Long-term NRF2-driven microglial repopulation mitigates microgliosis, neuronal loss and cognitive deficits in tauopathy
Tauopathies, including Alzheimer’s disease, feature chronic microglial reactivity that drives neuroinflammation and disease progression. Pharmacological microglial depletion and subsequent repopulation using colony- stimulating factor 1 receptor inhibitors have emerged as a potential therapeutic strategy to reprogram dysfunctional microglia. Despite promising short-term results, the long-term efficacy and pharmacological modulation of repopulated microglia remain poorly understood. Here, we investigated the long-term effects of microglial repopulation alone and in combination with the activation of the cytoprotective nuclear factor erythroid 2 p45–related factor 2 (NRF2) in an in vivo AAV-hTau P301L induced model. Integrating different behavioural, immunohistological and transcriptomic analysis, we evaluated cognitive function, tau pathology, neuronal survival and glial reactivity. We found that, whereas microglial repopulation alone did not significantly affect disease progression, NRF2-driven microglial replenishment sustained cognitive function, prevented hippocampal neuronal loss and restored microglial phenotype. Transcriptomic analyses further revealed that the combined treatment modulated tau- associated mitochondrial gene expression changes. These results highlight the importance of shaping the fate of self-renewed microglia and propose NRF2-mediated microglial repopulation as a potential pharmacological strategy for the treatment of tauopathiesThis work was supported by the Spanish Ministry of Science, Innovation and Universities (PDC2022-133809-I00 and PID2021- 125986OB-I00 to MGL; PID2021-122650OB-I00 to MIRF; PID2022- 139936OA-I00 to EN and FPU20/03747 fellowship to LV); and the General Council for Research and Innovation of the Community of Madrid (P2022/BMD-7230 to MGL and 2020-T1/BMD-19886 to ET
The study is structured around three core areas. The first examines various theoretical frameworks that support the fundamental distinction between criminal unity and concurrence of offences. Within the domain of concurrence, it further explores the need to differentiate the penal consequences associated with its various forms. The second part seeks to establish the criteria for distinguishing between the two principal forms of concurrence: ideal concurrence and real concurrence. The third and final part analyses the different doctrinal positions regarding how the rules for determining penalties should be structured in each of these forms.
Each section offers a critical assessment of the prevailing academic and jurisprudential debates, either by substantiating existing positions or by advancing alternative interpretations. In all instances, the primary objective is to achieve a regulatory approach that is as rational, coherent, and lenient as possible within the broader penal syste

Exploring efficient attention strategies in conformer-based sound event detection
Sound Event Detection (SED) requires models that can accurately localize and classify overlapping audio events within complex acoustic environments. Conformer-based architectures have demonstrated promising performance by leveraging self-attention to capture long-range dependencies. However, this global attention can be accumulated across layers, which can blur local temporal boundaries and reduce detection accuracy, especially for short or closely spaced events. While increasing the input sequence length can help recover temporal detail, the quadratic complexity of Conformers’ self-attention significantly increases computational costs. To address this, we propose integrating the Efficient Conformer architecture, which introduces subsampling along the input sequence length, effectively reducing the temporal dimension within blocks. This design enables processing longer input sequences at finer temporal resolution, enhancing localization accuracy without extending output length. Using the DCASE Challenge 2023 Task 4 benchmark, system performance is evaluated via the threshold-independent Polyphonic Sound Detection Score (PSDS), measuring both localization precision (PSDS1) and class robustness (PSDS2). Experiments on the DESED validation dataset demonstrate that the Efficient Conformer not only improves temporal resolution and long-range dependency modeling, but also outperforms standard Conformer and Convolutional Recurrent Neural Network (CRNN) baselines in PSDS2. Additionally, we explore lightweight attention mechanisms employing squeeze-and-excitation blocks to emulate frequency-axis translation invariance of Frequency Dynamic Convolutions (FDY). Our approach achieves performance comparable to heavier models like FDY+Conformer, while reducing computational cost by over 69%, showing promising results for Conformer-based systems in terms of precision and model efficiencyThis research was supported by FPI PRE2022-104808 funded by FSE, Spain as well as project PID2021-125943OB-I00 funded by MCIN/AEI/10.13039/501100011033/FEDER, UE and project PID2024-160789OB-I00 funded by MICIU/AEI/ 10.13039/501100011033/FEDER, U
