    PENGARUH PEMBERIAN EKSTRAK ETANOL BIJI MELON (Cucumis melo L.) PADA TIKUS PUTIH JANTAN GALUR WISTAR TERHADAP PENURUNAN KADAR KOLESTEROL TOTAL

    Hiperkolesterol merupakan salah satu penyebab penyakit jantung koroner (PJK). Senyawa yangmemiliki aktivitas sebagai antikolesterol salah satunya adalah foavonoid (Priatna, et al., 2015).Salah satu tanaman yang mengandung senyawa flavonoid yaitu biji melon (Kurniawati, 2015).Penelitian ini bertujuan untuk mengetahui aktivitas antikolesterol esktrak etanol biji melon danjuga untuk mengetahui dosis yang efektif sebagai antikolesterol. Uji aktivitas antikolesteroldilakukan pada tikus putih jantan galur wistar yang dibagi menjadi 6 kelompok perlakuan. Induksi hiperkolesterol menggunakan PTU 1% 10 mL/kg BB tikus dan kuning telur puyuh 10 mL/kg BB tikus. Kelompok kontrol normal hanya diberikan pakan standar, kelompok kontrol positif dengan pemberian tablet simvastatin dalam PGA 1%, kelompok kontrol negatif yaitu pemberian PGA 1% dan kelompok perlakuan yaitu pemberian esktrak etanol biji melon (Cucumis melo L.) dengan dosis 35,75; 71,50; dan 143 mg/200 gram BB tikus. Data dianalisis dengan menggunakan SPSS 23.0 menggunakan One Way ANOVA (α=0,05). Hasil menunjukkan bahwa ekstrak etanol biji melon (Cucumis melo L.) memiliki persentase penurunan kadar kolesterol total pada dosis 71,50 mg/200 gram BB tikus sebesar 10,24%; dan pada dosis 143 mg/200 gram BB tikus sebesar 38,19%. Aktivitas antikolesterol dosis 143 mg/200 gram BB tikus lebih baik dari dosis 35,75 dan 71,50 mg/200 gram BB tikus. Dosis 143 mg/200 gram BB tikus tidak terdapat perbedaan bermakna dengan kontrol positif dan kontrol normal, dan terdapat perbedaan bermakna dengan kontrol negatif, hal itu menunjukkan bahwa dosis 143 mg/200 gram BB tikus merupakan dosis yang terbaik.     Jeringau (Acorus calamus L.) and brotowali (Tinospora crispa L. Miers) are plants that empirically used to treat several diseases, especially those caused by bacteria. Empirical data shows the herba of jeringau can be used to treat scabies infection. Brotowali stems which have a bitter taste are known to have many benefits, including as an antipyretic, analgesic, antiparasitic, and antiseptic. Objective: This study aimed to determine the class of active compounds contained in the jeringau and brotowali and to determine the antibacterial activity of the extracts of jeringau herbs and brotowali stems against Streptococcus pyogenes. Methods: Phytochemical screening included alkaloid, tannin, flavonoid, polyphenol and saponin+ test based on the Harborne method and antibacterial activity test of jeringau herbs and brotowali stems extracts with Kirby-Bauer disc diffusion method against Streptococcus pyogenes. Results: The results showed that the jeringau herb contained alkaloids, polyphenols, and flavonoids with antibacterial activity values at a level of 2400 mg/mL of 4.26mm. While brotowali stems contain alkaloids, tannins, polyphenols, and flavonoids with antibacterial activity values at a level of 2400 mg/mL of 3.77mm. Conclusion: The ethanol extracts of jeringau herbs and brotowali stem had antibacterial activity against Streptococcus pyogenes at concentration of 2400mg/mL

    Coconut fruit (Cocos nucifera L.) can be utilized for treatment and beauty because it contains polyphenols, flavonoids, monoterpenes and seskuiterpene, phenolic acid. Several of these phytochemical compounds can function as antioxidants. Objective: This research aims to find out the activity of coconut fiber extract (Cocos nucifera L.) as an antidiabetic and the effective dose as an anti-diabetes. Methods: Coconut fibers was extracted by maceration with 96% ethanol solvent. Antidiabetic activity was Tested by the OGTT (Oral Glucose Tolerance Test) method. (Oral Glucose Tolerance Test). Measurement of blood glucose levels was tested on rats which were divided into 6 groups (normal, negative, positive, dose 1, dose 2 and dose 3), each group consisting of 4 rats. The The analysis uses the SPSS version 16.0 statistical test, namely Paired T Test with the condition that the data must be distributed normally and homogeneous with a 95% confidence level. Results: Based on the difference in the percentage of decrease in blood glucose levels is evidenced through the analysis of statistical using the paired sample T test. Stating, there is a significant difference, which at all doses can decrease blood glucose levels. This indicates the presence of anthocyanins as antioxidants. Conclusion: The results of coconut fiber extract (Cocos nucifera L.) test showed antidiabetic activity. Stating, test doses had antidiabetic activity, with the most effective dose being in dose 2 (500 mg/200 gram BW rats). The percentage of decreased levels at dose 2 is 25,0922%

