  Anthropogenic loads of inorganic nitrogen (N) derived especially from fertilizer use in agriculture, 
livestock and domestic wastes and sewage are significant sources of excess N in the environment, 
which can finally cause severe damage to the natural coastal environment and the marine ecosystems. 
Mangrove, located at the ecotone of the land and sea, might act as a buffer zone by filtering excess N 
from the runoff waters and stabilize the coastal environment and protect the marine ecosystems. 
Despite providing valuable ecosystem services to nature and human beings, mangroves are 
combatting enormous threats all in the territory. In the tropical and sub-tropical maritime nations, a 
significant part of the national economy is based on marine resources. Therefore, conservation and 
management of mangroves are vital for healthy marine ecology, proper growth of coral lives, and 
ecosystem sustainability. The aims of this thesis are to gain better understanding of likely origin and 
relative availability of N inflowing into the mangrove ecosystems of Okinawa and the corresponding 
responses of mangroves under the influences of different magnitudes of anthropogenic activities in 
watersheds. 
 
1. Foliar δ15N value and its relationship with land-use ratio in the mangrove watersheds 
We collected foliar samples of mangrove and back-mangrove species from fifteen mangrove 
watersheds on Iriomote, Ishigaki, and Okinawa islands. Land-use ratio delineation through ArcGIS 
revealed that Urauchi watershed was the most pristine environment with 97 % of forest covers and 
was used as a reference site in this study. The comparison of foliarδ15N values among watersheds 
on the three islands revealed that all the watersheds on Iriomote Is. were almost in a natural 
condition having δ15N less than 3 ‰, whereas those ranged from 5 to 14 ‰ in watersheds on 
Okinawa and Ishigaki islands, which was an indication of heavier human disturbances and changes 
in the natural ecosystems. In addition, Todoroki watershed on Ishigaki Is. showed the highest value

of δ15N (around 14 ‰), suggesting an intrusion of excess N in the environment through inland 
runoff from livestock farms. The results suggested that foliar δ15N values of mangroves were 
powerful ecological indicators for monitoring ecosystem conditions of Okinawa. Furthermore, the 
findings indicated that mangroves were potentially absorbing anthropogenic N from the ecosystems. 
 
2. Adaptation of acetone methods forδ15N-NO3- analysis of saline water 
For identifying N sources in NO3- in mangrove surface water samples, an adapted acetone method 
was developed for δ15N-NO3- analysis of saline water. Literature review on the pre-existing 
methodology for the determination of δ15N-NO3- suggested that widely used protocols have several 
drawbacks, such as sensitivity to the sample type (i.e., freshwater or saline water), requirement of a 
large volume of sample containing high concentration of NO3-, labor-intensive and time-consuming 
handling procedures, toxic substances, costly devices and especial bacterial culture, which is not 
available in many cases. Therefore, the adapted acetone method was developed for isolation of NO3- 
from saline water containing a low concentration of NO3- (4 µmol in an aliquot) forδ15N-NO3- 
analysis. We elucidated the usability of the adapted acetone method in the determination of 
δ15N-NO3- in saline water (containing less than 6 g of Cl- in sample aliquot) with an acceptable 
range of precision and accuracy (< 0.1 ‰). The adapted acetone method was based on a modification 
of a solvent mixture (acetone/NaI/n-hexane as 21/0.35/10 mL) of the previous acetone method to a 
new mixing ratio as acetone/NaI/n-hexane as 30/0.25/5 mL and bringing a change in sample 
reduction and drying processes. Consequently, the adapted acetone method successfully removed the 
contaminants from the saline samples, such as organic matter and excess chlorides, and resulted in a 
satisfactory analysis ofδ15N-NO3- in surface water samples collected from the mangrove 
watersheds. 
 
3. Nitrogen availability, sources of surface water NO3- and their relationships with foliar δ15N 
and land-use 
A conventional approach for measuring concentration of dissolved N (DIN, DON) in surface 
water and soil pore water in six watersheds on Ishigaki and Iriomote islands, was carried out to 
investigate the relative availability of N. Besides, the adapted acetone method was used to 
investigate the source of NO3- in the surface water samples. A larger concentration of DIN, especially 
NO3-, along with higher values ofδ15N-NO3- of the surface water samples, ranging from 7 to 14 ‰

