Aproximaciones ómicas para la biología y genética de Trypanosoma cruzi y la definición de biomarcadores en la enfermedad de Chagas

disease affects mainly rural areas from central and south America, where the insect
vector is endemic. However, this disease has spread to other continents due to
migration becoming a world health problem. It is a complex disease with great variety
of reservoirs and vectors, different ways of transmission and a high genetic variability.
According to the World Health Organization, about 6–7 million people worldwide are
estimated to be infected with Trypanosoma cruzi causing around 10-14 thousand
deaths per year, being the cardiac failure the most lethal complication.
Nowadays, there is not preventive vaccine or effective treatment against the chronic
phase of the disease. There are only two effective drugs for the acute phase, but
without demonstrated efficacy in chronic patients causing many adverse effects.
Therefore, it is necessary to further investigate this disease and the pathogenicity in
the hosts to improve the knowledge, discover new biomarkers and develop new
successful treatments. Consequently, the aims of this work were to characterize
critical molecular structures of the parasite, discover new biomarkers of cure or
progression, and understand the role of the Signaling Lymphocytic Activation
Molecule Family 1 receptor (SLAMF1) during the macrophage in vitro infection.
We analyzed and classified the complete repertoire of minicircle sequences of the Y
strain. Our results demonstrated that Trypanosoma cruzi minicircles are more
complex than previously described with a classification in four different groups
considering the size and number of their conserved and variable regions. Then, we
discovered new potential biomarkers for the chronic Chagas disease. We studied the
expression of circulating miRNAs detecting a set of four miRNAs that may serve as a
specific biomarker of chronic Chagas disease. Also, we described the principal
targets of those miRNAs and the different pathways in which they would be
implicated, pointing out their potential as future therapeutic approaches or
biomarkers. Furthermore, we described the metabolomic profile associated with
individuals with and without Chagas disease, depending on the presence or absence
of cardiac symptoms. We identified different metabolites with a promising profile of
cure biomarkers of Chagas disease, while others displayed a promising profile of
disease progression biomarkers.
We also analyzed by proteomics the role of SLAMF1 in the macrophage infection. We
detected different significant up or downregulated proteins involved in immune
regulation processes, being SLAMF1 and/or strain-dependent. These results have
opened new fields of research to elucidate the concrete role of SLAMF1 and to
discover new potential therapeutic approaches for the Chagas disease. Finally, we
characterized the parasite multi-gene family of the trans-sialidases, and its
interaction with the receptor SLAMF1. We performed a phylogenetic study obtaining
an aggrupation in five clusters, showing that the previous classification was not
accurate, and we did a docking analysis that confirmed their potential as ligands of
SLAMF1, displaying the most promising binding residue

Interacción entre las células dendríticas y las células CD8+ de memoria residente en inmunidad antiviral y antitumoral

