，腸管上皮細胞
内に誘導された小胞体ストレスを標的とするプロバイオティクスとして，以降はビフィズス
菌に焦点を当てることとした。 
＜ステップ B＞ 
液体培養系を用いた，ビフィズス菌株間での GOS 資化性の比較検討 
本ステップでは，GOS を含む液体培地にてビフィズス菌 7 菌種 14 菌株を培養し，各菌
株の ①GOS 利用性，②増殖性，および ③代謝産物である有機酸の産生プロファイルを比
較した。その結果，成人で頻繁に認められるビフィズス菌種（e.g. B. adolescentis）は，乳
幼児で頻繁に認められるビフィズス菌種（e.g. B. breve）と比較して高効率かつ優先的に
GOS を利用し，より高い比増殖速度を示した。高い GOS 資化性が認められたビフィズス
菌群では乳糖などの少糖類を糖源としたときと同様，ビフィズス菌の通常の糖代謝経路から
予測される酢酸：乳酸＝3：2 の比率にて有機酸が産生された一方，利用能が比較的低かっ
たビフィズス菌では乳酸の代わりに多量のギ酸の産生が認められた。以上の結果から，高い
GOS 資化性を示したビフィズス菌群は“無理なく”GOS を利用していることが示唆された。
続いて，GOS 資化性が高かった菌株の 1 つである B. adolescentis 基準株について遺伝子発
現解析した結果，-ガラクトシダーゼと LacS シンポーターから構成されるオペロンの発現
が GOS の利用に伴い顕著に誘導されることが明らかとなった。本オペロンは全ての試験ビ

フィズス菌株に保持されていた一方，そのアミノ酸配列の差異と GOS 利用能の間に相関が
認められた。以上の結果は，-ガラクトシダーゼ‐LacS オペロンがビフィズス菌の GOS
利用に関与すること，その配列を指標として高い GOS 資化能を有するビフィズス菌をスク
リーニング可能なことを示唆していた。 
以上，ステップ A および B より，成人で多く検出されるビフィズス菌群と GOS を併用
したシンバイオティクス素材を開発することで，小胞体ストレスに起因する腸疾患を効果的
に予防できることが期待された。今後は -ガラクトシダーゼ‐LacS オペロンをマーカーと
して候補菌株を選定し，動物およびヒトでの効果検証を進めたい。 
 
【総括と展望】 
生活習慣病の若年化とそれに伴う医療費の増額を抑制するためには，本疾患への効果的な
対処法を治療・予防双方の観点から検討することが理想である。本研究では in vitro 試験系
を活用し，それぞれにつき新たな抗生活習慣病素材を検討した。 
第 1 章と第 2 章では，水圏天然物をソースとし，生活習慣病に対する医薬品候補化合物
を探索した。その結果，①ヒト子宮頸がん由来 HeLa 細胞に対する穏やかな細胞傷害活性を
示す新規 1-グリセリルエーテルCeratodictyolsおよび ②前駆脂肪細胞を成熟細胞への分化
誘導することで糖尿病の改善に寄与することが期待される臭素化アセチレン酸の一群を単
離することに成功した。Ceratodictyols については，活性が微弱である点，構造が単純であ
る点から，医薬品の応用へは困難なことが推察された。一方，グリセリルエーテルは，化粧
品における乳化剤など，幅広い用途で利用される成分である。本研究の目的とは一致しない
ものの，新規かつシンプルな構造を有する本化合物が，今後こうした産業分野で活用される
ことも期待される。臭素化アセチレン酸については，独特なテトラヒドロフラン環構造を有
する Testufuran A において，高濃度下に限定されるものの，強力なアディポネクチン誘導
能が認められた。本化合物の誘導体化を行うことで，より低濃度で効果を導くことができる
可能性がある。 
第 3 章では最適なシンバイオティクス素材の組み合わせを検討した。主に
Bifidobacterium が種々の腸疾患の原因として注目される腸管上皮細胞内の小胞体ストレ
スに対する抑制能を有することを示した後（Step A），同菌株のうちプレバイオティクスで
あるガラクトオリゴ糖と相性の良いビフィズス菌株を検討した（Step B）。その結果，-ガ
ラクトシダーゼ－LacS オペロンのアミノ酸配列が Bifidobacterium の GOS 利用性とリン
クすることが明らかとなった。今後，本遺伝子座を指標として GOS と相性が良く，かつ安
全性の高いプロバイオティクス候補菌株を探索したいと考える。一方，実際のヒトにおける
シンバイオティクスの挙動は，腸内細菌叢や食事をはじめとするあらゆる宿主パラメータに
左右されることが見込まれる。前述の検討と並行して，今後は実際のヒト試験データを参照