Von Hippel-Lindau protein regulates dendritic cell activation in the thymus and periphery
En conclusión, la estabilización de HIF1α aumenta la eferocitosis y la presentación antigénica durante la maduración homeostática de las dendríticas, pero regula negativamente la maduración inmunogénica tras la estimulación con TLRsWork in the laboratory of David Sancho is funded by the CNIC; by the European Union’s Horizon 2020 research and innovation program under grant agreement ERC-2016-Consolidator Grant 725091; by MCIN PID2022-137712OB-I00, PID2019-108157RB, CPP2021-008310 and CPP2022-009762 MCIN/AEI/10.13039/501100011033 Unión Europea NextGeneration EU/PRTR; by Comunidad de Madrid (P2022/BMD-7333 INMUNOVAR-CM); by Scientific Foundation of the Spanish Association Against Cancer (AECC-PRYGN246642SANC); by Worldwide Cancer Research WWCR-25-0080; by European Union ERC-2023-PoC; by a research agreement with Inmunotek S.L.; and by “la Caixa” Foundation (LCF/PR/HR23/52430012 and LCF/PR/HR22/52420019).

NRF2 at the crossroads of parkinson’s disease and aging: mechanistic insights and translational perspectives
Parkinson's disease (PD) is a multifactorial neurodegenerative disorder characterized by dopaminergic neuronal loss, α-SYNUCLEIN aggregation, mitochondrial dysfunction, oxidative stress, and neuroinflammation. The transcription factor NRF2 (nuclear factor erythroid 2–related factor 2) orchestrates cellular defense mechanisms by controlling genes involved in antioxidant responses, detoxification, and proteostasis. Impaired NRF2 signaling in PD amplifies oxidative damage, protein misfolding, and inflammatory cascades, whereas NRF2 activation confers broad neuroprotection. This review summarizes evidence from cellular, animal, and human studies delineating NRF2 regulatory roles in redox homeostasis, mitochondrial integrity, and microglial activation. In preclinical models, NRF2 deficiency accelerates neurodegeneration, while pharmacological activation with agents such as dimethyl fumarate, sulforaphane, and synthetic triterpenoids mitigates dopaminergic loss and neuroinflammation. Human studies reveal altered NRF2 pathway components in PD brain and peripheral tissues, and genetic variants in NFE2L2 influence disease susceptibility and progression. Aging, PD's strongest risk factor, reduces NRF2 responsiveness through epigenetic and post-translational changes, promoting oxidative vulnerability and inflammaging. Environmental exposures, including pesticides and pollutants, further modulate NRF2 activity, compounding risk via cumulative “exposome” effects. Understanding NRF2 regulation provides mechanistic insight into PD pathogenesis and positions NRF2 activation as a promising therapeutic strategy for disease modification and healthy brain agin
Gapless superconductivity from extremely dilute magnetic disorder in 2H-NbSe2−S
Most superconducting materials exhibit a vanishing density of states at the Fermi level and Anderson’s theorem posits that thesuperconducting gap is robust against nonmagnetic disorder. Although dilute magnetic impurities lead to localized in-gap states, these states typically have no bearing on the material’s bulk superconducting properties. However, numerous experiments reveala finite density of states at the Fermi level in systems with an apparently negligible number of magnetic impurities. Here, usingscanning tunneling microscopy and self-consistent Bogoliubov-de Gennes calculations, we find that gapless superconductivityemerges in 2H-NbSe 2− S at remarkably low magnetic impurity concentrations. Furthermore, our density functional theorycalculations and in-gap quasiparticle interference measurements demonstrate that the Se-S substitution significantly modifies theband structure. This modification favours nesting and dictates the in-gap scattering for > 0, in stark contrast to the dominantcharge density wave interactions in pure 2H-NbSe 2. Our findings reveal an unusual superconducting response to disorder andhighlight the importance of incorporating material-specific band structures in the understanding