in the human affected watersheds compared to those of the reference site (less than 4 ‰), revealed 
that anthropogenically originated N were influencing the N dynamics in the systems. In addition, 
mangrove leafδ15N values were significantly correlated with the concentration of NO3- and 
δ15N-NO3- values of the surface water, and the total forested areas in the watersheds, suggesting that 
the nutrient concentrations influenced by the land-use were responsible for the particular isotopic 
signature of leaf and water nitrates. In contrast, the DIN concentration of soil pore-water samples 
was not significantly correlated with δ15N-NO3- and foliarδ15N values. Furthermore, along with 
DIN a considerable amount of DON was also observed in both surface and soil pore water in the 
mangrove ecosystems on Ishigaki Is., which was not shown in the case of mangrove watersheds on 
Okinawa Is. We assumed that higher concentration of DON in the study sites on Ishigaki Is. was due 
to livestock farms frequently found in the watersheds. 
 
4. Soilδ15N and its relationships with foliar δ15N, surface water δ15N-NO3- and land-use 
Two sets of samples, the former collected from mangrove watersheds on Okinawa and Iriomote 
islands in 2017 and 2018, and the latter in 2019, were used for analytical experiments on DIN, 
foliarδ15N of mangroves and back-mangroves, and the floor soils, and δ15N-NO3-of surface water. 
It was assumed that the results obtained from the two sets of samples would provide a better 
understanding of how mangroves responded to excess N, and whether the response would change in 
time. The concentration of DIN in the two sets of samples was distinct and relatively higher in the 
former set. Still, anthropogenic N inflowing into the mangrove ecosystems was evident from the 
elevated values of δ15N-NO3- of surface water samples in the former set, the higher soilδ15N values 
in the latter set, and their significant correlation with land-use ratio in the watersheds. The δ15N 
values of the soils collected from both the mangrove and back-mangrove vegetative areas were 
compared among the study watersheds, and significantly higher values (from 4.7 to 7.3 ‰) were 
observed in all the human-influenced watersheds than those in the reference site (0.9 ‰). 
Additionally, significant positive correlations between the foliar mangrove δ15N and the floor soil  
δ15N, between the foliarδ15N and the surface water δ15N-NO3-, and between the floor soilδ15N 
and the surface waterδ15N-NO3- indicated anthropogenically derived N uptake by the mangrove 
plants and reduction of excess N from the mangrove ecosystems.

主として農業で使用される肥料，畜産廃棄物，下水，生活排水に由来する無機窒素の人工
的負荷は，環境に放出された過剰窒素の重要な源で，最終的に沿岸環境や海洋生態系に深刻
な被害を引き起こす。陸と海との移行帯に位置するマングローブ生態系は，緩衝帯として，
陸からの流出水に含まれる過剰な窒素を取り除き，沿岸環境を安定化し，海洋生態系を保全
している可能性がある。自然や人類に貴重な生態系サービスを提供しているのにも関わらず，
マングローブは危機的状況にさらされている。国家経済の重要な部分を海洋資源に依存する
熱帯・亜熱帯の海洋国家において，マングローブの保全と管理は，健全な海洋生態やサンゴ
礁生物の生育ならびに生態系の持続性にとって重要課題である。 
本論文の主な目的は，人工的な影響度合の異なるマングローブ流域における，葉中窒素，
表流水中硝酸態窒素，および林床土壌中窒素の由来を推定することである。そこで本研究で
は，まずマングローブならびにバックマングローブ樹種の葉中窒素の安定同位体比，表流水
中硝酸イオンの窒素安定同位体比，ならびにマングローブならびにバックマングローブ樹種
が生育する土壌の窒素安定同位体比を測定した結果から，対象とした沖縄地域におけるマン
グローブ生態系による窒素削減について明らかにした。 
 
1. マングローブ流域における葉中窒素の安定同位体比と土地利用の関係 
西表島，石垣島および沖縄島に属する 15 のマングローブ流域から，マングローブ樹種葉
とバックマングローブ樹種葉を採取した。これらの流域を対象に ArcGIS を用いて土地利用
実態を解析した結果，浦内流域が森林被覆率 97％でもっとも清浄であったことから，本研
究における対照流域とした。上記 3 島の研究対象流域における葉中窒素の安定同位体比を比
較した結果，西表島のすべての流域ではほとんど自然状態に近かった（窒素安定同位体比は
3‰以下）のに対し，沖縄島および石垣島の流域における同値は 5～14‰と高く，自然生態
系に対する人為的攪乱や人為的変化がより大きいことを示した。加えて，石垣島の轟流域で
は最高値（約 14‰）が示されたが，このことは畜舎に由来する表流水を通じ，過剰窒素が
環境中に浸入していることを推測させた。これらの知見は，マングローブ葉中窒素の安定同
位体比が沖縄地域の生態系の現状をモニタリングする生態インディケーターとして有用で
あることを明らかにし，またマングローブは人為由来窒素を吸収する潜在力が強いことを示
唆した。 
 