and eliminating cancerous cells and cells infected with viruses, bacteria, or parasites. They
achieve this by identifying tumor or pathogen-derived peptides presented by major
histocompatibility complex (MHC) class I molecules, and directly kill target cells through the
release of cytotoxic molecules. Naı̈ve T cells require cognate antigen recognition along with costimulatory
signals and cytokines provided by the dendritic cells (DCs) for their optimal
activation. Once activated, CD8+ T cells can differentiate into effector or memory CD8+ T cells.
Effector cells directly engage in the immune response, while memory cells enhance
immunosurveillance and respond more rapidly to secondary infections, a hallmark of
immunological memory. Memory CD8+ T cells are also crucial for preventing relapse and
recurrence of tumors, being essential for the long-lived protective immunity in cancer.
Like other viruses, Vaccinia virus (VACV) can infect DCs, leading to direct presentation of
antigens via MHCI. Alternatively, DCs can acquire VACV antigens by phagocytosis of infected cells,
leading to cross-presentation of VACV antigens on MHCI to CTLs. Although most research has
focused on the role of DCs in priming T cells in lymphoid tissues, DCs can also affect T cell
responses within non-lymphoid tissues (NLTs) in a secondary response. This thesis explores the
crosstalk between resident memory CD8+ T (Trm) cells and DCs within tissues during a
secondary response and examines the pathophysiological consequences of this interaction in
antiviral and antitumor immunity.
To investigate this, we infected mice with VACV either in the skin, generating circulating central
CD8+ T (Tcm) cells and effector memory CD8+ T (Tem) cells, as well as Trm cells in the skin, or
intraperitoneally (i.p.), generating circulating memory CD8+ T cells but not skin Trm cells.
Subsequently, we reactivated memory CD8+ T cells with intradermal (i.d.) inoculation of VACV,
or the immunodominant peptide of VACV to mimic infection, or, alternatively, with tumors
containing a cognate antigen.
While non-inflamed skin typically lacks plasmacytoid dendritic cells (pDCs) –a DC subset
specialized in the secretion of large amounts of type I interferons (IFN-I)– this thesis shows that
reactivated Trm cells, independently of circulating memory CD8+ T cells, promotes the
infiltration and maturation of pDCs in the skin. In the tumor context, pDCs are also recruited in
the peritumoral skin upon memory CD8+ T reactivation. However, this Trm-dependent pDC
infiltration does not impact tumor control. In the viral context, pDCs infiltrate the skin upon
memory CD8+ T reactivation, and pDC depletion attenuates the Trm-mediated protection against
a secondary infection.
This Trm-dependent infiltration of pDCs correlates with activation of conventional type 1
dendritic cells (cDC1s) in an IFN-I-dependent manner, as revealed by blocking the IFNAR1
receptor, pDC depletion and transcriptomic analysis. Additionally, this analysis revealed that Trm
and Tem cells also upregulate IFN-I signaling pathways in response to Trm-dependent pDC
infiltration during a secondary response. Moreover, blockade of IFN-I signaling dampens
antigen-specific Trm-mediated protection in secondary infections with VACV or during a
heterologous infection with influenza A virus (IAV).
Thus, this thesis reveals that Trm do not act alone during a secondary response to viral
infections. Trm cells promote the recruitment and activation of pDCs, which produce IFN-I
boosting cDC1 activation and the antiviral response. These findings reveal the importance of the
cooperation between Trm and DCs in secondary infections. Antiviral therapies and vaccination
strategies that accentuate the crosstalk between Trm, pDCs and cDC1s could enhance viral
clearance and vaccine efficac
Lipopolisacáridos atípicos: Estructura, actividad biológica y posibles aplicaciones clínicas

Fecha de Lectura: 18-10-2024Esta tesis tiene embargado el acceso al texto completo hasta el 18-04-2026Lipopolysaccharides (LPS) are molecules found in the cell walls of Gram-negative bacteria. Traditionally, this molecule has been called endotoxin due to its presence in pathogenic bacteria and its toxic and inflammatory response in mammals. Rhizobiales is an order of Gram-negative bacteria whose LPS is associated with the formation of nodules in plant roots. In this work we have purified and characterised LPSs from the genera Ochrobactrum and Rhizobium that may be ligands of TLRs, pathogen-associated molecular pattern receptors (PAMPS) of the innate immune system.
We describe a new LPS purification protocol based on the classical phenol extraction method, in which we have successfully eliminated the presence of contaminations with β-1,2-cyclic glucans. Furthermore, thanks to mass spectrometry and nuclear magnetic resonance studies, we have characterised the most probable structure of the LPS of O. intermedium and partially of the LPS of R. rhizogenes K84 and R. vitis. We have defined that their atypical structure allows them to interact differently with TLR2 and/or TLR4 than toxic LPS. Both the structure of Lipid A and the core of atypical LPS alter TLR4 dimerization, because their VLFAC prevents proper binding of the TLR4/TLR4 dimer together with MD-2. Surprisingly, purified and separated Lipid A from O. intermedium has a higher affinity for TLR2.
These atypical LPS act as immunomodulators by activating macrophages inducing NF-B and MAPK signalling pathways and producing cytokines such as TNFα, IL-6, IL-10 and IL-12 both in vitro and in vivo acting differentially on TLR4 and TLR2, depending on the LPS, at levels several times lower than E. coli LPS and without its toxicity. We have shown that β-1,2-cyclic glucans have immunomodulatory activity in macrophages by inducing cytokine production via TLR2.
On the other hand, the immunomodulatory power of these atypical LPSs in the treatment of leishmaniasis reduces and delays the development of skin lesions in a mouse model of cutaneous leishmaniasis with L. major and splenomegaly in a mouse model of visceral leishmaniasis with L. infantum, due to their ability to promote the production of IL-12. We demonstrate that atypical LPSs, notably that of R. rhizogenes K84, are able to induce M2 macrophages in the diaphragm and activate the retinoic acid (RA) pathway both in vitro, in macrophage cell lines, and in vivo in pregnant rat fetuses in the nitrofen-induced CDH (Congenital Diaphragmatic Hernia) model resulting in a reversion from 70-80 % CDH+ fetuses to 30-40 % CDH+ with improvement of pulmonary hypoplasia and diaphragm repai
Cell phone use and its incidence on sociemotional skills in adolescents from rural educational institutions