    Kanker merupakan sel abnormal yang dapat bermetastasis dan menjalar ke organ lain. Pada penelitian sebelumnya tiourea diketahui memiliki aktivitas sebagai antikanker secara in silico dan in vitro. Tujuan penelitian untuk membuat senyawa kompleks besi (III)-tiourea secara in silico serta mengetahui profil farmakokinetik dan toksisitas. Metode yang digunakan ialah studi farmakokinetik dan toksisitas, molecular docking dan simulasi molecular dynamic. Pengujian delapan belas senyawa kompleks besi(III)-tiourea terhadap profil farmakokinetik dan toksisitas terdapat delapan senyawa yang memiliki absorpsi dan distribusi yang baik, sedangkan hasil toksisitas menunjukkan bahwa ada sebelas senyawa diprediksi tidak toksik . Hasil molecular docking dari ke delapan belas senyawa kompleks logam besi (III)-tiourea yang memiliki nilai energi bebas (ΔG) yang rendah, antara lain tiga senyawa pada reseptor kanker prostat (1T7F), tujuh belas senyawa pada reseptor kanker paru (2ITO) dan delapan belas senyawa pada reseptor kanker payudara (3ERT). Hasil molecular dynamic terbaik senyawa Bis-(3-methyl-1-(3-nitrobenzoyl) thiourea) besi-(III) pada reseptor kanker prostat (1T7F) selama 50 ns menunjukkan nilai RMSD dan RMSF yang rendah dan stabil. Senyawa kompleks besi (III)-tiourea memiliki profil farmakokinetik yang baik bahwa Bis-(3-methyl-1-(3-nitrobenzoyl) thiourea) besi (III) diprediksi menjadi kandidat yang paling berpotensi dengan afinitas dan kestabilan yang lebih baik namun memiliki toksisitas yang mutagenic

    In search of complementarities within and across platform ecosystems: Complementors’ relative standing and performance in mobile apps ecosystems

    Firms can benefit immensely from participating in digital platform ecosystems—specifically, from the shared technological assets and market opportunities offered by the platform owner. Yet, while aligning with the platform ecosystem rules, each member must decide whether to specialize in a given platform ecosystem or across multiple platform ecosystems to capture these benefits. We examine two common patterns through which platform ecosystem members (i.e., complementors) specialize within and across platform ecosystems, and the relative impact on their market performance. We look at the high relative standing of the complementary product as a reflection of complementors' specialization in the given product category or platform ecosystem. We then theorize that having products with high relative standing in a single product category and a single platform ecosystem, together, diminishes complementors’ market performance over time. Similarly, high relative standing in multiple platform ecosystems and multiple product categories, at the same time, adversely impacts the market performance. We find supportive evidence for our hypotheses, in a panel dataset of mobile app developers. This paper contributes to the burgeoning stream of research that investigates the trade-offs faced by complementors, suggesting that complementor strategies are more complex than simply trying to maximize market reach

    Classroom Simulations for Teaching Production Control in Non-repetitive Contexts: Insights for Theory and Practice

    When introducing production control solutions, most textbooks focus on a to-stock, repetitive context. In response, we extend the classical match and dice game, which focusses on a repetitive context,in order to teach complexitiesthat emerge in a non-repetitive context, such as the emergence of direct and indirect load, and to introduce appropriate production control solutions, e.g. Constant Work-In-Process (ConWIP)and Control of Balance by Card Based Navigation (COBACABANA). Twofield testsareused to prove the playability of our game and assess its teaching effectiveness using post-game scores and self-assessment. Both give support to our game and highlight that it helps students to understand complex knowledge.Applying the simulation as part of aniterative design process in two cases not only ensured the pragmatic validity of our design, i.e. that learning objectives were met, but also provided important insight on the production control systems taught. During the applicationwe encountered several issues that provide insights for the implementation of concepts as COBACABANA (or Workload Control) in practice. In terms of theory, applying the game showed that there is a change in mindset required when switching from Kanban to COBACABANA and physics synchronization is essential for logistics synchronization

    Space-Confined Synthesis of CoNi nanoalloy in N-doped Porous Carbon Frameworks as Efficient Oxygen Reduction Catalyst for Neutral and Alkaline Aluminum-air Batteries

    Exploring efficient and stable non-precious metal electrocatalysts for oxygen reduction reaction in neutral and alkaline solutions is of great importance for metal-air batteries. Herein, an efficient and stable electrocatalyst with CoNi nanoalloy (10-20 nm) uniformly embedded in porous N-doped carbon framework (CoNi-NCF) for neutral and alkaline primary Al-air batteries is prepared and studied. The CoNi-NCF electrocatalyst shows superior ORR performance in terms of half-wave potential of 0.91 V in 0.1 M KOH solution and 0.64 V in 3.5 wt% NaCl solution, outperforming those of the commercial Pt/C catalyst. Supported by in situ electrochemical Raman spectra and density functional theory calculations, the rich dual active sites of CoNi nanoalloy and porous N-doped carbon contributes to the outstanding ORR performance. Impressively, when employed as a cathode catalyst in both aqueous and solid flexible Al-air batteries, the CoNi-NCF-based Al-air batteries show the highest discharge performance, long-time durability and high flexibility, demonstrating the promising potential for practical application