2. 汽水試料を対象とした硝酸態窒素安定同位体比測定のためのアセトン法の改良 
好気的な環境においてもっとも活性の高い無機窒素は硝酸イオンで，また多くの人為的行
為は環境中の同イオン濃度を増進させることから，上記の結果に加え，流域中の硝酸態窒素
の由来を確認することが必要と考えられた。ところが，硝酸態窒素の安定同位体比を測定す
る主要でかつ広範に用いられている方法はいくつかの欠点がある。いくつかの方法は試料の

種類（例えば淡水か塩水か）に敏感で，またいくつかの方法は，大量の試料と高い硝酸態窒
素濃度，あるいは多くの労力や時間のかかる処理や有毒な薬剤を要求したり，さらには高価
な機材や特有の微生物培養を必要とする。そこで本研究では，低濃度の硝酸イオン（4 μ
mol）を含む汽水から硝酸イオンを単離するための方法（改良アセトン法）を開発した。そ
の結果，溶液中の塩化物イオン量が 6g 以下であれば，同改良法を硝酸イオンの単離法とし
て用い，硝酸態窒素の安定同位体比を測定した値の確度は 0.1‰以下で容認範囲であること
を確認した。同改良法は既往のアセトン法における溶媒混合比（アセトン/NaI/n-ヘキサン：
21/0.35/10 mL）を 30/0.25/5 mL に改変し，試料の還元と乾燥方法に変化を加えた。同改良
法は汽水試料から有機物や過剰な塩化物などの妨害物質を効率的に取り除き，マングローブ
流域から採取した表流水中硝酸態窒素の安定同位体比をより正確に測定することに成功し
た。 
 
3. 可給性窒素および表流水中硝酸イオンの由来と葉中窒素の安定同位体比および土地利
用との関係 
 石垣島と西表島に属する 6 つマングローブ流域を対象に，表流水と土壌溶液中の可給性窒
素量を確認するために，DIN（溶存性無機窒素）と DON（溶存性有機窒素）の濃度を測定
した。加えて，改良アセトン法を用い，表流水中硝酸態窒素の由来を推定した。人為影響の
強い流域では，高濃度の DIN，硝酸イオンが検出されたとともに，表流水中硝酸態窒素の
安定同位体比は，対照流域（4‰以下）に比べ 7～14‰と高く，人為活動に由来する窒素が
マングローブ生態系における窒素動態に強く影響していることが明らかになった。加えて，
マングローブ葉中窒素の安定同位体比は，表流水中の硝酸イオン濃度と硝酸態窒素の安定同
位体比，ならびに流域中の森林面積率と有意な正の相関関係が示されたことから，土地利用
の影響を受けた無機窒素濃度は，樹種葉や表流水の安定同位体比の原因となっていることが
推察された。他方，土壌溶液中の DIN 濃度は，表流水中の硝酸態窒素の安定同位体比やマ
ングローブ樹種葉中窒素の安定同位体比との間に有意な相関関係が示されなかった。さらに，
石垣島のマングローブ流域においては，沖縄島の流域での結果とは異なり，DIN に加え多
量の DON が表流水と土壌溶液の双方で検出されたが，このことは石垣島の調査流域内で散
見された畜舎に起因するものと推察された。 
 
4. マングローブ林床土壌の窒素安定同位体比と，樹種葉中窒素，表流水中硝酸態窒素の
安定同位体比ならびに土地利用との関係 
DIN 濃度，マングローブ，バックマングローブ樹種葉中窒素および林床土壌中窒素の安
定同位体比，および表流水中硝酸態窒素の安定同位体比を測定するために，2 回の時期に分
けて試料を沖縄島のマングローブ流域から採取した。第 1 回目の採取は 2017 年から 2018