し，これら交絡因子の影響を包括的に考慮した，個々人に最適な「テーラーメイド・シンバ
イオティクス」の検討にも挑戦したい。そのためには機械学習や人工知能をはじめとする計
算科学の知識が必要になると考えている。 
本研究では生活習慣病に対する方策を複数の角度から検討した。今後も未経験研究分野を
含め，あらゆる視点から人々の健康に寄与する食品ならびに医薬品素材の開発に従事したい。 
 
 
審 査 報 告 概 要 
 
近年，生活習慣病の若年化と医療費の増額が懸念され，本疾患に対する治療もしくは予防
素材の開発が求められている。そこで本研究では，in vitro 試験系を活用し，高い効能を有
する抗生活習慣病素材の探索および処方開発を検討した。第１章と第２章では海洋生物から
の医薬品候補化合物の探索をおこない，第３章では研究を２つのステップに分け，プロバイ
オティクスとプレバイオティクスの最適組み合わせからなるシンバイオティクス素材の開
発をおこなった。研究の結果，第１章と２章では海洋無脊椎動物から新規化合物１１種含む
計２２種の化合物の同定に成功した。第３章では，乳酸菌と比較してビフィズス菌が腸管上
皮内の小胞体ストレスに対する強い抑制効果を示すことを，Caco-2 細胞を用いた試験系に
より明らかにした。更に，一部のビフィズス菌株がプレバイオティクスの一種であるガラク
トオリゴ糖を特に効率よく利用できることを明らかにし，両者を組み合わせることで効果的
な抗生活習慣病効果が導かれる示唆を得た。 
これらの研究成果等を総合的に評価した結果，審査委員一同は博士（バイオサイエンス）
の学位を授与するに値すると判断した。

Capsicum species, popularly known as hot pepper, is a popular vegetable in the world. Hot pepper 
production worldwide in 2016 was 34.5 million tones with 17.79t ha-1 average (FAO, 2016). In 
many parts of the world, hot pepper is essentially valued for its pungency, nutrition and pigment 
contents of the fruits (Tian et al., 2014). Hot pepper is also an important horticultural crop in 
Afghanistan (Walters and Jha, 2016). It is widely utilized as an additive to many Afghan dishes. 
Afghanistan people eat fresh pepper as well. Northern provinces in Afghanistan are well known for 
hot pepper production, mostly being exporting to Pakistan as a fresh and dried product (Walters et al., 
2012). However, Afghanistan has been suffering from severe drought stress during the past decade, 
which affected germination, yield and quality of hot pepper as well as other horticultural crops. One 
of the short-term and most pragmatic approaches to alleviate drought stress is seed priming. Hence, I 
studied the effect of seed priming with polyethylene glycol (PEG 6000) on growth, quality and 
capsaicin concentration of hot pepper under drought conditions throughout seed, seedling and growth 
stages. 
 
1. Effect of Drought Stress on Growth, Quality and Capsaicin Concentration of Hot 
Pepper (Capsicum annuum) 
The results show that drought stress significantly retarded the growth and increased the ethylene 
production rate in comparison with daily irrigated control. Fruits harvested at 36 and 45 days after 
anthesis (DAA) accumulated higher concentration of glucose, fructose and sucrose in response to 
moderate and severe stress. In addition, capsaicin and dihydrocapsaicin content in fruits harvested at 
36 and 45 DAA also showed a significant increase under severe and moderate stress. The present 
findings revealed that drought stress might alter the primary and secondary metabolites of hot pepper

fruits. 
 
2. Effect of Seed Priming with Polyethylene glycol (PEG 6000) on Seed Viability, 
Germination of Hot Pepper (Capsicum annuum) 
Seed priming with high concentration of PEG (-1.2 and -1.6 MPa) significantly increased the seed 
viability and improved germination percentage. Higher concentration of PEG priming recorded 
significantly lower seed electrolyte leakage compared to lower concentration of PEG treatments. The 
effect of PEG priming can be attributed to the slowly achieved imbibition during hydration process 
and accelerated water uptake during rehydration process after sowing. The results suggest that seed 
priming with higher concentration of PEG might alleviate the deterioration in seed vigor, achieving 
the uniformed germination. The enhancement of root/shoot ratio by higher concentration of PEG 
treatment can also contribute to the adaptability in drought conditions. 
 
3. Effect of Seed Priming with Polyethylene glycol (PEG 6000) on Physiological 
Response of Hot Pepper (Capsicum annuum) Seedlings under Drought Conditions 
The results suggest that drought stress significantly increased leaf temperature and decreased 
stomatal conductance as well as photosynthetic rate in moderate and severe drought stress compared 
to mild treatment. Seedlings primed with higher concentration of PEG recorded increased 
photosynthetic rate under mild drought stress in accordance with the lesser degree of wilting. The 
decreased leaf stomatal size and increased number of stomata in the seedlings primed with higher 
concentration of PEG under drought stress might compensate the reduced stomatal conductance 
which both decrease photosynthesis and transpiration. 
 