    Using HOX Transcription Factors as Novel Diagnostic Biomarkers and HOX-PBX Interactions as Therapeutic Targets in Breast Cancer

    Triple Negative Breast Cancer (TNBC) represents 10-15% of all breast cancers, is characterised by its lack of hormone and HER2 receptor expression and exist as clinically high grade and aggressive tumours with poor prognosis. A major challenge of these tumours is the lack of effective targeted therapies and the resistance to conventional therapies. Homeobox (HOX) genes are a family of homeodomain-containing transcription factors, which are primarily known for establishing and maintaining the identity and fate of cells and tissues during normal embryogenesis and organ development. Recent studies have highlighted HOX genes to be dysregulated in most cancers including breast, ovarian, prostate, colorectal and melanoma, and possess an established role in driving key hallmarks in carcinogenesis, including the regulation of the cell cycle, apoptosis, angiogenesis, and metastasis. In this study, the HOX expression profiles between TNBC and all other breast cancer subtypes was evaluated relative to normal HOX expression and showed TNBC tumours possess unique and specific HOX expression profiles and were indicative of prognosis. Given their cancer-specific expression, targeting HOX proteins for cancer therapy would potentially provide a novel therapeutic option, as yet unexploited. Rather than targeting individual HOX genes, which is ineffective due to the functional redundancy of HOX genes, targeting multiple HOX proteins post-translationally becomes feasible by inhibiting the interaction between HOX and their PBX co-factors by disrupting protein-protein interactions.In this study, we evaluated the anti-tumour efficacy of a novel inhibitor of HOX protein function, HTL-001, which antagonizes the interactions between HOX and PBX proteins, thus preventing the binding of this complex to DNA to promote tumour growth. HTL-001 is a synthetic peptide comprised of a hexapeptide sequence, which resembles the interaction site between HOX proteins of paralogs 1-9 and PBX proteins. Targeting HOX/PBX dimers with HTL-001, induced apoptotic cell death in all breast cancer cell lines including MCF-7, ZR-75-1, MDA-MB-231, BT-20 and SK-BR-3, with the highest sensitivity seen in brain seeking MDA-MB-231-BR cells. HTL-001 also showed high synergistic anti-cancer effects in TNBC cell lines when combined with conventional chemotherapies and significantly translated these effects in vivo with significant inhibition of tumour growth by HTL-001 alone and in combination with Paclitaxel. The mechanism of action of HTL-001 was explored and the downstream effects on apoptosis was examined in detail. Disruption of HOX-PBX dimers by HTL-001 resulted in the upregulated expression of key apoptotic and anti-survival proteins that form an interconnecting network of signalling pathways initiated by ROS mediated ER stress which causes calcium influx and calpain activation to regulate caspase-independent activation of AIF, and release of immunogenic DAMPs. Mechanisms of synergy were evaluated to additionally show the main mechanism of synergistic apoptosis results from activating HTL-001 mediated signalling pathways and enhancing these by a chemotherapeutic agent. Resistance to HTL-001 was found to be mediated by autophagy and can be overcome by autophagy inhibitors. HTL-001 is currently the only therapy which targets HOX gene dysregulation and in doing so, may help address the current urgent unmet therapeutic need in TNBC through its unique mechanism of action and high levels of synergy with conventional anti-cancer agents

    Systematics of E2 strength in the sd-shell with the valence-space in-medium similarity renormalization group

    Background: Recent developments in {\it ab initio} nuclear theory demonstratepromising results in medium- to heavy-mass nuclei. A particular challenge formany of the many-body methodologies, however, is an accurate treatment of theelectric-quadrupole, E2E2, strength associated with collectivity. Purpose: Thevalence-space in-medium similarity renormalization group (VS-IMSRG) is aparticularly powerful method for accessing medium- and high-mass nuclei but hasbeen found to underpredict E2E2 strengths. The purpose of this work is toevaluate the isospin dependence of this underprediction. Methods: We perform asystematic comparison of valence-space in-medium similarity renormalizationgroup (VS-IMSRG) calculations with available literature. We make use ofisoscalar and isovector contributions to the E2E2 matrix elements to assessisoscalar and isovector contributions to the missing strength. Results: It isfound that the E2E2 strength is consistent throughoutTz=12T_z=\left|\frac{1}{2}\right|, Tz=1T_z=\left|1\right|,Tz=32T_z=\left|\frac{3}{2}\right| and Tz=2T_z=2 pairs within the sdsd-shell.Furthermore, no isovector contribution to the deficiency is identified.Conclusions: A comparison with toy-models and coupled-cluster calculations isused to discuss potential origins of the missing strength, which arises frommissing many-particle, many-hole excitations out of the model space. Theabsence of any significant isovector contribution to the missing E2E2 strengthindicates that the E2E2 strength discrepancy, and therefore any correction, islargely independent of the isospin of the nuclei in question
