    The Role of Interferon-β in the Treatment of Multiple Sclerosis

    Multiple sclerosis (MS) is a progressive neurodegenerative disorder that affects 140 out of every 100,000 individuals in North America. MS is believed to be mediated by T cells, in which the T cells directly attack or indirectly activate immune cells for attack on the myelin sheath of neurons, which can lead to blindness, paralysis, muscle weakness, and walking difficulties. Treatment of MS involves the use of immunomodulatory therapeutics, such as interferon-β (IFN-β), to decrease the progression of demyelination within the CNS. IFN-β has been linked to many different immunomodulatory roles in the pathology of MS. Of interest, IFN-β is believed to alter T-cell activation, cytokine production, and transmigration of immune cells. Studies have found that IFN-β increases co-stimulatory molecule production in dendritic cells and monocytes. IFN-β has also been found to alter various cytokine expressions within immune cells, such as T-cells and dendritic cells. In addition, IFN-β inhibits immune cell movement through the blood-brain barrier. These results taken together suggest a complex inhibitory system, propagated by IFN-β signaling, that may reduce the functionality of T cells and dendritic cells, such that T-cells have reduced ability to mediate neuronal attack in cases of MS

    Assessing Combinability of Phylogenomic Data Using Bayes Factors

    With the rapid reduction in sequencing costs of high-throughput genomic data, it has become commonplace to use hundreds of genes to infer phylogeny of any study system. While sampling a large number of genes has given us a tremendous opportunity to uncover previously unknown relationships and improve phylogenetic resolution, it also presents us with new challenges when the phylogenetic signal is confused by differences in the evolutionary histories of sampled genes. Given the incorporation of accurate marginal likelihood estimation methods into popular Bayesian software programs, it is natural to consider using the Bayes Factor (BF) to compare different partition models in which genes within any given partition subset share both tree topology and edge lengths. We explore using marginal likelihood to assess data subset combinability when data subsets have varying levels of phylogenetic discordance due to deep coalescence events among genes (simulated within a species tree), and compare the results with our recently described phylogenetic informational dissonance index (D) estimated for each data set. BF effectively detects phylogenetic incongruence and provides a way to assess the statistical significance of D values. We use BFs to assess data combinability using an empirical data set comprising 56 plastid genes from the green algal order Volvocales. We also discuss the potential need for calibrating BFs and demonstrate that BFs used in this study are correctly calibrated

    Relationship of Head Circumference and Epilepsy Severity in Infants with Tuberous Sclerosis Complex

    Tuberous sclerosis complex (TSC) is a rare genetic disorder, characterized by high incidence of seizures and tubers (benign cellular abnormalities that appear on brain imaging). Current literature reports an increased head circumference within the TSC population, but the implications, exact rate, or how big the increase is, remains unknown. We hypothesized that increased head circumference would correlate with epilepsy severity in infants with TSC. We examined clinical data from 121 infants diagnosed with TSC who were enrolled in the multi-site, prospective TSC autism center for excellence network (TACERN) study. We calculated each infant’s head circumference z-score from each study visit by comparing collected values with those reported by the World Health Organization of healthy child head circumference growth. The overall mean head circumference z-score across all study visits of all TACERN participants was 0.97 with no significant sex differences. The number of seizure types correlated with head circumference z-score are as follows: infants with no seizures had a mean z-score of 0.57, those with 1 seizure type had a mean z-score of 0.88, and those with 2 or more seizure types had a mean z-score of 1.31. Thus, our hypothesis was partially supported. This data suggests that a larger head circumference may serve as a biomarker for epilepsy in children diagnosed with TSC, enabling earlier treatment for epilepsy, and thus improved neurodevelopment