4. Effect of Seed Priming with Polyethylene Glycol (PEG 6000) on Quality and Capsaicin 
concentration of Hot Pepper (Capsicum annuum) under Drought Condition 
The results suggest that seed priming with higher concentration of PEG induced ethylene 
production in fruits harvested at 36 DAA under mild and moderate stress. Fruits harvested at 36 
DAA under drought stress with lower concentration of PEG priming produced higher concentration 
of capsaicin and dihydrocapsaicin compared to higher concentration of PEG priming treatment.  
 
Conclusion 
The findings of this research suggest that the possibility of PEG priming with -1.2 MPa and -1.6 
MPa treatments to maintain physiological activities of hot pepper under drought conditions, with 
improvements in seedling growth and fruit quality with rich sugars and capsaicinoids. The technique

of seed priming with PEG can be hopefully applicable to the small-scale farmers in Afghanistan. 
 
和文要旨 
アフガニスタンにおいてトウガラシはポピュラーな園芸作物として，人々に広く利用され，
アフガニスタン料理に欠かせない野菜の一つである。 
しかしながら，アフガニスタンは過去 10 年間の深刻な旱魃によって，トウガラシの発芽，
収量，果実品質が影響を受け，解決方法が模索されている。そこで本研究では乾燥ストレス
を軽減するための短期的かつ実用的なアプローチとしてポリエチレングリコール（PEG 
6000）を用いた種子プライミング処理に着目し，乾燥ストレス軽減のための種子プライミン
グ処理の至適条件を検討し，またその作用メカニズムについて調査を行った。 
その結果，高濃度の PEG（-1.2 および-1.6 MPa）を用いたプライミング処理は，種子の発
芽率を有意に増加させ，また細胞損傷の指標であるイオン溶出率が有意に低い値となった。
高濃度の PEG 処理はとくに根の生育を促進し，さらに軽度の乾燥ストレス下では光合成速
度の増加が見られた。また中度および重度の乾燥条件では高濃度の PEG 処理によって気孔
コンダクタンスが増加し，乾燥への順応性を高めたことが示唆された。これらの結果はサイ
ズの小さい気孔の数が増えていることに起因するものと考えられた。 
 果実品質については，高濃度の PEG を処理した区が，軽度および中度の乾燥ストレス下
で果実のエチレン生成を促進することが明らかとなった。また低濃度の PEG を処理した区
でカプサイシンおよびジヒドロカプサイシン含量の増加が見られ，辛味成分の増加に寄与す
ることが明らかとなった。 
 以上の結果，-1.2MPa および-1.6MP の濃度の PEG 種子プライミング処理がトウガラシの
苗の健全な発育に有効であり，乾燥ストレス耐性を高め，生産性および果実品質を向上させ
ることが示唆された。PEG を用いた種子プライミング処理は小規模な農家でも利用可能な技
術であり，アフガニスタンのような発展途上国における予測不可能な旱魃に対処するための
有用技術として貢献できる可能性が高い。 
 
 
審 査 報 告 概 要 
 
モハマド・ムスタファ・ハリス氏はアフガニスタンで重要な課題となっているトウガラシ
の乾燥ストレス軽減のために種子のプライミング技術の効果について検討し，ポリエチレン
グリコールによる 48 時間処理によって，乾燥条件下における健苗育成，光合成活性の維持，
カプサイシン濃度の向上に効果があることを見出した。ポリエチレングリコール処理は，葉
や果実のエチレン生成に影響を及ぼし，葉の気孔の数を増加させ，果実の糖含量やカプサイ

シン濃度を高めることが明らかとなった。ポリエチレングリコールによる種子のプライミン
グ処理は，簡便で安価な技術であり，アフガニスタンの小規模な農家においても利用可能な

論 文 内 容 の 要 旨 
 
The purpose of this study is to analyze the differences in relationship between vocational 
education and career education, and to have a clear idea of the structures involved with vocational 
education and career education and the proper way to implement them. Most of previous studies that 
analyzed the methods of career education are analyses of how to reflect the ideal career education in 
the present education system. They used career education models in the USA from the 1970s, and 
tried to transplant them to Japan and discussed how to develop them. However, the budding of career 
education in Japan has already been discussed by Soseki Natsume in terms of occupation and hobby 
in the Meiji Era. Career education in Japan originated in the vocational education system dating back 
to the Meiji Era, and the American-style educational concept has merged to continue to develop its 
unique style. Therefore, I came to the conclusion that the career/vocational education system that is 
unique to Japan may be established by revealing the relationship between career education and 
vocational education. As this idea is in the background, this study tried analyzing and suggesting the 
kind of career/vocational education that is necessary for young people before they become working 
people, based on the background of historical introduction and the approach at the educational site 
with the practical methodology in mind. 
This study consists of a total of 10 chapters from the Chapters I to Ⅹ. 
 