年にかけて，第 2 回目は 2019 年に行った。これらの測定結果から，マングローブの過剰窒
素に対するさらなる応答や，その応答の時期による違いに関する知見が得られた。DIN 濃
度は 2 回の調査期間により明瞭に異なり，第 1 回目で高かった。さらに，人為活動由来の
窒素がマングローブ生態系に流入していることが，第 1 回目の調査では表流水中硝酸態窒素
の安定同位体比により，また第 2 回目では林床土壌中窒素の安定同位体比により示された。
マングローブ，バックマングローブのそれぞれの植生域別で林床土壌中窒素の安定同位体比
を比較した結果，人間活動が関与する流域の値（4.7～7.3‰）は，対照流域の同値（0.9‰）
より有意に高かった。加えて，マングローブ葉中窒素とその林床土壌中窒素の安定同位体比
との間，ならびに林床土壌中窒素と表流水中窒素の安定同位体比との間に有意な正の相関関
係が示されたことから，マングローブ樹種は人為由来窒素を吸収し，マングローブ生態系か
ら過剰な窒素を削減していることが推察された。 
 
 
審 査 報 告 概 要 
 
熱帯・亜熱帯の陸海境界に発達するマングローブ生態系は，津波をはじめとする波浪や風
潮害から内陸を保護する，内陸からの土砂流出を軽減するなど，多様な生態系サービスを提
供している。本研究はそれらの機能のなかで，内陸で人為的に生成される窒素の吸収除去機
能について，窒素安定同位体研究手法を援用し，主に次の新知見を得た。1）マングローブ
葉中窒素の安定同位体比は流域生態系をモニタリングする生態インディケーターとして有
用であることを示した。2）高塩分濃度かつ低硝酸塩濃度の液体試料を対象とした窒素安定
同位体比測定のために，既往の測定方法を改良し，その精度が高いことを確認した。3）葉
中，土壌中，土壌孔隙水中，ならびに表流水中の窒素，および土地利用実態を関数とした相
関関係を調べた結果から，マングローブ樹種は人為由来窒素を吸収し，マングローブ生態系
から過剰な窒素を削減していることを明らかにした。以上のように本研究はマングローブ生
態系によるバイオフィルター機能の一端を明らかにするとともに，有用な分析法を開発した
貢献も大きいことから，審査員一同は，博士（国際農業開発学）の学位を授与する価値があ
ると判断した。

Afghanistan is regarded as one of the poorest countries due to the long lasting wars and conflicts, 
in addition to frequent attacks of drought or heat. The reconstruction of agricultural sector is, thus, 
the urgent priority. Vegetables are especially important to improve health condition of infants, 
elderlies, pregnant women, and so on which require highly nutrient dishes. The author investigated 
the current situation of tomato production and postharvest handlings in east provinces in 
Afghanistan, and tried to develop a new appropriate way to prolong postharvest freshness using 
green mature fruit being kept under higher temperatures. 
Tomato (Solanum lycopersicum L.) is an important crop in Solanaceae family which has been 
widely used in both fresh and processed consumption patterns (Harvey et al., 2003), and its 
production has increased to approximately 182 million tons by 2017 (FAOSTAT, 2019) and shares 
16% of total horticultural production in Afghanistan. Tomato contributes significant nutritional 
components to human health (Stommel, 2007), and has been documented as foods with potential 
chemo-preventive activities against several chronic diseases because of the high levels of lycopene 
and other bioactive compounds (Giovannucci, 2002). Maturity stage of tomato fruit at harvesting 
time is one of the important factors for storage life and final fruit quality (Alam et al., 2006). In 
developing countries, farmers usually harvest tomato fruit at the ripe stage, while it is considered that 
ripen fruit is easily damaged and resulted in shorter shelf-life (Watkins, 2006). Since tomato fruit is 
perishable (Javid Ullah, 2009), and contains a large quantity of water, it eventually leads to severe 
post-harvest losses in some cases. 
The shortage and instability of electricity and lack of proper cold storage are sometimes shelf life 
limiting factors for several fruits and vegetables in Afghanistan. In addition, because tomato is cold 
susceptible, low temperature storage might decline the fruit quality (Soto et al., 2005). 
One of the possible solutions for these challenges to minimize tomato fruit damage during