Fecha de Lectura: 11-10-2024Esta Tesis tiene embargado el acceso al texto completo hasta el 11-04-2026Esta Tesis se focaliza en identificar el nivel de incidencia del uso desmedido de canales virtuales de comunicación, y particularmente el celular, sobre las habilidades socioemocionales. Para ello se contó con la participación de estudiantes de los grados 9º, 10º y 11º de dos centros educativos del municipio de Duitama - Boyacá, Colombia, el equipo docente y las familias.
El enfoque metodológico utilizado ha sido de tipo mixto en dos fases, una cualitativa y otra cuantitativa. En primer lugar, se abordó el objetivo desde una línea cualitativa con perspectiva fenomenológica; para la recogida de información se utilizaron las técnicas de grupos focales en la que participaron 98 personas, observaciones no participantes a 151 estudiantes y 18 entrevistas semi-estructuradas. En una segunda fase cuantitativa y con el fin de conocer si las percepciones de esos participantes eran compartidas y generalizables a un mayor número de estudiantes, familias y profesorado se elaboró una encuesta que se administró a 92 jóvenes, 21 docentes y 63 docentes.
Los resultados de ambas fases señalan que el tiempo de uso del celular por parte de los adolescentes influye en sus habilidades socioemocionales tendiendo a minimizar la interacción con personas de su entorno. Estudiantes, docentes y familias coinciden en admitir que la preferencia de los jóvenes por la comunicación virtual afecta tanto a la dinámica familiar como a las relaciones sociales en el aula y, por tanto, las relaciones interpersonales. Asimismo, las conclusiones de este estudio permiten elaborar orientaciones generales para que docentes y familias puedan guiar a los jóvenes en la resignificación del uso del celula
Impacto del tratamiento con presión continua positiva en las comorbilidades metabólicas y vasculares vinculadas a la apnea obstructiva del sueño