ChapterⅠ Introduction 
 In the Introduction, the theme and the background of the study are outlined, and the awareness of 
the problems, as well as the prior research, are described.

ChapterⅡ Consideration of diversity of "Career" 
 Chapter Ⅱ explains that career is not a term bound by one definition, but implies various things 
due to changes in the external environment. Analysis of the external environment of a career is very 
important, and in that sense, thoroughly looking into the social situation, economic conditions, trends 
of educational policies and employment conditions, it is meant to be treated seriously. Since there is 
a strong tendency in Japan to think that career equals work, it has been found that businesses 
evaluate the disposition and skills of today’s students on their personality and friendship. They do 
not necessarily measure competencies with academic achievement and performance of the students. 
 
ChapterⅢ Losing substance of "Career" Environment 
 The trends of the Ministry of Education, Culture, Sports, Science and Technology (MEXT) in their 
education policies surrounding students is clarified in Chapter Ⅲ. The movement of the Ministry of 
Economy, Trade and Industry (METI) which is in charge of employment policy in Japan is analyzed 
as well. The necessity of career education for students is not only what is required for the students, 
but also there is the aspect that the competent authorities, such as the MEXT and METI, use career 
education as an indicator for measuring the achievement level of government policy implementation. 
As a result, it turned out that the substance of educational content is getting ruined. 
 
ChapterⅣ Initiations by Universities for "Career" of Their Students 
 The concept of career development initiated by universities for careers is analyzed in Chapter Ⅳ. 
At the same time, the report by the Central Education Council 2011 says that career development is 
“a process of realizing a way of life that fulfills our role in society”. “The process of realizing a way 
of living that as desired” does not clarify what it actually means, and there is little related research 
done thus far. In this study, the way of living as desired is determined as a track to follow to realize 
our goal which we set up ourselves. Whether or not you can realize your goal is a big problem if you 
are living independently. It became clear that the very track to follow to reach your goal is the gist of 
career development. 
 
ChapterⅤ Necessity of Career Counseling by Career Counselor 
 In Chapter Ⅴ, the establishment of a career center, placing counselors, and the role to be played by 
counseling, including creation of a curriculum of relevant subjects, is a university’s approach to 
career education. “Depending on the educational objectives of the universities and the faculties, etc., 
the universities need to create organic linkage internally and to establish suitable systems in order to 
cultivate required competencies of the students, so that they can improve their qualities after

graduation to become independent in society and at workplaces. Universities should prepare for 
implementation of the curriculum and the welfare guidance for the students. In other words, 
universities were impelled to practice guidance for social and vocational independence from inside 
and outside the education curriculum, aiming to foster a sustainable working life throughout life. In 
order to make such a thing possible, this study made clear that full-time career counselors are 
necessities for a university. It became clear from the interview with the university’s employment 
department staff that the required skill for a career counselor is not to listen to the students closely, 
but to play a professional role. 
 
ChapterⅥ Review of Career Counseling in Undergraduate Education 
 Chapter Ⅵ focused on career education in undergraduate education. Universities have their own 
degree awarding policy, and have been developing human resources within their frameworks. 
However, it became clear that, logistically, educating students in line with the university’s philosophy, 
from their acceptance to their graduation, is impossible in many cases, due to inconsistencies in the 
organization of the curriculum and implementation of their policies, or discrepancies revealed in the 
admission policy. What this means is that schools publicize various rhetoric on educating their 
students, but the reality is that they are not prepared for career education to realize it. Donald E. 
Super’s Career Rainbow theory was adopted as a career theory in the undergraduate course, but 
Krumboltz’s Planned Happenstance Theory is found to be useful. In other words, a career is like a 
tapestry composed of plans and contingencies, and we are forced to live autonomously in them. 
 
ChapterⅦ Vocational Education Including Financial Education 
 Chapter Ⅶ covers financial education as a part of vocational education which is a close field of 
career education. The author has revealed that the necessity of financial education is increasing more 
than ever when it comes to the career development of modern youth (IJMSBR, Volume7, Issue 8, 
2018). According to the survey conducted by the Central Council for Financial Services Information 
in 2016, the level of financial literacy of the students in Japan is significantly low compared to other 
OECD member countries (as of October 2016, 30 countries). This chapter empirically analyzes why 
financial education, which has shown development different from career education in the form of 
development of financial literacy, should be introduced to career education. 
The present situation and problems of specialized upper secondary school, which is the typical 
institution providing vocational education, are revealed. At the same time, questionnaires about the 
relationship between career education and vocational education were distributed in 637 major 
specialized upper secondary schools in Japan, and 284 effective responses were collected. The