transportation and to restart ripening in retail channels is to harvest tomato fruit at green mature 
stage. Lurie and Sabehat, (1997) reported that the storage of green mature tomato fruit at 38 0C prior 
to 2 0C storage could remove chilling injury development for up to 30 days without causing heat 
injury. Ogura (1975) stated that the storage of green mature tomato fruit at 33 0C for 5-33 days could 
prolong its postharvest freshness even when it was shifted to room temperature, though he did not 
show the physiological mechanism. 
Since the limited researchers have investigated on post-harvest handling and physiology of green 
mature tomato fruit under high-temperature storage, this study aims to evaluate the physiological 
characteristics of green mature tomato fruit to determine the most suitable temperature and duration 
of high temperature storage. 
The results suggest that the suppression of color development of green mature tomato fruit 
occurred during storage at higher temperatures; among the tested higher temperatures as 25, 29, 33, 
and 37 0C, the 37 0C inhibited the color formation most comparing to the other temperatures, even 
after shifting to 25 0C.  
The weight loss of the tomato fruit was the lowest at 25 0C, and the higher the temperature, the 
more the weight loss.  
The decreasing of tomato fruit firmness was faster when stored in higher temperatures, but it 
rather initiated to increase little by little after 1-month storage. The fruit peel seemed to be harder 
after losing some amount of water. It might be so-called “concentration effect,” where some water 
was lost and the fruit became harder.  
Ethylene production was inhibited by the storage under 33 0C for a variety of durations in 
comparison with 25 0C. The ethylene production was suppressed at higher temperatures, but 
whenever shifted to 25 0C, it rapidly increased in case of 10, 20, and 30 days storage at 33 0C.  
The result of respiration was similar to the findings of Ogura et al. (1976); 33 °C storage before 
shifting to 25 0C resulted in the suppression of respiration in tomato fruit.  
Brix value was lowest in the fruit stored at 25 0C and the higher the storage temperature, the 
higher the Brix value. It may well be attributed to the “concentration effect”; the more water was 
lost, the higher the relative sweetness.  
Effect of high-temperature storage on ascorbic acid of green mature tomato stored in 25, 29, 31, 
33 and 35 0C after harvest, the 33 0C storage showed the best among others.  
Ethylene production was inhibited by the storage under 33 0C in comparison with 25 0C along 
with the decreased level in ACC content and the deactivation of ACC oxidase.  
Based on the above results, the author suggests that to harvest green mature tomato fruit can be 
practical to bear long time transportation to sell them to far distant market, while it is better to 
harvest at the fully ripe stage tomato fruit to maximize nutritional value in case of local markets. 
This work might contribute to the harvesting operation in summer season and help the 
storekeepers to enhance their income and ensure food security through reduction in post-harvest 
losses.

文要要旨 
アフガニスタンは長年の戦争によって国土が荒廃し、世界の最貧国の一つに数えられている。
それゆえ農業基盤の再建は急務の課題であり、特に緑黄色野菜の供給が、乳幼児や高齢者、周産
期女性とはじめとする人々の健康維持にとって重要である。著者はアフガニスタンの園芸作物の
中で１６％のシェアを占めるトマトの栽培状況、収穫後の取扱いについてフィールド調査を行
い、高度な貯蔵施設を必要としない緑熟期トマトの高温貯蔵について研究を行った。緑熟期のト
マト果実は、機械的な損傷に対する抵抗性が高く、アフガニスタンのようなインフラストラクチ
ャーが未整備の途上国では輸送中のロスを軽減するのに有利である。 
著者はまず、緑熟期トマトの貯蔵性について、２５℃、２９℃、３３℃、３７℃、４１℃で貯
蔵実験を行い、３３℃貯蔵が最もすぐれていることを見いだした。 
 緑熟期トマトを３３℃で２０日以上貯蔵すると、エチレンの前駆体である ACC含量や ACCオキ
シダーゼ活性が低下し、エチレン生成が抑制されるが、これらの果実を２５℃に戻すと、エチレ
ン生成能力が回復し、果実の成熟が再開することが明らかとなった。ただし、７０日以上の長期
貯蔵では、２５℃に戻しても、エチレン生成能力は回復しなかった。 
果実の糖度と酸度は３３℃貯蔵の方が２５℃貯蔵よりも高くなったが、これは水分損失が大き
いことによる濃縮効果であると考えられた。３３℃貯蔵により、水分損失ははじめ大きいものの、
２０日以上の貯蔵である程度水分を失うと果皮の硬度が増加するため、２５℃貯蔵との差はわず
かであった。糖や有機酸含量が増加したのは、いわゆる濃縮効果によるものと考えられた。 
果実の外見に関しては、３３℃以上の高温で貯蔵した場合、２５℃に戻しても、赤色色素の蓄
積は抑制され、果実は黄色がかった色となり、カロチノイド合成系の中でも、高温で抑制される
経路とそうでない経路があると推察された。 
これらの研究の結果、輸送や貯蔵中における物理的損傷が少ない緑熟期トマトを収穫して高温
下で半年以内を目安に貯蔵を行い、市場価格や流通量にあわせて出荷することが実用的観点から
推奨されるといえる。本研究の成果は、低温貯蔵施設を有しない地方の小規模な農家でも利用可
能な技術であり、アフガニスタンのような発展途上国におけるトマトの収穫後貯蔵･輸送技術の
改善、農業所得の向上、栄養環境の改善に対する貢献が期待される。