Fecha de Lectura: 15-11-2024Esta tesis tiene embargado el acceso al texto completo hasta el 15-05-2026La apnea obstructiva del sueño (AOS) y el síndrome de Overlap (SO: AOS y enfermedad pulmonar obstructiva crónica, EPOC) se relacionan con la patogénesis de múltiples comorbilidades metabólicas y cardiovasculares. El tratamiento de elección es la terapia con presión positiva sobre la vía aérea (CPAP), pero no existe consenso acerca de su efecto sobre dichas alteraciones. El objetivo del presente trabajo es mejorar el conocimiento de los efectos del tratamiento con CPAP en pacientes con AOS, centrándose en el estudio de sus efectos sobre las comorbilidades metabólicas y cardiovasculares asociadas a esta enfermedad respiratoria.
MATERIALES Y MÉTODOS
Se incluyeron 57 pacientes diagnosticados de AOS (44 AOS y 13 SO) mediante poligrafía nocturna y espirometría, que accedieron a participar mediante la firma de un consentimiento previamente informado. Todos ellos mantuvieron la CPAP durante 18 meses. El 79% se sometieron a una auto-CPAP durante 3 noches consecutivas. A todos se les realizó una caracterización clínica global, y analítica con un amplio panel de parámetros bioquímicos y metabólicos, a partir de los cuales se calcularon diferentes índices predictivos de disfunción metabólica. Asimismo, se realizó una ecografía vascular de las arterias carótidas a 33 pacientes con AOS sin EPOC. Se construyeron modelos de regresión univariantes y multivariantes a partir de los parámetros analizados para determinar diferentes cambios clínicos y analíticos.
RESULTADOS
De los 57 pacientes incluidos, 44 presentaron diagnóstico de AOS y 13 de SO. Se trata de pacientes en su mayoría obesos o con sobrepeso (44%, respectivamente), que no modificaron el grado de obesidad global ni central tras la terapia con CPAP. Dicho tratamiento normalizó el índice de apneas e hipopneas (IAH) en la cohorte estudiada, de la que un 93% realizó un buen cumplimiento de la terapia con un uso superior a 4 horas por noche (uso medio de 6,48 horas por noche) y un 79% refirió una mejoría clínica de la sintomatología global. Un análisis de regresión logística binaria reveló que las horas de uso de la CPAP (con un mínimo de 4 horas) y la variación del índice predictivo LAP (ΔLAP) son las variables predictoras independientes de la mejora clínica del estado general. La presencia de SO redujo la posibilidad de experimentar mejoría clínica en comparación con aquellos pacientes AOS sin EPOC.
Impacto del tratamiento con presión continua positiva en las comorbilidades metabólicas y
vasculares vinculadas a la apnea obstructiva del sueño.
Tras el tratamiento con CPAP se observó una mejoría significativa de la función renal, definida por un aumento del filtrado glomerular, y del perfil lipídico, debido a una reducción significativa en los niveles de colesterol total, VLDL y triglicéridos. Asimismo, aunque el tratamiento con CPAP no afectó al perfil glucémico de los pacientes, sí redujo el grado de resistencia a la insulina en los pacientes con AOS resistentes a la hormona (HOMA-IR ≥ 3,5), que representaron un 60% de la población de estudio.
Del mismo modo, los pacientes diagnosticados de AOS con alta probabilidad de padecer esteatosis hepática al inicio del estudio, definida por un Fatty Liver Index (FLI) > 60, mejoraron significativamente el valor de este índice predictivo de esteatosis hepática tras el tratamiento con CPAP, proponiendo que la terapia reduce la probabilidad de progresión de la enfermedad hepática metabólica. Se calcularon otros índices de diagnóstico de disfunción cardiometabólica. Los valores de los índices predictivos LAP y VAI disminuyeron de manera significativa después del tratamiento con CPAP, sugiriendo que esta terapia reduce el riesgo cardiovascular asociado a la AOS. La reducción de los niveles de LDH y GOT, el aumento de los niveles de albúmina y la bajada en los valores de dos índices predictivos del riesgo cardiovascular (LAP y VAI) y dos de fibrosis hepática (FIB4 y APRI) encontrados en estos pacientes tras la terapia con CPAP, indican que estos biomarcadores podrían ser de utilidad para monitorizar la eficacia de la CPAP en pacientes con AOS. El tratamiento con CPAP demostró reducir significativamente el grosor de la íntima media (GIM) carotídeo en ambas arterias en pacientes con AOS sin EPOC, siendo la combinación de las variables edad y descenso del índice predictivo LAP la única que proporcionó un modelo de regresión logística binaria estadísticamente significativo, con una alta capacidad de determinar la probabilidad de que el GIM de la carótida izquierda fuera menor a 1 mm tras la CPAP.
CONCLUSIONES
Los resultados de este estudio sugieren un efecto positivo de la terapia con CPAP sobre diferentes alteraciones metabólicas asociadas a la AOS, reduciendo asimismo el riesgo cardiovascular de estos pacientes. Del mismo modo, se proponen diferentes biomarcadores sencillos o el reposicionamiento de algoritmos empleados para la predicción de las principales comorbilidades asociadas a la AOS para realizar el seguimiento de los pacientes con CPA
El pasacalle y la chacona: desfaciendo entuertos. Correlaciones entre ambas formas en la Europa de los Austrias y de los primeros Borbones (s. XVII)

pasacalle aparece indisolublemente unida; el porqué y el cómo no está muy claro, así
como su propia naturaleza. Ambas formas son cambiantes y viajeras y se han ido
desarrollando de distintas maneras en el siglo XVII a través de una Europa controlada de
una manera u otra por dos casas reales: los Habsburgo y los Borbones. En este trabajo
expondremos la correlación entre pasacalle y chacona en las zonas de influencia de ambas
dinastías, delimitando sus distintas (o comunes) funciones, desarrollando un estudio
desde su primera aparición conjunta en imprenta, así como sus antecedentes y evolución
en distintos territorios y épocas, para intentar clarificar sus diversas realidades, sus
afinidades y desavenencias, a lo largo del siglo XVII, y terminando en 1715 con el final de
la Guerra de Sucesión EspañolaOver the last few centuries, the perception of the chaconne and pasacalle forms appears

common) functions, developing a study from their first joint appearance in print, as well
as their background and development in different territories and periods, to try to clarify
their various realities, their affinities and disagreements, throughout the 17th century, and
ending in 1715 with the end of the War of the Spanish Successio