teachers who are in charge of vocational education in the specialized upper secondary schools were 
asked 34 questions in total:  
1. Questions about vocational education at the specialized upper secondary school, 2. Questions 
about career education at the specialized upper secondary school, 3. Questions about “task-oriented 
research” at the specialized upper secondary school. The results of the questionnaire show that the 
teachers do not make a clear distinction between vocational education and career education. 
According to the report, career education is defined as “Education stimulating career development 
aiming at social and career independence for necessary qualifications and attitudes which becomes 
competence based”. Vocational Education is defined as the “Education for young people to acquire 
professional knowledge, skills, qualifications and attitudes to be able to engage in continuous or 
specialized occupation”. Regardless of these definitions, it became obvious that the teachers at the 
education sites have some confusion and no clear distinction between vocational education and 
career education. 
 
ChapterⅧ Practice of Academic-Industrial Collaboration Activities in Career  
Education; Co-operative Education 
 In Chapter Ⅷ, based on the analysis, which is empirical analysis, on the contents of career 
education and vocational education, we examined what kinds of entities or parties should be 
organized to implement both kinds of education. In the discussion so far, one cannot deny that a 
self-righteous and discreet approach of the program has been taken because of the fact that both 
kinds of education have been implemented by educational institutions alone in many cases. In this 
chapter, we demonstrated how the industry-academia or industry-government-university 
collaborative activities will shine the way. In other words, it became clear that many achievements 
can be expected when businesses, schools, and government agencies engage in career education in a 
trinity according to the questionnaire analysis and interviews. The human resources development 
project by cooperative education in academic-Industrial collaboration is analyzed as the method of 
smooth operation of this approach. Cooperative education is a technique to further enhance a 
student’s independence and autonomy, which is also a characteristic function of internship, but it is 
also found out that students’ career design skills will be improved by implementing the program. 
Canada introduced career education later than the United States, and launched the Canadian 
Association for Co-operative Education (CAFCE) to promote career education which is more useful 
than internship in improving students’ career design skills. 60 or more educational institutions 
currently offer cooperative education.

ChapterⅨ New Career Education Adopting Project Management 
Chapter Ⅸ shows an implementation theory on how to carry out career education and vocational 
education. The education is not about reading through a textbook on a desk and learning it. It is 
education for yourself to think about your future life, to design a career for that, to establish the 
foundation you will be standing on independently, and it is a practical general-purpose education as 
well. The differences from the conventional education are observed, and the engineering approach, 
which is the education adopting the project management method, is found to be effective. This idea 
is recognized by the Ministry of Economy, Trade and Industry (METI), and was mentioned in the 
“Career Education Coordinator Training Guidelines” when the Career Education Private Coordinator 
Training and Evaluation System Development Project was formed in the Ministry in 2010. We have 
not seen these ideas for a while, but their usefulness is revealed again in this study. The whole picture 
can be overseen by looking at career education as one of the projects, and management of the entire 
process through the launching, planning, execution, effectiveness and evaluation is necessary. 
 
ChapterⅩ Conclusion: Relationship between Vocational Education and Career  
Education; Proposal to Introduce Industrial Education 
 In conclusion, the relationship between vocational education and career education is reorganized, 
and a new educational system related to career is presented. The approach to educational curriculum 
related to career should be integrated with financial education after clearing up the overlapping part 
of vocational education and career education. A new curriculum should be created with such a field 
classified according to the career development stage of every student. At the same time, as a 
methodology of implementation, the educational process should be monitored visually in adopting 
the project management method. This kind of education requires understanding industries 
comprehensively and being considered as a benefit for students when they make a decision about 
their career in a particular industry, but not so much for the education for career or vocational choice. 
This study named it “Industrial Education” in that sense. 
 However, there is a big problem in realizing industrial education. That is how to negotiate with the 
reality versus the idea. In other words, the question is how sincerely the teachers at educational sites 
can deal with career formation and creation for young people. In the career-path guidance and job 
guidance, the way to encourage students to seek advanced education or to get a job offer for the time 
being, or the extreme tendency to emphasize going to school even if the deviation of its reputation is 
only a little higher, or a leading company should be transformed to an approach with recognition of 
diversity and to support every student’s career design. Since these issues are not discussed in this 
study, future analysis would be the next step for this matter.