序 章 
本研究は，ヤムイモ（Dioscorea spp.）における生物的窒素固定の推定をもとに吸収窒素の
由来と共生微生物の存在を明らかにしたものである。 
ヤムイモはアフリカ，南アメリカ，アジア，熱帯太平洋諸国において，主作物として供給
されている。今後人口増加が予想される開発途上国を中心とした熱帯・亜熱帯地域において
ヤムイモの生産性を改善することは極めて重要である。しかし，近年アフリカにおけるヤム
イモの収量の増加はわずかであり，生産性の向上を目指した肥培管理技術の整備が必要であ
る。 
多くの先行研究はヤムイモの幅広い環境適応能力を指摘し，栽培環境や栽培方法及び品種
や系統によって生産に必要な施肥量が異なることを示している。低肥沃土壌条件下での適応
能力に優れたヤムイモ系統が存在する要因として，生物的窒素固定による独自の窒素吸収特
性が生育に関与していると推測された。これを実証するためにヤムイモへの窒素供給システ
ムの解明と共生窒素固定細菌の存在を明らかにすることを目指した。 
本論文では，ヤムイモの中でも主要な栽培種であるダイジョとホワイトヤムについて，窒
素固定能を明らかにし，植物体から分離した共生細菌について窒素固定活性の可否の推定と
近縁種細菌の決定を行った。さらに，窒素固定能の品種・系統間差異を明らかにして，アフ
リカの低肥沃土壌条件下に栽培可能な品種の育成並びに肥培技術の開発に資する基礎知見
を示した。 
 
第 2章 δ15N を用いたダイジョの窒素固定能の評価 
ヤムイモの幅広い環境適応能力や，系統によって施肥効果が異なる要因として窒素固定に
注目した研究はこれまでになく，大気由来の窒素の吸収についての詳細は不明である。本章

では，ヤムイモにおける窒素吸収源の推定および窒素固定能について検証することを目的に
ダイジョを沖縄県宮古島市で栽培し，植物体の重窒素自然存在比（δ15N）の測定を行った。 
ダイジョ A-19 系統を無施肥栽培し窒素吸収源の検証を行った結果，葉の δ15N は植えつけ
後 60 日目に+4.58‰を示したが生育に従って減少し，植えつけ後 180 日目には+2.33‰を示
したことからダイジョの植物体が空中窒素を取り込んでいることを確認した。さらに，植え
つけ後 120 日目においてカボチャを対照とし窒素固定寄与率を算出した結果，カボチャの
δ15N は+4.18‰であったのに対しダイジョの δ15N は+2.59‰と低く，ダイジョの窒素固定寄与
率は 38.04%であると推定された。また施肥をした場合，ダイジョ A-19 系統は肥料の種類に
関わらず与えた肥料の窒素成分を優先的に吸収するが，施肥の肥効が低下すると空気中の窒
素を固定している可能性が示された。 
 
第 3章 ダイジョにおける共生窒素固定細菌の単離と近縁種決定 
第 2 章では，ダイジョの生育に従って植物体の δ15N が減少することから，大気由来の窒
素を取り込んでいることを明らかにした。このことから，ダイジョ植物体に窒素固定菌が内
生している可能性が考えられた。そこで本章ではダイジョの共生窒素固定細菌の探索を目的
に，培養法を用いて無施肥栽培したダイジョ A-19 系統の植物体中に内生している細菌の単
離を行い，分離された細菌株についてアセチレン還元法（ARA 法）により窒素固定能の有
無を調べ，16S rRNA 遺伝子解析を行い属レベルでの近縁種の決定を行った。 
2014 および 2015 年に調査を行った結果，ダイジョ A-19 系統の茎および根から，計 13 菌
株の ARA を示す細菌を分離することに成功した。各分離株の 16S rRNA 遺伝子の部分配列
をシークエンスした結果，これらの分離株は Bacillus，Cohnella，Kosakonia，Azospirillum，
Pantoea，Dyella，Enterobacter 属に加え，根粒菌である Rhizobium および Ralstonia 属と近縁
であった。本実験により分離された窒素固定細菌と近縁な細菌種は，2014 年に分離された
細菌株と同じ属のものもあったが，系統樹解析の結果いずれも相同性が低かったことから，
種イモを介して継代的に感染することは確認できなかった。 
 