of a superconductor’s responseto even very low concentrations of magnetic impuritiesThis work was supported by the Spanish Research State Agency, AEI (PID2023-150148OB-I00, PID2023-149309OB-I00, PID2020-114071RB-I00, PDC2021-121086-I00, TED2021-130546BI00, CEX2023-001316-M, PID2024-162182NAI00 and CEX2024-001467-M), Comunidad de Madrid through project TEC-2024/TEC-380 “Mag4TIC” the EU through grant agreement No 871106, by the European Research Council PNICTEYES grant agreement 679080, ERC-2021-StG 101042680, 2D-SMARTiES and Pathfinder 4D-NMR project (No 101099676). Reactor design in carbon capture and utilization to produce cyclic carbonates using ionic liquids: Catalytic essays and techno-economic analysis
This work advances in the development of new carbon capture and utilization (CCU) technology to produce cyclic carbonates based on ionic liquids (ILs) through two main contributions: i) Experimental kinetic allowing rigorous reactor design and sizing at process scale for the first time; and ii) reliable techno-economic analysis (TEA) combined with life cycle assessment (LCA). The proposed CCU uses two ILs: CO2 is captured from a precombustion stream using aprotic heterocyclic anion-based ILs (AHA-ILs) as a CO2 chemical-physical adsorbent, and CO2 conversion occurs using halide-ILs as a catalyst to produce hexylene carbonate (Hex-C). A water-based liquid-liquid extraction strategy is proposed for IL/Hex-C separation. A multiscale research methodology, combining iterative computational-experimental approaches, is applied, integrating molecular modelling, catalytic tests, kinetic regression, phase equilibrium measurements, process simulation, TEA, and LCA. DFT/COSMORS screening selects [bmim][I] as optimal IL for the CO2 cycloaddition reaction to the epoxide, considering both catalytic activity and separation feasibility. Catalytic essays are then performed to identify the optimal operating pressure for the reactor, revealing that 20 bar minimizes energy consumption. Afterwards, the kinetic study allows designing the reactor, which was conveniently sized to assess reliable TEA and LCA. The results indicate that increasing the residence time leads to higher conversion rates, requiring a larger reactor but reducing the final costs of the process. Achieved key performance indicators (KPIs) are capital and operating expenses (CAPEX
and OPEX) of 161 per ton CO2 converted, respectively. In terms of Global Warming Potential (GWP), the proposed process emits approximately 0.34 kg of CO2 per kg of converted CO2 when considering only utilityrelated emissions, thus supporting the goal of emission reductionThe authors are grateful to Ministerio de Ciencia e Innovación of Spain (project PID2023-150532OB-I00) and Comunidad de Madrid (project TEC2024/BIO-17) for financial support, and Centro de Computación Científica de la Universidad Autónoma de Madrid for computational facilities. Rewiring receptor activation: Mechanistic insights into toggle switch modulation by 25CN-NBx compound
The development of new treatments for neuropsychiatric disorders relies on a deeper understanding of the molecular mechanisms behind psychedeliccompounds, particularly how they differentiate between hallucinogenic and antidepressant effects. In this study, we explore by molecular dynamics simulations a series of 25CN-NBx compounds bound to the 5-HT2A receptor. The simulations uncover that bulky substitutions on the N-benzyl ring cause a significant shift in the position of W336, a key toggle switch residue known to influence receptor activation and thought to play a crucial role in mediating psychedelic signaling. This result is confirmed through potential of mean force calculations along the toggle switch’s dihedral angle. End-state free energy calculations align closely with experimental data, showing that 25CN-NB-2-OH-3-Me and 25CN-NB-2-OH-5-MeO have the highest and lowest affinities, respectively, for the 5-HT2A receptor. Further analysis indicates that when W336 adopts its negative dihedral state, it establishes stronger van der Waals interactions, stabilizing its contacts with residues F332 and I163_key players previously linked to receptor activation. Our findings provide a framework for understanding receptor activation and can be extended to other G protein-coupled receptors where the toggle switch is central to signal activationThe authors acknowledges the support of the Comunidad de Madrid through the Attraction of Talent Program (2022-5A/ BMD-24244) and the Spanish Ministry of Science and Innovation through the project CNS2022-135720 funded by MCIN/AEI/10.13039/50110001103