審 査 報 告 概 要 
 
大学におけるキャリア教育の重要性は高まっているものの，キャリア教育と職業教育との
間に明確な区別がみられないのが現状である。そこで，本研究では，資料とアンケート調査
をもとに，これら教育の現状について検討した。第 1章～第３章では，キャリアという言葉
の多様性や問題点を浮き彫りにした。第４章～第６章では，大学でのキャリア教育における
キャリア・カウンセラーの有用性を明らかにした。学位授与方針へのキャリア教育の組み込
み方についても議論した。第７章では，職業教育における金融教育の取入れの有用性を示す
とともに，専門高校ではキャリア教育と職業教育に混乱がみられていることを明らかとした。
第８章では，産学連携などのコーオプ教育がキャリア教育の問題点の解決に必要かつ有用で
あることを明らかにした。第９章では，キャリア教育や職業教育を実施する場合，工学的な
アプローチが有効であることを明らかにした。第１０章では，前章までのキャリア教育と職
業教育の関係性を総合的に分析し，そこに金融教育を主導入し，産学連携教育としてのコー
オプ教育とプロジェクト・マネジメント手法，生き甲斐論等を加えた複合的な教育，すなわ
ち，新しい教育体系，“産業教育”を具体的に提案した。 
 このように，本論文はキャリア構造を解析し，生物産業界に入る若者に対しても重要性を
認めるものであり，これらの研究成果等を総合的に評価した結果，審査委員一同は博士（生
物産業学）の学位を授与する価値があると判断した。

「発酵」は古くから食材の保存性の向上や，嗜好性にあう性状に変化させるために食品の
加工・保蔵に利用されてきた。西洋の食文化における保存用食品はベーコン，ソーセージな
どの肉製品とバターやチーズのような乳製品が多いが，畜産物の活用が少ない伝統的な日本
の食生活においては，保存用食品の主な素材は野菜，海藻，魚である。野菜に塩を加えて貯
蔵すると「漬物」となり，広義の漬物には魚介類を原料とした製品もある。魚を食材とした
発酵食品に，福井県の郷土料理である「へしこ」がある。「へしこ」は，塩漬けにしたマサ
バを米糠に漬け込んで 1 年間の発酵熟成期間を経て製品となったもので，発酵後の米糠はペ
ースト状に変化し，米糠を取り除くことなく魚体と共に食されている。通常，米糠は非可食
部として扱われるが，「へしこ」の米糠が供される性状へ変化した要因には，米糠に糖質分
解酵素が作用していることが考えられた。「へしこ」に関する報告は，発酵過程の微生物叢
の変化，発酵熟成に伴う魚肉の遊離アミノ酸，有機酸，脂質などの一般成分の変化について
多くの報告がみられるが，米糠の性状変化に関する報告はみられない。 
そこで本研究では，伝統発酵食品の食品科学的研究として「へしこ」の米糠の性状と性状
変化に関与するアミラーゼ生産菌および生産酵素について明らかにすることを目的とした。
米糠の性状解析として，発酵熟成後の米糠の性状観察と，性状変化に寄与する要因としてア
ミラーゼ生産菌に着目し，発酵熟成期間後の米糠中のアミラーゼ生産菌の検索を行った。分
離した菌株から糖質分解酵素を生産する 5 種の菌株の推定を行ったところ，Oceanobacillus 
属細菌の 1 種である Oceanobacillus picturae と推定された菌株があり，この属の細菌は澱
粉の資化性に関する報告がみられないため供試菌株とした。さらにドラフトゲノム解析およ
びアミラーゼ活性を持つ菌体外生産酵素の解析を行い，目的タンパク質を選択しそれをコー
ドする遺伝子を導入した形質転換体を作成して組換えタンパク質の発現系を構築し，精製し
たリコンビナント酵素を得た。

本研究では，伝統発酵食品「へしこ」の米糠の性状解析とともに O. picturae に関する澱
粉の資化性を示した最初の報告であり，さらに O. picturae のアミラーゼ活性を持つ菌体外
生産酵素について，酵素科学的研究へ展開できる方法を確立した。以上の研究結果を報告す
る。 
 