第 4章 ナイジェリアのヤムイモにおける共生窒素固定細菌の単離と近縁種決定 
第 3 章の実験から，ダイジョが窒素固定能を持つ細菌と共生していることが確認された。
しかし，西アフリカのヤムイモ栽培種も同様の能力を持つかは未知である。そこで第 4 章で
は，アフリカ産のダイジョに加え，世界で最も生産量が多いホワイトヤムにおける窒素固定
の可能性を検討した。実験はナイジェリアの国際熱帯農業研究所（International Institute of 
Tropical Agriculture: IITA）で砂質栽培圃場および休閑森林土壌で栽培されたダイジョ 2 系統
およびホワイトヤム 8 系統を用いて ARA を示す細菌の分離を試みた。 
半流動培地を用いた植物切片の培養試料はホワイトヤム 8 系統すべてで茎，根ともに強い

ARA 活性を示した。ダイジョでは TDa02/1001,2003, 3001 では茎，根ともに活性を確認でき
たが，TDa00/1001,2003, 3001 では茎で認められたものの，根では認められなかった。このこ
とから同じ圃場で栽培されたヤムイモの系統間で共生細菌の種類が異なる可能性が示唆さ
れ，また，窒素固定細菌と共生度合いが低いヤムイモ系統が存在することが推察された。 
次に各ヤムイモ系統の茎，根それぞれの植物切片を接種した培地試料を用いて共生細菌の
単離を行った結果，ダイジョ TDa00/1001,2003, 3001 の根以外のすべての試料においてコロ
ニー形成を確認した。各分離細菌株の ARA 測定に供した結果，計 19 菌株で ARA を示し，
それらは Kosakonia，Pseudacidovorax，Klebsiella，Azospirillum，Pseudomonas，Mycobacterium，
Xanthobacter，Bosea，Rhizobium，Stenotrophomonas および Azoarcus 属と近縁であった。ホワ
イトヤムから窒素固定能を持つ細菌の分離に初めて成功した。 
 
第 5章 窒素の安定同位体を利用したホワイトヤムの窒素固定能の評価 
第 4 章でナイジェリア産のダイジョ，ホワイトヤムにおいても窒素固定能を持つ細菌との
共生が確認され，これらの細菌が植物生育促進能を持つことを期待された。ダイジョのよう
にホワイトギニアヤムも窒素固定が生育に関与している可能性が考えられたことから，ナイ
ジェリア産ホワイトヤム 6 系統について δ15N を用いて窒素固定能について調査した。 
ナイジェリアの IITA において，ホワイトヤム 6 系統を用いて圃場栽培を行った。葉の δ15N
を測定した結果，茎葉繁茂期における無施肥栽培の 6 系統の葉の δ15N は 1.38～3.49‰を示し
た。δ15Nは対照作物であるカボチャが 3.38‰で，ホワイトヤムの DrDRS074 および DrDRS020
系統がカボチャよりも低かった。窒素固定寄与率は 45.5～-4.7%と系統間で差がみられ，大
気由来窒素の集積量は DrDRS074 で最も多く 0.52 g/ plant DW であった。以上の結果から，
ホワイトヤムでは窒素固定能の系統間差異が認められた。さらに，茎葉繁茂期における窒素
固定寄与率と大気由来の窒素集積量は地上部乾物重と強い正の相関を示した。一方で，窒素
固定寄与率および大気由来の窒素集積量と 12 月の塊茎乾物重との間に有意な相関関係はな
かった。このことから，ホワイトヤムにおける窒素固定は特に茎葉繁茂期の地上部生育に寄
与していることが考えられた。 
以上の結果から，ダイジョ及びホワイトヤムの生育に窒素固定細菌の共生が関与すること
が明らかになり，ヤムイモの栽培における窒素固定細菌の利用に道を開くものと期待される。

際農業開発学）の学位を授与するに値すると判断した。