実験 1：「へしこ」米糠の性状解析およびアミラーゼ生産菌株の分離と菌種の推定 
実験 1-1：発酵前および発酵熟成後の米糠の性状解析 
へしこ（福井県福井市製造）製造に糠床として用いた米糠（コシヒカリ福井県産）と，発
酵熟成期間後（へしこ）の魚体周辺の米糠を測定用試料とした。一般成分分析およびナトリ
ウム含有量を測定した。さらに，米糠を脱脂したのち 60％エタノールに懸濁させ 200μm
のメッシュでろ過し，①残査をグルコアミラーゼおよびプロナーゼ処理した区分（区分Ⅰ：
非澱粉性多糖），②メッシュを通過したろ液を遠心分離し，沈殿をプロナーゼ処理した区分
（区分Ⅱ：澱粉性多糖および非澱粉性多糖の混在区分），③上澄み区分（区分Ⅲ：水溶性低
分子多糖），の 3 区分に分画した。区分Ⅰと区分Ⅱは走査型電子顕微鏡（SEM）観察，示差
走査熱量計（DSC）による熱分析およびアミロペクチンの鎖長分布測定，区分Ⅲは陰イオ
ン交換クロマトグラフィー（HPAEC－PAD），１H－NMR および 13C－NMR(周波数
400MHz)により糖の分析を行った。 
一般成分の変化は，水分，脂質，灰分，Na 含有量ともに増加し，炭水化物は著しく減少
していた。分析により発酵熟成後の米糠で炭水化物含量が減少していることが示された。ま
た SEM 観察により，発酵熟成後の米糠では，非澱粉性多糖の細胞壁の破損が顕著であり，
かつ澱粉粒には穴や欠損部が観察された。 
熱的性質では，区分Ⅰでは 79℃および 110℃に吸熱ピークが得られ，これは SEM 像より
細胞壁成分由来と考えられた。区分Ⅱでは，67℃および 78℃付近に吸熱ピークが得られ，
67℃のピークは澱粉由来，78℃付近は混在している他成分で，細胞壁成分由来であると推
察された。さらにアミロペクチンの単位鎖長分布においても，DP6-12 の短鎖の減少がみら
れたことから，発酵熟成期間に澱粉性多糖は澱粉分解酵素の作用を受けていることが明らか
となった。一方，区分Ⅲにおける糖組成の分析では，発酵熟成後の米糠から澱粉や細胞壁の
構成糖であるアラビノース，ガラクトース，グルコース，キシロースが同定され，以上の結
果より，米糠中の澱粉および非澱粉性多糖は発酵熟成過程で分解されていることが明らかと
なった。 
 
実験 1-2：発酵熟成後の米糠中のアミラーゼ生産菌株の分離と菌種の推定 
米糠中の澱粉や非澱粉性多糖の分解には微生物由来の糖質分解酵素の関与が示唆された
ため，発酵熟成期間を経て製造されたへしこ魚体周辺の米糠からアミラーゼ生産菌株を分離

した。分離されたアミラーゼ生産菌株の中から 5 株を選択し，16S rDNA により菌種を推定
した結果， Bacillus amyloliquefaciens (分類番号 A1 株), Paenibacillus amylolyticus (分
類番号 A3 株), Bacillus subtilis (分類番号 B1 株), Bacillus licheniformis (分類番号 B2 株), 
Oceanobacillus picturae (分類番号 B3 株)であることが推定された。推定された 5 菌株のう
ち，Bacillus 属細菌および Paenibacillus 属細菌は近縁種からも澱粉を資化できる菌種が多
いことが示されたが，Oceanobacillus picturae は，澱粉の資化性やアミラーゼに関する研
究報告がみられず，かつ，近縁種からも澱粉を資化できる菌種がみられないことが示された。 
 
実験 2：O. picturae (分類番号 B3株) のゲノム解析および，菌体外生産酵素の全長の予測 
実験 2-1：O. picturae (分類番号 B3 株) のゲノム解析 
O. picturae (分類番号 B3 株) を供試菌株とした。可溶性澱粉を含む液体培地にて培養し
た供試菌株の菌体からDNAを抽出し，次世代シーケンサで塩基配列を解析した。この結果，
92 コンティグ (約 3.9 Mbp) を決定し，3,889 の CDS を予測した。本菌を Oceanobacill-
us picturae Heshi-B3 と命名して，DDBJ へのゲノム配列登録を行った(BBXV01000001-
BBXV01000092)。 
 
実験 2-2：O. picturae Heshi-B3 の菌体外生産酵素の精製，および目的タンパク質のアミノ
酸配列分析と菌体外生産酵素の全長の予測 
供試菌株を可溶性澱粉を含む液体培地に植菌し，30℃で 72 時間静置培養して遠心分離で
菌体を除去した上清中の酵素を菌体外生産粗酵素溶液とした。その溶液中のアミラーゼ活性
を持つタンパク質の精製を目的として，①限外ろ過法，②硫安分画法，③疎水クロマトグラ
フィー (TOYOPEARL Butyl-650S)，および④陰イオン交換クロマトグラフィー (DEAE 
Sepharose Fast Flow) を行い，⑤遠心濃縮 (VIVASPIN 遠心濃縮チューブ) により濃縮し
た。その精製工程⑤の溶液を二次元電気泳動で分離後，PVDF 膜に転写した各タンパク質ス
ポットを切り出し，プロテインシーケンサを用いてエドマン分解法により N 末端のアミノ
酸配列を決定した。一方，精製工程⑤の溶液を二次元電気泳動で分離後，選択したタンパク
質スポットを切り出し，MALDI-TOF/MS によるペプチド断片の質量分析を行い，その質
量分析データから Mascot 検索で相同性の高いタンパク質を抽出した。 
その結果，複数のタンパク質スポットのうちの 1 つのスポットにおいて，N 末端から 7
残基までのアミノ酸配列を得ることができた。この情報について O. picturae Heshi-B3 と
対応する CDS を検索したところ，N 末端から 25～31 残基目のアミノ酸配列が完全一致す
るアノテーション名：cyclodextrin glucanotransferase precursor が得られた。この CDS
がコードするアミノ酸配列のうち N 末端から 24 残基目まではシグナル配列と予測され，シ
グナル配列切断後のタンパク質の分子量は約 7.6 万，等電点は 4.38 と推定され，二次元電

気泳動の結果と概ね一致した。 
また，MALDI-TOF/MS で解析されたペプチド断片の質量分析結果に対して Mascot 検
索による相同性の高いタンパク質は，O. picturae S1 の alpha-amylase と，O. picturae 
Heshi-B3 の CDS がコードする cyclodextrin glucanotransferase precursor であり，この
2 つのタンパク質のアミノ酸配列は完全に一致していた。さらに，この CDS がコードする
タンパク質の機能を探るために，NCBI サイトでの BLAST による相同性検索を行った。そ
の結果，Bacillus sp. 8SB 株由来の cyclodextrin glucanotransferase precursor と 72 %，
Bacillus trypoxylicola 由来のα-amylase と 70 % の相同性が認められた。従って，この
CDS がコードするタンパク質は糖質加水分解酵素または糖転移酵素の機能を持つことが示
唆されたため，この CDS をアミラーゼ遺伝子 HeshiAMY1 と命名した。 
 
実験 3：アミラーゼ遺伝子 HeshiAMY1の発現系の構築 
アミラーゼ遺伝子 HeshiAMY1 の塩基配列をもとに，シグナル配列を除いた HeshiAMY1 
をベクター (pBIC 3) に導入し， Brevibacillus choshinensis を宿主として，Ｎ末端に His
タグを融合させたリコンビナント HeshiAMY1 を菌体外に分泌させる発現系を構築した。
構築した発現系の培養上清のアフィニティークロマトグラフィー (Ni Sepharose 6 Fast 
Flow) でリコンビナント HeshiAMY1 を精製し，SDS－PAGE で単一バンドであることが
確認された。また，得られたリコンビナント HeshiAMY1 溶液を可溶性澱粉を基質として
作用したところ，還元糖が生成したことから糖質加水分解酵素または糖転移酵素であること
が確認された。 
 
まとめ 
本研究では，「へしこ」の米糠の性状変化を解析し，その性状変化には分離菌株の生産す
る菌体外生産酵素が寄与していることを示した。その分離菌株の 1 つは Oceanobacillus 
picturae と推定され，さらに本菌を Oceanobacillus picturae Heshi-B3 と命名して，DDBJ
へのゲノム配列登録を行った。また，供試菌株のゲノム解析結果と菌体外生産酵素のアミノ
酸配列から，菌体外に生産するアミラーゼのゲノム配列を予測してその発現系を構築し，
Brevibacillus choshinensis を宿主として，発現・精製したリコンビナント酵素
（HeshiAMY1）を得た。これらの成果は，伝統発酵食品の食品科学的研究として資する成
果でありさらに微生物学分野においても O. picturae Heshi-B3 に関する最初の澱粉の資化
性を示し，菌体外生産酵素についてコンビナント HeshiAMY1 の発現系を構築して酵素科
学的研究へ展開できる方法を確立し意義のある成果と考える。

審 査 報 告 概 要 
 
本研究では，「へしこ」の米糠の性状変化を解析し，魚体周辺の米糠からアミラーゼ生産
菌株を分離し，16S rDNA により Bacillus amyloliquefaciens (A1 株), Paenibacillus amylolyti-
cus (A3 株), Bacillus subtilis (B1 株), Bacillus licheniformis (B2 株), Oceanobacillus pictu-rae
 (B3 株)であることが推定され，そのうち Oceanobacillus 属の細菌は澱粉の資化性に関する
報告がなく，澱粉の資化性を示した最初の報告である。さらに本菌を Oceanobacillus pictur-
ae Heshi-B3 と命名して，DDBJ へのゲノム配列登録を行い供試菌株とした。また，供試菌
株の菌体外生産酵素を精製し，ゲノム解析結果と菌体外生産酵素のアミノ酸配列から，菌体
外に生産するアミラーゼのゲノム配列を予測してその発現系を構築し，Brevibacillus chosh- 
inensis を宿主として，発現・精製したリコンビナント酵素（HeshiAMY1）を得た。これら
の成果は，伝統発酵食品の食品科学的研究として資する成果でありさらに微生物学分野にお
いても O. picturae Heshi-B3 に関する最初の澱粉の資化性を示し，菌体外生産酵素について
コンビナント HeshiAMY1 の発現系を構築して酵素科学的研究へ展開できる方法を確立した
観点からも高く評価される。これらの研究成果等を詳細に検討した結果，審査員一同は博士
（食品栄養学）の学位を授与する価値があると判断した。
